Glycolate Oxidase Modulates Reactive Oxygen Species–Mediated Signal Transduction During Nonhost Resistance In Nicotiana Benthamiana And Arabidopsis, Clemencia M. Rojas, Muthappa Senthil-Kumar, Keri Wang, Choong-Min Ryu, Amita Kaundal, Kirankumar S. Mysore

Plants, Soils, and Climate Faculty Publications

In contrast to gene-for-gene disease resistance, nonhost resistance governs defense responses to a broad range of potential pathogen species. To identify specific genes involved in the signal transduction cascade associated with nonhost disease resistance, we used a virus-induced gene-silencing screen in Nicotiana benthamiana, and identified the peroxisomal enzyme glycolate oxidase (GOX) as an essential component of nonhost resistance. GOX-silenced N. benthamiana and Arabidopsis thaliana GOX T-DNA insertion mutants are compromised for nonhost resistance. Moreover, Arabidopsis gox mutants have lower H2O2 accumulation, reduced callose deposition, and reduced electrolyte leakage upon inoculation with hypersensitive response–causing nonhost pathogens. Arabidopsis gox mutants were not …

The Organic Complexation Of Iron In The Marine Environment: A Review, Martha Gledhill, Kristen N. Buck Feb 2012

The Organic Complexation Of Iron In The Marine Environment: A Review, Martha Gledhill, Kristen N. Buck

Marine Science Faculty Publications

Iron (Fe) is an essential micronutrient for marine organisms, and it is now well established that low Fe availability controls phytoplankton productivity, community structure, and ecosystem functioning in vast regions of the global ocean. The biogeochemical cycle of Fe involves complex interactions between lithogenic inputs (atmospheric, continental, or hydrothermal), dissolution, precipitation, scavenging, biological uptake, remineralization, and sedimentation processes. Each of these aspects of Fe biogeochemical cycling is likely influenced by organic Fe-binding ligands, which complex more than 99% of dissolved Fe. In this review we consider recent advances in our knowledge of Fe complexation in the marine environment and their …

Partial Characterization Of The Antimicrobial Activity Of Ccl28, Bin Liu Feb 2012

Partial Characterization Of The Antimicrobial Activity Of Ccl28, Bin Liu

Theses and Dissertations

This research focuses on the antimicrobial activity of the mouse chemokine CCL28. In addition to their well characterized chemotactic activity, many chemokines have been shown to be antimicrobial in vitro, including the mucosally expressed chemokine CCL28. I have investigated the primary sequence features required for antimicrobial activity, salt sensitive nature of killing/binding mechanism, and in vivo microbial interactions of CCL28. Through the use of protein mutation and expression techniques, I have shown that the holoprotein (108 amino acids) is necessary for full antimicrobial activity of CCL28. Furthermore, the C terminal region of CCL28 is essential for microbial killing as an …

MscL is a bacterial mechanosensitive channel that protects cells from lysis upon acute decrease in external osmotic environment. It is one of the best characterized mechanosensors known, thus serving as a paradigm of how such molecules sense and respond to stimuli. In addition, the fact that it can be genetically modified, expressed, isolated, and manipulated has led to its proposed use as a triggered nanovalve for various functions including sensors within microelectronic array chips, as well as vesicular-based targeted drug release. X-ray crystallography reveals a homopentameric complex with each subunit containing two transmembrane α-helices (TM1 and TM2) and a single …

Genetic Reporter System For Positioning Of Proteins At The Bacterial Pole, Kathryn R. Fixen, Anuradha Janakiraman, Sean Garritty, Daniel J. Slade, Andrew N. Gray, Nilay Karahan, Ann Hochschild, Marcia C. Goldberg

Publications and Research

Spatial organization within bacteria is fundamental to many cellular processes, although the basic mechanisms underlying localization of proteins to specific sites within bacteria are poorly understood. The study of protein positioning has been limited by a paucity of methods that allow rapid large-scale screening for mutants in which protein positioning is altered. We developed a genetic reporter system for protein localization to the pole within the bacterial cytoplasm that allows saturation screening for mutants in Escherichia coli in which protein localization is altered. Utilizing this system, we identify proteins required for proper positioning of the Shigella autotransporter IcsA. Autotransporters, widely …

Simultaneous Determinations Of Ascorbic Acid, Dopamine And Uric Acid At Ca/Gc Electrode, Ying Zhang, Wang Ren, Min-Jiao Li

Journal of Electrochemistry

The citric acid modified glassy carbon electrode (CA/GC) was constructed by potentiostatic technique and used for simultaneous determinations of ascorbic acid (AA), dopamine (DA) and uric acid (UA) by cyclic voltammetry (CV) and differential pulse voltammetry (DPV). The CA/GC electrode exhibits excellent electrocatalytic activity towards AA, DA and UA. The oxidation peaks are well separated with the potential differences between AA and DA, DA and UA, and AA and UA being 170, 130 and 300 mV, respectively, which is large enough to determine AA, DA and UA simultaneously. The catalytic peak currents obtained were linearly dependent on the concentrations of …

Ag Nanoparticles Self-Assembled Films On Iron Surface And Their Inhibition Properties, Zhe Zhang, Ming-Zi Jia, Le Ruan

Journal of Electrochemistry

The Ag nanoparticles were prepared in aqueous solutions through a chemical reduction method and transferred from water phase to toluene phase with the protection of 1-dodecanethiol (DDT). The polished iron electrode was immersed in the DDT-protected metal nanoparticles toluene solutions to form DDT/Ag nanoparticles self-assembled mixed films. Electrochemical methods such as electrochemical impedance spectroscopy and polarization curve measurement were used to study the inhibition properties of the self-assembled films in the 0.5 mol·L-1 H2SO4 solution. The appearance of elements such as S and Ag from the XPS data identified the presences of DDT and Ag nanoparticles.

Family Structure And Family Instability: Evaluating Their Influence On Adult Outcomes, Hsin-Yao Chiu Feb 2012

Family Structure And Family Instability: Evaluating Their Influence On Adult Outcomes, Hsin-Yao Chiu

Theses and Dissertations

Family structure is a widely used independent variable. However, in recent years researchers have questioned its predicting power. They also strive to improve its use. This study drew on Holman's (2001) theoretical model and examined the association between the family structure independent variables (Family Structure, Family Instability, and Parental Romantic Partners) and the outcome variables (Coming to Terms, Self-esteem, Maturity, and Depression). The analysis was conducted with a sample of 3,705 individuals (2,316 females and 1,389 males) randomly selected from the entire population that completed the Relationship Evaluation Questionnaire (RELATE). Results showed that Family Instability was a stronger predictor than …

Glycoinositolphospholipids From Leishmania Braziliensis And L. Infantum: Modulation Of Innate Immune System And Variations In Carbohydrate Structure, Rafael Ramiro Assis, Izabela Coimbra Ibraim, Fátima Soares Noronha, Salvatore J. Turco, Rodrigo Pedro Soares

Molecular and Cellular Biochemistry Faculty Publications

The essential role of the lipophosphoglycan (LPG) of Leishmania in innate immune response has been extensively reported. However, information about the role of the LPG-related glycoinositolphospholipids (GIPLs) is limited, especially with respect to the New World species of Leishmania. GIPLs are low molecular weight molecules covering the parasite surface and are similar to LPG in sharing a common lipid backbone and a glycan motif containing up to 7 sugars. Critical aspects of their structure and functions are still obscure in the interaction with the vertebrate host. In this study, we evaluated the role of those molecules in two medically important …

The Underclass In The Works Of Charles Dickens And Its Marxist Themes, Kukuh Prayitno Subagyo Feb 2012

The Underclass In The Works Of Charles Dickens And Its Marxist Themes, Kukuh Prayitno Subagyo

Bahasa dan Seni: Jurnal Bahasa, Sastra, Seni, dan Pengajarannya

In his novels, Dickens delivers important social messages through his empathetic characters living lives of misery, oppression, slavery, degradation, and exploitation under the so-called capitalistic system. These underclass people holler their plight of injustice and crises of the actual social milieu. From a Marxist perspective, these multiple crises are embedded in themes of exploitation, class consciousness, sacrifice, and revolution. Hence, the realism disguised under literary devices could hopefully accomplish dual purposes: both as a means of provoking a new kind of “social consciousness” among the bourgeoisie and as a means of raising the proletariat’s class consciousness.

Renegotiating Identities, Cultures And Histories: Oppositional Looking In Shelley Niro's "This Land Is Mime Land", Jennifer Danielle Mccall

USF Tampa Graduate Theses and Dissertations

My master's thesis explores the photographic series "This Land is Mime Land," which Shelley Niro made in 1992. Despite this work's complex form and structure, there are currently no sustained studies of this series alone, or books solely dedicated to Niro's art. Instead, "Mime Land" is often discussed in compilations that address a number of Native artists, Western feminist practices, or multiple works in Niro's oeuvre. My thesis fills this gap, as I closely investigate how "Mime Land" asks the viewer to look at visual culture, histories and Niro herself. Bell hooks's definition of the "oppositional gaze" - meaning a …
