Inference For Time To Event And Sojourn Time Data Under Right Censoring Using Reweighting Approaches., Jie Fan Aug 2010

Inference For Time To Event And Sojourn Time Data Under Right Censoring Using Reweighting Approaches., Jie Fan

Electronic Theses and Dissertations

In this dissertation research, we aim to solve problems of two types of survival data, clustered survival data with potentially informative cluster size and sojourn time data. The methods for these two types of data are different. However, both data have right censored observations, and we use reweighting approaches to deal with the censoring issue. In the first part of the dissertation research, we consider marginal AFT models for correlated survival data with potentially informative cluster size. Informative cluster size means that the size of the correlated groups may be predictive of their survival characteristics. Two competing proposals, cluster-weighted AFT …

Nitrogen Mustard Reloaded: Synthesis And Structural Studies Of Dna Interstrand Crosslinks, Angelo Guainazzi

Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)

Many of the drugs used in cancer chemotherapy target DNA to kill malignant cells. Some of them form DNA interstrand crosslinks (ICLs), which are extremely cytotoxic lesions that block essential metabolic process such as replication, transcription and recombination by forming covalent bonds between opposite strands of DNA. Despite the importance of chemotherapeutic agents that rely on ICLs for their efficacy, the mechanisms by which these lesions are repaired remains poorly understood. A major impediment in studying ICLs repair has been the limited availability of well-defined substrates. This dissertation describes the development of a new strategy for the synthesis of defined …

Modeling And Its Numerical Method For Micro-Scaled Diffusion -Reaction Systems In The Neuromuscular Junction, Abdul Khaliq

Doctoral Dissertations

The main subject of this research is the neuromuscular junction (NMJ). The NMJ is a biological structure composed of the interface between a neuron and a muscle cell. Currently, there is not a fully three dimensional model of diffusion-reaction processes occurring in the NMJ. Developing a useful predictive model of this structure will assist in the therapeutic efforts to restore and rehabilitate NMJ function to humans and in developing strategies to prevent damage to the NMJ. This research work developed 1D mass transport and full 3D reaction diffusion models. A new finite difference scheme is presented for solving 1D mass …

The Role Of Bcl-2 Modifying Factor (Bmf) In Airway Epithelial Cell Death, Amelia H. T. Unione Jul 2010

The Role Of Bcl-2 Modifying Factor (Bmf) In Airway Epithelial Cell Death, Amelia H. T. Unione

Biomedical Sciences ETDs

One of the components that obstruct the airway in asthma is the sudden and increased secretion of mucus from metaplastic mucous cells in small airways. Because previous studies have shown that the resolution of mucous cell metaplasia (MCM) during prolonged exposure of mice to allergen is mediated by IFN gamma and the Bcl-2 family of proteins, we investigated the regulation and the role of a pro-apoptotic BH3 domain-only protein, Bmf, in airway epithelial cells (AECs) treated with IFN gamma. We hypothesize that Bmf is crucial for IFN gamma-induced cell death. Our findings show that IFN gamma suppressed Bmf expression in …

Open Access Dissertations

Fish populations are regulated by both external environmental factors, e.g., water quality parameters and habitat, and internal reproductive biology and physiology processes. For many species and populations there is often ample external information, while critical internal, i.e., life-history trait (LHT), information is not available. For this study, I determined LHTs and energy allocation patterns for Epinephelus guttatus (red hind), E. striatus (Nassau grouper), and Mycteroperca venenosa (yellowfin grouper) harvested from The Bahamas. I determined age ranges, and how growth patterns and rates differed among the study species. The maximum ages were: 17, E. guttatus; 22, E. striatus; and 13, M. …

Development And Study Of Inhibitors Of Heat Shock Protein 70 Induction, Rongsheng Wang May 2010

Development And Study Of Inhibitors Of Heat Shock Protein 70 Induction, Rongsheng Wang

All Theses and Dissertations (ETDs)

Tumor and cancer cells that over express heat shock protein 70: HSP70) are found to be multidrug resistant and thermo tolerant, creating a hurdle to existing therapy. Although HSP70 is recognized as an increasingly important drug target, the protein structure of this highly conserved chaperone remains challenging for direct targeting. An alternative strategy is to inhibit the transcription of HSP70. Among known small molecule inhibitors of HSP70 induction, quercetin has very low toxicity and has the advantage of being easily modified for structure-activity studies. One part of the dissertation focuses on the identification of quercetin derivatives with improved specificity and …

Cell Wall/Surface Proteome Of Candida Albicans: Its Application In Rapid Identification Of Yeast Species By Mass Signature And Characterization By In Vitro And In Vivo Chemical Labelings, Jiang Qian

LSU New Orleans Theses and Dissertations

Candida albicans is an opportunistic fungal pathogen that may cause mucutaneous infection and/or disseminated candidasis if the host defense system is impaired (such as those in HIV patients). Cell surface of C. albicans is the frontier where initial interplay between host-pathogen takes place and therefore is of great importance in understanding the mechanism of hostpathogen interaction. MALDI-TOF-MS analysis of intact fungal cells yielded mass signatures for rapid species differentiation, strain grouping and yeast morphogenesis monitoring. Cell surface biotinylations at low temperature (4°C), enzymatic digestion of the intact fungal cell surface proteins ("whole cell shaving"), biotin-avidin affinity enrichment of biotinylated peptides, …

The development towards analytical methods for the quantification of the active …

Structural Equation Modeling With Time Series Data, Tianyi Zhang May 2010

Structural Equation Modeling With Time Series Data, Tianyi Zhang

Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)

Structural Equation Modeling (SEM), also commonly referred to as path analysis in the absence of latent variables, is a powerful multivariate analysis approach to explore and to confirm causal relationships. It imposes a structure on the covariance matrix and the imposed structure is subsequently validated by the data. In recent years, SEM has been extended to analyze autoregressive moving average (ARMA) time series data assuming time-constant path coefficients. The mechanism of ARMA-based SEM makes it the ideal procedure for the analysis of directional brain functional pathways based on the multi-subject, multivariate time series data generated through the functional magnetic resonance …

Peripheral Taste System In Inbred Mouse Strains, William Wes Shelton

Theses and Dissertations (ETD)

Taster and non-taster mice strains have been used extensively in animal taste research. Inbred mouse strains C57 and FVB have been proposed to be tasters, where as strains 129 and Balb/C are classified as non-tasters. Therefore, evaluating the peripheral taste system in these mice is of great importance to determine if there are morphological differences that might explain the physiological classification. The purpose of the present study was to determine if inbred mice that have been shown to be tasters have a larger number of fungiform papillae, have larger fungiform papillae and have larger taste buds. My work in this …

Energy Restriction During Development In Breeding Gilts: An Economic Analysis, Justin Cech May 2010

Energy Restriction During Development In Breeding Gilts: An Economic Analysis, Justin Cech

Department of Agricultural Economics: Dissertations, Theses, and Student Research

Swine production has become a low-margin business. As costs of production have increased, producers are continuing to increase efficiency in market pig production and gilt development. Restricting energy intake during gilt development could have a positive impact on a producer’s bottom line, but few studies have economically analyzed production differences caused by energy restriction. This study utilized gilt development and market pig production data from biological studies that included a 2x2 factorial arrangement of half-sibling maternal lines (LWxLR and L45X) entering two gilt development programs. In one program, gilts were fed on an ad libitum basis. In the other, gilts …

Ampa Receptor Allosterism: Measurement Of The Conformational Changes In The Ligand Binding Domain Of A Functional Receptor, Jennifer Gonzalez-Mcgehee May 2010

Ampa Receptor Allosterism: Measurement Of The Conformational Changes In The Ligand Binding Domain Of A Functional Receptor, Jennifer Gonzalez-Mcgehee

Dissertations and Theses (Open Access)

Ionotropic glutamate receptors are important excitatory neurotransmitter receptors in the mammalian central nervous system that have been implicated in a number of neuropathologies such as epilepsy, ischemia, and amyotrophic lateral sclerosis. Glutamate binding to an extracellular ligand binding domain initiates a series of structural changes that leads to the formation of a cation selective transmembrane channel, which consequently closes due to desensitization of the receptor. The crystal structures of the AMPA subtype of the glutamate receptor have been particularly useful in providing initial insight into the conformational changes in the ligand binding domain; however, these structures are limited by crystallographic …

The carbohydrate portions of natural products play a crucial role in their pharmacological properties. These properties can vary from improved water solubility, improved membrane transport properties, improved tissue targeting, as well as improved target binding. Herein we describe our efforts to use de novo synthesis to help elucidate the various roles carbohydrates play in biologically active glycosylated natural products.;The O'Doherty group has developed a method that uses asymmetric synthesis for the de novo synthesis of carbohydrates. In contrast to traditional carbohydrate methods, this de novo approach builds the desired functionality and stereochemistry within each sugar. This methodology relies on a …

The Role Of The Tumor Suppressor Kinase Lkb1 In Xenobiotic Metabolism, Rebecca Ann Kohnz May 2010

The Role Of The Tumor Suppressor Kinase Lkb1 In Xenobiotic Metabolism, Rebecca Ann Kohnz

Stony Brook Theses and Dissertations Collection, 2006-2020 (closed to submissions)

The liver kinase-B1 (LKB1) is a serine/threonine kinase that has numerous roles in human disease. LKB1 was originally identified as a tumor suppressor kinase that, when inactivated, is responsible for the human inherited disorder Peutz-Jeghers syndrome which is characterized by benign gastric polyposis and increased cancer predisposition. More recently, inactivating LKB1 somatic mutations in humans have been found in 15-35% of non-small cell lung carcinomas and in 20% of cervical carcinomas. LKB1 functions as a master kinase upstream of 14 different substrates kinases and these substrates have critical roles in cell growth, polarity, and metabolism. Previous studies show that in …

Extracellular Matrix Proteins: Implications For Angiogenesis, Kent Edward Williams May 2010

Extracellular Matrix Proteins: Implications For Angiogenesis, Kent Edward Williams

All-Inclusive List of Electronic Theses and Dissertations

The extracellular matrix (ECM) is an essential requirement for maintaining permanent shape and rigidity in multicellular organisms. The ECM serves two main functions: scaffolding and signaling. Insoluble collagen and soluble proteoglycans, glycosaminoglycans (GAGs), and glycoproteins allow for water retention and flexibility. The signaling role of the ECM is essential for a multitude of events including vascular development and angiogenesis. Via interactions with vascular endothelial cells, proteins of the ECM can induce or repress angiogenesis.
