The Endogenous Soluble Vegf Receptor-2 Isoform Suppresses Lymph Node Metastasis In A Mouse Immunocompetent Mammary Cancer Model, Masa-Aki Shibata, Jayakrishna Ambati, Eiko Shibata, Romulo J. C. Albuquerque, Junji Morimoto, Yuko Ito, Yoshinori Otsuki

Ophthalmology and Visual Science Faculty Publications

BACKGROUND: Cancer metastasis contributes significantly to cancer mortality and is facilitated by lymphangiogenesis and angiogenesis. A new splicing variant, endogenous soluble vascular endothelial growth factor receptor-2 (esVEGFR-2) that we recently identified is an endogenous selective inhibitor of lymphangiogenesis. To evaluate the antimetastatic potential of esVEGFR-2, gene therapy with vector expressing esVEGFR-2 (pesVEGFR-2) or endostatin (pEndo) as a positive control was conducted on murine metastatic mammary cancer.

METHODS: Syngeneic inoculated metastatic mammary cancers received direct intratumoral injection of pesVEGFR-2, pEndo or pVec as control, once a week for six weeks. In vivo gene electrotransfer was performed on the tumors after each …

An Examination Of The Relationship Between Self-Compassion And Burnout In Practicing Psychotherapists : A Project Based Upon An Independent Investigation, Jennifer Ann Thurlow Nov 2010

An Examination Of The Relationship Between Self-Compassion And Burnout In Practicing Psychotherapists : A Project Based Upon An Independent Investigation, Jennifer Ann Thurlow

Theses, Dissertations, and Projects

Burnout is particularly a problem among people whose work involves responding to human suffering; therefore, without adequate personal resources to draw from, psychotherapists are especially at risk. Self-compassion is a powerful coping skill, thus therapists may be protected from burnout through the practice. The purpose of this study was to examine how self-compassion is related to burnout in practicing psychotherapists. It was hypothesized that higher total self-compassion scores would be associated with lower scores on the burnout subscales of emotional exhaustion and depersonalization and that higher scores of total self-compassion would be associated with higher scores on the burnout subscale …

Genetic Evaluation Of American Shad Alosa Sapidissima Restoration Success In James River, Virginia, Aaron Aunins Nov 2010

Genetic Evaluation Of American Shad Alosa Sapidissima Restoration Success In James River, Virginia, Aaron Aunins

Theses and Dissertations

The American shad Alosa sapidissima has experienced severe declines throughout its native range due to habitat degradation, fragmentation, and over-fishing. Hatchery supplementation is often used for stock restoration, but the effects of supplementation on population structure and genetic diversity are rarely assessed. This study employed molecular markers to evaluate how supplementation of the James River American shad population with Pamunkey River origin larvae since 1994 has impacted genetic diversity and population structure. Population genetic parameters of other major Chesapeake Bay tributaries (Susquehanna, Rappahannock, Potomac, and Nanticoke) also were characterized. Prior to stocking, the James and Pamunkey populations exhibited subtle genetic …

Chronic Spontaneous Activity Generated In The Somata Of Primary Nociceptors Is Associated With Pain-Related Behavior After Spinal Cord Injury, Supinder S Bedi, Qing Yang, Robyn J Crook, Junhui Du, Zizhen Wu, Harvey M Fishman, Raymond J Grill, Susan M Carlton, Edgar T Walters

Faculty, Staff and Student Publications

Mechanisms underlying chronic pain that develops after spinal cord injury (SCI) are incompletely understood. Most research on SCI pain mechanisms has focused on neuronal alterations within pain pathways at spinal and supraspinal levels associated with inflammation and glial activation. These events might also impact central processes of primary sensory neurons, triggering in nociceptors a hyperexcitable state and spontaneous activity (SA) that drive behavioral hypersensitivity and pain. SCI can sensitize peripheral fibers of nociceptors and promote peripheral SA, but whether these effects are driven by extrinsic alterations in surrounding tissue or are intrinsic to the nociceptor, and whether similar SA occurs …

Racial/Ethnic Differences In Bone Mineral Density Of Young Adults, Linda D. Wilkin, Tristan D. Sims, Matthew C. Jackson, Bryan L. Haddock

International Journal of Exercise Science

An estimated 1.5 million people suffer a bone disease-related fracture every year. Most work investigating bone mineral density (BMD) focuses on post-menopausal females but a report from the Surgeon General in 2004 stated that of particular concern are men, racial and ethnic minorities, poor individuals, individuals with disabilities, and individuals living in rural areas. The purpose of this study was to examine the racial/ethnic differences in bone mineral density of young adults and to investigate any correlations with variables suggested to influence BMD. BMD was assessed at a younger age than most studies based on the assumption that osteoporosis is …

DNA methyltransferase 1 (DNMT1) is the primary enzyme that maintains DNA methylation. We describe a previously unknown mode of regulation of DNMT1 protein stability through the coordinated action of an array of DNMT1-associated proteins. DNMT1 was destabilized by acetylation by the acetyltransferase Tip60, which triggered ubiquitination by the E3 ligase UHRF1, thereby targeting DNMT1 for proteasomal degradation. In contrast, DNMT1 was stabilized by histone deacetylase 1 (HDAC1) and the deubiquitinase HAUSP (herpes virus–associated ubiquitin-specific protease). Analysis of the abundance of DNMT1 and Tip60, as well as the association between HAUSP and DNMT1, suggested that during the cell cycle the initiation …

Halo Ion Trap Mass Spectrometry: Design, Instrumentation, And Performance, Miao Wang Nov 2010

Halo Ion Trap Mass Spectrometry: Design, Instrumentation, And Performance, Miao Wang

Theses and Dissertations

New ion trap mass spectrometry (ITMS) instrumentation, the toroidal IT and halo IT, were developed to meet the significant growth in on-site analysis applications. The miniature toroidal IT mass analyzer was operated with radio frequency (RF) trapping voltages of 3 kVp-p or less. Despite its reduced dimensions, it has roughly the same ion trapping capacity as conventional 3D quadrupole ITs. Unit-mass resolved spectra for n-butylbenzene, xenon, and naphthalene were obtained. The desired linear mass scale was obtained using conventional mass-selective instability scan combined with resonance ejection. The halo IT was also based on toroidal trapping geometry and microfabrication technology, consisting …

Dendroaspis Natriuretic Peptide For Treatment Of Cancer, David L. Vesely Nov 2010

Dendroaspis Natriuretic Peptide For Treatment Of Cancer, David L. Vesely

USF Patents

Disclosed is a method of inhibiting the growth of a cancer cell using Dendroaspis natriuretic peptide (DNP), isolated from the Green Mamba snake venom with similar structure to ANP, with or without four cardiac natriuretic peptides i.e., atrial natriuretic peptide (ANP), vessel dilator, long acting natriuretic peptide (LANP), and kaliuretic peptide. Dose-response curves revealed a significant (p<0.0001) decrease in human glioblastoma cells with each ten-fold increase in concentration from 1 μM to 100 μM of four of the cardiac peptide hormones. There was an 75%, 68%, 67%, and 65% elimination within 24 hours of glioblastoma cells secondary to vessel dilator, kaliuretic peptide, ANP, and LANP, respectively (p<0.0001) while DNP had no significant effect at 1 μM (2% decrease), and 10 μM (7%), but 100 μM caused a (17%) decrease (p<0.05). Three days after treatment with these peptide hormones, the cancer cells began to proliferate again. These same hormones decreased DNA synthesis from 65% to 87% (p<0.00001).

Immobilization Of Saccharomyces Cerevisiae In Rice Hulls For Ethanol Production, Edita Martini, Dian Andriani, Sriramulu Gobikrishnan, Kyeong Eop Kang, Surn-Teh Bark, Changshin Sunwoo, Bambang Prasetya, Don-Hee Park

Makara Journal of Technology

The whole cell immobilization in ethanol fermentation can be done by using natural carriers or through synthetic carriers. All of these methods have the same purpose of retaining high cell concentrations within a certain defined region of space which leads to higher ethanol productivity. Lignocellulosic plant substance represents one of highly potential sources in ethanol production. Some studies have found that cellulosic substances substances can also be used as a natural carrier in cell immobilization by re-circulating pre-culture medium into a reactor. In this experiment, rice hulls without any treatment were used to immobilize Saccharomyces cerevisiae through semi solid state …
