Arenaviruses are responsible for acute hemorrhagic fevers worldwide and are recognized to pose significant threats to public health and biodefense. Small molecule compounds have recently been discovered that inhibit arenavirus entry and protect against lethal infection in animal models. These chemically distinct inhibitors act on the tripartite envelope glycoprotein (GPC) through its unusual stable signal peptide subunit to stabilize the complex against pH-induced activation of membrane fusion in the endosome. Here, we report the production and characterization of the intact transmembrane GPC complex of Junín arenavirus and its interaction with these inhibitors. The solubilized GPC is antigenically indistinguishable from the …

Surface-Enhanced Raman Imaging Of Intracellular Bioreduction Of Chromate In Shewanella Oneidensis, Sandeep P. Ravindranath, Kristene Henne, Dorothea Thompson, Joseph Irudayaraj Feb 2011

Surface-Enhanced Raman Imaging Of Intracellular Bioreduction Of Chromate In Shewanella Oneidensis, Sandeep P. Ravindranath, Kristene Henne, Dorothea Thompson, Joseph Irudayaraj

Birck and NCN Publications

This proposed research aims to use novel nanoparticle sensors and spectroscopic tools constituting surface-enhanced Raman spectroscopy (SERS) and Fluorescence Lifetime imaging (FLIM) to study intracellular chemical activities within single bioremediating microorganism. The grand challenge is to develop a mechanistic understanding of chromate reduction and localization by the remediating bacterium Shewanella oneidensis MR-1 by chemical and lifetime imaging. MR-1 has attracted wide interest from the research community because of its potential in reducing multiple chemical and metallic electron acceptors. While several biomolecular approaches to decode microbial reduction mechanisms exist, there is a considerable gap in the availability of sensor platforms to …

Students Studying Students: An Assessment Of Using Undergraduate Student Researchers In An Ethnographic Study Of Library Use, Allyson Washburn, Sheila C. Bibb Feb 2011

Students Studying Students: An Assessment Of Using Undergraduate Student Researchers In An Ethnographic Study Of Library Use, Allyson Washburn, Sheila C. Bibb

Faculty Publications

This paper reports on the use of undergraduate students enrolled in an Applied Anthropology course as researcher for a library use study at Brigham Young University's (BYU) Harold B. Lee Library (HBLL). This is a common practice at BYU, but has not been reported extensively in the literature. The study was carried out by the authors with the assistance of undergraduate students, the students being the researchers and was conducted in order that the HBLL could determine student ideas for reconfiguring some newly opened space in the Periodicals room. Using students assisted the library as well as met a curricular …

Evidence For Mesenchymal-Epithelial Transition Associated With Mouse Hepatic Stem Cell Differentiation, Bin Li, Yun Wen Zheng, Yuuki Sano, Hideki Taniguchi Feb 2011

Evidence For Mesenchymal-Epithelial Transition Associated With Mouse Hepatic Stem Cell Differentiation, Bin Li, Yun Wen Zheng, Yuuki Sano, Hideki Taniguchi

Faculty Publications

Mesenchymal-epithelial transition events are related to embryonic development, tissue construction, and wound healing. Stem cells are involved in all of these processes, at least in part. However, the direct evidence of mesenchymal-epithelial transition associated with stem cells is unclear. To determine whether mesenchymal-epithelial transition occurs in liver development and/or the differentiation process of hepatic stem cells in vitro, we analyzed a variety of murine liver tissues from embryonic day 11.5 to adults and the colonies derived from hepatic stem/progenitor cells isolated with flow cytometry. The results of gene expression, immunohistochemistry and Western blot showed that as liver develops, the expression …

Hay Drying, Preservatives, Conditioning, Ash Content, Dan Undersander Feb 2011

Hay Drying, Preservatives, Conditioning, Ash Content, Dan Undersander

Kentucky Alfalfa and Stored Forage Conference

Drying forage for hay has always been a challenge. While we cannot control the weather we can manage cut forage to maximize drying. The purpose of this paper is to give a few principles of hay and silage making and discuss machinery available relative to these principles. Then we will also talk about minimizing ash in hay to optimize the total digestible nutrients of the forage.

Coupled Folding And Specific Binding: Fishing For Amphiphilicity, Vikas P. Jain, Raymond S. Tu Feb 2011

Coupled Folding And Specific Binding: Fishing For Amphiphilicity, Vikas P. Jain, Raymond S. Tu

Publications and Research

Proteins are uniquely capable of identifying targets with unparalleled selectivity, but, in addition to the precision of the binding phenomenon, nature has the ability to find its targets exceptionally quickly. Transcription factors for instance can bind to a specific sequence of nucleic acids from a soup of similar, but not identical DNA strands, on a timescale of seconds. This is only possible with the enhanced kinetics provided for by a natively disordered structure, where protein folding and binding are cooperative processes. The secondary structures of many proteins are disordered under physiological conditions. Subsequently, the disordered structures fold into ordered structures …

Lea Proteins During Water Stress: Not Just For Plants Anymore, Steven C. Hand, Michael A. Menze, Mehmet Toner, Leaf Boswell, Daniel Moore Feb 2011

Lea Proteins During Water Stress: Not Just For Plants Anymore, Steven C. Hand, Michael A. Menze, Mehmet Toner, Leaf Boswell, Daniel Moore

Faculty Publications

Late embryogenesis abundant (LEA) proteins are extremely hydrophilic proteins that were first identified in land plants. Intracellular accumulation is tightly correlated with acquisition of desiccation tolerance, and data support their capacity to stabilize other proteins and membranes during drying, especially in the presence of sugars like trehalose. Exciting reports now show that LEA proteins are not restricted to plants; multiple forms are expressed in desiccation-tolerant animals from at least four phyla. We evaluate here the expression, subcellular localization, biochemical properties, and potential functions of LEA proteins in animal species during water stress. LEA proteins are intrinsically unstructured in aqueous solution, …
