Physiological Experimentation With The Crayfish Hindgut: A Student Laboratory Exercise, Ann S. Cooper, Bonnie Leksrisawat, Allison B. Gilberts, A. Joffre Mercier, Robin L. Cooper

Biology Faculty Publications

The purpose of the report is to describe dissection techniques for preparing the crayfish hindgut and to demonstrate how to make physiological recordings with a force transducer to monitor the strength of contraction. In addition, we demonstrate how to visually monitor peristaltic activity, which can be used as a bioassay for various peptides, biogenic amines and neurotransmitters. This preparation is amenable to student laboratories in physiology and for demonstrating pharmacological concepts to students. This preparation has been in use for over 100 years, and it still offers much as a model for investigating the generation and regulation of peristaltic rhythms …

Modeling Biological Membranes With Circuit Boards And Measuring Electrical Signals In Axons: Student Laboratory Exercises, Martha M. Robinson, Jonathan M. Martin, Harold L. Atwood, Robin L. Cooper

Biology Faculty Publications

This is a demonstration of how electrical models can be used to characterize biological membranes. This exercise also introduces biophysical terminology used in electrophysiology. The same equipment is used in the membrane model as on live preparations. Some properties of an isolated nerve cord are investigated: nerve action potentials, recruitment of neurons, and responsiveness of the nerve cord to environmental factors.

Secondary Minerals From Extrapedogenic Per Latus Acidic Weathering Environments At Geomorphic Edges, Eastern Nebraska, Usa, Robert Matthew Joeckel, K.D. Wally, B.J. Ang Clement, P.R. Hanson, J.S. Dillon, S.K. Wilson

Conservation and Survey Division: Faculty and Staff Publications

Acidic weathering of the sulfidic Upper Cretaceous Carlile and Pierre Shales in Nebraska has led to the precipitation of the Al sulfate–hydroxide minerals aluminite, alunite, “basaluminite”/felsöbányaite (e.g.,), the aluminum hydroxides gibbsite and bayerite, and the rare Al phosphate hydroxide vashegyite. Kaolinite has also been produced as a result of this acidic weathering. These minerals do not appear as neoformed constituents in any extant soils in the region, and their existence underscores the ability of pyrite oxidation to produce major changes in mineralogy on a Holocene to Recent time scale. Jarosite, hydronium jarosite, gypsum, halotrichite, and melanterite also appear as secondary …

Comparison Of Two Storage Methods For The Analysis Of Cholinesterase Activities In Food Animals., Ka Askar, Ac Kudi, Aj Moody

School of Biological and Marine Sciences

Cholinesterases (ChE) are specialized carboxylic ester hydrolases that catalyse the hydrolysis of choline esters. They are classified into either acetylcholinesterase (AChE) or butyrylcholinesterase (BChE). Determination of ChE in the tissues is the appropriate tool for the diagnosis of organophosphorus and carbamate exposures. In general, a significant inhibition was seen in both AChE and BChE activities after 6 months of freezing at -80°C and after 3 months of freezing at -20°C. Linear regression of mean AChE and BChE was observed in all individual samples during the months of the two freezing methods. Bland and Altman plot of the ratios of the …

Traversing Swanton Road, 3rd Ed., James A. West

Academic, Government & Associated Organizational Research Resources

Situated at the northwest end of Santa Cruz County and occupying circa 30 square miles of sharply contrasted terrain, the Scott Creek Watershed concentrates within its geomorphological boundaries, at least 10-12% of California's flora, both native and introduced. Paradoxically, the use/abuse that the watershed has sustained over the past 140+ years, has not necessarily diminished the biodiversity and perhaps parallels the naturally disruptive but biologically energizing processes (fire, flooding, landslides and erosion), which have also been historically documented for the area for +60 years. This is an early edition of an extensive document by James A. West. Please visit the …

2010 Vegetation Survey Of The Fort Niobrara National Wildlife Refuge (Final Report To The Nebraska Game And Parks Commission, U.S. Fish And Wildlife Service, And The Sandhills Prairie Refuge Association), Robert F. Steinauer

Nebraska Game and Parks Commission: White Papers, Conference Presentations, and Manuscripts

Fort Niobrara National Wildlife Refuge (FNNWR) is located on the Niobrara River in north central Cherry County, NE. There is little detailed information available documenting the vegetation of FNNWR. Tolstead (1942) studied the grasslands of northern Cherry County, (including FNNWR) with a focus on describing the dominant species of grassland types and determining the relationships between vegetation distribution and environmental variables and management practices. Churchill et al (1988) described the vascular flora of The Nature Conservancy’s nearby Niobrara Valley Preserve. Fiest et al (2010) conducted vascular plant surveys on portions of the FNNWR in 2009 and compiled a partial vascular …

Suppression Subtractive Hybridization Pcr Isolation Of Cdnas From A Caribbean Soft Coral, Jose V. Lopez, Angela Ledger, Lory Z. Santiago-Vázquez, Mihai Pop, Dan D. Sommer, Llanie K. Ranzer, Robert A. Feldman, Russell G. Kerr

Marine & Environmental Sciences Faculty Articles

Transcriptomic studies of marine organisms are still in their infancy. A partial, subtracted expressed sequence tag (EST) library of the Caribbean octocoral Erythropodium caribaeorum and the sea fan Gorgonia ventalina has been analyzed in order to find novel genes or differences in gene expression related to potential secondary metabolite production or symbioses. This approach entails enrichment for potential non-"housekeeping" genes using the suppression subtractive hybridization (SSH) polymerase chain reaction (PCR) method. More than 500 expressed sequence tags (ESTs) were generated after cloning SSH products, which yielded at least 53 orthologous groups of proteins (COGs) and Pfam clusters, including transcription factors …

High Prevalence Of Obesity Among Inner-City Adolescent Boys In The Bronx, New York: Forgetting Our Boys, Carmen R. Isasi, Amy Whiffen, Yolanda Florez, Katherine Freeman, Judith Wylie-Rosett, Eleanor Campbell

Publications and Research

We examined sex differences in overweight and obesity in a sample of 1,619 inner-city adolescents. Participants were enrolled from 11 public schools in the Bronx, New York. The prevalence of overweight and obesity was 21.7% and 22.5%, respectively; prevalence of obesity was significantly higher among adolescent boys than adolescent girls (24.9 vs 20.1%). Childhood obesity is a public health concern in the United States, and the higher prevalence of obesity in adolescent boys requires additional attention.

Zebrafish embryos are emerging as models of glucose metabolism. However, patterns of endogenous glucose levels, and the role of the islet in glucoregulation, are unknown. We measured absolute glucose levels in zebrafish and mouse embryos, and demonstrate similar, dynamic glucose fluctuations in both species. Further, we show that chemical and genetic perturbations elicit mammalian-like glycemic responses in zebrafish embryos. We show that glucose is undetectable in early zebrafish and mouse embryos, but increases in parallel with pancreatic islet formation in both species. In zebrafish, increasing glucose is associated with activation of gluconeogenic phosphoenolpyruvate carboxykinase1 (pck1) transcription. Non-hepatic Pck1 protein is …

Controlling Weeds In Horse And Cattle Pastures, Byron Sleugh, Robert Masters Jan 2011

Controlling Weeds In Horse And Cattle Pastures, Byron Sleugh, Robert Masters

Forage Symposium at the Kentucky Cattlemen’s Convention

Weed control is one of the most talked about concerns of forage-animal systems. Producers will sometimes pay more attention to some pests than others and more so in some crops than others. For example, a producer may have a very low threshold for weed infestation in their corn, soybean or tobacco field but will have a very high tolerance for weeds in their pastures. This is usually because the “direct” dollar value of the forage is not seen while we can “see” how a reduction in soybean or corn yield will affect the bottom line of our operation. Vegetation manipulation …

Forages: Back To Basics, Garry D. Lacefield

Forage Symposium at the Kentucky Cattlemen’s Convention

Over the past fifteen years, we have covered many forage-related topics ranging from the species we grow through marketing our final products. Through all of these themes, we have attempted to address the “basics” in a practical way. We will continue that theme today as we deal with some very important-practical topics.

A Generalized Approach For Testing The Association Of A Set Of Predictors With An Outcome: A Gene Based Test, Benjamin A. Goldstein, Alan E. Hubbard, Lisa F. Barcellos

U.C. Berkeley Division of Biostatistics Working Paper Series

In many analyses, one has data on one level but desires to draw inference on another level. For example, in genetic association studies, one observes units of DNA referred to as SNPs, but wants to determine whether genes that are comprised of SNPs are associated with disease. While there are some available approaches for addressing this issue, they usually involve making parametric assumptions and are not easily generalizable. A statistical test is proposed for testing the association of a set of variables with an outcome of interest. No assumptions are made about the functional form relating the variables to the …

The synthesis of both proinflammatory leukotrienes and anti-inflammatory lipoxins requires the enzyme 5-lipoxygenase (5-LOX). 5-LOX activity is short-lived, apparently in part because of an intrinsic instability of the enzyme. We identified a 5-LOX-specific destabilizing sequence that is involved in orienting the carboxyl terminus, which binds the catalytic iron. Here, we report the crystal structure at 2.4 angstrom resolution of human 5-LOX stabilized by replacement of this sequence.
