This study tested a hypothetical model for predicting both graduate GPA and graduation of University of Louisville Kent School of Social Work Master of Science in Social Work (MSSW) students entering the program during the 2001-2005 school years. The preexisting characteristics of demographics, academic preparedness and culture shock along with the subjective experiences of academic stability and academic performance were studied. A hierarchical multiple regression analysis was used to determine the best predictors of final GPA. The best predictors were age, undergraduate GPA, differences between undergraduate and graduate institution size, continuity index, and the course completion ratio. A hierarchical logistic …

This thesis explores pattern juxtaposition using traditional American quilt block designs as the compositional basis for nine works pieced from screen printed paper. In the 1970s, the quilt was celebrated for its apparent connection to Modernist design and was reclaimed by many feminist artists including Miriam Schapiro as a symbol of the exclusion of women's crafts from the arts hierarchy. The tension between these two perspectives continues today in the contemporary art quilt movement. Reconciliation Of Il-1ss Loss With Trif-Biased Tlr4 Signaling By Monophosphate Lipid A., Chelsea A. Embry

Monophosphoryl Lipid A (MPLA), a derivative of LPS endotoxin, is a TLR4 agonist that displays as little as 0.1-1% as much toxicity as its parent molecule while retaining immunostimulatory properties. We discovered that MPLA activates a TRIF-biased pattern of TLR4 signaling, resulting in reduced production of MyD88-dependent pro-inflammatory factors, and credited TRIF-bias for MPLA's reduced toxicity. A contemporary study showed that MPLA fails to promote maturation of the potent inflammatory cytokine IL-1ß. This dissertation seeks to reconcile MPLA's TRIF-biased signaling with IL-1 ß loss, and to determine the ultimate cause of MPLA's reduced toxicity compared to LPS. We find that …

The purpose of this study was to examine the phenomenon of teenage prescription drug abuse in Jefferson County, Kentucky to gain a better understanding of the environment and characteristics of a teenager who abuses prescription drugs, and to gain insight into this situation through the perspectives of adult professionals who regularly work with teenagers. In-depth interviews were conducted with five professionals, and data analyses were performed to determine recurring themes. There were four primary emergent themes, including: profiling of a teenage prescription drug abuser; factors influencing likelihood of drug abuse; portfolio of mental, emotional, and social issues; and treatment processes. …

The initial goals of this study include locating and identifying the taxonomic groups mentioned in Moffitt's (1993) (i.e. life-course persistent offenders, adolescent-limited offenders) using data from the National Longitudinal Survey 1997 (NLSY97). Further, this study compares the social demographics with the predictions of Moffitt (1993,1994) as her theory describes race, particularity those of African-American offenders. This study also examines the role of parental and peer relationships and their effect on the offender disparity among the typologies defined by Moffitt (1993). A phenomenological study of the lived experience of mothering during incarceration was conducted at a women's multi-custody level prison in Kentucky. The purpose of the study was to explore and describe the experience of mothering among incarcerated women to address the gap within this body of literature. Semi-structured interviews, observation, and field notes from 24 participants were collected and analyzed to provide an in-depth understanding of the mothering experience for the participants. Using black feminist perspective and standpoint this study explored factors that affect black women's matriculation and retention in graduate degree programs by examining how experiences and opportunities connected to race, class, and gender inform their decision to pursue and persist through graduate education. Specifically the study sought to investigate how the outlined factors affect the decision to pursue and matriculate through a graduate degree program. "Regen-Kanon" is the fourth of six pieces or "sound images" in the orchestral cycle Spharen, composed by York Holler between 2001 and 2006. The first four movements, namely "Wolkengesang" ("Song of the Clouds"), "Windspiel" ("Wind Chimes"), "Erdschichten" ("Layers of Earth"), and "Regen-Kanon" ("Rain Canon") are related to Holler's experiences of particular landscapes and consequently to associated impressions of nature, whereas the fifth and sixth movements, "Feuerwerk" ("Fireworks") and "Spharentrauer" ("Sorrow of the Spheres") convey different kinds of images. Mixed-effects model is an efficient tool for analyzing longitudinal data. The random effects in mixed-effects model can be used to capture the correlations among repeated measurements within a subject. The time points are not fixed and all available data can be used in mixed-effects model provided data are missing at random. This work presents a novel comprehensive effort to understand the fluid dynamics within orbiting dishes and the effects of the resulting oscillating fluid flow on shear and endothelial cellular responses within the dishes. High dynamic range imaging involves imaging at a bit depth higher than the typical 8-12 bits offered by standard video equipment. We propose a method of imaging a scene at high dynamic range, 14+ bits, to detect motion correlated with changes in the measured optical signal. Features within a scene, namely edges, can be tracked through a time sequence and produce a modulation in light levels associated with the edge moving across a region being sampled by the detector. Juvenile delinquency with co-occurring substance abuse and mental health disorders has become an increasing problem within the United States. In part this can be attributed to the excessive number of delinquent youth entering the juvenile justice system with untreated substance abuse and/or mental health disorders-thus requiring juvenile justice to become a default system for substance abuse and mental health intervention and treatment services. Also, the films were analyzed to see if correlation exists between overt religious content and IMDb.com user ratings. The overarching goal of the Pathway Semantics Algorithm (PSA) is to improve the in silico identification of clinically useful hypotheses about molecular patterns in disease progression. By framing biomedical questions within a variety of matrix representations, PSA has the flexibility to analyze combined quantitative and qualitative data over a wide range of stratifications. The resulting hypothetical answers can then move to in vitro and in vivo verification, research assay optimization, clinical validation, and commercialization. Band 4.1B is a cytoskeletal adaptor protein that regulates various cellular behavior; however, the mechanisms by which Band 4.1B contributes to intracellular signaling are unclear. This project addresses in vivo and in vitro functions for Band 4.1B in integrin-mediated cell adhesion and signaling. Band 4.1B has been shown to bind to β8 integrin, although cooperative functions of these two proteins have not been determined. Here, functional links between β8 integrin and Band 4.1B were investigated using gene knockout strategies. Ablation of β8 integrin and Band 4.1B genes resulted in impaired cardiac morphogenesis, leading to embryonic lethality by E11.5. These embryos …

The human nucleotide excision repair (NER) pathway resolves a range of lesions in DNA including those caused by harmful UV light, environmental mutagens, and agents of chemotherapy. The pathway involves the concerted action of over 30 proteins that recognize the damage, excise it within an oligonucleotide, and fill in the resulting gap, restoring the DNA to its original form. Computational systems biology is an emerging field built upon advances in biological experimentation, computational tools, and joint interdisciplinary efforts from physical sciences to engineering. In this thesis, I solve two important problems in the developmental genetics of Drosophila melanogaster by means of two different computational systems approaches, one stochastic and the other deterministic. In the first problem I use exact simulations of a spatially extended Master Equation to analyze intrinsic fluctuations in the gradient of the Bicoid morphogen. The combustion of fossil fuels has increased CO2 in the world's oceans and decreased pH. Ocean acidification may alter the growth, survival, and diversity of marine organisms that synthesize shells due to decreased carbonate ion availability. I examined the response of larvae and early juveniles from three species of commercially and ecologically valuable bivalve shellfish (Mercenaria mercenaria, Argopecten irradians, and Crassostrea virginica) exposed to past, pre-industrial levels (~250 ppm), present (~390 ppm), and future (~750, 1500 ppm) levels of CO2. Under higher CO2 concentrations, all three species experienced decreased growth, survival, and metamorphosis with C. virginica being the least affected. …

Myelin is essential for proper human neurophysiology, but the precise molecular mechanisms underlying the myelination process remain poorly understood. One potential effector molecule, the b1 integrin, has been suggested to play a critical role in CNS myelination. In support of this, a6b1 integrin has been shown to modulate oligodendrocyte development in vitro and the loss of an integrin ligand, laminin, causes myelin defects in vivo. Here, we use transgenic mice (b1-CNSko) engineered to excise the b1 integrin gene at the neural progenitor stage to study how b1 integrin modulates oligodendrocyte development and myelination. Notch is an essential cell surface receptor whose proper functioning is critical during development in metazoans. Notch can be modified with two unique forms of O-linked glycosylation: O-fucose and O-glucose. Protein O-fucosyltransferase1 (Pofut1, OFUT1 in flies) and Protein O-glucosyltransferase (Poglut, Rumi in flies) are the glycosyltransferases that add O-fucose and O-glucose monosaccharides to Notch, respectively. Loss of Pofut1/Ofut1 or Poglut/rumi in mice or flies phenocopies Notch-null phenotypes, suggesting both forms of O-glycosylation are essential for Notch function. Finite mixture models emerge in many applications, particularly in biology, psychology and genetics. This dissertation focused on detecting associations between a quantitative explanatory variable and a dichotomous response variable in a situation where the population consists of a mixture. That is, there is a fraction of the population for whom there is an association between the quantitative predictor and the response and there is a fraction of individuals for whom there is no association between the quantitative predictor and the response. This research is a collaboration between the Department of Mechanical Engineering at Stony Brook University and the Department of Environmental Sciences at Brookhaven National Laboratory. It is proposed to use 14 MeV neutrons tagged by the associated particle neutron time-of-flight technique (APnTOF) to identify the fillers of unexploded ordnances (UXO). The ultimate goal is to construct a prototype portable neutron interrogation probe that will search for UXO in a target volume, locate targets in three dimensions, and accurately identify the major elemental constituents. We are interested in predicting the time dependent behavior of biochemical networks such as interaction between proteins. These networks are represented by a system of chemical reactions. In the forward problem, we have knowledge of all the reactions and the associated rate constants. We want to determine the joint probability density of the populations of all the molecular species at any time instant.
