Thermodynamics Of Dna Hybridization On Surfaces, Terry J. Schmitt, Thomas A. Knotts Iv May 2011

Thermodynamics Of Dna Hybridization On Surfaces, Terry J. Schmitt, Thomas A. Knotts Iv

Faculty Publications

Hybridization of single-stranded DNA (ssDNA) targets to surface-tethered ssDNA probes was simulated using an advanced coarse-grain model to identify key factors that influence the accuracy of DNA microarrays. Comparing behavior in the bulk and on the surface showed, contrary to previous assumptions, that hybridization on surfaces is more thermodynamically favorable than in the bulk. In addition, the effects of stretching or compressing the probe strand were investigated as a model system to test the hypothesis that improving surface hybridization will improve microarray performance. The results in this regard indicate that selectivity can be increased by reducing overall sensitivity by a …

Discovery Of A Functional Ecdysone Response Element In Brugia Malayi, Tracy Enright May 2011

Discovery Of A Functional Ecdysone Response Element In Brugia Malayi, Tracy Enright

USF Tampa Graduate Theses and Dissertations

The aim of this study was to determine whether functional ecdysone response elements (EcREs) exist within the genome of Brugia malayi, a parasitic nematode that causes lymphatic filariasis. The hypothesis that EcREs exist in B. malayi stemmed from previous demonstration of a functional ecdysone response system in B. malayi (Tzertzinis et al., 2010). Real-time PCR (qPCR) experiments were conducted to measure gene expression levels for twelve genes proximal to five putative EcREs in 20-OH ecdysone treated and untreated B. malayi embryos. Seven genes showed consistent upregulation with 20-OH ecdysone treatment. Each of the five putative EcREs had at least one …

Berle And Veblen: An Intellectual Connection, Charles R.T. O'Kelley May 2011

Berle And Veblen: An Intellectual Connection, Charles R.T. O'Kelley

Seattle University Law Review

For O’Kelley’s contribution to Berle II, he chose to highlight the need for further research into the connection between The Modern Corporation and Private Property and the work of Thorstein Veblen, the father and intellectual inspiration of institutional economics. While the connection between Veblen and Modern Corporation is indisputable, the meaning of that connection has not been explored in any depth. O’Kelley provides a framework for thinking about the nature of Veblen’s influence on Modern Corporation and suggests that further research on Veblen’s influence would be most fruitful. O’Kelley then provides an initial account of how Veblen’s work may …

Facilitating Service Co-Production: A Dramaturgical Perspective, Zagros Hatami May 2011

Facilitating Service Co-Production: A Dramaturgical Perspective, Zagros Hatami

Nordes Conference Series

Unlike products, the production and consumption of service occurs simultaneously with service users acting as co-producers of service. This role is significant as the quantity, quality and experience of service is often reliant on the quality of user efforts. Thus, service designers need to consider the co-productive roles various service actors are required to play at the time of service consumption. This awareness allows designers to facilitate this role taking process by setting the stage for users as well as other service actors to successfully play their part in the production of service. As service interactions are dyadic social interactions, …

Research And Design Practice: An Exploratory Update Of Donald Schön, Peter Ullmark May 2011

Research And Design Practice: An Exploratory Update Of Donald Schön, Peter Ullmark

Nordes Conference Series

The Reflective Practitioner and Educating the Reflective Practitioner, the two most influential books by Donald Schön from 1983 and 1987, have so far been regarded as a self-evident platform for design research. The ideas put forward have been much discussed but not basically questioned. However, during the last years the conditions for design practice has changed fundamentally. To find out if the understanding of Schön can be developed to match the new situation or if other approaches are necessary, a new critical discussion is necessary. This article is a first and explorative attempt to identify important issues for such an …

Conformational-Dependent And Independent Rna Binding To The Fragile X Mental Retardation Protein, Xin Yan, Robert B. Denman May 2011

Conformational-Dependent And Independent Rna Binding To The Fragile X Mental Retardation Protein, Xin Yan, Robert B. Denman

Publications and Research

The interaction between the fragile X mental retardation protein (FMRP) and BC1 RNA has been the subject of controversy. We probed the parameters of RNA binding to FMRP in several ways. Nondenaturing agarose gel analysis showed that BC1 RNA transcripts produced by in vitro transcription contain a population of conformers, which can be modulated by preannealing. Accordingly, FMRP differentially binds to the annealed and unannealed conformer populations. Using partial RNase digestion, we demonstrate that annealed BC1 RNA contains a unique conformer that FMRP likely binds. We further demonstrate that this interaction is 100-fold weaker than that the binding of eEF-1A …

Time Variant Cross Correlation To Assess Residence Time Of Water And Implication For Hydraulics Of A Sink‐Rise Karst System, V. Bailly-Comte, Jonathan B. Martin, E. J. Screaton

KIP Articles

Transport rates and residence time in the subsurface are critical parameters for understanding water‐rock interactions for efficient contaminant remediation. This paper presents a methodology for assessing flow and transit time of water through hydrological systems, with specific applications to karst systems and implication for hydraulics of a conduit system surrounded by a porous and permeable intergranular matrix. A time variant cross‐correlation function analysis is applied to bivariate time series that characterize mass transfer, assuming a stationary system using sliding windows of various sizes. We apply the method to 1 year long temperature records in the Santa Fe River (north central …

Dft Study Of Co2 Reduction To Hydrocarbons On Cu Surfaces, Li-Hui Ou, Sheng-Li Chen May 2011

Dft Study Of Co2 Reduction To Hydrocarbons On Cu Surfaces, Li-Hui Ou, Sheng-Li Chen

Journal of Electrochemistry

CO2 reduction on Cu(111) single crystal surfaces was studied using DFT calculations on the reaction energies and the minimum energy paths. The results indicated that the possible reaction paths for CO2 reduction on Cu(111) surface are CO2(g) + H* → COOH* → (CO +OH)*, (CO + H)* → CHO*, CHO + H → CH2O* → (CH2+O)*, CH2* + 2H* → CH4 or 2CH2* → C2H4. On Cu(111) surface, the reaction rate is controlled by steps of CH2O* → (CH2 …

Induced Resistance Studied In Maize With The Fall Armyworm, Spodoptera Frugiperda (J.E. Smith) (Lepidoptera: Noctuidae) And The Chemical Inducer Actigard, Stephen D. Danielson

Department of Entomology: Faculty Publications

Induced resistance has been defined as the "qualitative or quantitative enhancement of a plant's defense mechanisms against pests in response to extrinsic physical or chemical stimuli." (Kogan and Paxton, 1983). Physical stimuli can include infection by a pathogenic organism (Jorgensen et al, 1996; Morris et al, 1998) and feeding by insects (Felton and Eichenseer, 1999). Chemical stimuli include such organic compounds as salicylic acid (Hammerschmidt and Smith-Becker, 1999), jasmonic acid (Staswick and Lehman, 1999), and the commercially available Actigard® 50WG (Tally et al., 1999) (Syngenta Crop Protection, P.O. Box 18300, Greensboro, NC 27419). Actigard® 50WG is registered in the U.S. …

Adl-Specific Versus Standard Aquatic Exercise In Older Persons, David A. Edwards May 2011

Adl-Specific Versus Standard Aquatic Exercise In Older Persons, David A. Edwards

Open Access Dissertations

With aging there is a decrease in a person’s ability to perform activities of daily living (ADL) which may be most effectively addressed using training patterns that are biomechanically similar to ADL. Since aquatic exercise offers the opportunity to provide resistance with a high level of safety, the pool may afford the ideal environment for ADL-specific training in an aging population. Purpose: The purpose of this investigation was to compare a traditional aquatic exercise program (TRAD) to an aquatic program tailored to target ADL (ADLspec). Methods: Eighteen independently living individuals (68.7 + 7.5 years) were randomly assigned to a TRAD …

Additions To The Staphylinidae (Coleoptera) Of The Cayman Islands, J. H. Frank, J. Klimaszewski, D. S. Chandler

Insecta Mundi

In 1947, 20 species of Staphylinidae were reported from the Cayman Islands as a result of an Oxford University expedition there in 1938 which made extensive use of a light trap. The list is here expanded to 62 species based on collections by R. R. Askew, G. E. Ball, E. A. Dilbert, B. K. Dozier, E. J. Gerberg, P. J. Fitzgerald, M. C. Thomas, and R. H. Turnbow since 1970, all of whom also used light traps except for a collection or two by flight intercept trap.

Examination Of Nak-Associated Protein-1 (Nap1) Homo And Hetero-Interactions In The Interferon Pathway”, Richard Call May 2011

Examination Of Nak-Associated Protein-1 (Nap1) Homo And Hetero-Interactions In The Interferon Pathway”, Richard Call

Theses and Dissertations

Double stranded RNA (dsRNA), the genomic material of some viruses and a replication intermediate in others, is recognized by multiple signaling receptors that initiate the anti-viral response1. Viruses have developed mechanisms to circumvent the anti-viral response by targeting components of the signaling pathway. An example of one such pathway is the TLR3 signaling pathway, which contains a kinase complex that activates interferon regulatory factor 3 (IRF3), leading to production of type I interferons. The kinase complex consists of a scaffold protein, NAK-associated protein 1 (NAP1), and two kinases, TANK-binding kinase 1 (TBK-1) and IκB kinase epsilon (IKKε). A fourty residue …

Variations In Mre11/Rad50/Nbs1 Status And Dna Damage-Induced S-Phase Arrest In The Cell Lines Of The Nci60 Panel, Kristen M. K. Garner, Alan Eastman May 2011

Variations In Mre11/Rad50/Nbs1 Status And Dna Damage-Induced S-Phase Arrest In The Cell Lines Of The Nci60 Panel, Kristen M. K. Garner, Alan Eastman

Dartmouth Scholarship

The Mre11/Rad50/Nbs1 (MRN) complex is a regulator of cell cycle checkpoints and DNA repair. Defects in MRN can lead to defective S-phase arrest when cells are damaged. Such defects may elicit sensitivity to selected drugs providing a chemical synthetic lethal interaction that could be used to target therapy to tumors with these defects. The goal of this study was to identify these defects in the NCI60 panel of cell lines and identify compounds that might elicit selective cytotoxicity.

Ab Initio Description Of Nonlinear Dynamics Of Coupled Microdisk Resonators With Application To Self-Trapping Dynamics, Hamidreza Ramezani, Tsampikos Kottos, V. Shuvayev, L. Deych

Physics & Astronomy Faculty Publications

An ab initio approach is used to describe the time evolution of the amplitudes of whispering gallery modes in a system of coupled microdisk resonators with Kerr nonlinearity. It is shown that this system demonstrates a transition between Josephson-like nonlinear oscillations and self-trapping behavior. Manifestation of this transition in the dynamics of radiative losses is studied.

BACKGROUND: Neuronal iron accumulation is thought to be relevant to the pathogenesis of Parkinson's disease (PD), although the mechanism remains elusive. We hypothesized that neuronal iron uptake may be stimulated by functional mitochondrial iron deficiency. OBJECTIVE: To determine firstly whether the mitochondrial toxin, 1-methyl-4-phenylpyridinium iodide (MPP(+)), results in upregulation of iron-import proteins and transporters of iron into the mitochondria, and secondly whether similar changes in expression are induced by toxins with different mechanisms of action. METHODS: We used quantitative PCR and Western blotting to investigate expression of the iron importers, divalent metal transporter, transferrin receptor 1 and 2 (TfR1 and …

Videogames, Experiential Reality, Ethics, And Gamers, Sean Naubert May 2011

Videogames, Experiential Reality, Ethics, And Gamers, Sean Naubert

Senior Theses

“Videogames are a waste of time and rob people of a chance at a real life.” This has long been the status quo regarding videogames and gamers. Videogames, Experiential Reality, Ethics, and Gamers rallies against this by undermining the philosophical assumption that comes with the claims against gaming, namely that reality is solely and completely physically determined.

On this quest, “legends and philosophical clans” shall be called upon and united to besiege the conventional notion that there is a sole physical reality. This notion is cut down like so many orcs, leaving the Individual Champion as the lone warrior left …
