Purification And Characterization Of Blue And Green Chromoprotein Pigments From The Integument Of Male Darters In The Genus Etheostoma, Katelyn Boone

Unlike most other vertebrates, many species in the genus Etheostoma do not utilize structural refraction to display blue or green color. Instead, blue and green mating coloration exhibited by male rainbow darters (E. caeruleum) and male greenside darters (E. blennioides) results from the presence of true chromoprotein pigments. This study was conducted in order to extract, purify, characterize, and compare these novel pigments. Pigments were extracted in aqueous buffer and partially purified by ammonium sulfate fractionation and gel filtration chromatography. Final purification consisted of preparative non-denaturing polyacrylamide gel electrophoresis for E. caeruleum and hydroxyapatite chromatography for E. blennioides. Isolation of …

Brain-based initiatives and school readiness mandates in education have prompted researchers to examine the biological mechanisms associated with learning in the hope that understanding empirical evidence can maximize learning potential. Current research has examined working memory skills in relationship to early learning. The function of working memory is best examined in the context of childhood developmental theory using the framework of Baddeley and Hitch's model. Understanding developmental working memory components in regards to early reading and math skill acquisition is important to designing appropriate interventions. The work of Jacques Lacan (1901-1981) provided clinicians with three main diagnostic ontological structures: neurosis, psychosis, and perversion. All three diagnostic structures indicate fundamentally different ways of solving the problems of alienation, separation from the primary caregiver, and of castration, or having limits set by the law on one's jouissance. Of the three, perversion remains by far the least understood and least discussed. To date, Lacan's diagnostic system--not to mention his corpus in general--has been largely ignored by psychotherapists in the United States. Despite the sparseness of the currently available data, there is accumulating evidence of information processing impairment in post-traumatic stress disorder (PTSD). Studies of event-related potentials (ERPs) are the main tool in real time examination of information processing. In this paper, we sought to critically review the ERP evidence of information processing abnormalities in patients with PTSD. We also examined the evidence supporting the existence of a relationship between ERP abnormalities and symptom profiles or severity in PTSD patients. An extensive Medline search was performed. Rhodococcus equi is an animal pathogen that was initially isolated from horses and is being increasingly reported as a cause of infection in humans with impaired cellular immunity. However, this pathogen is underestimated as a challenging antagonist and is frequently considered to be a mere contaminant despite the potential for life-threatening infections. Most case reports have occurred in immunocompromised patients who have received organ transplants (for example kidney, heart, bone marrow) or those with human immunodeficiency virus infection. Abstract
Background
Aberrant Wnt/β-catenin signaling is associated with breast cancer even though genetic mutations in Wnt signaling components are rare. We have previously demonstrated that Rad6B, an ubiquitin conjugating enzyme, stabilizes β-catenin via polyubiqutin modifications that render β-catenin insensitive to proteasomal degradation. Rad6B is a transcriptional target of β-catenin, creating a positive feedback loop between Rad6B expression and β-catenin activation.

Bladder cancer is one of the most common malignancies and causes hundreds of thousands of deaths worldwide each year. Bladder cancer is strongly associated with exposure to environmental carcinogens. It is believed that DNA damage generated by environmental carcinogens and their metabolites causes development of bladder cancer. Nucleotide excision repair (NER) is the major DNA repair pathway for repairing bulk DNA damage generated by most environmental carcinogens, and XPC is a DNA damage recognition protein required for initiation of the NER process. Background
Representatives of Cetacea have the greatest absolute brain size among animals, and the largest relative brain size aside from humans. Despite this, genes implicated in the evolution of large brain size in primates have yet to be surveyed in cetaceans.
Results
We sequenced ~1240 basepairs of the brain development gene microcephalin (MCPH1) in 38 cetacean species. Alignments of these data and a published complete sequence from Tursiops truncatus with primate MCPH1 were utilized in phylogenetic analyses and to estimate ω (rate of nonsynonymous substitution/rate of synonymous substitution) using site and branch models of molecular evolution. The unfolded protein response (UPR) is a cascade of intracellular stress signaling events in response to an accumulation of unfolded or misfolded proteins in the lumen of the endoplasmic reticulum (ER). Cancer cells are often exposed to hypoxia, nutrient starvation, oxidative stress and other metabolic dysregulation that cause ER stress and activation of the UPR. Depending on the duration and degree of ER stress, the UPR can provide either survival signals by activating adaptive and antiapoptotic pathways, or death signals by inducing cell death programs. Cetacea (dolphins, porpoises, and whales) is a clade of aquatic species that includes the most massive, deepest diving, and largest brained mammals. Understanding the temporal pattern of diversification in the group as well as the evolution of cetacean anatomy and behavior requires a robust and well-resolved phylogenetic hypothesis. Although a large body of molecular data has accumulated over the past 20 years, DNA sequences of cetaceans have not been directly integrated with the rich, cetacean fossil record to reconcile discrepancies among molecular and morphological characters.

Phyloinformatic analyses involve large amounts of data and metadata of complex structure. Collecting, processing, analyzing, visualizing and summarizing these data and metadata should be done in steps that can be automated and reproduced. This requires flexible, modular toolkits that can represent, manipulate and persist phylogenetic data and metadata as objects with programmable interfaces.

Background
Abdominal aortic aneurysm (AAA) is a complex disorder with multiple genetic risk factors. Using affected relative pair linkage analysis, we previously identified an AAA susceptibility locus on chromosome 19q13. This locus has been designated as the AAA1 susceptibility locus in the Online Mendelian Inheritance in Man (OMIM) database.
Methods
Nine candidate genes were selected from the AAA1 locus based on their function, as well as mRNA expression levels in the aorta. A sample of 394 cases and 419 controls was genotyped for 41 SNPs located in or around the selected nine candidate genes using the Illumina GoldenGate platform. …

Background
Large-scale RNAi-based screens are playing a critical role in defining sets of genes that regulate specific cellular processes. Numerous screens have been completed and in some cases more than one screen has examined the same cellular process, enabling a direct comparison of the genes identified in separate screens. Surprisingly, the overlap observed between the results of similar screens is low, suggesting that RNAi screens have relatively high levels of false positives, false negatives, or both.

Intersubtype HIV-1 recombinants in the form of unique or stable circulating recombinants forms (CRFs) are responsible for over 20% of infections in the worldwide epidemic. Mechanisms controlling the generation, selection, and transmission of these intersubtype HIV-1 recombinants still require further investigation. All intersubtype HIV-1 recombinants are generated and evolve from initial dual infections, but are difficult to identify in the human population. Luminescence studies were conducted on tetracyanoplatinate complexes that contain up to four different ligands simultaneously, resulting in very complex structural features. Most of the complexes of (terpy)-Eu-platinum tetra cyanide complexes (terpy = terpyridine) crystallize with low one-, and two-dimensional structural types. The effects of anion variation on the intermolecular energy transfer (IET) mechanism are discussed in detail. Temperature dependent luminescence studies were also conducted on terbium based compounds that contain different ligands resulting in complex structural features. The system also displays dual donor feature where both the organic ligand (terpyridine) and the platinum tetracyanide (TCP) metal complex act as donors. …

Sweet sorghum (Sorghum bicolor (L.) Moenich) is a promising alternative energy crop. Biophysical crop models are advanced agronomic tools designed to predict crop growth for given conditions and to supplement field experiments. No crop model has been developed for sweet sorghum. A method was developed for the simultaneous determination of indole-3-acetic acid (IAA) and kinetin, based on the oxidation of both phytohormones at a pencil-lead graphite electrode. The electrochemical behaviors were examined by cyclic, linear sweep, and square-wave voltammetry in the pH range of 2-12. In Britton-Robinson buffer solution (pH 6), the oxidation peaks of IAA and kinetin are well separated by square-wave voltammetry. By adding an anionic surfactant (sodium dodecylsulfate) in a concentration of 50 \mu M, the sensitivity of kinetin was increased significantly without reducing the resolution. A set of ester and amide derivatives of some acidic NSAIDs, including ibuprofen, ketoprofen, and mefenamic acid (1-3), were synthesized and evaluated for their in vivo analgesic and antiinflammatory activity using the p-benzoquinone-induced writhing test and the carrageenan-induced paw edema model, respectively. Among the synthesized compounds, ester derivatives of ketoprofen showed especially potent analgesic and antiinflammatory activity as compared to the parent drug. In vitro chemical stability studies revealed that ester and amide derivatives were chemically stable in simulated gastric (pH 1.2) and intestinal fluids (pH 6.8). In mammals zinc is the second most abundant essential trace metal. Since Zn2 is a small, hydrophilic, and a highly charged ion, it cannot be transported across the plasma or intracellular organelle membrane by passive diffusion. Different types of cells require a different constant concentration of zinc at all times. Presence of excess free Zn ions can be toxic to the cell. All cells must have tightly regulated homeostatic mechanisms in order to preserve healthy levels and proper compartmentalization of zinc. Cigarette smoking during pregnancy has been associated with negative child behavioral outcomes. Though many studies have found links between maternal smoking during pregnancy and behavior problems in children, few if any have looked into potential moderating factors of that relationship and few have examined the effects of second-hand smoking. This study examined child temperament at 6 months and sex as potential moderators of the relationship between maternal smoke exposure and both internalizing and externalizing behaviors at 24 month, 36 months and first grade. Determining fathers' perceptions of community norms on discipline is paramount to understanding how to develop social work interventions that target fathers' parenting behaviors. Understanding why a father engages in one form of discipline over another in child rearing and understanding how fathers view their parenting role is important when discussing implementation of parenting programs. This study was conducted using qualitative focus groups comprised of men ranging from late adolescents to mid-adulthood. The majority of the men were lower economic status, African American fathers recruited from a human services agency in Detroit. Several sets of neuropathological criteria have been used for the postmortem diagnosis of Alzheimer's Disease (AD), but few studies have examined these criteria in the oldest old. For this study, we examined a sample of centenarians, all of whom received AD assessments using four different neuropathological criteria: Khachaturian, Braak and Braak, CERAD, and NIA-R. Findings indicate that NIA-R criteria differed significantly from CERAD and Khachaturian criteria. In addition, NIA-R and CERAD criteria predicted performance on the MMSE and three FOME indices; Braak and Braak criteria predicted performance on the MMSE and one FOME index. Consumption of trans-fatty acids (tFA) and saturated fatty acids (SFA) have been associated with higher incidence of coronary heart disease. Experimental and observational data suggests that consumption of PHVO containing tFA, like SFA leads to increased total cholesterol and LDL-cholesterol (LDL-C) but compared to tFA, SFA raises HDL-cholesterol (HDL-C). An attempt to eliminate PHVO from the food supply has resulted in the need to find alternatives which despite their higher dietary fat saturation as compared to naturally occurring vegetable oils, may be preferable to PHVO. In this dissertation, we consider solutions of hybrid systems in which both continuous dynamics and discrete events coexists. One
of the main ingredients of our models is the two-time-scale formulation. Under broad conditions, asymptotic expansions are developed for the solutions of the systems of backward equations for switching diffusion in two classes of models, namely, fast switching systems and fast diffusion systems. To prove the validity of the asymptotic expansions, uniform error bounds are obtained.

This dissertation presents innovate methodologies for locating, extracting, and separating multiple incoherent sound sources in three-dimensional (3D) space; and applications of the time reversal (TR) algorithm to pinpoint the hyper active neural activities inside the brain auditory structure that are correlated to the tinnitus pathology. Specifically, an acoustic modeling based method is developed for locating arbitrary and incoherent sound sources in 3D space in real time by using a minimal number of microphones, and the Point Source Separation (PSS) method is developed for extracting target signals from directly measured mixed signals. Previous work by our research group demonstrated that quantitative chemical analysis of analytes, such as mercury and chromium species, in environmental matrices could be successfully carried out without using calibration curves and with correction for species interconversion by using EPA Method 6800A. This method encompasses isotope dilution mass spectrometry (IDMS) and speciated isotope dilution mass spectrometry (SIDMS), both of which are described in detail in chapter 1. Polymeric excipients are generally the least well-characterized components of pharmaceutical formulations. The aim of this dissertation work is to facilitate the quality-by-design (QbD) approach to pharmaceutical formulation and manufacturing by evaluating the inter-grade and inter-batch variability of pharmaceutical-grade polymeric excipients. Sodium alginate, a widely used polymeric excipient, was selected for evaluation using appropriate analytical methods and test conditions, especially rheological methods. The materials used were six different grades of sodium alginate and an additional ten batches of one of the grades.
