The Crispr/Cas Adaptive Immune System Of Pseudomonas Aeruginosa Mediates Resistance To Naturally Occurring And Engineered Phages, Kyle C. Cady, Joe Bondy-Denomy, Gary E. Heussler, Alan R. Davidson, George A. O'Toole

Dartmouth Scholarship

Here we report the isolation of 6 temperate bacteriophages (phages) that are prevented from replicating within the laboratory strain Pseudomonas aeruginosa PA14 by the endogenous CRISPR/Cas system of this microbe. These phages are only the second identified group of naturally occurring phages demonstrated to be blocked for replication by a nonengineered CRISPR/Cas system, and our results provide the first evidence that the P. aeruginosa type I-F CRISPR/Cas system can function in phage resistance. Previous studies have highlighted the importance of the protospacer adjacent motif (PAM) and a proximal 8-nucleotide seed sequence in mediating CRISPR/Cas-based immunity. Through engineering of a protospacer …

A Medicaid Long-Term Care Eligibility Primer: Basic Eligibility Rules

Marquette Elder's Advisor

Determining Medicaid eligibility for elders who need nursing home care is central to the practice of elder law. The purpose of various Medicaid rules, which vary from state to state, can seem incomprehensible- even for those who provide excellent Medicaid planning services.

Electroconvulsive Therapy (Ect): An Introduction For Elder Law Professionals, Deirdre Johnston, Doug Eitel, W. Vaughn Mccall

Marquette Elder's Advisor

Electroconvulsive therapy (ECT), colloquially called "shock therapy," is often denounced as barbaric by members of the lay population. However, it is actually a highly efficacious treatment for depression, which can be particularly common and severe in the elderly. This discussion illuminates the potential benefits and risks of this controversial treatment.

Ethics, Law, And The Science Of Fish Welfare, Colin Allen Aug 2012

Ethics, Law, And The Science Of Fish Welfare, Colin Allen

Between the Species

Fish farming is one of the fastest growing sectors of agriculture, attracting considerable attention to the question of whether existing farming regulations and animal welfare laws are adequate to deal with the expanding role of fish in feeding humans. The role of fish as model organisms in scientific research is also expanding -- a majority of research biology departments now keep zebrafish for the purposes of genome biology, and they are used widely used for basic neuroscience research. However, due to their diversity and distance from mammalian biology, fish pose difficult questions for the application of legal and ethical principles …

Effectiveness Of Phosphorus Fertilizers In Hydroponics And Glasshouse Settings With Moderate And High Organic Matter Soils, Jeffrey Sean Christian Summerhays Aug 2012

Effectiveness Of Phosphorus Fertilizers In Hydroponics And Glasshouse Settings With Moderate And High Organic Matter Soils, Jeffrey Sean Christian Summerhays

Theses and Dissertations

Phosphorus (P) is poorly soluble in most soils and, thus, has poor plant uptake efficiency. AVAIL® and Carbond P (CBP) are new fertilizer products shown to increase P use efficiency (PUE) and increase crop yields when grown in P limiting soils. Carbond P has specifically been seen to increases P uptake and crop yields in soils low in P, although effectiveness in regards to soil organic matter is unknown. The objectives of these studies were to determine if the mode of action for these products is related to physiological response, to determine if Carbond P is toxic to plant roots …

Characterization And Variable Expression Of The Cslf6 Homologs In Oat (Avena Sp.), Melissa A. Coon

Theses and Dissertations

(1,3;1,4)-β- D-glucan (β-glucan) is a plant cell wall hemicellulose and a main component of endosperm cell walls. The Cellulose Synthase F family of genes is involved in the synthesis of β-glucan. In this study full-length genomic sequences of CslF6 were obtained from multiple Avena species. Three unique alleles were found in each A. sativa line. Comparisons of these alleles to diploid Avena species allowed for identification of the genomic origin of each allele. The A and D genome alleles had identical amino acid sequences while the C-genome had 13 different amino acids. Global expression of CslF6 was completed at three …

A Confidence-Prioritization Approach To Data Processing In Noisy Data Sets And Resulting Estimation Models For Predicting Streamflow Diel Signals In The Pacific Northwest, Nathaniel Lee Gustafson Aug 2012

A Confidence-Prioritization Approach To Data Processing In Noisy Data Sets And Resulting Estimation Models For Predicting Streamflow Diel Signals In The Pacific Northwest, Nathaniel Lee Gustafson

Theses and Dissertations

Streams in small watersheds are often known to exhibit diel fluctuations, in which streamflow oscillates on a 24-hour cycle. Streamflow diel fluctuations, which we investigate in this study, are an informative indicator of environmental processes. However, in Environmental Data sets, as well as many others, there is a range of noise associated with individual data points. Some points are extracted under relatively clear and defined conditions, while others may include a range of known or unknown confounding factors, which may decrease those points' validity. These points may or may not remain useful for training, depending on how much uncertainty they …

Examining How Middle School Science Teachers Implement A Multimedia-Enriched Problem-Based Learning Environment, Min Liu, Jennifer Wivagg, Renata Geurtz, Shih-Ting Lee, Hyeseung Maria Chang

Interdisciplinary Journal of Problem-Based Learning

This study examined how a group of ten middle school teachers implemented a technology enriched problem-based learning (PBL) environment. The goal was to understand their motivation, document their implementation techniques, and identify factors that teachers considered important in using technology-based PBL tools in their teaching. The analysis identified four factors that provided the impetus for teachers to consider the adoption of technology-based PBL instruction. These factors are (1) the PBL program addresses the teachers’ curricular needs and implementing it has campus administrative and technical support, (2) the method is aligned with teachers’ pedagogical beliefs, (3) the PBL program offers a …

Mitochondrial Involvement In Neurodegeneration And Aging, Alicia M. Pickrell Aug 2012

Mitochondrial Involvement In Neurodegeneration And Aging, Alicia M. Pickrell

Open Access Dissertations

Mitochondrial DNA (mtDNA) damage and oxidative phosphorylation deficiency have been associated with neurodegenerative disease and aging. I hypothesized that different neuronal populations handle the respiratory chain dysfunctions differently and that different cell types also mediate different responses to mtDNA damage. We used a transgenic mouse model under the tetracycline response system to temporally and spatially control the expression of a mitochondrial-targeted restriction endonuclease, mito-PstI. Using different neuronal drivers to induce the expression of mito-PstI, we were able to elicit mtDNA double strand break damage causing an OXPHOS deficiency in vivo (Chapter 2 and 3). We then were able to follow …

Schmidtea Mediterranea Planaris As A Novel Animal Model For Investigating The Dynamics Of Nervous System Development In Fetal Alcohol Spectrum Disorder, Jesse Lowe

Theses and Dissertations

Fetal Alcohol Spectrum Disorder (FASD) is a prevalent developmental disease that is caused by excess in utero exposure to ethanol. The importance of timing and dosage of ethanol has been elucidated by previous investigations conducted with humans, rodents, and other model organisms, but challenges associated with determining functional deficiencies have necessitated the introduction of a novel animal model for investigating the dynamics of FASD. In this thesis, we propose that the freshwater flatworm planarian species, Schmidtea mediterranea, has the capability to fulfill this need. Planaria have the ability to completely regenerate fragments into intact, functional animals, and in these studies, …

Biological Sciences Faculty Research and Publications

Huntington's disease (HD) is a devastating neurodegenerative disorder with no disease-modifying treatments available. The disease is caused by expansion of a CAG trinucleotide repeat and manifests with progressive motor abnormalities, psychiatric symptoms, and cognitive decline. Expression of an expanded polyglutamine repeat within the Huntingtin (Htt) protein impacts numerous cellular processes, including protein folding and clearance. A hallmark of the disease is the progressive formation of inclusions that represent the culmination of a complex aggregation process. Methylene blue (MB), has been shown to modulate aggregation of amyloidogenic disease proteins. We investigated whether MB could impact mutant Htt-mediated aggregation and neurotoxicity. MB …

Fluorescene Imaging Of Mouse Sperm / Noorain H., H. Nooraain Aug 2012

Fluorescene Imaging Of Mouse Sperm / Noorain H., H. Nooraain

Student Works (2010-2019)

The protocol for detecting acrosome in mouse sperm was developed by using two types of fluorescent dyes namely fluorescein conjugated Pisum sativum agglutinin (FITC-PSA) and rhodamine conjugated Lens culinaris agglutinin (TRITC-LCA). The results showed that mean percentages of acrosome intact sperm incubated in vitro were considerably low. After one-hour incubation there were 43.20±2.83%, 31.81±8.44% and 30.90±4.15% for ICR, C57/6J and F1 sperm, respectively. Intact acrosome in ICR sperm was significantly reduced (p<0.05) to 16.95±4.23% after two-hour incubation, however, insignificant reduction (p>0.05) was found for C57BL/6J and F1 sperm, which were 30.03±2.06% and 20.93±3.81%, respectively. The results showed that both dyes FITC-PSA and TRITC-LCA suitably stained the mouse sperm …

Preliminary Assessment Of The Effect Of Plant Protease Inhibitors On The Expression Of Heterologous Genes In Plant Cells / Nassim Khalighi, Nassim Khalighi Aug 2012

Preliminary Assessment Of The Effect Of Plant Protease Inhibitors On The Expression Of Heterologous Genes In Plant Cells / Nassim Khalighi, Nassim Khalighi

Student Works (2010-2019)

Despite the successful development of pant-based expression systems for the production of a wide range of recombinant proteins in recent years, there remain serious limitations in the final yield accumulation of the foreign protein in the plant cells used. Plant protease activity represents a significant barrier to efficient and hence economical recombinant protein production. Inadequate information is available on specific plant proteases and its role in foreign protein degradation. Some strategies have been reported recently to overcome endogenous plant protease effects on foreign protein integrity and activity in plants including the co-expression of recombinant proteases to reduce the endogenous protease …

Α2-Adrenoceptor And 5-Ht3 Serotonin Receptor Ligands As Potential Analgesic Adjuvants, Genevieve Alley Aug 2012

Α2-Adrenoceptor And 5-Ht3 Serotonin Receptor Ligands As Potential Analgesic Adjuvants, Genevieve Alley

Theses and Dissertations

There continues to be a need for more effective analgesics. The α2-adrenoceptor (AR) agonist clonidine is an analgesic whose use is severely limited by undesirable side effects. meta-Chlorophenylguanidine (MD-354), an agent developed in our laboratory, selectively potentiates the antinociceptive effects of clonidine in a biphasic manner. Mechanistic studies suggest that both 5-HT3 receptor and α2-AR mechanisms are involved. To further evaluate mechanisms underlying the analgesia-potentiating effect of clonidine by MD-354, pharmacological studies using more established 5-HT3 receptor agonists: meta-chlorophenylbiguanide (mCPBG) and centrally-acting SR57227A, and non-selective α2-adrenoceptor ligand TDIQ, administered alone and in combination with clonidine, were conducted in mouse antinociceptive …

Investigating Synergy Between Ribonucleotide Reductase Inhibitors And Cmv Antivirals, Sukhada Bhave Aug 2012

Investigating Synergy Between Ribonucleotide Reductase Inhibitors And Cmv Antivirals, Sukhada Bhave

Theses and Dissertations

Cytomegalovirus (CMV) infections remain a significant problem in congenitally infected infants and immunocompromised individuals. Modest antiviral activities of currently approved drugs coupled with dose-limiting toxicities restrict effectiveness and promote development of resistance. The potential for ribonucleotide reductase (RR) inhibitors hydroxyurea (HU), Didox, and Trimidox to synergize, through reduction of nucleotide pools, with the deoxynucleotide analog Ganciclovir (GCV) was examined. A yield reduction assay that utilizes luciferase expressed by a recombinant virus as a surrogate measure of viral infectious units was developed and used to determine effective dose ranges for each drug. RR inhibitors exhibited intrinsic anti-CMV activities on their own …
