The hypothesis for this research was: Herpes simplex virus type 1 (HSV-1) infection of murine keratinocyte cell lines (HEL-30 and PAM-212) treated with mitotic inhibitors leads to silencing of virus replication. At 0.1 (Multiplicity of infection) MOI, PAM-212 keratinocytes showed some cell lysis and viral plaques were seen in HEL-30 keratinocyte cultures. PAM-212 keratinocytes, infected at 0.01 MOI, were more susceptible to the lytic effect of HSV-1 than were HEL-30 cells. In this study, 5-fluoro-2'-deoxyuridine (FUDR) treatment of both keratinocyte cell lines permitted an increase survival of HSV-1- infected keratinocytes especially for PAM-212 cell line which was further examined for …

A procedure has been developed for calculating permeability (k) from the Kozeny-Carmen equation, a procedure that links ideas from percolation theory with the ideas of Koltermann and Gorelick (1995) and Esselburn et al. (2011). The approach focuses on the proportion of coarser pores that are occupied by finer sediments and defines a threshold proportion. For proportions below this threshold, the unoccupied coarser pores percolate. Following the ideas of Koltermann and Gorelick (1995), the effective grain-size term in the Kozeny-Carman equation is calculated using the geometric mean if below the threshold proportion, and with the harmonic mean if above. Early attachment disruption is thought to promote later onset of depressive illness through a process involving sensitization. Maternal separation in guinea pig pups (~21 days of age) produces depressive-like behavior and core body temperature fluctuations that appear to be mediated by proinflammatory activity. These responses are enhanced during repeated separations over several days. Here, enhanced depressive-like behavior and core body temperature responses were observed from the early pre-weaning to the periadolescent period (~10-40 days of age) and persisted for more than a week. The greatest temperature response was observed during the final separation. These results demonstrate persisting sensitization of behavioral …

In the current study I examined the relationships between personality, culture, and the decision to pursue self-interests. Specifically, I proposed that conscientiousness, extraversion, idiocentrism, masculinity, and power distance would be positively related to the decision to pursue self-interests whereas agreeableness would be negatively related to the pursuit of self-interests. I also proposed an exploratory examination of interactions between personality and individual cultural values in predicting the decision to pursue self- interests. As predicted, idiocentrism and masculinity significantly predicted the decision to pursue self-interests. In addition, masculinity moderated the effects of both agreeableness and neuroticism on the pursuit of self-interests. Also, …

Darters are small benthic-feeding fish. I examined reasons why some darter species are rarer than others based on geographic range, habitat specificity, and local population size using the Ohio EPA database and field research. The Qualitative Habitat Evaluation Index (QHEI) represented habitat quality. I examined drainage area and gradient at the landscape-scale, riparian and channel characteristics at the reach-scale, and substrate, cover, and riffle/pool characteristics at the microhabitat scale. Some rare species occur in few rivers in Ohio, but throughout a basin, while other rare species occur only in moderate-sized rivers. Most rare species occur at only a few sites …

Causal attributions can play an integral part in how employees respond to events in the work environment (Bowling and Beehr, 2006; Dasborough and Ashkanasy, 2002; Perrewé and Zellars, 1999). Causal attributions of a work behavior or event include locus of causality (i.e., self-directed, supervisor-directed, organization-directed), stability, and intentionality (i.e., altruistic, self-serving). In the current study, I examined the consequences of subordinates' causal attributions on responses to emotional and instrumental supervisor support. As expected, emotional and instrumental supervisor support were positively associated with job satisfaction, supervisor satisfaction, organizational citizenship behaviors, and organizational commitment assessed 30 days later. Cortical bone is a porous structure. The presence of these pores creates the possibility of a local overstressed area that has the likelihood of premature failure. Some failure modes of the vertebral endplates, for example subsidence which occurs at rates as high as 77 percent, can be better predicted with further understanding of failure mechanisms and the ability to predict those mechanisms. A probabilistic assessment of the pore size and its contribution to the fracture toughness has not been investigated in the cortical shell of the vertebral endplates. This study investigated the relationship between individuals' reasons for leaving their former employers (e.g., inadequate pay) and relevant attitudinal variables (e.g., attitudes regarding pay and other compensation). The role of attitudes as a mediator of the relationship between reasons and intentions was also investigated. Finally, the relationship between reasons, attitudes, and reemployment in the same industry was investigated. Results from an archival sample of former employees (n = 5044) from 8 small to medium-sized companies in a variety of industries indicated mixed support for the hypotheses. Implications and directions for future research were discussed.

These organisms play many roles in human health and well-being. However, shifts in the composition of the intestinal microbiota have been associated with diseases such as irritable bowel syndrome, inflammatory bowel disease, and colon cancer. Osmotic brain edema or chronic treatment with desipramine alters brain water permeability. In this study we investigated aquaporin 4 expression and distribution in these two conditions. Brain edema development was induced by intraperitoneal water injection. Blood serum osmolality decreased from 296 ± 1 mOsm to 278 ± 2 mOsm within 15 min. Cerebral cortex water content increased from 79.8 ± 0.2 % to 81.3 ± 0.5% during 120 min of this hyposmotic exposure. Aquaporin 4 immunostaining intensity at the astrocytic endfeet increased in water injected animals from 2.6 ± 0.04 intensity unites (IU) to 3.2 ± 0.21 IU, while total …

Protein kinase inhibition by staurosporine causes apoptotic volume decrease involving potassium (K) channels in immortalized human B3 lens epithelial cells (LECs). Here, the effect of two pro-apoptotic protein kinase C (PKC) inhibitors [12-O-tetradecanoyl-phorbol-13-acetate (TPA) and chelerythrine (CET)] were studied on membrane K transport in a fetal human LEC line (FHL124) by western blotting, immunofluorescence, ion flux, ATP, apoptosis, and biotinylation assays. Long term TPA exposure (0-6 h) inhibited 75% of Na-K-2Cl cotransport (NKCC). In contrast, short term (0-20 min) exposure to 50 μM CET reduced Na/K pump and NKCC by >90% and >70%, respectively, without retrieval from the membrane into …

Human endometrium is a high dynamic tissue that contains stem/progenitor cells. These endometrial stromal progenitor cells (hESCs) have been differentiated into a number of mesodermal lineages. There is limited information on differentiating hESCs into neurons, and no information on differentiating hESCs into megakaryocytes (MKs) and platelets (PLTs). The aim of this work was to investigate the possibility of differentiating hESCs into two distinct lineages: MKs, with subsequent PLT formation, and neurons.

Currently, there is no research that assesses how individual's perceptions of personality and other characteristics in gay male and lesbian applicants affect employment decisions. I examined individuals' perceptions of personality in gay and lesbian applicants and the effect of these perceptions on employment outcomes. I hypothesized that individuals would prescribe gender atypical traits to gay male and lesbian applicants, and that qualification ratings for these applicants would depend on job-type match with the applicants' gender atypical traits. Emissions from coal-fired utilities are the major anthropogenic source of mercury (Hg) to the atmosphere. Because emitted Hg may be deposited near the source, there are concerns on levels of toxic monomethylmercury (MeHg) in local biota. We investigated the potential impact of a large Hg-emitting (> 500 kg Hg y-1) coal-fired power station on MeHg levels in mosquitoes near the Conesville power station in central Ohio. Renin angiotensin system (RAS) plays a pivotal role in the development and progression of diabetic nephropathy. Previous studies on frame-of-reference effects have focused on domain-specific personality measures as predictors of performance in North America. The current study expands on this research by comparing work-specific and general personality as predictors of CWBs and OCBs in an East Asian culture (i.e., China). Consistent with the literature on frame-of-reference effects in personality assessment, I found that three Big Five personality dimensions, including agreeableness, conscientiousness, and emotional stability, were significantly related with OCBs and CWBs. Lithium-ion battery anodes with a nanostructure of randomly dispersed carbon nanofibers (CNFs), carbon nanotubes (CNTs), and nanoparticles of tin-oxide or silicon were fabricated and tested in order to develop high capacity, easily manufactured anodes. In these anodes, a mesh of CNTs and CNFs form a conductive network within which the nanoparticles of tin-oxide are suspended. The CNT network directs electron flow to and from the nanoparticles while accommodating their volume changes. We describe a methodology that combines urinary metabolomics with a tissue-specific stressor administration to enhance assessment of tissue function. Kidney function in rats was mildly compromised with a sub-acute dose of D-serine and stressed with furosemide. NMR-based metabolomics analyses showed no detectable effects due to D-serine alone; but furosemide or D-serine + furosemide groups, classified separately from each other, and from control. Furosemide alone caused a ca. 2-fold increase in glucose, lactate, choline, and a 30% decrease in TCA intermediates (p≤0.05). D-serine suppressed these effects and produced a 1.7-fold increase in a p-phenolic acid-derivative of tyrosine (PAdY) relative to control …

The hypothesis for this study was: IFN-β expression differs between murine fibroblasts (A2R1 and L929) of different mouse strains upon infection of HSV-1. Fluorescent microscopy was used to examine localization of IFN-β in the different murine fibroblast and keratinocyte cell lines after 6 hours of infection with herpes simplex virus type-1 (HSV-1). Because keratinocytes cell lines HEL-30 and PAM-212 grew in clusters, staining patterns in individual cells could not be determined. AMP-activated protein kinase (AMPK) regulates cellular metabolism and promotes ATP production when energy is depleted. Evidence suggests that AMPK may be involved in oxygen sensing by carotid body cells, which are responsible for regulating the breathing rate to maintain proper blood oxygen levels. There are two isoforms of the catalytic alpha subunit, AMPKalpha1 and AMPKalpha2, which could be involved in oxygen sensing. Here, the production of a short hairpin RNA (shRNA) targeting both catalytic isoforms of AMPK in human, mouse, and rat is described. The Role Of Deliberate Behavior In Expert Performance: The Acquisition Of Information Gathering Strategy In The Context Of Emergency Medicine, Frank Eric Robinson

Effective performance in dynamic domains requires experts to assess problems and implement solutions within the constraints of their work system, but these processes are not well accounted for in the expertise literature. I observed resident and attending emergency physicians to determine the behaviors that distinguish doctors as they manage patients and what contextual variables may affect these behaviors. Factor analyses revealed three types of behavior: goal establishment behavior, goal enactment behavior, and acknowledging uncertainty. Multilevel analyses indicated that doctors' experience and both the local and global context of care impact these behaviors. More experienced doctors appear to be more sensitive …

Organophosphorus nerve agents are amongst the most deadly chemical compounds ever synthesized. Sarin is an organophosphate (OP) ester that irreversibly forms a phosphoester bond at the active site of acetylcholinesterase and thereby induces a rapid and lethal cholinergic crisis. It remains an active threat to vulnerable civilian populations due to its ease of synthesis and known use by rogue nations and terrorist groups. Death is the most extreme consequence of sarin toxicity. Current treatments fail to provide protection against progressive cognitive impairments years after mild exposure. Nowadays, 24% of the nanomaterial-based consumer products contain silver nanoparticles (AgNPs) and exploit the well-known antimicrobial properties of silver. Although AgNPs have a wide range of biomedical research and industrial applications, very little is known about their toxicity. The main goal of this study is to synthesize, characterize and manipulate Creighton colloidal AgNPs for future cytotoxicity studies. These "naked" AgNPs were free from chemically aggressive capping/stabilizing agents, reaction byproducts or organic solvents. How long-tail liability claims such as asbestos bodily injury claims and environmental property damage claims are allocated among multiple triggered policy years can result in the shifting of tens or hundreds of millions of dollars from one party to another. In recent years, insurers have argued that clauses commonly titled, “Prior Insurance and Non-Cumulation of Liability” (referred to herein as “Non-Cumulation Clauses”), which are found in commercial liability policies, should be applied to reduce or eliminate their coverage responsibilities for long-tail liability claims by shifting their coverage responsibilities to insurers that issued policies in earlier policy years.
