The Occurrence Of An Abdominal Fauna In An Articulated Tapir (Tapirus Polkensis) Recovered From The Miocene Gray Fossil Site Northeast Tennessee., Shannon M. Mcconnell

The analysis of samples recovered from the abdominal area of an articulated tapir from the Late Miocene (4.5-7 million bp) Gray Fossil Site, revealed a rich palyno-fauna comprised of about 94% egg/oocyst-like structures and 6% pollen and other palynomorphs. In addition, a tight grouping of six hickory nuts (Carya) was recovered from the same area suggesting that the samples represent the abdominal contents. The analysis of a sample from immediately outside the tapir produced a sample with 98% pollen and less than 0.5% egg-like structures. This study examined the epigeic spider fauna in Kings Mountain National Military Park. The aim of this study is to make this information available to park management for use in the preservation of natural resources. Pitfall trapping was conducted monthly for one year in three distinct habitats: riparian, forest, and ridge-top. The study was conducted from August 2009 to July 2010. One hundred twenty samples were collected in each site. Overall, 289 adult spiders comprising 66 species were collected in the riparian habitat, 345 adult comprising 57 species were found in the forest habitat, and 240 adults comprising 47 species …

The Fontan circulation is the result of a series of operations performed to save the lives of children born with univentricular circulations. The Fontan procedure achieves venous return to the pulmonary circulation without a ventricular power source. The load on the heart is reduced to normal, and these patients can lead a normal life into adulthood, although late complications continue to prevent normal lifespan. A unique feature of the Fontan circulation is the dependency of inferior vena cava flow on respiration. In the past Accelerated Vehicle Retirement (AVR) programs have been implemented to combat the economic and environmental costs of automobile dependent societies. Seventy-five such programs have been implemented worldwide since 1990. This thesis examines correlations among factors affecting program performance, the relationship of articulated program objectives to program performance, and how factors affecting program performance influence environmental concerns. The purpose of this study is to propose a historically appropriate rehabilitation plan for the Biltmore Estate's Ramble based upon Olmsted's original design intent. In order to ascertain Frederick Law Olmsted's original design intent for the Ramble, an extensive review of Victorian Era garden design, cultural landscapes, the U.S. Horticultural Industry, and possible design influences on Olmsted as he designed the site was performed. Using primary archival materials and secondary sources, the developmental history of the Ramble from 1890 to present was studied. The 2002 Farm Bill included a provision requiring various agricultural commodities to comply with country of origin labeling (COOL). Proponents of COOL argued that the policy would support U.S. producers and benefit U.S. consumers. This argument was based on the assumption that U.S. consumers prefer domestic products to imported products and hence the demand for U.S. products would increase following the implementation of COOL. Murashige and Skoog 1962 performed one-factor-at-a-time (OFAT) experimentation in order to regenerate tobacco callus on semi-solid agar medium. This work became an established medium for tissue culture micropropagation and experimentation. Micropropagation is done in niche markets with herbaceous perennials, among other crops, and the optimization of various inputs to produce maximal responses is a necessary step towards process development. This current study of macronutrient factors simultaneously altered media volume, amount of tissue (plants per vessel), sucrose, nitrogen (as NO3- and NH4+ ions), and K+ in a d-optimal design space with only 55 experimental units (including 5 true replicates). The effectiveness of a narrow grass hedge in reducing runoff nutrient loads following manure application was examined in this study. Beef cattle manure was applied to 0.75 m wide by 4.0 m long plots established on an Aksarben silty clay loam located in southeast Nebraska. Manure was added at rates required to meet none or the 1, 2, or 4 year nitrogen requirements for corn. Runoff water quality was measured during three 30 min simulated rainfall events. Manure application rate significantly affected dissolved phosphorus (DP) and total phosphorus (TP) loads in runoff on the plots without a grass hedge. However, …

Female preferences are frequently invoked to explain the widespread occurrence of elaborate male ornaments, yet empirical data demonstrating such preferences are sometimes equivocal or even contradictory. In the wolf spider Schizocosa stridulans, despite evidence of strong female choice, prior research has been unable to link the conspicuous sexually dimorphic foreleg ornamentation of males to their mating success. We conducted three experiments aimed at determining the function of this previously enigmatic ornamentation. Background: Recombinant DNA technologies have played a pivotal role in the elucidation of structure-function relationships in hemoglobin (Hb) and other globin proteins. Here we describe the development of a plasmid expression system to synthesize recombinant Hbs in Escherichia coli, and we describe a protocol for expressing Hbs with low intrinsic solubilities. Since the alpha- and beta-chain Hbs of different species span a broad range of solubilities, experimental protocols that have been optimized for expressing recombinant human HbA may often prove unsuitable for the recombinant expression of wildtype and mutant Hbs of other species.

Phylogenetic reconstructions provide a means of inferring the branching relationships among members of multigene families that have diversified via successive rounds of gene duplication and divergence. Such reconstructions can illuminate the pathways by which particular expression patterns and protein functions evolved. For example, phylogenetic analyses can reveal cases in which similar expression patterns or functional properties evolved independently in different lineages, either through convergence, parallelism, or evolutionary reversals. MITOSTATIN, a novel putative tumor suppressor gene induced by decorin overexpression, is expressed in most normal human tissues but is markedly down-regulated in advanced stages of mammary and bladder carcinomas. Mitostatin negatively affects cell growth, induces cell death and regulates the expression and activation levels of Hsp27. In this study, we demonstrated that ectopic expression of Mitostatin in PC3, DU145, and LNCaP prostate cancer cells not only induced a significant reduction in cell growth, but also inhibited migration and invasion. OBJECTIVES: To identify renal clear cell carcinoma-associated marker proteins. METHODS: Twelve patients with renal cell carcinoma (RCC) were collected and processed in the Department of Urology, Renmin Hospital, Wuhan University, China, between January 2008 and September 2009. Two-dimensional polyacrylamide gel electrophoresis and matrix assisted laser desorption ionisation time-of-flight mass spectrometry (MALDI-TOF-MS) were employed to investigate differentially expressed protein spots between RCC tissues and adjacent normal tissues, then reverse transcription polymerase chain reaction and western blot were employed to confirm the proteomic results. OBJECTIVES: To investigate the drug susceptibility pattern of isolated Mycobacterium tuberculosis (M. tuberculosis) against conventional anti-tuberculosis drugs in Dhaka, Bangladesh. METHODS: Sputum samples from 101 suspected new and previously treated patients were collected and M. tuberculosis was identified by microscopic observation and Ziehl-Neelsen staining. Drug susceptibility was performed against 4 anti-tuberculosis drugs, and the obtained data was analyzed. In this study we developed a theory grounded in data from women who continued healthy eating behaviors after a weight management program. Participant recruitment was guided by theoretical sampling strategies for focus groups and individual interviews. Inclusion criteria were: African American or Caucasian women aged 30+ who lost > 5% of their body weight in a weight management program > year ago. Participants > 5% below their baseline weight were maintainers (n = 9); those above were non-maintainers (n = 14). We asked open-ended questions regarding healthy eating behaviors. The binding of drugs with serum proteins can affect the activity, distribution, rate of excretion, and toxicity of pharmaceutical agents in the body. One tool that can be used to quickly analyze and characterize these interactions is high-performance affinity chromatography (HPAC). This review shows how HPAC can be used to study drug-protein binding and describes the various applications of this approach when examining drug interactions with serum proteins. Cultured juvenile blue crabs ( Callinectes sapidus Rathbun) were held for up to 77 days and monitored for survival, growth and development in low salinity water containing various compositions of ions. ~rabs were isolated in separate containers and placed in thirty gallon aquaria containing 2ppt of ion solution. Temperature, ambient, humidity, and nitrite levels were monitored daily. Molts were measured to determine growth, and molt increments were recorded to determine rate of development. We report evidence that Amblyomma maculatum tick populations are well established in southeastern Virginia. We found that 43.1% of the adult Gulf Coast ticks collected in the summer of 2010 carried Rickettsia parkeri, suggesting that persons living in or visiting southeastern Virginia are at risk for infection with this pathogen.

We examined the effects of the introduced gypsy moth (Lymantria dispar) on seven species of North American woodpeckers by matching spatially explicit data on gypsy moth outbreaks with data on breeding and wintering populations. In general, we detected modest effects during outbreaks: during the breeding season one species, the Red-headed Woodpecker (Melanerpes erythrocephalus), increased over pre-outbreak levels, while during the winter one species, the Yellow-bellied Sapsucker (Sphyrapicus varius), increased and one, the Downy Woodpecker (Picoides pubescens), decreased from pre-outbreak levels. The reaction mechanisms employed during non-ribosomal peptide (amide) synthesis in the non-ribosomal modular enzyme complex and in the enzyme glutaminyl cyclase were also used for spontaneous peptide (amide) synthesis in a buffered, abiotic solution. Analytical analyses revealed that spontaneous peptide (amide) bond formation occurred under mild prebiotic conditions during both the thioester-based production of phenylalanine peptides and peptidyl thioesters, and the spontaneous cyclization of glutamine t-butyl ester (Gln-tbe) into pyroglutamic acid t-butyl ester. Internal standard-based quantitative MS determined that 4.9 x 10-5 sec-1 was the rate constant for spontaneous Gln-tbe cyclization in imidazole buffer, pH 7, at 50°C, and that …

Predaceous diving beetles (Coleoptera: Dytiscidae) are a model group of organisms for testing coexistence and exclusion mechanisms because numerous species of the family interact in isolated, aquatic habitats, and are ubiquitous throughout landscapes. Two morphologically similar species, laccophilus fasciatus rufus and Laccophilus proximus, co-occur in numerous habitats in South Mississippi, but are hypothesized to have strong interspecific interactions. I investigated several possible mechanisms that may promote such coexistence, including segregation in habitat domain, differences in behavior, and differences in dispersal that may allow co-occurrence between these two species of beetle. The Gulfcoast ticks transmit disease-causing pathogens to humans and animals. Rickettsia parkeri is notable among the pathogens transmitted by A. maculatum to humans. Heavy infestations of A. maculatum on animal ears cause them to become thickened and curled, a condition commonly called "gotch ear." The tick's multifunctional salivary glands are vital to their biological success and likely also play a critical role in transmission of disease; tick saliva contains a broad array of secretory products that facilitate prolonged tick attachment and feeding; disrupting tick blood feeding or inactivating key tick salivary proteins presents a novel strategy for tick-borne disease prevention. …
