Objectives: 1.) To develop and characterize an optimal formulation of oil-in-water nano-emulsions of trifluoperazine for parenteral delivery. 2.) To perform short term stability testing of the optimal formulation and monitor the potency using high performance chromatography (HPLC).

Materials and Methods: Emulsions containing soybean oil, water, trifluoperazine hcl as an amphiphilic drug, phospholipon 90 and Tween 80 as surfactant blend were prepared using the Thin-layer hydration method. Z-average, polydispersity index, zeta potential of emulsions were determined. A fully randomized 2X2X2X2X2 statistical design was developed using JMP software. This disturbance of mood impacts many facets of the individual's life, including the ability to participate in and benefit from educational experiences. School psychologists are challenged to find effective treatments for adolescents experiencing depression that can be delivered in a school setting. Interpersonal Psychotherapy for Adolescents (IPT-A; Mufson, Dorta, Moreau, & Weissman, 2004) has been found effective for adolescents in clinical settings as well as in urban, school-based clinics. Although the experience of anxiety among children and adolescents is well documented in the literature, relatively little empirical study has focused on this topic. Existing research is essentially void of studies into gender-specific aspects of anxiety development and maintenance or race-based differences in anxiety symptomatology. The current study sought to fill this gap by examining racial differences in anxiety disorder symptom structure and trajectories among pre-adolescent female youth. The disease malaria originates from the transmission of the parasite Plasmodium to humans by female anopheline mosquitoes. Estimates put the number of deaths at 1-3 million people annually and this number will increase without the establishment of new control strategies. There is currently no vaccine and the effectiveness of insecticides and drugs are thwarted by the gain of resistances for both the insect and parasite. An alternative genetic engineering approach to combating malaria is presented here. The Tumor Necrosis Factor (TNF) Receptor Associated Factor 6 (TRAF6) is an intracellular signal transducers, being responsible for mediating many of the activation events initiated by TNF receptor (TNFR) and Toll-like/Interleukin-1 and 18 receptor (TIR) families, in which TRAF6 plays central roles in numerous biological processes including innate and adaptive immunity, osteoclastogenesis and bone development, CD40 signaling, neuronal cell development, and cancer cell progression.

This study explored the potential of near-infrared spectroscopy in the determination of pharmaceutical pellet characteristics and to predict desired process endpoints during fluidized bed drug suspension layering and coating operations. Various strengths of diltiazem HCl pellets were prepared via a tangential-spray rotogranulation process and subsequently coated Eudragit RS30D in a Wurster column. In-line and at-line near-IR process monitoring methods were evaluated. Quantitative calibrations for potency, applied polymer solids and dissolution were developed using several different regression models. Both in-line and at-line determinations of pellet potency were effectively accomplished with average standard errors of prediction of 1.11% and 0.63%, respectively. Near-IR …

This thesis describes the design and synthesis of 1,4,6-trisubstituted benzimidazoles as CDK5 inhibitors and the progress toward the total synthesis of tubulysin D.

Tubulysin is a natural product with potential anti-cancer activity. The two gamma-amino acids in the Tuv and Tup fragments of tubulysin were synthesized. Evans' oxazolidinone chemistry was employed for the stereoselective synthesis of Cbz-beta-homovaline in Tuv fragment. A crystal structure of the Tup fragment was obtained.

Multiple sclerosis (MS) is an autoimmune disease of the central nervous system (CNS) that affects approximately 400,000 people in the United States every year and results in neurological deficits. Inflammatory events associated with MS include activation of auto-reactive T cells and migration of these and other immune cells into the CNS, leading to a coordinated attack upon oligodendrocytes and demyelination. Most current therapies only treat the symptoms of disease, not the cause, which is still in large part unknown. To address a controversy in the literature concerning whether monkeys show an aversion to inequity, individuals of a New World monkey species, cotton top tamarins (Saguinus oedipus) were tested in an offering task and in a bartering task. At issue was whether the monkeys rejected rewards because of a violation of expectancy of the preferred reward, or whether they rejected rewards because of a sensitivity to socially mediated inequity. The data from both tasks indicated that the subjects were more likely to reject when preferred rewards were presented, either because of another animal eating the reward (the social condition) or …

—Humans seem unique in their consideration of others’ goals, motivations, intentions, and needs. But the human form of social awareness did not spring from nowhere; certain mechanisms shared across primates formed the foundation from which these processes derived. A review of recent nonhuman primate research points to particular ancestral mechanisms, including an interest in images moving in synchrony with self, a mirror neuron system that responds in the same way to actions made by the self and by others, and inherited social tolerance that provided the bases for social thinking. Contaminated sediments are a significant problem that has adverse effects on human, animal and plant health. Heavy metal contamination, with dissolved metals being the most bioavailable form, has been occurring since man first started living in communities around water bodies. Pore water provides contaminated metals a medium for transport from the sediment to the water body. Determining the toxic metal concentration in pore water is essential to any remediation plan. This research focused on what accelerates or hinders virus-particle association, how the addition of turbidity affects both chlorine and ultraviolet light disinfection, and if sonication can disrupt virus-particle associations exposing viruses to disinfection methods.

A decrease in pH, from 7--4, was found to accelerate association by 2-logs. Calcium increased association by 2.74-logs, and total organic carbon decreased association by 0.36-logs.

An addition of 5 NTU, the maximum turbidity level allowed by USEPA for unfiltered drinking water supplies, affected both chlorine and UV light disinfection showing differences in the rate of kill per dose. The association of viruses and particles can …

This thesis describes the design and development of two software tools to support comparative genomics research. These tools were specifically developed to support the analysis and assembly of sequence data produced from innovative new DNA sequencing technology from 454 Life Sciences using the PicoTiterPlate(TM) device. This research investigates thermal properties of fractured bedrock for the purpose of better understanding the sustainability of standing column well (SCW) geothermal heating systems. The three objectives are to quantify effective thermal conductivity and heat capacity of the fracture network; measure heat exchange between the fluid and the fractured surfaces; and estimate time of thermal breakthrough into a pumping well. Single and dipole well tests are performed to meet these objectives. Single well data is compared with an analytical heat flow model to estimate thermal conductivity and heat capacity.
