Research was conducted to implement remote setting technology for the commercial- scale production of oysters on alternative cultch material in Louisiana. Genetic research has created an enhanced oyster stock for commercial cultivation. Since the Louisiana oyster industry relies on natural seed production for product, they are unable to utilize these new advances and are never guaranteed a reliable source of oyster seed. Remote setting is one way of producing a consistent supply of genetically improved oysters. Cultch material for oyster settlement is a limiting resource. The Louisiana Gulf Coast is losing land at an alarming rate for a combination of reasons. At the same time carbon dioxide is accumulating in the atmosphere and the oceans by the increased burning of fossil fuels. Therefore, bioengineered oyster dominated artificial reefs have been developed in an effort to mitigate erosion while storing carbon in the oyster shells. These reefs support the growth of a native oyster species, Crassostrea virginica, which are considered ecological engineers and serve to improve water quality, protect coastal wetlands, capture and store carbon, and enhance the economy. The most common toxins detected following freshwater harmful algal blooms are microcystins, a group of hepatotoxins produced by cyanobacteria (e.g., Microcystis and Anabaena spp.). Preference for filter-feeding prey, including bivalves, makes the blue crab, Callinectes sapidus, vulnerable to microcystin contamination and makes the commercially important crab a potential vector of microcystins. I used a combination of field and laboratory studies to determine blue crab vulnerability to microcystin contamination and consequent impacts on crab physiology. Samples collected from a hyper-eutrophic freshwater lake, Lac des Allemands, Louisiana, were analyzed for cyanobacterial abundances and microcystins in surface water and blue crabs using light-microscopy …

Mycobacterium tuberculosis (Mtb) is the causative agent of tuberculosis (TB) and infects nearly one-third of the world's population. The same TB chemotherapeutic regimen has been administered for more than 50 years, and can be improved by developing innovative methods to discover new anti-TB drugs. This work describes a newly established high-throughput screen (HTS) platform technology based on Mycobacterial-Protein Fragment Complementation (M-PFC) to identify small molecule inhibitors of protein-protein interactions in mycobacteria. M-PFC HTS employs both whole cell and target-based approaches, and offers flexibility in selecting an appropriate protein interaction target for HTS. The ability to differentially manipulate available genetic information in order to generate diverse cellular identities represents an innovation of complex multicellular eukaryotic organisms. Cis-acting modules that regulate transcription play extremely important roles in ensuring lineage-specific expression of genes that define cellular identities. In our studies, using Interferon-gamma (IFN-gamma, a cytokine encoded by the gene Ifng), a cytokine encoded by T, NK and NKT cells as a model, we have identified and characterized the roles of key cis regulatory elements that facilitate T lineage specific induction as well as repression of Ifng transcription. Metastatic disease accounts for the overwhelming majority of cancer related deaths. More specifically, breast cancer remains one of the leading causes of death in women and breast cancer cells metastasize to bone more than any other secondary site. Upon arriving within the bone microenvironment, breast cancer cells interact with bone marrow cells, leading to changes in bone biology that favor growth of the cancer cells. Additionally, some cancer cells are capable of direct cellular communication with cells at metastatic sites via dysregulation of a family of proteins known as connexins. Prostate cancer is the most common male malignancy and the second leading cause of cancer related death in the United States. Despite recent advances in the detection, diagnosis, and treatment of prostate cancer, there is a need for more effective therapies. Unfortunately, most conventional therapeutic modalities, such as androgen ablation therapy, frequently result in androgen-independent cancers. These cancers are typically more aggressive, metastatic, and resistant to chemotherapeutic agents than androgen-dependent prostate cancer. Following a radiological or nuclear disaster, radiation dose assessment is imperative to minimize morbidity and mortality through rationally directed medical intervention. Current methods of retrospective dosimetry are not amenable to mass exposure scenarios and remain limited to monitoring of clinical symptoms (nausea/vomiting and lymphocyte depletion) and cytogenetic analysis. The goal of this study was to identify radiation biomarkers capable of qualitative (non-irradiated/irradiated) and quantitative (dose) assessment of radiation exposure. Initial analyses revealed 17 radiation-responsive cytokine/chemokine genes in blood samples from 6 pediatric cancer patients undergoing fractionated total body irradiation (TBI). The pore size, shape, surface morphology, and pore connectivity are among the most important factors that affect the scaffold's mechanical properties and biointegration. Exposure to air pollutants are known to induce airway inflammation, decrease lung function, and worsen existing pulmonary conditions such as asthma. Additionally, air pollutant exposure has also been shown to impact cardiovascular health. Although these pollutant-induced cardiovascular effects were considered to be mediated by the direct action of pollutants upon the cardiovascular tissue and/or the indirect release of pro-inflammatory factors into the circulation, the relative contribution of the two pathways remained equivocal. Aspergillus flavus is an opportunistic fungal pathogen that infects peanuts, cotton, corn and tree nuts. Aspergillus flavus is a major problem globally due to the production of acutely toxic and carcinogenic aflatoxins. Louisiana climatic conditions lead to annual threats of corn aflatoxin contamination. The purpose of this study was to determine the specific ability of different strains of A. flavus to infect corn. Research conducted over two years evaluated EPTC at 2.0, 2.9, 3.9, and 4.9 kg ai/ha incorporated on pre-formed sugarcane beds using a Lilliston® rolling cultivator and a hipper/bedder. Averaged across EPTC rates 30 days after application, bermudagrass was controlled 62% when incorporated with a rolling cultivator compared with 43% for the hipper/bedder. For johnsongrass and purple nutsedge, incorporation implements were equally effective and control with EPTC 30 days after application averaged 49 and 30%, respectively. Averaged across incorporation methods, control of all weeds 30 days after EPTC application was maximized at 3.9 kg/ha, but control was no greater than when …

In Louisiana, the extensive loss and fragmentation of coastal marshes has prompted inquires into the impacts these processes may have on estuarine-dependant nekton. To date, research on nekton response to marsh loss and fragmentation has been limited to landscape-level studies which focus on the relationship between nekton productivity and the availability of marsh edge. These studies have relied on the assumption that marsh edges provide the same level of support to nekton regardless of the degree of surrounding marsh loss or fragmentation. This study investigates whether the Grade Level Expectations (GLEs) as outlined by the Louisiana Department of Education prepare students for introductory college level chemistry. A sample of 135 students in Dr. Watkins fall 2009 CHEM 1201 classes were tested for correlations between knowledge of the GLEs (chemical concept inventory pre-test and post-test score and normalized gain) and performance in CHEM 1201 (average exam score and final exam score). No significant correlations were found between any of these variables. Crawfish and shrimp byproducts are an excellent source of astaxanthin. The antioxidant-rich natural astaxanthin dispersed in alpha linolenic acid-rich flaxseed oil (FO) may provide healthier functional food options for US consumers. The goals of this study were to extract astaxanthin from crawfish and shrimp byproducts and to develop astaxanthin dry powders using microencapsulation technology. Astaxanthin extracted with FO from crawfish (FOCA) and shrimp (FOSA) byproducts were stable at 30 and 40 Ž, but had substantial degradation at 50 and 60 Ž during four 4 h heating. Evidence suggests that general self-efficacy, one’s beliefs about his or her global abilities, and social self-efficacy, one’s beliefs in his or her ability to navigate social situations, are strongly connected to levels of social anxiety. PLGA (polylactic-co-glycolic acid) nanoparticles containing the hydrophobic antifungal Itraconazole (ITZ) were developed to address the need for more efficient means of treating fungal infections. PLGA-ITZ nanoparticles were synthesized using an oil-in-water emulsion evaporation method. The nanoparticles morphology (TEM), size and size distribution, zeta potential (DLS), encapsulation efficiency (UV-VIS), release profile, and antifungal activity were characterized. The blank NPs and loaded PLGA-ITZ NPs were spherical with diameters of 201±5 nm, 232±1 nm and 223±36 nm at 0%, 12.5% and 25% loadings, respectively. PURPOSE: This study was designed to examine the effects of sun-dried raisins versus sports jellybeans on endurance performance in trained cyclists/triathletes. METHODS: Ten healthy males (18-33 y) completed one water-only acclimatization (WAT) exercise trial and two randomized exercise trials. Each trial consisted of a 120-minute constant intensity glycogen depletion period followed by a 10-km time trial (TT). During each experimental trial, participants consumed isocaloric amounts of sun-dried raisins (SDR) or Sports Jelly Beans(SJB) in 20-minute intervals. North American grassland birds have been declining at an alarming rate. Winter habitat for grassland species in the southeastern U.S. generally occurs within forests subject to management. I studied wintering grassland bird communities in De Soto National Forest in southern Mississippi. My objectives were to: 1) Assess bird communities and vegetation structure in upland forest stands that were salvage-logged following Hurricane Katrina, in stands managed for Red-cockaded Woodpeckers (Picoides borealis, RCW), and in stands containing bogs; and 2) Determine the extent of the association between vegetation and occurrence of the common wintering grassland birds. As a multi-million dollar industry, production of the eastern oyster Crassostrea virginica is significant to the economy of the nation’s coastal regions. The cost of commercial oyster aquaculture has higher inputs than the oyster fishery, but larger, better quality oysters can be produced for culture by the stable availability of triploid individuals.

Water chemistry, habitat quality, and channel stability interact to influence a stream’s biological integrity. The goal of this project was to assess how channel stability, together with other physicochemical stream measurements, are associated with the structure and abundance of resident macroinvertebrate communities. I recorded multiple physicochemical parameters and calculated a Pfankuch habitat stability index monthly for a year at potential reference streams in southeastern Louisiana. I assessed the relationships of channels stability with measured physicochemical parameters with multiple regression and principle components analysis.
