Assessing Plasticity : The Populations Responsible For The Epithelial Engraftment On Marrow-Derived Cells, Justin Brent Cohen Sep 2010

Assessing Plasticity : The Populations Responsible For The Epithelial Engraftment On Marrow-Derived Cells, Justin Brent Cohen

Yale Medicine Thesis Digital Library

Bone marrow-derived cells (BMDCs) have significant plasticity allowing them to give rise to various non-hematopoietic cell types including epithelial cells of the lung, liver, gut, and skin. These findings offer tremendous possibilities for the use of cell therapy to treat tissue injury and disease. However, the specific populations of cells responsible for this phenomenon are still unclear with considerable controversy over the mechanism of this transformation. In this study, we sought to compare the epithelial engraftment ability of two populations enriched for hematopoietic stem cells (HSCs), specifically the fractionated, lineage-depleted, and homed subset (FLH) and the lineage-Sca-1+c-Kit+ sorted population (LSK), …

Pattinson** and Brian W. Forsyth* *Department of Pediatrics, Yale University School of Medicine, New Haven, CT **Maternal and Infant Health Care Strategies Research Unit, University of Pretoria, Pretoria, South Africa. Maternal and child care in South Africa is sub-optimal, contributing to the maternal mortality and high infant mortality rates in the country. About a third of these deaths are due to modifiable factors, some of which are related to poor communication between healthcare …

Microrna-21 Dysregulates The Expression Of Mef2c In Neurons In Monkey And Human Siv/Hiv Neurological Disease., Sowmya V. Yelamanchili, A Datta Chaudhuri, L N. Chen, Huangui Xiong, Howard S. Fox

Journal Articles: Pharmacology & Experimental Neuroscience

MicroRNAs (miRNAs) play important roles in regulating a plethora of physiological and pathophysiogical processes including neurodegeneration. In both HIV associated dementia in humans and its monkey model SIV encephalitis we find miR-21, a miRNA largely known for its link to oncogenesis, to be significantly upregulated in the brain. In situ hybridization of the diseased brain sections revealed induction of miR-21 in neurons. MiR-21 can be induced in neurons by prolonged N-methyl-D-aspartic acid receptor stimulation, an excitotoxic process active in HIV and other neurodegenerative diseases. Introduction of miR-21 into human neurons leads to pathological functional defects. Furthermore, we show that miR-21 …

An Evaluation Of Pmtct And Follow-Up Infant Hiv Testing In Kwazulu-Natal, South Africa, Juliana Chen Sep 2010

An Evaluation Of Pmtct And Follow-Up Infant Hiv Testing In Kwazulu-Natal, South Africa, Juliana Chen

Yale Medicine Thesis Digital Library

ABSTRACT AN EVALUATION OF PMTCT AND FOLLOW-UP INFANT HIV TESTING IN KWAZULU- NATAL, SOUTH AFRICA. Juliana Chen. Edendale Hospital, Pietermaritzburg, KwaZulu-Natal, South Africa. (Sponsored by Brian Forsyth, Department of Pediatrics, Yale University School of Medicine). Background: The primary cause of pediatric HIV infection is mother-to-child transmission, which can be reduced significantly by interventions that include highly active antiretroviral therapy (HAART) and antiretroviral (ARV) prophylaxis. Prevention of mother-to-child transmission (PMTCT) programs serve as critical entry points for the provision of HIV testing and treatment, though often remain poorly implemented and underutilized. Such programs are especially critical in resource-limited settings that have …

Perspectives On Intern Well-Being: The Importance Of Education, Support, And Professional Satisfaction, Heather Speller Sep 2010

Perspectives On Intern Well-Being: The Importance Of Education, Support, And Professional Satisfaction, Heather Speller

Yale Medicine Thesis Digital Library

The purpose of this qualitative study was to explore intern's perspectives on how the professional environment impacts their well-being. In-depth, semi-structured interviews were conducted in March and April of 2009 with seventeen interns from residency programs in a variety of specialties at an urban teaching hospital. Investigators coded interview transcripts line-by-line, and identified recurrent themes through an iterative process of analyzing tagged quotations. Three themes (each with three sub-themes) characterized aspects of the professional environment that interns perceived as impacting their well-being: 1) high-quality education (workload, work hours, and quality and quantity of teaching), 2) professional development and satisfaction (making …

Drug Off-Target Effects Predicted Using Structural Analysis In The Context Of A Metabolic Network Model, Roger L. Chang, Lei Xie, Philip E. Bourne, Bernhard O. Palsson Sep 2010

Drug Off-Target Effects Predicted Using Structural Analysis In The Context Of A Metabolic Network Model, Roger L. Chang, Lei Xie, Philip E. Bourne, Bernhard O. Palsson

Publications and Research

Recent advances in structural bioinformatics have enabled the prediction of protein-drug off-targets based on their ligand binding sites. Concurrent developments in systems biology allow for prediction of the functional effects of system perturbations using large-scale network models. Integration of these two capabilities provides a framework for evaluating metabolic drug response phenotypes in silico. This combined approach was applied to investigate the hypertensive side effect of the cholesteryl ester transfer protein inhibitor torcetrapib in the context of human renal function. A metabolic kidney model was generated in which to simulate drug treatment. Causal drug off-targets were predicted that have previously been …
