Association Of Glpq With Serum-Resistance In Salmonella Typhimurium, Kristie Carlotta Mitchell Nov 2011

Association Of Glpq With Serum-Resistance In Salmonella Typhimurium, Kristie Carlotta Mitchell

Master's Theses and Doctoral Dissertations

Salmonella typhimurium is a leading cause of infectious human gastroenteritis and systemic disease when it escapes into the bloodstream. The human serum complement system is central in fighting systemic bacterial infections. S. typhimurium exhibits resistance to killing by complement (serum-resistance), a trait conferred by several known virulence genes. This study investigates a potential chromosomal complement-resistance gene in S. typhimurium tagged by TnphoA insertional transposon mutagenesis. S. typhimurium strain EM876 has a chromosomal TnphoA insertion and reduced serum-resistance. Inverse PCR and subsequent DNA sequence analysis reveal the insertion to be in the gene glpQ. glpQ codes for a …

Positive Cooperative Mechanistic Binding Of Proteins At Low Concentrations: A Comparison Of Poly (Sodium N-Undecanoyl Sulfate) And Sodium Dodecyl Sulfate, Susmita Das, Monica R. Sylvain, Vivian E. Fernand, Jack N. Losso, Bilal El-Zahab, Isiah M. Warner

Faculty Publications

The interactions of the negatively charged achiral molecular micelle, poly (sodium N-undecanoyl sulfate) (poly-SUS), with four different proteins using intrinsic and extrinsic fluorescence spectroscopic probes, are studied. A comparison of poly-SUS with the conventional surfactant, sodium dodecyl sulfate (SDS), and the monomeric species, SUS, is also reported. In this work, we observed that poly-SUS preferentially binds to acidic proteins, exhibiting positive cooperativity at concentrations less than 1. mM for all proteins studied. Moreover, it appears that the hydrophobic microdomain formed through polymerization of the terminal vinyl group of the monomer, SUS, is largely responsible for the superior binding capacity of …

No Accession-Specific Effect Of Rhizosphere Soil Communities On The Growth And Competition Of Arabidopsis Thaliana Accessions, Anna G. Aguilera, Adán Colón-Carmona, Rick Kesseli, Jeffrey S. Dukes Nov 2011

No Accession-Specific Effect Of Rhizosphere Soil Communities On The Growth And Competition Of Arabidopsis Thaliana Accessions, Anna G. Aguilera, Adán Colón-Carmona, Rick Kesseli, Jeffrey S. Dukes

Biology Faculty Publication Series

Soil communities associated with specific plant species affect individual plants' growth and competitive ability. Limited evidence suggests that unique soil communities can also differentially influence growth and competition at the ecotype level. Previous work with Arabidopsis thaliana has shown that accessions produce distinct and reproducible rhizosphere bacterial communities, with significant differences in both species composition and relative abundance. We tested the hypothesis that soil communities uniquely affect the growth and reproduction of the plant accessions with which they are associated. Specifically, we examined the growth of four accessions when exposed to their own soil communities and the communities generated by …

Sweep, Step, Pulse, And Frequency-Based Techniques Applied To Protein Monolayer Electrochemistry At Nanoparticle Interfaces, Debbie S. Campbell-Rance, Tran T. Doan, Michael C. Leopold

Chemistry Faculty Publications

Protein monolayer electrochemistry (PME), a strategy using synthetic platforms to study the electron transfer (ET) properties of adsorbed proteins, has been successfully applied to proteins adsorbed at monolayer-protected gold cluster (MPCs) assembled films, an adsorption interface shown to be an effective alternative, compared to traditional self-assembled monolayer (SAM) films, for the immobilization and study of ET proteins. Within PME studies, cyclic voltammetry (CV) remains the most commonly applied electrochemical technique in spite of several limitations that occur when the sweep technique is used at either platform. In particular, CV for PME at MPC films results in analysis complications stemming from …

Describing Connections Between Science Content And Future Careers: Implementing Texas Curriculum For Rural At-Risk High School Students Using Purposefully-Designed Field Trips, Tommye Hutson, Susan Cooper, Tony Talbot

The Rural Educator

The state of Texas has an ‘essential knowledge’ component in some high school science courses indicating that students be able to describe connections between academic science content and future jobs or training through effective exposure to course content. The participants in this study were from a small rural high school in central Texas. Each was labeled as ‘at-risk’ and self-identified an inability to describe those types of connections after earning credit in more than one science course with that ‘essential knowledge’ component. A career-focused field trip to a local vocational/technology training center was designed to address that particular deficit. This …

Traversing Swanton Road, 6th Ed., James A. West

Academic, Government & Associated Organizational Research Resources

Situated at the northwest end of Santa Cruz County and occupying circa 30 square miles of sharply contrasted terrain, the Scott Creek Watershed concentrates within its geomorphological boundaries, at least 10-12% of California's flora, both native and introduced. Paradoxically, the use/abuse that the watershed has sustained over the past 140+ years, has not necessarily diminished the biodiversity and perhaps parallels the naturally disruptive but biologically energizing processes (fire, flooding, landslides and erosion), which have also been historically documented for the area for +60 years. This is an early edition of an extensive document by James A. West. Please visit the …

Oxidative Stress And Heme Oxygenase-1 Regulated Human Mesenchymal Stem Cells Differentiation, Luca Vanella, Christopher Sanford Jr., Dong Hyun Kim, Nader G. Abraham, Nabil Ebraheim

Biochemistry and Microbiology

This paper describes the effect of increased expression of HO-1 protein and increased levels of HO activity on differentiation of bone-marrow-derived human MSCs. MSCs are multipotent cells that proliferate and differentiate into many different cell types including adipocytes and osteoblasts. HO, the rate-limiting enzyme in heme catabolism, plays an important role during MSCs differentiation. HO catalyzes the stereospecific degradation of heme to biliverdin, with the concurrent release of iron and carbon monoxide. Upregulation of HO-1 expression and increased HO activity are essential for MSC growth and differentiation to the osteoblast lineage consistent with the role of HO-1 in hematopoietic stem …

The Persistence Of "Dr. Watts" In The Core Culture African American Churches In The Memphis Area, Brent Virgil Buhler

Electronic Theses and Dissertations Archive

In the mid-eighteenth century, Rev. Samuel Davies introduces slaves to an now archaic form of English Hymnody, which became known as "Dr. Watts." Isaac Watts, Charles and John Wesley, John Newton, and Horatius Bonar, among others wrote many of these hymns. Nearly three hundred years later "Dr. Watts" continues atweekly worship services as an essential component in stabilizing, strengthening, and bringing peace and comfort to many core culture African Americans. The traditionis under-studied inmuch of theUnited States, especially in the Memphia Area. The purpose of this thesis is to confirm the musical characteristics, performance practices, and examine some of the motivations that …
