Characterization Of Lysophosphatidic Acid Subspecies Using A Novel Hplc Esi-Ms/Ms Method, Eric Mayton
Characterization Of Lysophosphatidic Acid Subspecies Using A Novel Hplc Esi-Ms/Ms Method, Eric Mayton
Theses and Dissertations
Lysophosphatidic acid (LPA) is a bioactive lipid with a plethora of biological functions, including roles in cell survival, proliferation, and migration. Although high-performance liquid chromatography electrospray ionization tandem mass spectrometry (HPLC ESI-MS/MS) technology has been used to measure the levels of LPA in human blood, serum and plasma, current methods cannot readily detect the minute levels of LPA from cell culture. In this study, a novel HPLC ESI-MS/MS method with enhanced sensitivity was developed which allows accurate measurements of LPA levels with a limit of quantitation at approximately 10 femtomoles. The method was validated by quantitation of LPA levels in …

Examining The Relationships Among Injury Outcomes, Velocity And Grip Strength During Dynamic Platform Perturbation, Braden Cripe
Examining The Relationships Among Injury Outcomes, Velocity And Grip Strength During Dynamic Platform Perturbation, Braden Cripe
Master's Theses (2009 -)
Studies examining the relationship between inputs and outputs for simulated models of dynamically perturbed horizontal platforms are scarce. Most of these scenarios include a standing operator with upper extremity grip, oftentimes subjected to lateral impulses, which may lead to occupant injury. While the detailed study of these collision scenarios is sparse, the prevalence of their application is great.
This thesis aims to identify how two input parameters, velocity change (ÄV) and grip strength, affect injury assessment reference values (IARVs). This is accomplished by using Mathematical Dynamic Modeling (MADYMO) software to simulate the scenarios defined by those inputs. In the simulation, …

A Comparative Study On Anaerobic Capacity, Fatigue Index, And Recovery Speed Between Racket Sport Players (Tennis, Badminton, Table Tennis), Iman Najmuddin Abbas
Modern Sport
The tournament games, including badminton, tennis, and table tennis, rely on anaerobic metabolism for energy production during performance. This energy production, primarily fueled by phosphate compounds like ATP-PC, supports rapid, intense muscle work. Anaerobic capacity is linked to the quantity of these compounds and is influenced by training, with performance levels peaking during periods of maximum effort (5-10 seconds) and sustaining for up to 15 seconds in untrained individuals. However, trained athletes can maintain this performance level for up to 25-30 seconds. This study aims to identify significant differences in anaerobic power and fatigue index among tennis, badminton, and table …
Understanding Of Functions Of Selenoproteins And Dietary Selenium By Using Animal Models, Marina V. Kasaikina
Understanding Of Functions Of Selenoproteins And Dietary Selenium By Using Animal Models, Marina V. Kasaikina
Department of Biochemistry: Dissertations, Theses, and Student Research
Selenium (Se) is a trace element that is incorporated into proteins in the form of the 21st amino acid, selenocysteine (Sec). Se supplementation was reported to have beneficial roles in prevention of cardiovascular and muscle disorders, cancer prevention and enhancement of the immune function. However, recent studies also showed that excessive dietary Se increases the risk of development of type 2 diabetes mellitus. Thus, better understanding of Se and selenoprotein functions is required. We used three approaches to address this problem.
First, we used high-throughput sequencing to examine composition of the gut microflora in mice maintained on selenium-deficient, selenium-sufficient, …
Ischemia Impairs Vasodilation In Skeletal Muscle Resistance Artery, Kyle Remington Struthers
Ischemia Impairs Vasodilation In Skeletal Muscle Resistance Artery, Kyle Remington Struthers
Master's Theses
Functional vasodilation in arterioles is impaired with chronic ischemia. We sought to examine the impact of chronic ischemia and age on skeletal muscle resistance artery function. To examine the impact of chronic ischemia, the femoral artery was resected from young (2-3mo) and adult (6-7mo) mice and the profunda femoris artery diameter was measured at rest and following gracilis muscle contraction 14 days later using intravital microscopy. Functional vasodilation was significantly impaired in ischemic mice (14.4±4.6% vs. 137.8±14.3%, p<0.0001 n=8) and non-ischemic adult mice (103.0±9.4% vs. 137.8±14.3%, p=0.05 n=10). In order to analyze the cellular mechanisms of the impairment, a protocol was developed to apply pharmacological agents to the experimental preparation while maintaining tissue homeostasis. Endothelial and smooth muscle dependent vasodilation were impaired with ischemia, 39.6 ± 13.6% vs. 80.5 ± 11.4% and 43.0 ± 11.7% vs. 85.1 ± 10.5%, respectively. From this data, it can be supported that smooth muscle dysfunction is the reason for the observed impairment in arterial vasodilation.

Characterization And Implementation Of A Decellularized Porcine Vessel As A Biologic Scaffold For A Blood Vessel Mimic, Aubrey N. Smith
Master's Theses
Every 34 seconds, someone in the United States suffers from a heart attack. Most heart attacks are caused by atherosclerotic build up in the coronary arteries, occluding normal blood flow. Balloon angioplasty procedures in combination with a metal stent often result in successful restoration of normal blood flow. However, bare metal stents often lead to restenosis and other complications. To compensate for this problem, industry has created drug-eluting stents to promote healing of the artery wall post stenting. These stents are continually advancing toward better drug-eluting designs and methods, resulting in a need for fast and reliable pre-clinical testing modalities. …

Low back pain is one of the most costly ailments in the world today. A key biological structure studied in the research of low back pain is the facet joint. The facet joint is thought to both play a role in the causation of low back pain through its response to noxious loading and in its relief through beneficial loading caused by spine manipulation. The purpose of these studies is to use a computational model to predict the biomechanical response of the facet joints to various loading parameters. This was accomplished using a linear 3D Finite Element model of the …
Cross-Talk Of Retinoic Acid And Adrenergic Hormone Signaling May Influence Development Of Cardiac Conduction And Rhythmicity In Utero, Sabikha Alam
HIM 1990-2015
Stress hormones, adrenaline and noradrenaline, have been shown to be critical for heart development. Mice lacking dopamine greek lower case letter beta]-hydroxylase (Dbh), an enzyme responsible for synthesis of these adrenergic hormones, die during mid-gestation due to cardiac failure. Prior research showed that adrenergic cells are found within the electrical conduction system of the heart, and adrenergic deficiency leads to slowed cardiac conduction during embryogenesis. Microarray analysis of wild-type (Dbh+/+) and knockout (Dbh-/-) mouse hearts revealed significant differences in expression of retinoic acid (RA) signaling genes. RA signaling has also been shown to be critical for heart development. These data …

Biophysical Studies Of Gene Sequence G-Quadruplexes And I-Motifs, Jamie Marie Dettler
Theses and Dissertations
The treatment and/or prevention of cancer by selective down regulation of cancer causing gene (oncogene) transcription would represent a significant advance in the area of anticancer drug design. Non-canonical higher order DNA structures formed in oncogene promoter regions are novel targets for the modulation of oncogene expression. An obvious advantage of selectively targeting oncogene expression would be that general cytotoxicity would be minimized and the negative side effects of current chemotherapy approaches could be minimized or eliminated. To provide a foundation for the design of drugs that target oncogene promoter G-quadruplexes and i-Motifs, the basic understanding is required of the …

Comparative Analysis Of The Magnitude Of Oxidative Stress Following Acute High Fat Feeding And Acute Strenuous Exercise, Cameron Grant Thomas Mccarthy
Electronic Theses and Dissertations Archive
Two prevalent origins of oxidative stress in Western society are the ingestion of certain nutrients and exercise. The purpose of this investigation was to compare the magnitude of increase in oxidative stress following acute feeding and acute exercise. Twelve exercise-trained men consumed a high-fat meal or performed one of three exercise bouts, in a random order, cross-over design. Blood samples were assayed for malondialdehyde (MDA), hydrogen peroxide (H2O2), trolox equivalent antioxidant capacity (TEAC), and triglycerides (TAG). A significant condition effect was noted for MDA (p=0.01) and H2O2 (p<0.0001), with values highest for the meal condition. A trend was also noted for TAG (p=0.07), with values highest for the meal condition. These results illustrate that the magnitude of oxidative stress following a mealis significantly greater than the magnitude of oxidative stress elecitied after an acutebout of strenuous exercise, in a sample of healthy, exercise-trained men.
