Gold Nanoparticle Interactions And Impact Upon A Common Biofilm Source: Legionella Pneumophila , Amber Stojak Dec 2009

Gold Nanoparticle Interactions And Impact Upon A Common Biofilm Source: Legionella Pneumophila , Amber Stojak

All Theses

There exists widespread concern of pathogenic bacteria colonizing and establishing biofilms in water systems providing a direct exposure route to the public. Legionella are widely distributed in human-made and natural environments, and colonization lends the potential to infect human hosts that may lead to the development of Legionnaire's Disease. Silver has been used in cooling towers and water filtration systems for removal of planktonic bacteria and biofilms; however permanent removal of L. pneumophila biofilms with metal ions is impossible when established biofilms are in hard to access places such as dead end piping or inside a protozoan host. Bulk metals …

Cloning And Expression Of Porcine Dicer And Argonaute-2, Heather Stowe Dec 2009

Cloning And Expression Of Porcine Dicer And Argonaute-2, Heather Stowe

All Theses

In vitro-produced embryos exhibit aberrations in development, but the reasons for these developmental problems are unknown. Recently, a class of small non-coding RNA called microRNA (miRNA) has been described and reported to have roles in normal mammalian embryonic development. These miRNAs are encoded in the genome, transcribed by RNA pol II and processed into fragments approximately 22 nt in length by ribonuclease enzymes, the final one being a protein called Dicer. miRNA work through the RNA-induced silencing complex (RISC), of which the argonaute gene family are key proteins. Argonaute-2 (Ago2) has been identified as the only member possessing endonuclease activity, …

Immune Regulation Of The Host Autophagic Response And The Elimination Of Mycobacterium Tuberculosis, Sergio A. De Haro Dec 2009

Immune Regulation Of The Host Autophagic Response And The Elimination Of Mycobacterium Tuberculosis, Sergio A. De Haro

Biomedical Sciences ETDs

Autophagy is an immune effector response against a variety of intracellular pathogens, including Mycobacterium tuberculosis. Mycobacteria survive in macrophages by blocking maturation of the phagosome. Induction of autophagy by the T helper 1 (Th1) cytokine IFN-γ, or amino acid starvation, enables macrophages to overcome the phagosome maturation block and impedes mycobacterial survival. This study investigates the role of T helper 2 (Th2) cytokines Interleukin 4 (IL-4) and Interleukin 13 (IL-13) in autophagy in macrophages and their ability to eliminate intracellular mycobacteria. The Th2 cytokines abrogated autophagy and autophagy-induced transfer of mycobacteria into lysosomal compartments, thus enhancing mycobacterial survival within infected …

Dissolved Organic Matter Influences The Timing Of Embryonic Development Of The Purple Sea Urchin, Strongylocentrotus Purpuratus., Corbin J. Hodges

Master's Theses

Marine dissolved organic matter (DOM) comprises one of the largest carbon reservoirs on earth and has long been considered a potential energy source for marine invertebrates. The importance of DOM transport has been adequately demonstrated for unicellular organisms, where DOM can meet 100% of an organisms energy needs, but the effects of DOM uptake for marine metazoans are less well understood. In this study, three general areas involving the influence of DOM transport to marine invertebrates were explored. First, we assessed the effects of using seawater exposed to high intensity ultraviolet radiation (UVR) on the study organism; embryos of the …

Regulation Of Energy Stores And Feeding By Neuronal And Peripheral Creb Activity In Drosophila., Koichi Iijima, Lijuan Zhao, Christopher Shenton, Kanae Iijima-Ando

Department of Biochemistry and Molecular Biology Faculty Papers

The cAMP-responsive transcription factor CREB functions in adipose tissue and liver to regulate glycogen and lipid metabolism in mammals. While Drosophila has a homolog of mammalian CREB, dCREB2, its role in energy metabolism is not fully understood. Using tissue-specific expression of a dominant-negative form of CREB (DN-CREB), we have examined the effect of blocking CREB activity in neurons and in the fat body, the primary energy storage depot with functions of adipose tissue and the liver in flies, on energy balance, stress resistance and feeding behavior. We found that disruption of CREB function in neurons reduced glycogen and lipid stores …

Cloning And Characterization Of A Novel Oocyte-Specific Gene Fbos Encoding An F-Box Protein In Rainbow Trout (Oncorhynchus Mykiss), Lei Wang

Graduate Theses, Dissertations, and Problem Reports (ETD)

Oocyte-specific genes play critical roles in oogenesis, folliculogenesis and early embryonic development. Through analysis of expressed sequence tags (ESTs) from a rainbow trout oocyte cDNA library, we identified a novel transcript which is represented by multiple ESTs derived only from the oocyte cDNA library. Analysis of tissue distribution of the novel Fbos gene by reverse transcription polymerase chain reaction revealed that the novel gene is only expressed in unfertilized oocytes but not in 10 somatic tissues examined. Northern blot analysis showed the transcript size is approximately 2.1 kb. The full-length cDNA sequence for the novel gene Fbos was obtained by …

Graduate Theses, Dissertations, and Problem Reports (ETD)

Gamma-Glutamyldehydroalanylglycine (EdAG) is a dethiolated, electrophilic metabolite of glutathione (GSH) derived from the Phase II conjugation of GSH with busulfan catalyzed by glutathione S-transferase (GST). It was demonstrated in this dissertation that EdAG could be successfully synthesized and shown to be a metabolite of busulfan from in vitro metabolism by enzymes in human liver cytosol. The electrophilic EdAG was reactive toward the cellular thiols cysteine and glutathione (GSH), and toward cysteine residues in reduced bovine insulin B-chain and bovine serum albumin in vitro , as well. EdAG was demonstrated to be a reversible noncompetitive inhibitor of GSTs, specifically GSTA1-1 at …

The Evaluation Of A Nutrition Education And Fitness Program With A Contest Component Among College Students Using The Re-Aim Framework, Michelle L. Bartlett Dec 2009

The Evaluation Of A Nutrition Education And Fitness Program With A Contest Component Among College Students Using The Re-Aim Framework, Michelle L. Bartlett

Graduate Theses, Dissertations, and Problem Reports (ETD)

Only recently have practitioners and researchers targeted the population of college students in studying interventions aimed at increasing health behaviors. There is evidence that a proportion of college students are making attempts on their own to remedy weight gain and poor physical fitness with little or no guidance, which is reflected in consistently rising obesity rates in this population. Thus, the need for organized and effective interventions is illustrated. The impact of an intervention can be determined through evaluation research. Although the RE-AIM model has not yet been used to evaluate single-site, university-sponsored, college-student weight-loss/fitness programs, it provided a useful …

Mechanism By Which Dietary Polyunsaturated Fat Regulates Lipogenic Gene Expression, Alison Bloom Kohan Dec 2009

Mechanism By Which Dietary Polyunsaturated Fat Regulates Lipogenic Gene Expression, Alison Bloom Kohan

Graduate Theses, Dissertations, and Problem Reports (ETD)

Primary hepatocytes treated with non-esterified polyunsaturated fatty acids have been used as a model for describing the inhibitory effects of dietary polyunsaturated fats on lipogenic gene expression. This model resembles starvation or uncontrolled diabetes but not dietary fat ingestion, which is delivered to liver via chylomicron remnants. Chylomicron remnants enriched with polyunsaturated fatty acids stimulate a unique pattern of intracellular signaling. These remnants enhance p38 mitogen activated protein kinase (MAPK) activity but do not inhibit insulin signaling or induce extracellular-signal related kinase (ERK) or AMP-activated protein kinase (AMPK) activation as do non-esterified polyunsaturated fatty acids. Polyunsaturated fatty acids-enriched remnants and …

Prevalence And Biological Control Of Salmonella Contamination In Rendering Plant Environments And The Finished Rendered Meals, Brandon Kinley Dec 2009

Prevalence And Biological Control Of Salmonella Contamination In Rendering Plant Environments And The Finished Rendered Meals, Brandon Kinley

All Dissertations

Although the rendering process serves as invaluable means for the disposal of inedible animal by-products, the finished products often harbor pathogenic and opportunistic microorganisms such as Salmonella and enterococci, respectively. The temperatures used during the rendering process far exceed the heat tolerance threshold of most bacterial species, so cross-contamination from the environment and/or from the incoming raw material is the proposed source of the contamination. Research has demonstrated that the raw material coming into the rendering facility is highly contaminated with pathogenic bacteria including Salmonella. While not in a rendering facility, studies have also demonstrated that bacteria such as Salmonella …
