Co-Localization Of Amanitin And A Candidate Toxin-Processing Prolyl Oligopeptidase In Amanita Basidiocarps, Hong Lou, Heather E. Hallen-Adams, John S. Scott-Craig, Jonathan D. Walton
Department of Food Science and Technology: Faculty Publications
Fungi in the basidiomycetous ge nus Amanita owe their high mammalian toxicity to the bicyclic octapeptide amatoxins such as ∝-amanitin. Amatoxins and the related phallotoxins (such as the heptapeptide phalloidin) are encoded by members of the “MSDIN” gene family and synthesized on ribosomes as short (34 - 35 amino acid) proproteins. Anti-amanitin antibodies and confocal microscopy were used to determine the cellular and subcellular localization of amanitin accumulation in basidiocarps (mushrooms) of the Eastern North American destroying angel (Amanita bisporigera). Consistent with previous studies, amanitin is present throughout the basidiocarp (stipe, pileus, lamellae, trama, and universal veil), but …

Fusarium graminearum (teleomorph: Gibberella zeae) is an Ascomycete fungal plant pathogen which infects a range of agriculturally important crops, including wheat, barley, and maize. A random plasmid insertion mutagenesis approach was used to analyze the pathogenicity of the PH-1 strain, for which full genomic information is available. Fungal transformants were initially screened for their ability to infect wheat ears. From a total of 1,170 transformants screened, eight were confirmed to be highly reduced in pathogenicity toward wheat ears and roots. These were designated disease-attenuated F. graminearum (daf) mutants. The in vitro growth rate and appearance of each daf …

Convergent Evolution Of Sequestrate Forms In Amanita Under Mediterranean Climate Conditions, Alfredo Justo, Ingo Morgenstern, Heather E. Hallen-Adams, David S. Hibbett
Department of Food Science and Technology: Faculty Publications
The systematic position of secotioid (Torrendia) and gasteroid (Amarrendia) forms within the agaricoid Amanita lineage (Agaricales, Basidiomycota) was studied with molecular (nLSU, ITS) data. Secotioid and gasteroid forms occur in four independent clades nested within agaricoid forms. One clade corresponds to the secotioid T. pulchella from southern Europe and northern Africa. The others correspond to Torrendia and Amarrendia species from Australia. Mediterranean climatic conditions are postulated as a force driving the convergent evolution of these secotioid and at least one of the gasteroid forms in geographically distant areas. Species formerly placed in Torrendia and Amarrendia are …

The Burmese python (Python molurus bivittatus) is established in Everglades National Park and neighboring areas in south Florida. Beyond its substantial ecological impacts to native fauna in south Florida, concerns have been raised as to its potential to occupy other parts of the USA, even as far north as Washington, DC. During a recent period of cold weather, seven of nine captive Burmese pythons held in outdoor pens at our facility in north-central Florida died, or would have died absent our intervention. This cold-induced mortality occurred despite the presence of refugia with heat sources. Our findings cast doubt …
Importance Of Resource Selection And Social Behavior To Partitioning Of Hostile Space By Sympatric Canids, Todd Atwood, Eric Gese
Importance Of Resource Selection And Social Behavior To Partitioning Of Hostile Space By Sympatric Canids, Todd Atwood, Eric Gese
United States Department of Agriculture Wildlife Services: Staff Publications
Investigations into mechanisms of resource partitioning are particularly suited to systems where nascent interactive behaviors are observable. Wolf (Canis lupus) recolonization of the Greater Yellowstone Ecosystem provided such a system, and we were able to identify behaviors influencing the partitioning of resources by coyotes (Canis latrans) and wolves. We observed coyote–wolf interactions immediately after wolf recolonization, when reemergent behaviors mediating the outcome of competitive interactions were detectable and mechanisms of spatial avoidance were identifiable. Although coyotes used the same space as wolves, they likely minimized risk of encounter by making adaptive changes in resource selection based …

Human monkeypox has never been reported in Ghana, but rodents captured in forested areas of southern Ghana were the source of the monkeypox virus introduced into the United States in 2003. Subsequent to the outbreak in the United States, 204 animals were collected from two commercial trapping sites in Ghana. Animal tissues were examined for the presence of orthopoxvirus (OPXV) DNA using a real-time polymerase chain reaction, and sera were assayed for antibodies against OPXV. Animals from five genera (Cricetomys , Graphiurus , Funiscirus, and Heliosciurus ) had antibodies against OPXV, and three genera (Cricetomys , Graphiurus …

Iptakalim: A Potential Antipsychotic Drug With Novel Mechanisms?, Tao Sun, Changjiu Zhao, Gang Hu, Ming Li
Department of Psychology: Faculty Publications
Iptakalim is a novel putative adenosine triphosphate (ATP)-sensitive potassium (KATP) channel opener. In the brain, iptakalim is thought to act on the neuronal and astrocytic plasma membrane and/or mitochondrial KATP channels. Because iptakalim demonstrates an action on the regulation of dopamine and glutamate release in the forebrain regions, we examined its potential antipsychotic efficacy in several preclinical tests. First, we show that iptakalim is effective in reducing amphetamine- and phencyclidine-induced hyperlocomotion as well as selectively disrupting conditioned avoidance responding. Next, we show that combined iptakalim and amphetamine treatment produces a reduction on prepulse inhibition of acoustic startle …
Aging And The Vulnerability Of Speech To Dual Task Demands, Susan Kemper, Ralynn Schmalzried, Lesa Hoffman, Ruth Herman
Aging And The Vulnerability Of Speech To Dual Task Demands, Susan Kemper, Ralynn Schmalzried, Lesa Hoffman, Ruth Herman
Department of Psychology: Faculty Publications
Tracking a digital pursuit rotor task was used to measure dual task costs of language production by young and older adults. Tracking performance by both groups was affected by dual task demands: time on target declined and tracking error increased as dual task demands increased from the baseline condition to a moderately demanding dual task condition to a more demanding dual task condition. When dual task demands were moderate, older adults’ speech rate declined but their fluency, grammatical complexity, and content were unaffected. When the dual task was more demanding, older adults’ speech, like young adults’ speech, became highly fragmented, …
Reference Place Conditioning Procedure With Cocaine: Increased Sensitivity For Measuring Associatively Motivated Choice Behavior In Rats, Carmela M. Reichel, Jamie L. Wilkinson, Rick A. Bevins
Reference Place Conditioning Procedure With Cocaine: Increased Sensitivity For Measuring Associatively Motivated Choice Behavior In Rats, Carmela M. Reichel, Jamie L. Wilkinson, Rick A. Bevins
Department of Psychology: Faculty Publications
Place conditioning is widely used to study the conditioned rewarding effects of drugs. In the standard version, one reward (cocaine) is compared to no reward (saline). A modified variant of this task, “reference-conditioning” procedure, compares two potentially rewarding stimuli (high versus low cocaine dose). There has been little research on the utility of this procedure. Experiment 1 used the standard protocol with saline administered before confinement to the reference compartment of a place-conditioning chamber. On alternating days, saline, 2.5, 5, 7.5, 10, or 20 mg/kg cocaine was administered before confinement to the opposite compartment. In Experiments 2 and 3, reference-compartment …
An Examination Of Questionnaire Evaluation By Expert Reviewers, Kristen Olson
An Examination Of Questionnaire Evaluation By Expert Reviewers, Kristen Olson
Department of Sociology: Faculty Publications
Expert reviews are frequently used as a questionnaire evaluation method but have received little empirical attention. Questions from two surveys are evaluated by six expert reviewers using a standardized evaluation form. Each of the questions has validation data available from records. Large inconsistencies in ratings across the six experts are found. Despite the lack of reliability, the average expert ratings successfully identify questions that had higher item nonresponse rates and higher levels of inaccurate reporting. This article provides empirical evidence that experts are able to discern questions that manifest data quality problems, even if individual experts vary in what they …

Abandoned Mine Drainage In The Swatara Creek Basin, Southern Anthracite Coalfield, Pennsylvania, Usa: 1. Stream Water Quality Trends Coinciding With The Return Of Fish, Charles A. Cravotta Iii, Robin A. Brightbill, Michael J. Langland
United States Geological Survey: Staff Publications
Acidic mine drainage (AMD) from legacy anthracite mines has contaminated Swatara Creek in eastern Pennsylvania. Intermittently collected base-flow data for 1959–1986 indicate that fish were absent immediately downstream from the mined area where pH ranged from 3.5 to 7.2 and concentrations of sulfate, dissolved iron, and dissolved aluminum were as high as 250, 2.0, and 4.7 mg/L, respectively. However, in the 1990s, fish returned to upper Swatara Creek, coinciding with the implementation of AMD treatment (limestone drains, limestone diversion wells, limestone sand, constructed wetlands) in the watershed. During 1996–2006, as many as 25 species of fish were identified in the …

One of the goals of this Symposium was to bring together agricultural and conservation users of rodenticides to discuss the impacts of rodenticides on the environment, examine the current regulatory climate governing their use, and identify ways that users can reduce or eliminate these impacts. Some of the presentations in today’s symposium highlighted specific impacts, and the preceding talk described the scenario of what can happen if an issue related to pesticide impacts ends up in the courts. The three agencies that were represented on this panel (USDA, USFWS, and EPA) have regulatory oversight and enforcement authority for the use …

Superabundant Food At Catfish Aquaculture Facilities Improves Body Condition In American White Pelicans, D. Tommy King, Jerold Belant, Brent Harrel, James Glahn
United States Department of Agriculture Wildlife Services: Staff Publications
Few studies have investigated the use of aquaculture-produced foods by piscivorous birds. American White Pelicans (Pelecanus erythrorhynchos) were collected from four locations (two aquaculture, one riverine and one coastal) in the southeastern United States during winter and spring 1998-1999 to assess the contribution (biomass, frequency of occurrence) of aquaculture-produced foods and their effect on body condition. Pelican diets reflected opportunistic foraging across locations. Diet near catfish ponds consisted mostly of Channel Catfish (Ictalurus punctatus). Diets along the Mississippi River had similarly high biomass of catfish, but otolith counts suggested lower use of catfish. Diets near crawfish ( …

In 1977, scientific surveys indicated that bowhead whales (Balaena mysticetus) in the Beaufort Sea were in trouble, with fewer than 1,000 individuals remaining. The International Whaling Commission took action to put a moratorium on native hunts in order to protect the species. Yet local Inuit hunters didn't see what the fuss was about. Their own estimates, gleaned from time and experience, put bowhead numbers at 7,000. The Inuits also disputed western scientists' contentions that whales couldn't swim under offshore ice and that they did not feed during migration. Researchers responded to these criticisms by developing a new survey …

In-Situ Stress And Fracture Characterization For Planning Of An Egs Stimulation In The Desert Peak Geothermal Field, Nevada, Stephen H. Hickman, Nicholas C. Davatzes
United States Geological Survey: Staff Publications
An integrated study of natural fracture geometry, fluid flow and stress was conducted in Desert Peak well 27-15 in preparation for development of an Enhanced Geothermal System (EGS) through hydraulic stimulation. This stimulation will be carried out at depths of ~3000 to 3500 ft in units comprised of silicified rhyolite tuffs and metamorphosed mudstones at ambient temperatures of ~180 to 195° C. Our previous analyses of borehole image logs from this well showed that the current minimum horizontal principal stress, Shmin, is oriented 114 ± 17º and that numerous fractures in the planned stimulation interval are optimally oriented for normal …

Hydrogeophysical Methods For Analyzing Aquifer Storage And Recovery Systems, Burke J. Minsley, Jonathan Ajo-Franklin, Amitabha Mukhopadhyay, Frank Dale Morgan
United States Geological Survey: Staff Publications
Hydrogeophysical methods are presented that support the siting and monitoring of aquifer storage and recovery (ASR) systems. These methods are presented as numerical simulations in the context of a proposed ASR experiment in Kuwait, although the techniques are applicable to numerous ASR projects. Bulk geophysical properties are calculated directly from ASR flow and solute transport simulations using standard petrophysical relationships and are used to simulate the dynamic geophysical response to ASR. This strategy provides a quantitative framework for determining site-specific geophysical methods and data acquisition geometries that can provide the most useful information about the ASR implementation. An axisymmetric, coupled …

Locations And Magnitudes Of Historical Earthquakes In The Sierra Of Ecuador (1587–1996), Céline Beauval, Hugo Yepes, William H. Bakun, José Egred, Alexandra Alvarado, Juan-Carlos Singaucho
United States Geological Survey: Staff Publications
The whole territory of Ecuador is exposed to seismic hazard. Great earthquakes can occur in the subduction zone (e.g. Esmeraldas, 1906, Mw 8.8), whereas lower magnitude but shallower and potentially more destructive earthquakes can occur in the highlands. This study focuses on the historical crustal earthquakes of the Andean Cordillera. Several large cities are located in the Interandean Valley, among them Quito, the capital (∼2.5 million inhabitants). A total population of∼ 6 million inhabitants currently live in the highlands, raising the seismic risk. At present, precise instrumental data for the Ecuadorian territory is not available for periods earlier than …

United States Geological Survey: Staff Publications
Feral cats (Felis catus) have spread throughout anthropogenic and insular environments of the world. They now threaten many species of native wildlife with chronic depredation. Knowledge of feral cat population dynamics is necessary to understand their ecological effects and to develop effective control strategies. However, there are few studies worldwide regarding annual or lifetime survival rates in remote systems, and none on Pacific islands. We constructed the age distribution and estimated survival of feral cats in a remote area of Hawai‘i Island using cementum lines present in lower canine teeth. Our data suggest annual cementum line formation. A …

Subspecies Status And Population Genetic Structure In Piping Plover (Charadrius Melodus), Mark P. Miller, Susan M. Haig, Cheri L. Gratto-Trevor, Thomas D. Mullins
United States Geological Survey: Staff Publications
Piping Plover (Charadrius melodus) is a migratory shorebird that is listed as endangered in Canada and the U.S. Great Lakes and as threatened throughout the rest of its breeding and winter range. We undertook a comprehensive molecular-genetic investigation to (1) address subspecific taxonomy, (2) characterize population genetic structure, and (3) infer past bottlenecks and demographic processes in this species. Analyses included individuals from 23 U.S. states and Canadian provinces and were based on mitochondrial DNA sequences (580 base pairs, n = 245) and 8 nuclear microsatellite loci (n = 229). Our findings provide support for separate Atlantic and …

Reply To Comments Of Nolan And Cook, Larry V. Benson, Timothy R. Pauketat, Edward R. Cook
United States Geological Survey: Staff Publications
We appreciate and would like to respond to the comments made by Nolan and Cook. We make three points in our reply, which are directed at clarifying our position and responding to a few of Nolan and Cook's assumptions.
First, as a matter of clarification, the Palmer Drought Severity Index (PDSI) is not a measure of summer rainfall as Nolan and Cook (2010) imply. Instead it is a theoretical measure of soil-moisture, the value of which evolves over several months in response to fluxes of precipitation, evaporation, and runoff. In practice, PDSI is usually heavily weighted toward the precipitation side …

Accurate trace-metal and strontium-isotope analyses of archaeological corn cobs require that metal contaminants be removed prior to chemical analysis. Archaeological cobs are often coated with construction debris, dust, or soilwhich contains mineral particles. In addition, most archaeological cobs are partially or completely burned and the burned parts incorporate mineral debris in their hardened residual structures. Unburned cobs are weak ion exchangers and most metals within a cob are not firmly bound to cob organic matter; therefore, immersing cobs in acids and rinsing them in deionized water to remove mineral contaminants may result in the undesirable loss of metals, including strontium, …

Used a population-based sample (Georgia Centenarian Study, GCS), to determine proportions of centenarians reaching 100 years as (1) survivors (43%) of chronic diseases first experienced between 0–80 years of age, (2) delayers (36%) with chronic diseases first experienced between 80–98 years of age, or (3) escapers (17%) with chronic diseases only at 98 years of age or older. Diseases fall into two morbidity profiles of 11 chronic diseases; one including cardiovascular disease, cancer, anemia, and osteoporosis, and another including dementia. Centenarians at risk for cancer in their lifetime tended to be escapers (73%), while those at risk for cardiovascular disease …

The creation and use of a globally available database of DNA sequences from a standardized gene region has been proposed as a tool for species identification, assessing genetic diversity and monitoring the legal and illegal trade in wildlife species. Here, we contribute to the Barcode of Life Data System and test whether a short region of the mitochondrial cytochrome c oxidase subunit 1 (COX1) gene would reliably distinguish among a suite of commonly hunted African and South American mammal and reptile species. We used universal primers to generate reference barcode sequences of 645 bp for 23 species from five vertebrate …

African dust additions explain the origin of terra rossa soils that are common on the carbonate-platform island of Mallorca, Spain. Mineralogical and geochemical analyses indicate that Quaternary carbonate eolianites on Mallorca have a very high purity, usually composed of more than 90% carbonate minerals (calcite, dolomite, and aragonite). In contrast, terra rossa soils developed on these eolianites have lower carbonate contents and contain higher concentrations of quartz and other silicates. Analyses of immobile trace elements indicate that the non-carbonate fractions of the eolianites have distinctive Zr/Hf, La/Yb, Cr/Sc and Th/Ta values that differ from the superjacent terra rossa soils. These …

Assessing The Potential For Luminescence Dating Of Basalts, S. Tsukamoto, G. A.T. Duller, A. G. Wintle, Daniel R. Muhs
United States Geological Survey: Staff Publications
The possibility of dating basalt using luminescence was tested on four samples with independent age control from Cima volcanic !eld, California, with the ultimate aim of assessing whether the technique could be used to date sediments on the surface of Mars. Previous analysis of these samples had demonstrated that the infrared stimulated luminescence (IRSL) signal is most suitable for dating as it showed the lowest fading rate among various luminescence signals. In this study, changes in equivalent dose as a function of preheat are described. The ages for the two youngest Cima samples agree with the independent ages based on …

Monetary Policies In A Small Open Economy Model With Labor Mobility And Remittances, Diana Rose Del Rosario
Dissertations and Theses Collection (Open Access)
This paper presents a model of a small open economy that allows for international labor mobility, thereby endogenizing migrant transfers or remittances. The resulting model is calibrated to the Philippine economy, of which labor migration and remittance inflows are key forces that drive the economy’s growth. The model’s impulse response functions illustrate that the presence of these features generates a different set of dynamics from the standard small open economy model (without labor mobility). Depending on the source of the shock, labor mobility and remittances can either exacerbate or cushion the impact of the shock on the economy. A temporary …

In this paper we examine how stock returns in China respond to monetary policy announcements made by PBC in a short term around announcement day. We employ a nonparametric event-study method to investigate such reactions. We arrive at the following conclusions. Firstly, there is information leakage of monetary policy changes, which is verified by significant changes in stock returns before monetary policy announcement and quitness of stock market after announcement. Secondly, financially constrained and financially unconstrained firms respond quite similarly to monetary policy shocks, which disobeys credit channel of monetary policy transmission in the short run. Thirdly, reserve ratio changes …

I propose that various measures of mutual funds’ performance are more consistent with their investment capability when mutual funds present low idiosyncratic risks. This paper finds conditional predictor for funds’ returns: alpha predicts returns positively for low idiosyncratic risk funds. It suggests that mutual funds which showed high alpha and low idiosyncratic risk in the past may be capable in investment. Their performance is consistently higher than funds with low idiosyncratic risk and low alpha. On the other hand, the performance of high idiosyncratic risk funds is more likely to reverse in the future: expected returns are low for high …

This thesis studies a model presented by Neumeyer & Perri (2005), which aims to explain the strong countercyclicality of interest rates and net exports in emerging market economies. The model accomplishes this by decomposing interest rates into an international rate and a country risk component, and by making labor demand sensitive to movements in these rates via a working capital constraint imposed on the representative firm. Moreover, it proposes two approaches to determining the stochastic processes for these interest rates: the independent country risk case and the induced country risk case. The induced country risk model calibrated to Argentine data …
