Synthesis, Antiviral And Contraceptive Activities Of Nucleoside–Sodium Cellulose Sulfate Acetate And Succinate Conjugates, Hitesh K. Agarwal, Anil Kumar, Gustavo F. Doncel, Keykavous Parang

Biomedical and Pharmaceutical Sciences Faculty Publications

Chemical conjugates between sodium cellulose sulfate (CS), displaying contraceptive and HIV-entry inhibiting properties, and nucleoside reverse transcriptase inhibitors (NRTIs) (3′-azido-2′,3′-dideoxythymidine (AZT), 3′-fluoro-2′,3′-dideoxythymidine (FLT), or 2′,3′-dideoxy-3′-thiacytidine (3TC)) were designed to simultaneously provide contraceptive and anti-HIV activity. Two linkers, acetate and succinate, were used to conjugate the nucleoside analogs with CS. The conjugates containing cellulose sulfate-acetate (CSA) (e.g., AZT–CSA and FLT–CSA) were found to be more potent than CS and other conjugates (e.g., AZT–succinate–CS, and FLT–succinate–CS). The presence of both sulfate and the acetate groups on cellulose were critical for generating maximum anti-HIV activity. In addition to showing equal potency against wild-type …

Pregnane X Receptor Is Required For Interleukin-6-Mediated Down-Regulation Of Cytochrome P450 3a4 In Human Hepatocytes, Jian Yang, Chunshu Hao, Dongfang Yang, Deshi Shi, Xiulong Song, Xiaofei Luan, Gang Hu, Bingfang Yan

Biomedical and Pharmaceutical Sciences Faculty Publications

Cytochrome P450 3A4 (CYP3A4) is the most abundant cytochrome P450 enzyme in human liver and metabolizes more than 60% of prescribed drugs in human body. Patients with liver conditions such as cirrhosis show increased secretion of cytokines (e.g., interleukin-6) and decreased capacity of oxidation of many drugs. In this study, we provided molecular evidence that cytokine secretion directly contributed to the decreased capacity of oxidative biotransformation in human liver. After human hepatocytes were treated with IL-6, the expression of CYP3A4 decreased at both mRNA and protein levels, so did the CYP3A4 enzymatic activity. Meanwhile, the repression of CYP3A4 by IL-6 …

Watershed Modeling Using Gis Technology: A Critical Review Jan 2010

Watershed Modeling Using Gis Technology: A Critical Review

Journal of Spatial Hydrology

Understanding and managing water resource problems involves complex processes and interactions within the watershed surface and subsurface. The imposition of total maximum daily load (TMDL) regulations on the pollutant influx to a watershed has created a strong demand for new assessment tools. The spatial scales relevant to transport of the pollutants may span many orders of magnitude, ranging from field plots to regional hydrological systems. As the demand for and development of watershed modeling capabilities have evolved, geographic information systems (GIS) in tandem with remote sensing technologies have played an essential role supporting both data collection and analysis. This paper …

Biological Sciences Faculty Research & Creative Works

Coal and gold mining are important economic activities in New Zealand, and the West Coast of the South Island and Southland, for example, have long histories of mining. The process of mineral extraction inevitably results in environmental impacts, but few tools exist to help mining companies and regulators assess and predict environmental impacts of mining operations. This framework has been developed as part of a collaborative research programme with key mining partners to assist with planning of future mine developments on the West Coast and in Southland. This framework focuses on water quality issues associated with coal and gold mining, …

Termite Raiding By The Ponerine Ant Pachycondyla Analis (Hymenoptera: Formicidae): Behavioural And Chemical Ecology, Ahmed Yusuf Abdullahi

All PhD Dissertations

The ant Pachycondyla analis (formerly Megaponera foetens, commonly known as the Matabele ant) is a widespread ponerine in sub-Saharan Africa. It feeds solely on termites of economic importance belonging to the sub-family Macrotermitinae. These termites are captured during organised raids on their nests and galleries. Previous studies mostly concentrated on certain aspects of the raiding behaviour and trail laying pheromones in this species. Thus the detailed raiding behaviour and chemicallymediated communication between P. analis and its prey are virtually unknown. The aim of this study was to undertake detailed behavioural studies on termite raiding behaviour of P. analis, and to …

Receptor-Dependent Phagocytosis Of Clostridium Sordellii By Human Decidual Macrophages, Tennille D. Thelen Jan 2010

Receptor-Dependent Phagocytosis Of Clostridium Sordellii By Human Decidual Macrophages, Tennille D. Thelen

Master's Theses and Doctoral Dissertations

Clostridium sordellii is an emerging pathogen associated with highly-lethal female reproductive tract (FRT) infections following childbirth, abortion, or cervical instrumentation. Gaps in our understanding of the pathogenesis of C. sordellii infections present major challenges to the development of better preventive and therapeutic strategies against this problem. We sought to determine the mechanisms whereby uterine DMs phagocytose this bacterium and tested the hypothesis that human DMs utilize class A scavenger receptors (CASRs) to internalize unopsonized C. sordellii. In vitro phagocytosis assays with human DMs incubated with pharmacological inhibitors of CASRs (fucoidan, polyinosinic acid, and dextran sulfate) revealed a role for …

Background Sooty (Puffinus griseus) and short-tailed (P. tenuirostris) shearwaters are abundant seabirds that range widely across global oceans. Understanding the foraging ecology of these species in the Southern Ocean is important for monitoring and ecosystem conservation and management. Methodology/Principal Findings Tracking data from sooty and short-tailed shearwaters from three regions of New Zealand and Australia were combined with at-sea observations of shearwaters in the Southern Ocean, physical oceanography, near-surface copepod distributions, pelagic trawl data, and synoptic near-surface winds. Shearwaters from all three regions foraged in the Polar Front zone, and showed particular overlap in the region around 140°E. Short-tailed shearwaters …

Niche Partitioning Among And Within Sympatric Tropical Seabirds Revealed By Stable Isotope Analysis, H S. Young, D J. Mccauley, R Dirzo, R D. Dunbar, Scott A. Shaffer

Faculty Publications, Biological Sciences

The low productivity and unpredictable nature of resources in tropical waters would appear to make resource partitioning among predators difficult. Yet, stable isotope data from the present study suggest that substantial resource partitioning occurs among tropical seabird communities, both between and within species. In the present study, we compared δ13C and δ15N levels among 8 sympatric tropical seabird species. For a subset of these species, we also examined isotopic levels across years, breeding periods, and sexes. When breeding and non-breeding periods were both considered, we found that all species occupied distinct isotopic niches across at least one time period. Resource …

The Medicalization Of Hyperactivity And Inattentiveness : A Social History And Theoretical Perspectives On Adhd, Justin Douglas Brock Jan 2010

The Medicalization Of Hyperactivity And Inattentiveness : A Social History And Theoretical Perspectives On Adhd, Justin Douglas Brock

Theses, Dissertations and Capstones

This study investigates the rise of the Attention-Deficit/Hyperactivity Disorder diagnosis. It approaches the topic from a medicalization perspective and frames it under the therapeutic state as proposed by Nicholas Kittrie in 1971. It asks three questions: how has society, and particularly, the medical community changed to allow the medicalization of hyperactivity and social control of active children? How has the continuing diagnosis of hyperactivity in children expanded to include attention deficit hyperactivity disorder in both children and adults? And what forces are behind the continuously inclusive diagnosis and why? In an attempt to answer these questions the history of the …

Acanthamoeba Casstellanii : Intercellular Location Of Metacaspase, Danielle Maria Desser Jan 2010

Acanthamoeba Casstellanii : Intercellular Location Of Metacaspase, Danielle Maria Desser

Theses, Dissertations and Capstones

Acanthamoeba castellanii, classified as a free living protist, are found in abundance in fresh water, filtered water, and soil, feeding upon bacteria in their environment. The pathogenic strain of this organism can cause either Acanathamoeba keratitis that targets eyes, or more rarely, granulomatous amoebic encephalitis, with immunocompromised individuals most at risk. Acanthamoeba castellanii are characterized by their two different stages: the trophozoite, which is the growing and dividing form, and the cyst form, which is the dormant stage. The cyst stage is highly resistant to harsh environmental conditions due to the double layer cell wall made largely of cellulose. It …

High Gene Flow Due To Pelagic Larval Dispersal Among South Pacific Archipelagos In Two Amphidromous Gastropods (Neritomorpha: Neritidae), Eric D. Crandall, Jonathan R. Taffel, Paul H. Barber Jan 2010

High Gene Flow Due To Pelagic Larval Dispersal Among South Pacific Archipelagos In Two Amphidromous Gastropods (Neritomorpha: Neritidae), Eric D. Crandall, Jonathan R. Taffel, Paul H. Barber

School of Natural Sciences Faculty Publications and Presentations

The freshwater stream fauna of tropical oceanic islands is dominated by amphidromous species, whose larvae are transported to the ocean and develop in the plankton before recruiting back to freshwater habitat as juveniles. Because stream habitat is relatively scarce and unstable on oceanic islands, this life history would seem to favor either the retention of larvae to their natal streams, or the ability to delay metamorphosis until new habitat is encountered. To distinguish between these hypotheses, we used population genetic methods to estimate larval dispersal among five South Pacific archipelagos in two amphidromous species of Neritid gastropod (Neritina canalis …

Hydrodynamic Forces Affect Larval Zebra Mussel (Dreissena Polymorpha) Mortality In A Laboratory Setting, Thomas Horvath, Lori Crane

School of Natural Sciences Faculty Publications and Presentations

Mortality of zebra mussel, Dreissena polymorpha, larvae was quantified in a laboratory experiment that was designed to assess the role played by both intensity and duration of the exposure to hydrodynamic forces. Larvae were collected in a plankton net and distributed in 100-ml aliquots to 125-ml Erlenmeyer flasks. The flasks were spun on an orbital shaker at different speeds, 100 rpm and 400 rpm, to change the intensity of the hydrodynamic forces experienced by larvae inside the flasks. Actual shear forces were not quantified. A parallel set of control flasks were not spun. Flasks were spun for 1, 24 and …
