Good grapes are required to make good wine. Quality wines start in quality vineyards. Vineyard design, installation, and operation, including site location, trellis options, vine selection, and canopy management, along with when to harvest are discussed. While winemaking is the final phase in the pursuit of the winemaker's passion, it is but a simple formula. The beginning of a good wine is in a good vineyard. This study includes a financial projection for a 10-acre vineyard as well as for starting an onsite, complementary sized winery. Discovery And Characterization Of An Antibiotic From The Soil Bacterium Bacillus Sp., Thomas S. Barber

Many important antibiotics have become nearly obsolete due to the rise of antibiotic resistant pathogens. Rhodococcus, an actinomycete related to the prolific antibiotic producing genus Streptomyces, harbors over 30 genes for secondary metabolism that could be involved in antibiotic production. Several antibiotics have already been reported for Rhodococcus, suggesting the genus may be a good source for new inhibitory compounds. Fifty four soil bacteria were isolated using enrichment culture techniques (including 37 Rhodococcus) and screened for antibiotic producers. BTHX2, a species of Bacillus was found to have activity against Micrococcus luteus and Rhodococcus erythropolis. BTHX2 …

Severe neuromuscular disorders can produce locked-in syndrome (LIS), a loss of nearly all voluntary muscle control. A brain-computer interface (BCI) using the P300 event-related potential provides communication that does not depend on neuromuscular activity and can be useful for those with LIS. Currently, there is no way of determining the effectiveness of P300-based BCIs without testing a person's performance multiple times. Additionally, P300 responses in BCI tasks may not resemble the typical P300 response. The concentrations of heavy metals (Cd, Cu, Fe, Ni, Pb, Zn) were measured in water, and fish samples caught from the pond at Fishery Park in Unicoi County, TN. The water samples were collected once a week for 8 weeks. The amounts of metals in the muscle tissues, gill, and liver of the two fish species were measured. This was to determine if any correlation exists between the metal contents in water and those in the fish samples. The power distribution infrastructure in the United States is deteriorating at a rapid rate exposing infrastructure wiring and creating potential shock hazards. Periodic road and sidewalk maintenance projects can also expose wiring and create energized objects. In urban settings inadvertently energized objects include: lamp posts, bus shelters, metal street curbs, sign posts, transformer vaults, and manhole covers as well as concrete and asphalt pavement. Every year electric shocks occur when people and domestic animals (such as dogs and cats) make incidental contact with these energized objects. The transition to high school has been identified by researchers as a pivotal point in students' lives. The addition of a Freshman Academy in some schools has targeted the 9th grade year to ease students' transition to high school and increase the likelihood of academic success.

One purpose of this study is to compare student outcomes of schools that have implemented a Freshman Academy with schools that have not. Little is known about the behavioral patterns of honey bee queens. To determine if mated honey bee queens possess diel rhythmicity in behavior, we observed them in glass-sided observation hives using three types of observation regimes: focal studies consisting of 2-hour and 24-hour continuous observations as well as scan-sampling of multiple queens. All behaviors (active: walking, inspecting, egg-laying, begging for food, feeding, and grooming self; inactive: standing) occurred at all times of day and night, but no queen showed consistent diel rhythmicity in any of the individual behaviors. The intent of this qualitative study was to understand the impact of job-embedded professional development coaches on teacher practice. Coaches in this study worked with teachers to plan lessons, model lessons with feedback, and disaggregate data. This qualitative study was conducted using interviews of coaches to gain their perceptions of the work they did with teachers and how this work influenced teacher practice. Teachers were interviewed to determine their perceptions of the impact coaches' work had on their classroom practices. In particular, it is unknown to what degree broadband internet access is available in the counties that Walters State considers the service area of the college.

This study was designed to investigate Southern Appalachian, first-generation students' expectations of higher education. Research indicates that many first-generation students drop out of college after only 1 semester; however, little research exists concerning the expectations and experiences of first-generation college students from Southern Appalachia.

The study employs a qualitative methodology based in the tradition of grounded theory to highlight students' experiences while encouraging the emergence of data-driven theory based on what the researcher heard. Thus, the entire study is couched in the interpretivist philosophy of research.

Binding of inorganic phosphate (P_i) in ATP synthase catalytic sites is a crucial step for the synthesis of adenosine-5'-triphosphate (ATP). ATP is the fundamental means of cellular energy in almost every organism, and in order to gain insight into the regulation of ATP catalysis, critical amino acid residues responsible for binding P_i must be identified. Here, we investigate the role of highly conserved α-subunit VISIT-DG sequence residues αSer-347, αGly-351, and αThr-349 in P_i binding. Mutations αS347A/Q, αG351Q, αT349A/D/R, βR182A, and αT349R/βR182A were generated via site directed mutagenesis. Results from biochemical assays showed that αSer-347 is required …

This study spatially analyzes emergency 911 call-for-service records from January 1, 2000 through December 31, 2009 for motor vehicle accidents inside the corporate limits of Johnson City, Tennessee. Records were compared according to the land use classification for adjacent properties, roadway type, and traffic signal proximity. Data were evaluated through ArcGIS software using proximity analysis, point pattern analysis, and hotspot analysis.

Motor vehicle accidents evaluated during this study consist of accident locations reported to the Washington County Emergency Communications District concerning property damage, personal injury, and fatalities.
