Minimal Membrane Docking Requirements Revealed By Reconstitution Of Rab Gtpase-Dependent Membrane Fusion From Purified Components, Christopher Stroupe, Christopher M. Hickey, Joji Mima, Amy S. Burfeind, William Wickner

Dartmouth Scholarship

Rab GTPases and their effectors mediate docking, the initial contact of intracellular membranes preceding bilayer fusion. However, it has been unclear whether Rab proteins and effectors are sufficient for intermembrane interactions. We have recently reported reconstituted membrane fusion that requires yeast vacuolar SNAREs, lipids, and the homotypic fusion and vacuole protein sorting (HOPS)/class C Vps complex, an effector and guanine nucleotide exchange factor for the yeast vacuolar Rab GTPase Ypt7p. We now report reconstitution of lysis-free membrane fusion that requires purified GTP-bound Ypt7p, HOPS complex, vacuolar SNAREs, ATP hydrolysis, and the SNARE disassembly catalysts Sec17p and Sec18p. We use this …

TRYPTOPHAN DEPLETION BY INDOLAMINE 2,3-DIOXYGENASE (IDO) CONTRIBUTES TO MEDIAL IMMUNOPRIVILEGE IN GRAFT ARTERIOSCLEROSIS. Amanda M. Silverio, Madison C. Cuffy, and George Tellides. Interdepartmental Program in Vascular Biology and Transplantation and the Departments of Surgery and Immunobiology, Yale University, School of Medicine, New Haven, CT. Introduction: Since blood vessels and their endothelial lining are major stimulators and targets of the graft rejection response, the study of the differential immune properties of vascular cells is needed for the proper application of human vessels in the transplant setting. Experimental models of atherosclerosis and graft arteriosclerosis show leukocytes selectively infiltrating the intima and adventitia …

Living Kidney Donor Informed Consent Practices Vary Between U.S. And Non-U.S. Centers, Ami Parekh Oct 2009

Living Kidney Donor Informed Consent Practices Vary Between U.S. And Non-U.S. Centers, Ami Parekh

Yale Medicine Thesis Digital Library

Living kidney donation rates are increasing in the United States and internationally. Major consensus statements on the care of living kidney donors recommend communicating all potential health and psychosocial risks to donors. We evaluated the degree of international variation in the process of informed consent of potential donors during their evaluation. Transplant professionals attending the 2006 World Transplant Congress responded to a survey assessing their informed consent processes, donor evaluation and risk communication to living donors. US based respondents were compared to non-US respondents. There were 221 respondents from 177 transplant centers and 40 countries (48% US respondents). Across US …

Use Of Antidepressant Medications And The Subsequent Course Of Depressive Symptoms Among Older Adults, Janet Jalal Abou Oct 2009

Use Of Antidepressant Medications And The Subsequent Course Of Depressive Symptoms Among Older Adults, Janet Jalal Abou

Yale Medicine Thesis Digital Library

Antidepressant medications are commonly prescribed for older adults with depressive symptoms who may not have a major depressive disorder. Yet, the effect of antidepressants on depressive symptoms over time in this population is largely unknown. We sought to determine whether the use of antidepressant medications is associated with a reduction in the severity of depressive symptoms over time. Participants included 754 community-dwelling adults, aged 70+ years, who were followed at 18-month intervals for 90 months. Depressive symptoms were assessed using the 11-item CESD scale, with a higher score indicating worse depressive symptoms. A linear mixed effects model, adjusted for demographic …

Failure To Ferment Dietary Resistant Starch In Specific Mouse Models Of Obesity Results In No Body Fat Loss, June Zhou, Roy J. Martin, Richard T. Tulley, Anne M. Raggio, Li Shen, Elizabeth Lissy, Kathleen Mccutcheon, Michael J. Keenan Oct 2009

Failure To Ferment Dietary Resistant Starch In Specific Mouse Models Of Obesity Results In No Body Fat Loss, June Zhou, Roy J. Martin, Richard T. Tulley, Anne M. Raggio, Li Shen, Elizabeth Lissy, Kathleen Mccutcheon, Michael J. Keenan

Faculty Publications

Resistant starch (RS) is a fermentable fiber that decreases dietary energy density and results in fermentation in the lower gut. The current studies examined the effect of RS on body fat loss in mice. In a 12 week study (study 1), the effect of two different types of RS on body fat was compared with two control diets (0% RS) in C57Bl/6J mice: regular control diet or the control diet that had energy density equal to that of the RS diet (EC). All testing diets had 7% (w/w) dietary fat. In a 16 week study (study 2), the effect of …
