The use of vaccinia virus (VACV) as a vaccine resulted in the eradication of smallpox in 1979. Characteristics that contribute to the effectiveness of VACV as a vaccine and viral vector include its ability to elicit strong, long-lived humoral and cell-mediated immune responses as a live-replicating virus and to accept large inserts of DNA into its genome. However, adverse events associated with its use as the smallpox vaccine have constrained it from being more widely utilized in vaccines and therapies. Scholars analyzing the intersection of federalism and disaster law and policy have primarily focused on the difficulties federalism poses for interjurisdictional coordination of disaster response. Though scholars have highlighted that rising disaster risks and costs are associated with “land-use planning that exacerbates, rather than mitigates, disaster risk,” a more holistic analysis of land-use-related disaster law and policy is needed. This Article provides a more comprehensive framework within which to analyze prospective mitigation or prevention of disaster risk and costs through a rebalancing—or reconstituting—of the respective roles of the federal and state governments in land-use planning. Rev. B. Fay Mills was a popular, late nineteenth century Protestant evangelist whose fame approached that of the eminent Gospel preacher, Dwight L. Moody. Preaching to audiences in large urban settings, Mills’ revivals captured headlines and significant column space as he preached sermons of individual salvation from sin from the perspective of Christian orthodoxy. Yet, just as he was reaching the very top of the field of itinerant evangelists, he changed his message to reflect his growing interest in and association with the Social Gospel movement. Research indicates that sustainability higher education (SHE) has been promoted since the 1970s but has not achieved satisfactory progress in meeting original goals. Reflecting the evasive nature of sustainability as a goal, SHE programs appear stunted and there is little overall guidance with regard to curricula development. This dissertation addresses this issue by conducting a comprehensive literature research and sampling of those in sustainability post-graduate programs in an effort to determine an articulable set of core thinking and learning elements to assist in implementing SHE programs. The data were collected from the teachers about their perceptions regarding the female principals with whom they work. This qualitative study was conducted by interviewing 8 teachers from 3 elementary schools in northeast Tennessee. The teachers were interviewed to understand their perceptions of effective leadership practices exhibited by female principals.

Most nuclease enzymes can hydrolyze phosphoester bonds (in DNA and RNA) by using metal ions cofactors that coordinate and activate water molecules in the enzymes' active sites. However, there are some hydrolase enzymes (including nucleases) that can function without the aid of metal ions. 2,6-Di(1H-imidazol-2-yl)phenol, a model compound for hydrolase enzyme, was synthesized by the reaction between ethylenediamine and dimethyl-3-carboxysalicylate, initially resulting in the formation of diimidazoline. The diimidazoline was subsequently aromatized to the diimidazole by dehydrogenation over palladium. The overall reaction yield was low; therefore, other dehydrogenation transformation reactions were tried but all failed to improve the …

This research is done to determine the distribution of flavonoids in diverse varieties of onions (commercial and wild onions). Reflux extraction method was done on dried onions and the abstracts were analyzed subsequently by high performance liquid chromatography (HPLC) and gas chromatography-mass spectrometry (GCMS). Statistical calculations were done to see if there are significant differences in the varieties of onions studied from the chromatographic profiles obtained. Phylogenetic analyses suggest that a novel DNA sequence (NS1) found in a boreal forest soil-clone library belongs to the fungal kingdom but does not fall unambiguously within any known class. In order to determine if NS1 codes for an authentic ribosomal RNA (rRNA) gene-copy, I modeled ribosomal RNA secondary structure for four gene regions. Such analyses have never been used on environmental ribosomal sequences before. It appears that NS1 does code for an authentic gene-copy and is not a biological or lab artifact. I also elucidated the habitat preferences, horizon preferences, and fine-scale spatial structure of NS1 using molecular methods. …

While climate systems are known be nonlinear, most statistical tools used to study climate are linear. Two nonlinear analyses are introduced for indicating predictability in climate studies: Hurst analysis and Renyi analysis, the advantages of which are illustrated by applying both to characterize Alaska climate time series 'dynamics' or temporal evolution. These methods are also applied to reanalysis and model data to compare with the observational analysis. A needs assessment was conducted identifying the need for a training program in both culture and communication. Research questions explored the need for a training program in communication and cultural diversity. Few studies have investigated landslides in frozen ground. As Alaska's population grows and climate warms, we can expect an increase in landslide frequency. This thesis presents mapping and subsurface analysis of a slide occurring in discontinuous permafrost near Chitina, Alaska. The active slide intersects the McCarthy Road and occurs within an underlying clayey glaciolacustrine diamicton deposit in an area previously used as a dump. The multifaceted investigation included landslide feature mapping, seismic surveys, tree-ring sampling, and a slope stability analysis to back-calculate residual soil strength properties. Listeria monocytogenes, the causative agent of listeriosis can contaminate cold smoked salmon (CSS) and make it a high-risk ready-to-eat food product. Due to the lack of a critical control point in the cold-smoking process, a post-process elimination is essential to ensure product safety. The goal of this project was to improve safety of vacuum-packaged CSS supplemented with commercially refined liquid smoke (LS). In this work, state-of-the-art existing simulation models of East Barrow and Walakpa natural gas fields with associated gas hydrates were rebuilt, tuned with additional data (some of the data used were from the Mt. Elbert Well, which significantly improved earlier models), and updated in terms of production data and history matching. The nascent polypeptide-associated complex (NAC) is a highly conserved protein complex known to play an important role in the development of metazoan organisms, but its molecular function is not well understood. Recent evidence from experiments using Saccharomyces cerevisiae as model supported the hypothesis that the NAC is either a chaperone or a component of the cytosolic chaperone network that interacts with nascent peptides emerging from the ribosome.
