We previously reported that Dot1a*AF9 complex represses transcription of the epithelial Na+ channel subunit α (α-ENaC) gene in mouse inner medullary collecting duct mIMCD3 cells and mouse kidney. Aldosterone relieves this repression by down-regulating the complex through various mechanisms. Whether these mechanisms are sufficient and conserved in human cells or can be applied to other aldosterone-regulated genes remains largely unknown. Here we demonstrate that human embryonic kidney 293T cells express the three ENaC subunits and all of the ENaC transcriptional regulators examined. These cells respond to aldosterone and display benzamil-sensitive Na+ currents, as measured by whole-cell patch clamping. We also …

Increasing urbanization of many regions of the world has resulted in the decline of suitable habitat for wild flora and fauna. Green roofs have been suggested as a potential avenue to provide patches of good-quality habitat in highly developed regions. In this study, we surveyed green roofs in Toronto, Ontario, Canada for bee diversity and abundance to determine their potential as quality habitats in an urban area for these important pollinators. By comparing various biodiversity measures between green roofs and ground-level sites, we show that green roofs provide habitat to many bee species. Gardens have become increasingly important places for growing nutritional food, for conserving biodiversity, for biological and ecological research and education, and for community gathering. Gardens can also be designed with the goal of attracting aesthetically pleasing wildlife and pollinators, like birds and butterflies, but other important garden visitors, like bees, can also be drawn to specially planned and modified gardens. A community garden in San Luis Obispo, California provided the setting for modification with the goal of attracting native bee pollinators by planting known bee-attractive plants. Women at the completion of treatment for breast cancer experience relief along with continued physical and psychological distress. This study explored the effectiveness of two forms of a Relaxation Intervention in providing psychosocial support to women at the completion of treatment for breast cancer. The two conditions included (1) Music-Assisted Relaxation (MAR), in which contemporary, sedative music was paired with standard, spoken relaxation directives, and (2) Relaxation Alone (RA), in which only spoken relaxation directives were used. The T cell function of a proliferation inducing ligand (APRIL or TNFSF13) remains unclear. By comparing APRIL-/- mice with wild type (WT) mice, we have investigated the roles of APRIL in Th1, Th2 and Th17 responses. With regard to APRIL in Th1 responses, cultured APRIL-/- CD4+ T cells showed increased IFN-gamma production under non-polarizing, but not under Th1 polarizing, conditions. No difference in antigen-specific IgG2a levels existed between APRIL-/- and WT mice immunized with ovalbumin (OVA) and complete Freund's adjuvant (CFA) which induces Th1 polarization. Our data indicate that APRIL represses Th1 responses only under non-polarizing conditions. As for APRIL …

How do neighboring neurons differ? How are they similar? How do they relate to each other? Retinal ganglion cells (RGCs) and amacrine cells are born during the same developmental window, from the same population of progenitor cells, and some amacrine cells even migrate to the same retinal layer where RGCs reside, the ganglion cell layer. Amacrine cells are presynaptic to RGCs, whose axons subsequently project through the optic nerve and carry visual information to the brain. Problem: An evaluation of a smoking cessation educational intervention for direct care RNs. Design: A randomized controlled trial. Methods: The study was conducted at a large Midwestern academic medical center. Direct care Registered nurses (dcRN’s) employed at least .7 FTE and spending 80% of their time in direct patient care on general medicine or general surgery units were recruited. Recruitment occurred between April and May, 2009. Outcome data was abstracted from charts of patients receiving care from the intervention group. The purpose of this research study was to determine if transformational learning is an outcome of the experiences of training for and completing a marathon. Transformational learning has gained visibility and greater acceptance in the learning community in recent years due in large part to the efforts of Jack Mezirow and associates. Metal ions like Ca2+, Mn2+, Tb3+, Eu3+ etc., can bind at the active site and have varying effects on the enzymatic activity. In a crystal structure with DNA, two Ca2+ ions were bound to the active site of each PvuII monomer, and the metals share the ligands. Ca2+ ions promote DNA binding and not cleavage in many nucleases. We explored the role of Mg2+ and Ca2+ binding at the metal binding sites of PvuII to elucidate the role of each metal site. We monitored the cleavage kinetics of PvuII in presence of mixed metals. Research has demonstrated that parental divorce and family functioning are associated with children’s socieomotional and psychological adjustment well into their adult years. Research has also demonstrated that sexual attitudes are becoming more liberal (cf., Harding & Jencks, 2003; Leiblum, Wiegel, & Brickle, 2003). The purpose of this research was to examine family functioning and parental divorce status in relation to attachment styles and sexual attitudes among college students (n = 387). The participants completed the Brief Sexual Attitudes Scale (BSAS), The Experiences in Close Relationships-Revised Scale (ECR-R), and Family Relationship Index (FRI). The ulu is an enduring object in the lives of Inuit women which has multiple meanings as both a tool and symbol of traditional subsistence activity. While it continues to be recognized as a symbol of identity for Inuit women across the Arctic, it has received little attention by Western scientists and academics. "I examined how headwater streams are ecologically linked with the terrestrial environment and upstream waters. I examined relationships between fish (rainbow and cutthroat trout), invertebrates, and habitat in 15 headwater streams in two ecoregions (wet, dry) and timber harvest scenarios (logged, unlogged) in the Wenatchee River sub-basin in the eastern Cascade Mountain Range, Washington state, USA. Fish biomass, density, and size were not related to ecoregion or to logging history. Invertebrate drift manipulations in 13 streams influenced fish movement (fish moved downstream in sites that were not supplemented with food) and diet (fish consumed less prey when drifting invertebrates were …

Mammalian predation, avian predation, female body condition and food availability on the breeding ground are likely the main factors influencing nesting success in tundra-nesting waterfowl. These driving factors are mediated by the primary life history characteristics; incubation behavior, female body size, nesting associations, and nest site selection. I created a conceptual model illustrating how these factors are interrelated and how they impact nest success through a variety of pathways to better understand the evolution of a species' nesting strategy and patterns observed in the field. "The Arctic fresh water hydrological cycle is dominated by the melting of the seasonal snow cover and scattered precipitation events during the summer months. Predicting and characterizing potential hydrological response is an important component for engineering infrastructure for the appropriate climatic conditions. A semi-distributed Swedish conceptual model, HBV, has been applied to the Imnavait basin, located in the headwaters of the Kuparuk River on the North Slope of Alaska, to examine runoff during spring and summer months. After more than 20 years of only mechanical brush cutting, ADOT&PF evaluated the use of herbicides to manage vegetation that interferes with line-of-sight and maintenance of the roadway. While researchers have investigated herbicide effectiveness and attenuation in more-temperate climates, little study has focused on cold regions. The purpose of this project was to measure the effectiveness and attenuation of two different selective auxin-type herbicides, 2, 4 dichlorophenoxyacetic acid (2,4-D), and 3,5,6-trichloro-2-pyridinyl acetic acid (triclopyr) in two subarctic climates; an extremely cold continental climate and a maritime climate. I noticed variations in such curricula while developing the University of Alaska Fairbanks course, Fundamentals of Fly Fishing. "The growth of Seward Peninsula sockeye salmon (Oncorhynchus nerka) from Salmon and Glacial lakes is related to their physical environment. Dermal scales collected over many years were measured to document the annual age specific growth of smolts and adults. The effect of fertilization on fry growth was examined using the first year of growth. The growth histories of Salmon Lake sockeye salmon were compared to Glacial Lake sockeye salmon through smolting and in the marine environment. "The goal of this study was to characterize Kittlitz's murrelet (Brachyramphus brevirostris) foraging habitat relative to prey availability and oceanography in Kenai Fjords National Park, a glacial-marine system. I conducted oceanographic, hydroacoustic, trawl, beach seine, and marine bird surveys monthly from June-August in 2007 and 2008. High sediment load from glacial river runoff shaped the marine ecosystem, and this appeared critically important to Kittlitz's murrelets at sea. Submerged moraines influenced inner fjord habitat that was characterized by cool, fresh, stratified, and silt-laden waters. This silty glacial runoff limited light availability to chlorophyll near tidewater glaciers, but zooplankton abundance was enhanced …

Salmon returning to Lake Clark, Alaska are a valuable subsistence, commercial and ecological resource, and are an important component of the larger Kvichak River escapement. Average escapement to the Kvichak River declined sharply during 1996-2005, prompting studies to investigate age and size at maturity, key life history traits of salmon linked to reproductive success and survival. We examined potential factors which may influence sockeye salmon Oncorhynchus nerka age and size at maturity: spawning habitat and ocean environment, and examined variation in both traits over time. Inconnu (Stenodus leucichthys) is present throughout much of the Yukon River drainage in Alaska, but only five spawning areas have been identified. Spawning habitat requirements are therefore thought to be very specific; however, the physical qualities of these habitats have only been characterized in general terms. The Sulukna River is one of five identified inconnu spawning areas within the Yukon River drainage. A systematic sampling design was used in September and October of 2007-2008 to define Sulukna River spawning locations. "Steller's eiders (Polysticta stelleri) and other sea duck species have undergone population declines in recent decades, and the causes for the declines remain largely unknown. As part of a study to investigate the role of disease in sea duck population declines, I further characterized Steller's eider humoral immunity by quantifying total serum immunoglobulin Y (IgY). Baseline values of total serum IgY were determined for a captive flock of Steller's eiders housed at the Alaska SeaLife Center using species-specific assays. "Increasing numbers of sea kayakers in Kenai Fjords National Park, Alaska prompted a study to evaluate human disturbance on harbor seals. Harbor seal behavior recorded during the molt from 2004-2006 via remotely controlled cameras and direct field observations were used to evaluate effects of human activities. Behaviors of the seals observed in the presence and absence of kayakers/walkers were contrasted by method of collection, year, presence of humans, presence of a guide, and guide training. Results demonstrated that harbor seals abandoned the ice and were more alert when kayakers were present than when humans were absent. Harbor seals became progressively …

Extent of larder hoarding differs among species of kangaroo rats, and limited information is available for food stored in burrows by Ord's kangaroo rat (Dipodomys ordii)—the most widespread species of Dipodomys. I excavated 9 burrows recently used by adult D. ordii during summer in the Sandhill Region of Nebraska. I observed only small quantities of food stored in burrows. Eight of 9 burrows contained segments of sand dropseed (Sporobolus cryptandrus) in limited quantities. These segments consisted of upper leaves with fruits (containing seeds) housed in sheaths. My observations represent the first documentation of D. ordii storing …
