Integrins are important cell surface receptors that transmit bidirectional signals across the membrane. It has been shown that a conformational change of the integrin β-subunit headpiece (i.e. the β I domain and the hybrid domain) plays a critical role in regulating integrin ligand binding affinity and function. Previous studies have used coarse methods (a glycan wedge, mutations in transmembrane contacts) to force the β-subunit into either the open or closed conformation. Here, we demonstrate a detailed understanding of this conformational change by applying computational design techniques to select five amino acid side chains that play an important role in the …

Induction Of Microrna-221 By Platelet-Derived Growth Factor Signaling Is Critical For Modulation Of Vascular Smooth Muscle Phenotype, Brandi N. Davis, Aaron C. Hilyard, Peter H. Nguyen, Giorgio Lagna, Akiko Hata
Biology
The platelet-derived growth factor (PDGF) signaling pathway is a critical regulator of animal development and homeostasis. Activation of the PDGF pathway leads to neointimal proliferative responses to artery injury; it promotes a switch of vascular smooth muscle cells (vSMC) to a less contractile phenotype by inhibiting the SMC-specific gene expression and increasing the rate of proliferation and migration. The molecular mechanism for these pleiotropic effects of PDGFs has not been fully described. Here, we identify the microRNA-221 (miR-221), a small noncoding RNA, as a modulator of the phenotypic change of vSMCs in response to PDGF signaling. We demonstrate that miR-221 …

Primer pheromones play key roles in regulating division of labor, which is one of the most fundamental and defining aspects of insect sociality. Primer pheromones are chemical messengers that transmit hormone-like messages among colony members; in recipients these messages can either induce or suppress phenotypic caste differentiation. Here, we investigated soldier-caste-derived chemicals as possible primer pheromones in the lower termite Reticulitermes flavipes, a species for which no primer pheromones have yet been identified. We determined that soldier head extracts (SHE), when provided to totipotent workers along with the insect morphogenetic juvenile hormone (JH), significantly enhanced soldier caste differentiation. The Igniting Streams of Learning in Science (ISLS) academy engages 11th and 12th grade students and their teachers in a year long learning community (LC) experience. ISLS transforms attitudes about science in school curricula, local communities, college classrooms, and government agencies. While the goals of ISLS are ambitious: (1) to increase student interest in teaching high school science; (2) to increase the number of students pursing Science, Technology, Engineering, and Math (STEM) majors in college, and (3) to connect students to their local environments using methods of scientific investigation, the challenges for STEM education have never been greater. Background: Single nucleotide polymorphisms (SNPs) in the dhfr and dhps genes are associated with sulphadoxine-pyrimethamine (SP) treatment failure and gametocyte carriage. This may result in enhanced transmission of mutant malaria parasites, as previously shown for chloroquine resistant parasites. In the present study, we determine the association between parasite mutations, submicroscopic P. falciparum gametocytemia and malaria transmission to mosquitoes. Methodology/Principal Findings: Samples from children treated with SP alone or in combination with artesunate (AS) or amodiaquine were genotyped for SNPs in the dhfr and dhps genes. Gametocytemia was determined by microscopy and Pfs25 RNA-based quantitative nucleic acid sequence-based amplification (Pfs25 QT-NASBA). …
