This paper presents a software tool (the Investment Comparison Tool), a methodology (the Investment Comparison Methodology), and a decision support system (the Investment Comparison System) to aid decision makers with Research and Development Investment allocations. The Investment Comparison System (ICS) can be used vertically within an organization and horizontally across organizations at multiple portfolio investment levels. The ICS is applicable to any R&D environment ranging from Industry Research Environments to Federal Agencies. Choosing the appropriate neuroimaging phenotype is critical to successfully identify genes that influence brain structure or function. While neuroimaging methods provide numerous potential phenotypes, their role for imaging genetics studies is unclear. Here we examine the relationship between brain volume, grey matter volume, cortical thickness and surface area, from a genetic standpoint. Four hundred and eighty-six individuals from randomly ascertained extended pedigrees with high-quality T1-weighted neuroanatomic MRI images participated in the study. Surface-based and voxel-based representations of brain structure were derived, using automated methods, and these measurements were analysed using a variance-components method to identify the heritability of these traits …

During cell proliferation, protein degradation is strictly regulated by the cell cycle and involves two complementary ubiquitin ligase complexes, the SCF (Skp, Cullin, F-box) and APC/C (Anaphase Promoting Complex/Cyclosome) ubiquitin ligases. SCF ligases are constitutively active and generally target only proteins after they have been selected for degradation, usually by phosphorylation. In contrast, APC/C complexes are themselves activated by phosphorylation and their substrates contain a targeting signal known as degron, a consensus amino acid sequence such as a D-Box. SCF complexes degrade proteins during the G1 phase. However, as DNA synthesis begins, the SCF complexes are degraded and APC/C complexes …

The rising temperature of the world's oceans has become a major threat to coral reefs globally as the severity and frequency of mass coral bleaching and mortality events increase. In 2005, high ocean temperatures in the tropical Atlantic and Caribbean resulted in the most severe bleaching event ever recorded in the basin. Methodology/Principal Findings: Satellite-based tools provided warnings for coral reef managers and scientists, guiding both the iming and location of researchers' field observations as anomalously warm conditions developed and spread across the greater Caribbean region from June to October 2005. Background. The gut is a major reservoir for human immunodeficiency virus (HIV) in patients receiving antiretroviral therapy (ART). We hypothesized that distinct immune environments within the gut may support varying levels of HIV.

Methods. In 8 HIV-1-positive adults who were receiving ART and had CD4⁺ T cell counts of >200 cells/μL and plasma viral loads of <40 copies/mL, levels of HIV and T cell activation were measured in blood samples and endoscopic biopsy specimens from the duodenum, ileum, ascending colon, and rectum.

Results. HIV DNA and RNA levels per CD4⁺ T cell were higher in all 4 gut sites compared with those in the blood. HIV DNA levels increased from the duodenum to the rectum, whereas the median HIV …

The vitelline membrane (VM), the oocyte proximal layer of the Drosophila eggshell, contains four major proteins (VMPs) that possess a highly conserved “VM domain” which includes three precisely spaced, evolutionarily conserved, cysteines (CX⁷CX⁸C). Focusing on sV23, this study showed that the three cysteines are not functionally equivalent. While substitution mutations at the first (C123S) or third (C140S) cysteines were tolerated, females with a substitution at the second position (C131S) were sterile. Fractionation studies showed that sV23 incorporates into a large disulfide linked network well after its secretion ceases, suggesting that post-depositional mechanisms are in place to …

Tumor necrosis factor alpha (TNF-α) is a critical mediator of inflammation, and its production is tightly regulated, with control points operating at nearly every step of its biosynthesis. We sought to identify uncharacterized TNF-α 3' untranslated region (3'UTR)-interacting proteins utilizing a novel screen, termed the RNA capture assay. We identified CARHSP1, a cold-shock domain-containing protein. Knockdown of CARHSP1 inhibits TNF-α protein production in lipopolysaccharide (LPS)-stimulated cells and reduces the level of TNF-α mRNA in both resting and LPS-stimulated cells. mRNA stability assays demonstrate that CARHSP1 knockdown decreases TNF-α mRNA stability from a half-life (t(1/2)) of 49 min to a t(1/2) …

Protein phosphorylation is a post translational modification of proteins in which a serine, a threonine or a tyrosine residue is phosphorylated by an enzyme, kinase. Phosphorylation of proteins is a reversible and very important regulatory mechanism that occurs in both prokaryotes and eukaryotes. Phosphorylation turns many protein enzymes on and off, preventing or causing many diseases such as diabetes, cancer and rheumatoid arthritis. The phosphorylation on tyrosine residues of proteins is essential for transmission of signals for cell growth, proliferation and differentiation. Chromatin Unfolding By Cdt1 Regulates Mcm Loading Via Opposing Functions Of Hbo1 And Hdac11-Geminin, Philip G. Wong

The efficiency of metazoan origins of DNA replication is known to be enhanced

by histone acetylation near origins. Although this correlates with increased MCM

recruitment, the mechanism by which such acetylation regulates MCM loading is

unknown. We show here that Cdt1 induces large-scale chromatin decondensation that is

required for MCM recruitment. This process occurs in G1, is suppressed by Geminin, and

requires HBO1 HAT activity and histone H4 modifications. HDAC11, which binds Cdt1

and replication origins during S-phase, potently inhibits Cdt1-induced chromatin

unfolding and re-replication, suppresses MCM loading, and binds Cdt1 more efficiently

in the presence of Geminin. We also …

Vertebrates display a wide variety of parental care behaviours, including the guarding of offspring pre and post nutritional independence as well as the direct provision of nutrients during the early development period. The Amazonian cichlid Symphysodon spp. (discus fish) is unusual among fish species, in that both parents provide offspring with mucus secretions to feed from after hatching. This extensive provision of care, which can last up to a month, imposes a physiological demand on both parents and gives rise to conflict between the parent and offspring. Hypercalcaemia, a common medical problem, can be a manifestation of various diseases. When severe, it can represent a medical emergency. Correct diagnosis is important to prevent unnecessary investigations and parathyroidectomies. We here present five patients with severe hypercalcaemia, most misdiagnosed for months or years before being referred to Sultan Qaboos University Hospital, Oman. We report how clinical examination, evaluation of fasting serum calcium, phosphate, creatinine and 24-hour urine calcium levels together with a review of their radiographics predicted the pathophysiology of the disorder and guided investigative procedures before hormone assays results were available.

Medical curriculum revisions all over the world in the last 20 years have increased the numberof teaching hours in communication skills. This article describes a concrete communication skills teaching programme focused on general practice. Since the spring of 1992, more than 2,000 physicians from Denmark,Sweden and Finland have attended courses within this programme in Kalymnos, Greece. The training mainly takes place in groups of 8 doctors and 1 supervisor. The skills training is based on video feedback using the Window Supervision Method described here. We have identified several classical pitfalls for the doctor which this training programme seeks to address. …

In 1991 the American Society of Testing and Materials (ASTM) introduced the first version of a standard called ASTM E1394-91 for communication between centralised clinical analysers and host systems. For nearly 20 years this low key standard has been used as the basis for analyser host communications. A minor revision of the standard (ASTM1394-97) was published in 1998. This work gives a brief summary of the development of lab messages that led to the introduction and continued use of the standard. This essay constructs a comprehensive view of Dewey’s approach to history, the historical method, and history education. Drawing on Dewey’s approach to the subject at the University of Chicago Laboratory School (1896-1904), Dewey's chapter on the historical method in Logic: A Theory of Inquiry (1938), and a critique of Dewey’s philosophy of history that appeared in the American Historical Review and the published response to this attack by Dewey’s colleagues (1954), the author argues that Dewey consistently approached history in genetic and historicist terms.
