A Conserved Two-Component Regulatory System, Pids/Pidr, Globally Regulates Pigmentation And Virulence-Related Phenotypes Of Burkholderia Glumae, Hari Sharan Karki, Inderjit Kaur Barphagha, Jong Hyun Ham

Burkholderia glumae is a rice pathogenic bacterium that causes bacterial panicle blight. Some strains of this pathogen produce dark brown pigments when grown on casamino-acid peptone glucose (CPG) agar medium. A pigment-positive and highly virulent strain of B.glumae, 411gr-6, was randomly mutagenized with mini-Tn5gus, and the resulting mini-Tn5gus derivatives showing altered pigmentation phenotypes were screened on CPG agar plates to identify the genetic elements governing the pigmentation of B.glumae. In this study, a novel two-component regulatory system (TCRS) composed of the PidS sensor histidine kinase and the PidR response regulator was identified as an essential regulatory factor for pigmentation. Inferring The Evolutionary History Of Divergence Despite Gene Flow In A Lizard Species, Scincella Lateralis (Scincidae), Composed Of Cryptic Lineages, Nathan D. Jackson, Christopher C. Austin

Although recent radiations are fruitful for studying the process of speciation, they are difficult to characterize and require the use of multiple loci and analytical methods that account for processes such as gene flow and genetic drift. Using multilocus sequence data, we combine hierarchical cluster analysis, coalescent species tree inference, and isolation-with-migration analysis to investigate evolutionary relationships among cryptic lineages of North American ground skinks. We also estimate the extent that gene flow has accompanied or followed diversification, and also attempt to account for and minimize the influence of gene flow when reconstructing relationships. Cryopreservation of sperm from Xiphophorus fishes has produced live young in three species: X. hellerii, X. couchianus, and X. maculatus. In this study, the goal was to establish protocols for sperm cryopreservation and artificial insemination to produce live young in X. variatus, and to identify needs for repository development. The objectives were to: 1) collect basic biological characteristics of males; 2) cryopreserve sperm from X. variatus, 3) harvest live young from cryopreserved sperm, and 4) discuss the requirements for establishment of sperm repositories. The Borrelia burgdorferi BpaB proteins of the spirochete's ubiquitous cp32 prophages are DNA-binding proteins, required both for maintenance of the bacteriophage episomes and for transcriptional regulation of the cp32 erp operons. Through use of DNase I footprinting, we demonstrate that BpaB binds the erp operator initially at the sequence 5′-TTATA-3′. Electrophoretic mobility shift assays indicated that BpaB also binds with high affinity to sites located in the 5′ noncoding regions of two additional cp32 genes. Characterization of the proteins encoded by those genes indicated that they are a single-stranded DNA-binding protein and a nuclease, which we named SsbP and NucP, …

Hannah Tapfield King (1807-1886), converted to The Church of Jesus Christ of Latter-day Saints in 1850 in Cambridge, England, and emigrated with her family to Utah. She was a prolific and popular writer of poetry, essays, and educational pieces for Utah's newspapers, and she had a reputation as a woman of refinement. From her autobiography one can see her complete devotion to the Church. She and her family sacrificed a comfortable middle class life in England running the family farm, and they suffered considerably in Utah's desert. Her husband, Thomas King, eventually joined the Church but was never active or …

The domestic dog offers a unique opportunity to study complex disorders similar to those seen in humans, but within the context of the much simpler genetic backgrounds of pure breeds, which represent closed populations. We performed a whole-genome search for genetic risk factors of adult-onset deafness in the Border Collie, a breed of herding dog that relies on acute hearing to perceive and respond to commands while working. Adult-onset deafness in Border Collies typically begins in early adulthood and is similar to age-related hearing loss in humans. Maximal Force Characteristics Of The Ca2+-Powered Actuator Of Vorticella Convallaria, Sangjin Ryu, Matthew J. Lang, Paul Matsudaira

The millisecond stalk contraction of the sessile ciliate Vorticella convallaria is powered by energy from Ca2+ binding to generate contractile forces of ~10 nN. Its contractile organelle, the spasmoneme, generates higher contractile force under increased stall resistances. By applying viscous drag force to contracting V. convallaria in a microfluidic channel, we observed that the mechanical force and work of the spasmoneme depended on the stalk length, i.e., the maximum tension (150–350 nN) and work linearly depended on the stalk length (~2.5 nN and ~30 fJ per 1 mm of the stalk). Protein phosphatase activity acts as a primary determinant of the extent and duration of phosphorylation of cellular proteins in response to physiological stimuli. Ser/Thr protein phosphatase-1 (PP1) belongs to the PPP superfamily, and is associated with regulatory subunits that confer substrate specificity, allosteric regulation, and subcellular compartmentalization. In addition, all eukaryotic cells contain multiple heat-stable proteins that originally were thought to inhibit phosphatase catalytic subunits released from the regulatory subunits, as a fail-safe mechanism. "The past is certainly a distant land and getting there is a difficult and imperfect undertaking" (Brown, Hirschman & Maclaran (2006). This paper explores motivations behind how consumers reach that "distant land." Over 1,000 respondents of a variety of ethnic groups show very different stories and diaspora timelines, but personal identity and connection with place are always top ranked motivations for interest in ancestors. How might groups, who may suffer from a lack of identity, fit into these findings when 'personal identity' is the number one reason why consumers engage in genealogy and legacy tourism? This dissertation examines the occurrence of catalogs and lists in the literary works of several twentieth-century authors, including James Joyce, poet James Schuyler, novelist and cultural historian James McCourt, the postmodern fabulist Thomas Pynchon, and the French experimental prose author Georges Perec. The dissertation seeks to trace how each author makes use of catalogs in his work, how catalogs form a central part of his style and subject matter, and how his use of catalogs can be read against the biographical, historical, and social contexts surrounding his life and work. The Thai Journal of Veterinary Medicine
The safety of ethanolic extract from the seeds of Moringa oleifera was evaluated for its oral toxicity by acute toxicity test and subchronic toxicity study in experimental animals. The acute toxicity test in mice showed that the extract at doses ranging from 5.1 to 10.0 g/kg caused toxic signs and dose dependent mortality. Oral administration of the extract in rats at doses of 100, 500 and 1000 mg/kg/day for 90 consecutive days revealed that male rats treated with the extract at 1000 mg/kg had a significant decrease in RBC but this alteration was within rats reference range. This study was conducted to investigate Chikungunya virus (CHIKV) infection in mammal and avian byusing mice and baby chickens as model animals. Thailand 2010 and Ross/186 strain (reference strain) were used inthis study. 104, 106 and 108 CID50 of CHIKV were inoculated to four- and six-week-old mice. Blood was collected andtested for virus for seven days. 108 CID50 of CHIKV was inoculated to two-week-old mice. Blood was collected andtested for virus for five days. The primary goal in development of at-risk species conservation assessments is to compile biological and ecological information that may assist conservation practitioners in making decisions regarding species of interest. The Nebraska Natural Legacy Project recognizes the plains harvest mouse (Reithrodontomys montanus griseus) as a Tier I at-risk species of high conservation need. Some general management recommendations are made here regarding the plains harvest mouse; however, conservation practitioners will need to use professional judgment to make specific management decisions based on objectives, location, and a multitude of variables. This paper examines how new Hmong textile forms that started in Ban Vinai refugee camp have served as dialogical performance, spiritual and political agency, and transnational commodity in the diasporas. These textiles create a hybrid social language that draws from a diversity of linguistic consciousnesses in a culture where there was not a universally understood writing system until the mid-20th century. Traditional paj ntaub (flower cloth) garments in Laos were a primary indicator of Hmong identity and can be seen as an alternate text. Northern Californian has a long history of protest and political involvement. For Linda Gass, Gyongy Laky and Linda MacDonald the passion they bring to political issues is reflected in their artwork and the paths their lives have followed. Based on personal interviews and research, this paper will shed light on their views and the artistic means used to express them. Each of these artists is engaged in political activism in her own way. In each case we are drawn in by the form, beauty or humor of the textiles.
