The occurrence of melanism (darkening of the background coloration) is documented in 13 felid species, in some cases reaching high frequencies at the population level. Recent analyses have indicated that it arose multiple times in the Felidae, with three different species exhibiting unique mutations associated with this trait. The causative mutations in the remaining species have so far not been identified, precluding a broader assessment of the evolutionary dynamics of melanism in the Felidae. The KH-type splicing regulatory protein (KSRP) promotes the decay of AU-rich element (ARE) containing mRNAs. Although KSRP is expressed in the developing and mature nervous system, very little is known about its role in regulating gene expression in the brain. In this study, we utilized in vitro binding and decay studies to examine whether KSRP regulates the stability of the GAP-43 transcript, an ARE-containing neuronal mRNA whose protein product plays a role in axonal growth and synaptic plasticity. The focus of the work in this dissertation is to elucidate the efficacy of cannabinoid 2 receptor agonists, specifically targeting the spinal cord, for the treatment of chronic neuropathic pain. Chemical structures for all cannabinoid compounds utilized are displayed (Supplemental Figure A-1). As an orientation for the reader, the organization of this dissertation consists of five components: chapters 1 through 5. In the first chapter, a background is developed to acquaint one with the topics of chronic pain, and the use of cannabinergic compounds for chronic pain relief. Coarctation of the aorta (CoA) is associated with substantial morbidity despite treatment. Mechanically induced structural and functional vascular changes are implicated; however, their relationship with smooth muscle (SM) phenotypic expression is not fully understood. Using a clinically representative rabbit model of CoA and correction, we quantified mechanical alterations from a 20-mmHg blood pressure (BP) gradient in the thoracic aorta and related the expression of key SM contractile and focal adhesion proteins with remodeling, relaxation, and stiffness. Activation of γ-globin gene expression in adults is known to be therapeutic for sickle cell disease. Thus, it follows that the converse, alleviation of repression, would be equally effective, since the net result would be the same: an increase in fetal hemoglobin. A GATA-1-FOG-1-Mi2 repressor complex was recently demonstrated to be recruited to the -566 GATA motif of the Aγ-globin gene. We show that Mi2β is essential for γ-globin gene silencing using Mi2β conditional knockout β-YAC transgenic mice. In addition, increased expression of Aγ-globin was detected in adult blood from β-YAC transgenic mice containing a T>G HPFH point mutation …

Today complimentary medicine is being increasingly sought out. Ayurveda and TCM, are among the oldest systems of medicine and have been developed for over thousands of years in India and China respectively. This paper details the philosophies, medical theories, anatomy, diagnosis, and treatments of both of these systems, including a comparison. Both of these modalities of healing operate with a microcosm – macrocosm paradigm. This makes them fundamentally similar, and compatible with each other. Ayurveda uses Tridoshic theory to apply treatments ranging from diet, massage, meditation, yoga among other therapies to bring Vata, Pitta, and Kapha into balance. TCM is …

Two trials (T1, T2) were conducted to determine the effects of enrichment: A) broiler chick addition on early mortality and B) providing environmental enrichments on leg strength and welfare of tom turkeys. Trial 1A utilized 248 1d poults and 8 3d broiler chicks. Four pens of 32 poults were placed for the control. Four pens of 30 poults and two 3d broiler chicks were placed for the trt group. Trial 2A utilized 296 1d poults and 24 3d broiler chicks. Four trts were randomly divided among 16 pens (20 birds/pen). Trt 1 brooders were placed with assistance finding resources. Trt …

Replacing the amount of corn in finishing diets with ethanol byproducts, such as distillers grains, has been well researched and improves performance. However, research when including soybean hulls (SBH) in feedlot finishing diets with distillers grains is limited. Two feedlot experiments were conducted to evaluate the effects of SBH on animal performance and carcass characteristics in finishing diets. Dietary concentrations of SBH used were 0, 12.5, 25, and 37.5% of diet DM, in both experiments. Modified distillers grains plus solubles was included in the diet at 25% for Exp.1 while Exp. 2 contained an inclusion level of 40% wet distillers …

Recently, producers in dryland wheat farming regions have made a shift from the typical winter wheat fallow rotation to a no-till system paired with cover crops. Cover crops have been shown to minimize these problems associated with the conventional fallow and possibly provide a source of forage. A 2-year grazing study was conducted to evaluate forage quality and utilization of cover crops (CC) planted to replace fallow in no-till wheat systems compared to crested wheatgrass pastures (CWP). Bovine herpes virus 1 (BHV-1) is a significant viral pathogen in cattle that induces a myriad of clinical symptoms. These symptoms include: conjunctivitis, upper respiratory tract infections, genital disorders, and abortions. BHV-1 infection can also lead to transient immune-suppression, which predisposes cattle to secondary bacterial infection leading to life-threatening pneumonia referred to as bovine respiratory disease (BRD). Following acute infection, BHV-1 establishes latency in sensory neurons within trigeminal ganglia. Reactivation of the virus can occur periodically, resulting in virus transmission. The latency-related (LR) RNA is the only abundantly expressed transcript in latently infected sensory neurons and it encodes several proteins, …

Johnson approaches The Turducken as a travesty of The Seagull, examining six iconic scenes from The Seagull, in order to explore the satirical effect of the altered scenes. In December of 2008, Bedlam Theatre of Minneapolis presented The Turducken, "a holiday dinner theater spectacular inspired by Anton Chekhov's The Seagull." Playwright Josef Evans takes Chekhov's 1895 work and turns the classic piece into a musical and farcical satire. The plot of The Turducken follows the plot of The Seagull, and some scenes …

In his article "Barthelme's 'Paraguay,' the Postmodern, and Neocolonialism," Daniel Chaskes explores the analytic opportunities afforded by conjoining globalizing critical approaches with a story by an author who has often been circumscribed by the postmodern rubric. Donald Barthelme's "Paraguay," written the summer after Nelson Rockefeller's fact-finding mission to South America in 1969, provides a chance to consider modes of anti-colonial critique in Barthelme's work. It also offers examples of a more self-reflective criticism aimed at the U.S. counterculture and the indeterminacies of postmodernism. Educating students to relate harmoniously with people from different backgrounds has become an important agenda for student affairs professional because of the increasingly diverse nature of the American society. The purpose of this study was to assess how American and international college students develop mature interpersonal relationship skills. All the participants were from a mid-sized comprehensive institution in the Southern United States. The two main constructs measured were tolerance and quality of relationships. Attempts were also made to identify other predictors that may influence mature interpersonal relationships: gender, age, educational level, and academic achievement as measured by GPA. Co-Space refers to interactive virtual environment modelled after the real world in terms of look-and-feel, functionalities and services. We have developed a 3D virtual world named Nan yang Technological University (NTU) Co-Space populated with virtual human characters. Three key requirements of realistic virtual humans in the virtual world have been identified, namely (1) autonomy: Agents can function on their own, (2) interactivity: Agents can interact naturally with players, and (3) personality: Agents can exhibit human traits and characteristics. Manifested through paintings and found objects from a piano, my thesis negotiates a series of missions through an evolution of creative methodology in the interest of discovering new potentials. At the same time generating labeled data is expensive and time consuming. A United States interagency avian influenza surveillance plan was initiated in 2006 for early detection of highly pathogenic avian influenza viruses (HPAIV) in wild birds. The plan included a variety of wild bird sampling strategies including the testing of fecal samples from aquatic areas throughout the United States from April 2006 through December 2007. Although HPAIV was not detected through this surveillance effort we were able to obtain 759 fecal samples that were positive for low pathogenic avian influenza virus (LPAIV). We used 136 DNA sequences obtained from these samples along with samples from a public influenza sequence database for …

Evaluations of the local effects of global change are often confounded by the interactions of natural and anthropogenic factors that overshadow the effects of climate changes on ecosystems. Long-term watershed and natural elevation gradient studies at the Hubbard Brook Experimental Forest and in the surrounding region show surprising results demonstrating the effects of climate change on hydrologic variables (e.g., evapotranspiration, streamflow, soil moisture); the importance of changes in phenology on water, carbon, and nitrogen fluxes during critical seasonal transition periods; winter climate change effects on plant and animal community composition and ecosystem services; and the effects of anthropogenic disturbances and …
