BACKGROUND: The p53 protein family coordinates stress responses of cells and organisms. Alternative promoter usage and/or splicing of p53 mRNA gives rise to at least nine mammalian p53 proteins with distinct N- and C-termini which are differentially expressed in normal and malignant cells. The human N-terminal p53 variants contain either the full-length (FL), or a truncated (ΔN/Δ40) or no transactivation domain (Δ133) altogether. The functional consequences of coexpression of the different p53 isoforms are poorly defined. Here we investigated functional aspects of the zebrafish ΔNp53 ortholog in the context of FLp53 and the zebrafish Δ133p53 ortholog (Δ113p53) coexpressed in the …

A Novel Method For Evaluating The Release Of Fermentable Sugars From Cellulosic Biomass, Rafael Jiménez-Flores, Gina Fake, Jennifer Carroll, Elizabeth Hood, John Howard

Efficient hydrolysis of cellulosic biomass into free sugars for the production of bioethanol would allow utilization of an abundant and renewable natural resource. However, complex microcrystalline cellulose in biomass resists digestion. A preferred method to deconstruct cellulose is using enzyme cocktails, but the presence of multiple enzymes with synergistic outcomes for this process complicates direct biochemical analysis as a reliable indicator of deconstruction. Methods to detect release of free sugars rather than direct measurement of enzymatic activity are available but are time-consuming, and do not reflect the production of metabolic inhibitors of microbial growth which may interfere with the subsequent …

D-Aspartate (D-Asp) is an endogenous compound found in the central nervous system (CNS) of a variety of organisms. Despite its prevalence, however, relatively little understood of its physiological role. The prevailing theory is that D-Asp is an alternate agonist of N-methyl-D-aspartate receptor (NMDAR) channels. The goal of this work was to characterize the currents activated by D-Asp in neurons Aplysia californica, focusing on cells of the buccal S cluster (BSC). First, a general electrophysiological characterization was carried out, examining ion permeability, agonist dose-response, and the kinetics of activation, inactivation, and desensitization. Mechanism Of Allosteric Modulation Of The Cys-Loop Receptors, Yongchang Chang, Yao Huang, Paul Whiteaker
Translational Neuroscience
The cys-loop receptor family is a major family of neurotransmitter-operated ion channels. They play important roles in fast synaptic transmission, controlling neuronal excitability, and brain function. These receptors are allosteric proteins, in that binding of a neurotransmitter to its binding site remotely controls the channel function. The cys-loop receptors also are subject to allosteric modulation by many pharmaceutical agents and endogenous modulators. By binding to a site of the receptor distinct from the neurotransmitter binding site, allosteric modulators alter the response of the receptors to their agonists. The mechanism of allosteric modulation is traditionally believed to be that allosteric modulators …

Information Visualization is an area of research concerned with the presentation of abstract data in a visual format. It has many diverse applications in fields of software engineering, information sciences, biology, chemistry, and medical, among others. This thesis is primarily concerned with the application of information visualization to the display of directed graphs and to the display of multivariate data for analysis. Two novel applications will be presented that are both advancements in the field of information visualization. Even though some strains are pathogenic, most E. coli strains still show evidence of being one of God’s “very good” creations.

Escherichia coli is frequently in the news (Fig. 1).
