Background Successful reprogramming of a somatic genome to produce a healthy clone by somatic cells nuclear transfer (SCNT) is a rare event and the mechanisms involved in this process are poorly defined. When serial or successive rounds of cloning are performed, blastocyst and full term development rates decline even further with the increasing rounds of cloning. Identifying the "cumulative errors" could reveal the epigenetic reprogramming blocks in animal cloning. Results Bovine clones from up to four generations of successive cloning were produced by chromatin transfer (CT). Critical to understanding the widespread symbolism and imagery pointing to Jesus Christ in the New Testament is an exegetical grasp of the content--that is, an understanding of the historical, literary, and theological context of the language. The image of water recurs frequently throughout the New Testament Gospels as a symbol of the Savior's purity, cleansing power, true doctrine, and so forth. Similarly, blood is used often to reflect the sacred mission of Christ and the price of our salvation. Urban environments often support depauperate insect faunas, although they can serve as suitable habitats for some taxa. The potential value of urban wetlands as habitat for regional dragonfly populations has not been well studied. Landscape patterns of natal habitat use by lentic dragonflies were studied at small wetlands in Rhode Island, U.S.A. during three field seasons (2004, 2005, 2006). Dragonfly exuviae were collected along defined perimeter paths on six (2004) or five (2005, 2006) site visits per season (May- October). The exuviae were identified in the laboratory, and the number collected per species/hour was tallied for each field season. Three …

Iron-sulfur proteins are ubiquitous in biological processes. Here we report that NO, a physiological free radical, can effectively inhibit cell growth of Escherichia coli in minimal medium under anaerobic growth conditions. Fractionation of the cell extracts obtained from NO-exposed cells shows a broad distribution of the protein-bound dinitrosyl-iron complexes (DNICs) formed by NO. On the other hand, the cell growth of E. coli can be restored when NO-exposed cells are either supplemented with the branched-chain amino acids (BCAAs) anaerobically or returned to aerobic growth conditions. It turns out that dihydroxyacid dehydratase (IlvD), an iron-sulfur enzyme essential for the BCAAs biosynthesis, …

Chemical manufactures are often associated with a negative publicity due to chemical spills that can cause human health problems and environmental pollution. Innovative methods such as phytoremediation in lieu of traditional remediation methods are being researched to determine environmentally friendly options for remediation. Phytoremediation was studied as an alternative remediation method for removal of chemicals in a contaminated groundwater plume in Louisiana. The main constituents of concern in the plume were cumene and 4-cumylphenol. Current regulatory testing provides the basis for determining acceptable levels of pollutants in the environment, yet these acceptable levels of contaminants have resulted in undesirable consequences to organisms. The purpose of this dissertation was to test the hypothesis that biomarkers of cellular stress could be detected from sub-lethal exposure to pesticides in sperm and early life stages of broadcast spawning invertebrates. Exposures were conducted on oyster (Crassostrea virginica), mussel (Mytilus galloprovincialis and Dreissena polymorpha), and sea urchin (Lytechinus variegatus) embryos and larvae for 4 and 24 h to Bayluscide® and Roundup®. The Louisiana dairy production region (LDPR) is located in southeast Louisiana and includes five parishes (Tangipahoa, Washington, Livingston, St. Helena, and St. Tammany). It is home to approximately 90% of the state’s dairy industry and is a major contributor to nonpoint source pollution (NPS) in the area’s watershed through nitrogen, phosphorus, and sediment effluents. While point source (PS) effluents are easily identified and subject to federal and state regulations, NPS mitigation efforts must address uncertainties such as location of the NPS as well as stochastic parameters such as rainfall and its effect on nutrient and sediment flow. A comparison of equilibrium and non-equilibrium dissolution of tetrachloroethylene (PCE) was conducted to ascertain how PCE saturation, individual blob properties (volume, surface area, sphericity), and PCE occupied pores are affected by two distinct dissolution regimes. One-dimensional columns were imaged at various dissolution stages using high resolution (~10 um) synchrotron x-ray tomography (XT) and image subvolumes were analyzed using a series of grain, pore network structure, and blob analysis algorithms. An analysis of algorithm-generated data was conducted to determine grain and pore statistics, PCE saturation and individual blob properties, and correlations between PCE blobs and pore network structure. Since the cloning of Dolly there has been little change in the efficiency of nuclear transfer (NT). Research is beginning to investigate the characteristics of donor cells. Adiposetissue is an abundant source of adult-derived cells that have displayed “stemness” in-vitro(Gimble et al., 2003). The overall goal of this research was to define the in-vitro characteristicsof bovine adipose-derived adult stem cells (ADAS) for the use in NT. The red-cockaded woodpecker (Picoides borealis; hereafter RCW) is a cooperative breeder endemic to open old-growth pine savannah ecosystems in the southeastern United States (Jackson 1994). The RCW was listed as federally endangered in 1973, after a population decline due to habitat loss. Habitat fragmentation produces isolated populations of RCWs, which in turn ultimately limits the success of the species. RCW biologists and managers counteract effects of fragmentation by aggregating recruitment clusters and translocation. The goal of this study was to systematically examine group differences among those with intellectual disabilities (ID) and comorbid Autism Spectrum Disorders (ASD) and epilepsy in an adult population through a detailed exploration of the characteristics that these disorders present in the areas of psychopathology, behavior problems, and impaired social behavior. Previous studies indicating that individuals with ID have comorbid ASD and neurological conditions tend to stop short of addressing these disorders’ impact on the full range of psychosocial issues, particularly in adult samples. Equine Herpesvirus-1 (EHV-1) is an important ubiquitous enzootic equine pathogen, causing significant economic losses to the horse industry. Despite extensive vaccination protocols, EHV-1 continues to be a major cause of epidemic abortion, perinatal mortality, respiratory disease and neurologic disease. EHV-1 infections are usually dealt with by using management practices that limit spread of the disease and secondary complications, providing symptomatic relief to infected horses, but no specific treatment is available. The aim of this paper is to compare and contrast the philosophies of Martin Heidegger and John Dewey regarding metaphysics and science. Concerning metaphysics, there are two main areas of attention, the historical emergence of metaphysics in the philosophical tradition, and the abandonment of the metaphysical notions of substance and subjectivity. After examining these two areas of both thinkers’ philosophies, we then turn to their philosophy of science, demonstrating how the scientific thought of both thinkers is inseparable from their metaphysical critiques. This work was done in order to deepen our molecular level understanding of how soil organic matter (SOM) is assembled in a whole soil and to provide further insight on the effect of SOM assembly on the uptake and release of hydrophobic organic compounds. Various techniques, including ultraviolet absorbance, fluorescence, and total carbon analysis, demonstrate that hydration/solvation of SOM is kinetically controlled. Initial wetting of a soil releases the hydrophobic moieties that are located at the outer surface of SOM, and longer wetting times exfoliate more hydrophobic quinone type moieties that are present in the middle layer, which in turn …

Processing biomass in near- and supercritical water has garnered increasing attention because of its ability to accept a variety of wet feedstocks, energy efficiency, and ability to regulate the solubility and separation of components. Very few studies of biomass hydrothermal conversion provide a comprehensive evaluation of multiple process parameters and various additives and their effects on a single product phase. This research examined the influence of temperature, residence time, biomass concentration, and particle size on volatiles production from the hydrothermal conversion of sugarcane bagasse. Supercritical fluid extraction (SFE) of oxeye daisy (Chrysanthemum leucanthemum L.) seed, heartwood of Port-Orford cedar (POC) (Chamaecyparis lawsoniana), Alaska yellow cedar (AYC) (Chamaecyparis nootkatensis), and Eastern red cedar (ERC) (Juniperus virginiana L), and Chinese tallow (Sapium sebiferum L. Roxb. ) seed was investigated in this study and compared with other extraction methods. For the oxeye daisy seed extraction, Soxhlet extraction (SE) with hexane, microwave assisted extraction (MAE), and supercritical fluid extraction (SFE) were conducted. Scanning probe microscopy (SPM) characterizations are becoming more prevalent for surface investigations due to their capabilities for obtaining structural information and physical measurements. New capabilities of SPM for studying and controlling nanoscale processes are emerging as valuable assets in research. A fundamental understanding of the interactions of surface reactions provides essential information for developing workable applications for nanotechnology. The development of photocatalytic reactor is essential for the successful application of heterogeneous semiconductor in environmental study, which has been shown to be photoactive and effective to oxidate organic pollutant and photoreduce CO2 to useful compounds. Recognition of point mutations in a codon 12 of the K-ras gene, most frequently observed, is considered to be useful in the early diagnosis of several types of the human cancers. We have developed a multifunction, disposable, microfluidic module which detects low-abundant point mutations in human genomic DNA in modular architecture. Each functional component including a microfluidic PCR reactor, a passive diffusional micromixer reactor, and a microfluidic LDR reactor was separately designed and fabricated. In western Amazonia, large numbers of frugivorous bats regularly visit natural forest clearings known locally as collpas (also called clay licks or mineral licks). Bats arrive at collpas to drink water that has accumulated in depressions created by larger mammals that consume soil. Although collpa visitation by bats appears relatively common in western Amazonia, little is known about its causes and its ecological implications. In this dissertation I describe general and seasonal patterns of collpa visitation by frugivorous bats in the Peruvian Amazon, and I investigate potential explanations for this unique behavior. Our genetic inheritance begins with one genome copy from each parent. The presence of one or more errors in either the maternal or the paternal genome can lead to genetic disease or disruption of the embryonic program and potential loss of the offspring. Chronic inflammation is involved in the adipose tissue dysfunction through regulation of endocrine and storage function of adipocytes. As a representative proinflammatory cytokine, TNF-α was reported to inhibit expression of adiponectin. However, the mechanism of inhibition remains to be identified. Here, we provide experimental evidence that TNF-α inhibits adiponectin at both transcriptional and posttranscriptional levels. Amyloid-beta (Aβ), a 39 to 43 amino acid long peptide, is the primary species identified in senile plaques associated with Alzheimer’s disease (AD) and has been implicated in the neurotoxicity associated with AD. It is believed that Aβ toxicity is mediated through the interaction with neuronal membranes. A variety of evidence indicates that 1) Aβ may bind to the cell surface sialic acids, 2) the affinity of this interaction is higher if the gangliosides or sialic acids on the cell surface are clustered, 3) the removal of the surface sialic acids attenuate Aβ toxicity. For over a century, nucleation for all systems was thought simplistically to be a process that advances through the formation of critical clusters with a well-defined composition. Our results show intriguing nucleation mechanisms that challenge the aforementioned notion. We employed the simple TraPPE-UA (transferable potential for phase equilibria – united atom) force field and the AVUS-HR approach (a combination of aggregation-volume-bias Monte Carlo, umbrella sampling, and histogram reweighting), to investigate the homogeneous vapor-to-liquid nucleation of various nucleating systems. The main objective of this work was to assess the effect of 10% (w/v) polyvinylpyrrolidone (PVP) on the pattern of intracellular ice formation (IIF) in human adipose tissue derived adult stem cells (ASCs) in the absence of serum and other cryoprotective agents (CPAs). Passage 1 (P1) ASCs were cultured, washed and suspended in either 1x PBS (Phosphate Buffered Saline) or 10% w/v solution of PVP in 1x PBS. Seasonal growth characteristics and shading ability of six sugarcane varieties were compared during the second production year. For each variety, trend analysis of data showed a significant linear trend. Differences in shoot emergence among the varieties were observed beginning in early April and as the season progressed, shoot production and shoot height increased for ‘L 97-128’, ‘Ho 95-988’, ‘L 99-226’, and ‘L 99-233’ but lagged for ‘HoCP 96-540’ and ‘LCP 85-384’. The marshland upwelling system (MUS) was designed to treat domestic wastewater from coastal dwellings where conventional methods are inadequate due to high water tables, poor hydraulic soil conditions, anaerobic soils, and saline groundwater. Currently there is no adequate treatment system available and coastal dwellings are contributing to water quality problems. This study focused on determining the treatment effectiveness of the MUS for organic matter and understanding the specific processes involved in nitrogen treatment.

A model plant, Arabidopsis thaliana, duplicates its chromosomes without cell division, in a process known as endoreplication. The primary objective of this study was to identify genes and proteins that specifically accumulate in endoreplicated nuclei in Arabidopsis thaliana. Analysis of nuclei sorted by fluorescent activated cell sorter (FACS) revealed that ploidy levels and nuclear volume were positively correlated. However, the protein amounts among 2C, 4C, and 8C nuclei had no significant differences. This indicated a decrease of the protein concentration in endoreplicated nuclei. In contrast, the RNA amount in the 8C nuclei was 1.3- and 1.4-fold higher than that in …
