The effect of the plant-derived nonpsychotropic cannabinoid, cannabidiol (CBD), on the function of hydroxytryptamine (5-HT)3A receptors expressed in Xenopus laevis oocytes was investigated using two-electrode voltage-clamp techniques. CBD reversibly inhibited 5-HT (1 μM)-evoked currents in a concentration-dependent manner (IC50 = 0.6 μM). CBD (1 μM) did not alter specific binding of the 5-HT3A antagonist [3H]3-(5-methyl-1H-imidazol-4-yl)-1-(1-methylindol-3-yl)propan-1-one (GR65630), in oocytes expressing 5-HT3A receptors. In the presence of 1 μM CBD, the maximal 5-HT-induced currents were also inhibited. The EC50 values were 1.2 and 1.4 μM, in the absence and presence of CBD, indicating that CBD acts as a noncompetitive antagonist of 5-HT3 …

Metolachlor is one of the most commonly used herbicides in the United States of America. Numerous studies have shown that exposure to this herbicide results in decreased cell proliferation, growth and reproductive ability in different organisms. However, pervious studies used high concentrations of metolachlor to see molecular effects. In the study presented here, we investigated the effects of metolachlor on human cells. Previous cell proliferation analysis from our laboratory has shown that metolachlor is capable of significantly inhibiting the growth of normal human fibroblasts and HepG2 cells at 1.6 parts per billion (ppb) and 50 ppb. John Berryman's The Dream Songs encompasses an extensive and complex negation of characterization and structure, offering a method of interpretation that focuses on the negations and the absences expressed in and presented by the song cycle. Through his nonlinear account of his life and through the historical and literary allusions he uses to express himself, Henry obscures any effort for readers to discover a stable, absolute identity. Past studies on aggression have typically focused on physical aggression (Crick & Grotpeter, 1995). Recently, however, researchers have started to look at a subtle form of aggression known as relational aggression (Crick & Grotpeter, 1995). Mutations affecting the mitochondrial electron transport chain cause numerous neurodegenerative disorders in humans and affect longevity in other organisms. A natural model system to study the relationship between mitochondrial function and aging within an evolutionary or population genetic context has been lacking. Natural populations of Caenorhabditis briggsae nematodes were recently found to harbor mitochondrial genetic variation with likely functional consequences for aging. Specifically, C. briggsae isolates containing high frequencies of a deletion mutation affecting the mitochondrial NADH dehydrogenase 5 (ND5) gene were found to have reduced reproductive fitness and lifespan and elevated levels of mutagenic superoxide. This thesis explores the obstacles in establishing a consistent and effective response framework for humanitarian catastrophe and the importance of maintaining a sustained dialogue to this end. It does so by recognizing the underlying conflict between two positions: the norm of non-intervention of states into the affairs of others, and the protection of individuals caught in the middle of violent conflict. The importance of working towards a resolution of this conflict is illustrated through the case study of Darfur, where a divided international community led an insufficient response to the crisis that can ultimately be judged as a failure. Diisocyanates are very reactive low molecular weight chemicals that are widely used in the manufacture of polyurethane products. Diisocyanate exposure is one of the most commonly reported causes of occupational asthma. Although diisocyanates have been identified as causative agents of respiratory diseases, the specific mechanisms by which these diseases occur remain largely unknown.

Tandem mass spectrometry was used to unambiguously identify the binding site of isocyanates within four model peptides (Leu-enkephalin (Leu-enk, YGGFL), Angiotensin I (DRVYIHPFHL), Substance P-amide (RPKPQQFFGLM-NH2), and Fibronectin-adhesion promoting peptide (FAPP, WQPPRARI)). Trace gases can have a significant impact on the Earth's climate, and the analysis of changes in these gases and an understanding of how much of these changes are a result of human activity is important for understanding global climate change. Methane (CH4) is the second only to CO2 in radiative forcing over the last 200 years, and its concentration in the atmosphere has more than doubled since 1750. Two new cancer therapies apply bioelectric principles. These methods target tumor structures locally and function by applying millisecond electric fields to deliver plasmid DNA encoding cytokines using electrogene transfer (EGT) or by applying rapid rise-time nanosecond pulsed electric fields (nsPEFs). EGT has been used to locally deliver cytokines such as IL-12 to activate an immune response, resulting in bystander effects. NsPEFs locally induce apoptosis-like effects and affect vascular networks, both promoting tumor demise and restoration of normal vascular homeostasis. EGT with IL-12 is in melanoma clinical trials and nsPEFs are used in models with B16F10 melanoma in vitro and in …

Au cours des dernières années, plusieurs auteurs ont remis en cause le niveau de protection accordé aux travailleurs par la valeur seuil reconnue pour le béryllium (Be) et ses sels (VEMP Québec, TLV (TWA) ACGIH). La prévalence des effets associés au Be suggère que les risques sont en fonction de sa spéciation. Par ailleurs, il semble que les particules fines constituent la fraction d’intérêt pour l’occurrence de tels effets. Although contemporary practitioners have rejected the confrontational style and perceived aggressiveness of earlier family therapists in favor of a more "collaborative" stance, confrontation, as a technique, is still widely used in almost all forms of psychotherapy, including family therapy. The present process study explored what makes confrontation more or less effective in motivating clients to recognize and reevaluate counterproductive ways of interacting. Vestibular schwannomas (VS) arising sporadically or in patients with neurofibromatosis type 2 (NF2) consistently lack expression of Merlin, a tumor suppressor. Conventional treatment options include surgery and radiotherapy but there is no validated medical option. Recent evidence suggests that Merlin deficiency may result in abnormal activation of receptor tyrosine kinases (RTKs) and downstream signaling, promoting tumor growth. Although small-molecule RTK inhibitors are widely available for clinical use, no such therapy has been validated in patients with VS. As human activities continue to generate accelerating levels of carbon dioxide emissions, the world’s oceanic resources are threatened by variability in seawater chemistry, known as ocean acidification. Recent increases in atmospheric carbon dioxide have resulted in decreased carbonate ion concentrations and ocean pH levels, leading to increasingly acidic waters. The exact consequences of these chemical changes on ecosystems and individual species are difficult to predict; however, research has shown that economically valuable calcifying species will experience reduced reproductive fitness and population declines. This thesis presents a new methodology based on directly measuring user behavior and making decisions based on experimental results. I have built and tested a tool which will enable researchers to use the methodology to determine whether particular financial statement presentations are more beneficial than others. The purpose of this study was to examine the efficacy of continuous resistance training (3 days/wk) compared to interrupted resistance training where 20-24 h separated an exercise bout (i.e. 6 days/wk) for enhancing bone mineral density (BMD) in growing male rats. The total volume of work performed per week between the two resistance training programs was equivalent by design. Young male rats were randomly divided into Control (Con, n = 9), 3 days/wk resistance trained group (RT3, n = 9), and 6 days/wk resistance trained group (RT6, n = 9). Renewable energy credits ("RECs")--commodities representing a megawatt-hour of renewable electricity but tradable separately from the electricity itself-developed to encourage renewable energy investment and to allow individuals and corporations without direct access to renewable energy to subsidize its construction. RECs can be sold voluntarily or applied to state-imposed renewable energy purchase obligations. These state mandates, known as renewable portfolio standards, have contributed dramatically to the demand for RECs. Sickle cell disease (SCD) is a pleiotropic genetic disorder of hemoglobin that has profound multiorgan effects. The low prevalence of SCD ( approximately 100,000/US) has limited progress in clinical, basic, and translational research. Lack of a large, readily accessible population for clinical studies has contributed to the absence of standard definitions and diagnostic criteria for the numerous complications of SCD and inadequate understanding of SCD pathophysiology. In recent years, elk have begun recolonizing areas east of the Rocky Mountains that are largely agroforested ecosystems composed of privately owned land where management of elk is an increasing concern due to crop and forage depredation and interspecific disease transmission. We used a Geographic Information System, elk use locations (n = 5013), random locations (n = 25,065), discrete-choice models, and information-theoretic methods to test hypotheses about elk resource selection in an agro-forested landscape located in the Pine Ridge region of northwestern Nebraska, USA.
