Bevacizumab Effect On Topotecan Pharmacokinetics In A Murine Orthotopic Rhabdomyosarcoma Xenograft Model, Zaifang Huang

Increasing evidence suggests that inhibition of vascular endothelial growth factor (VEGF) can transiently normalize tumor vasculature, thereby improving delivery of systemic chemotherapy. Bevacizumab (BEV), an anti-VEGF antibody, has been shown to transiently normalize tumor vasculature by increasing tumor vessel maturity, decreasing tumor vessel permeability, and increasing tumor oxygenation in an Rh30 orthotopic rhabdomyosarcoma xenograft model. However, the effects of BEV on the pharmacokinetics of TPT and the antitumor activity of TPT have not been evaluated. Polymorphic Variation Of The Dgat1 Gene In Cal Poly Holstein And Jersey Breeds And The Correlation Of Milk Fat Content, And Genetic Values, Andrea M. Laubscher

The selection of dairy cattle is dictated by improvement of yield and composition of milk. Genotyping tests give breeders the opportunity to use previously unavailable information to increase the frequency of beneficial alleles in their herds. A QTL influencing milk yield and composition is located in the centromeric end of chromosome 14. The polymorphism being analyzed is a K232A substitution (lysine to alanine amino acid change) in exon VII of the diacylglycerol acyl transferase 1 (DGAT1) gene. DGAT1 is a key acyltranferase in triglyceride biosynthesis. DGAT1 encodes an enzyme which catalyzes the final step of mammalian triglyceride synthesis from carbohydrates. …

Interest in consuming unprocessed foods has led to assumptions that raw milk has health benefits over pasteurized milk. This study was designed to evaluate the belief that raw milk is nutritionally superior. Eighteen mice were randomly assigned to one of two groups: raw milk or pasteurized milk. Mice were grouped into breeding trios and given a fresh supply of milk every four hours between 7 a.m. and 11 p.m. for 55 days. Milk consumption was tracked at each feeding by measuring the amount of milk that was provided and the amount of milk that was left from the previous feeding. …

Proteorhodopsin (pR) is an intrinsic membrane protein with an important role in solar-energy storage of the biosphere. Earlier work in our lab has shown that polyhistidine-tagged pR can be purified by means of selective precipitation with citrate under specific conditions, as can a number of mutants based on this His6-tagged pR. Purification of a heterologously-expressed trans-membrane protein by a simple salt such as citrate is novel. However, such a phenomenon leads to several questions: How does citrate cause pR precipitation? Does the polyhistidine-tag assist in such a precipitation? Is this precipitation pH-specific? Acute strength exercise elicits a transient oxidative stress, but the factors underlying the magnitude of this response remain unknown. The purpose of this investigation was to determine whether muscle-fiber type relates to the magnitude of blood oxidative stress after eccentric muscle activity. Eleven college-age men performed 3 sets of 50 eccentric knee-extensions. Blood samples taken pre-, post-, and 24, 48, 72, and 96 hr postexercise were assayed for comparison of muscle damage and oxidative-stress biomarkers including protein carbonyls (PCs). The Freedom of Information Act ("FOIA" or "the Act") serves an important purpose in informing the citizenry of its government's actions. Congress created it as a disclosure statute, the Supreme Court has decided FOJA in favor of disclosure, and the D. C. Circuit has kept that goal of disclosure by refusing to broaden its exemptions. Despite this history of consistent treatment, the Third Circuit attempted to change the game with its holding in AT&T v. FCC. The Third Circuit held that corporations may have personal privacy under FOIA Exemption 7(C). This paper will argue that this holding contradicted three FOIA …

Objective: Assess the impact of the rural food environment on the eating behaviors and BMI of rural low-income children. Method: Statewide (Maine, 2009) household survey of parents of children on Medicaid (n=1722), oversampled in six rural communities, resulting in n=272 for six target communities. Food environment measured using modified Nutrition Environment Measures Survey in Stores (NEMS-S) for 46 retail food outlets. Multi-variate analysis assessed factors affecting home food environment, child's eating behavior and BMI. Results: Home food behaviors (how often: family eats together, child eats breakfast, vegetables served) and parent food consumption were significantly associated with children's healthy eating behaviors. …

Nutritional and energetic needs of female mammals depend upon size or reproductive investment and shape individual activity budgets and behavioral patterns. To maximize nutritional and energetic intake females may increase time allocated to foraging or access resources through risk-prone behaviors, represented as aggression or leadership. Conversely, to minimize nutritional and energetic spending females may engage in risk-averse activities such as resting or nonaggressive social interactions. Females with the highest needs should exhibit activities and behaviors that facilitate the greatest nutritional and energetic returns or highest metabolic savings. This article reviews the development of universal women’s human rights since 1970. It begins by discussing how the international feminist movement influenced the development of women’s legal human rights, and continues by reviewing three debates in the literature on women’s rights. The first debate is whether human rights as originally formulated were actually men’s rights; the second debate is about the relationship between culture and women’s rights; and the third considers the effects of globalization on women’s rights. Refugees face different circumstances than other immigrants regarding housing in initial resettlement in the U.S. Refugees have no choice of their initial residence as this is determined in advance by the resettlement agency. Refugees who belong to minority ethnic groups and who have little education or skills may experience discrimination and hostility from local citizens. Resettlement areas that are high in population density, rental units, minorities, crime, unemployment, inadequate transportation, and low income may present additional barriers to cultural and economic assimilation.

The first objective of this study was to identify and explain the genetic gain achieved in commercial beef cattle operations with the use of genomic predictions versus the use of visual appraisal and EPDs. I argue that the American Congress of Obstetricians and Gynecologists (ACOG), as an organization and through its individual members, can and should be a far greater ally in the prevention of violence against women. Specifically, I argue that we need to pay attention to obstetrical practices that inadvertently contribute to the problem of violence against women. While intimate partner violence is a complex phenomenon, I focus on the coercive control of women and adherence to oppressive gender norms. Aluminum (Al) toxicity is the major stress in acidic soil that comprises about 50% of the world's arable land. The complex molecular mechanisms of Al toxicity have yet to be fully determined. As a barrier to Al entrance, plant cell membranes play essential roles in plant interaction with Al, and lipid composition and membrane integrity change significantly under Al stress. Here, we show that phospholipase Dγs (PLDγs) are induced by Al stress and contribute to Al-induced membrane lipid alterations. RNAi suppression of PLDγ resulted in a decrease in both PLDγ1 and PLDγ2 expression and an increase in Al resistance. Genetic …

The translation of mRNAs without in-frame stop codons results in the following problems: the ribosomes are stalled and sequestered on the defective mRNA, the incomplete polypeptides produced are potentially toxic to the cells, and the defective mRNA can result in more futile translational cycles. Bacteria have developed a unique system, called trans-translation to resolve all of the above-mentioned problems. The key players of the trans-translation system are tmRNA and its protein cofactor SmpB. One part of my work showed that the system mediates the degradation of non-stop mRNAs by recruiting RNase R to stalled ribosomes through the interaction between the …

Background. Dietary supplements are widely used among United States senior citizens for various indications. Potential dangers with supplement use include the lack of regulation by the Food and Drug Administration (FDA) and the possibility for drug-supplement interactions. Epigenetic mechanisms may play an important role in the developmental programming of adult-onset chronic metabolic diseases resulting from suboptimal fetal nutrition, but the exact molecular mechanisms are incompletely understood. Given the central role of the liver in metabolic regulation, we investigated whether chronic maternal dietary protein restriction has long-term effects on liver gene expression in the offspring. Plants are frequently under attack from pests and deploy various defense mechanisms to fight off predators. Many plant defenses are induced following herbivory or pathogen infection. An investment in defense could potentially lead to a diversion of metabolic resources away from primary metabolism, including reduction in plant commitment to photosynthesis. Jump ability is important in volleyball; therefore analysis of factors that influence jump performance is necessary to ensure maximal jump ability. Purpose: To analyze how strength characteristics associated with jumping are affected by percent body fat, lean body mass and free fat mass. Methods: Data from eleven female NCAA DI volleyball players as part of an ongoing athlete monitoring program collected from 2007-2010 was analyzed. Developmental biology is expanding into several new areas. One new area of study concerns the production of adult-onset phenotypes by exposure of the fetus or neonate to environmental agents. These agents include maternal nutrients, developmental modulators (endocrine disruptors), and maternal care. In all three cases, a major mechanism for the generation of the altered phenotype is chromatin modification. Nutrient conditions, developmental modulators, and even maternal care appear to alter DNA methylation and other associated changes in chromatin that regulate gene expression. This brings a new, under-appreciated, dimension of gene regulation into developmental biology, and it also demonstrates the poverty of …
