Reactive Oxygen And Nitrogen Species Induce Protein And Dna Modifications Driving Arthrofibrosis Following Total Knee Arthroplasty., Theresa A Freeman, Javad Parvizi, Craig J Della Valle, Marla J Steinbeck Nov 2009

Reactive Oxygen And Nitrogen Species Induce Protein And Dna Modifications Driving Arthrofibrosis Following Total Knee Arthroplasty., Theresa A Freeman, Javad Parvizi, Craig J Della Valle, Marla J Steinbeck

Department of Orthopaedic Surgery Faculty Papers

BACKGROUND: Arthrofibrosis, occurring in 3%-4% of patients following total knee arthroplasty (TKA), is a challenging condition for which there is no defined cause. The hypothesis for this study was that disregulated production of reactive oxygen species (ROS) and nitrogen species (RNS) mediates matrix protein and DNA modifications, which result in excessive fibroblastic proliferation.

RESULTS: We found increased numbers of macrophages and lymphocytes, along with elevated amounts of myeloperoxidase (MPO) in arthrofibrotic tissues when compared to control tissues. MPO expression, an enzyme that generates ROS/RNS, is usually limited to neutrophils and some macrophages, but was found by immunohistochemistry to be expressed …

Nonhomologous End-Joining: Tdp1-Mediated Processing, Atm-Mediated Signaling, Amy Hawkins Nov 2009

Nonhomologous End-Joining: Tdp1-Mediated Processing, Atm-Mediated Signaling, Amy Hawkins

Theses and Dissertations

This thesis investigates two separate features of nonhomologous end-joining (NHEJ) DNA repair: end processing, and DNA repair kinase signaling. DNA end processing was investigated in a mouse model of hereditary spinocerebellar ataxia with axonal neuropathy (SCAN1), a congenital neurodegenerative disease. SCAN1 is caused by a homozygous H493R mutation in the active site of tyrosyl-DNA phosphodiesterase (TDP1). To address how the H493R mutation elicits the specific pathologies of SCAN1 and to further elucidate the role of TDP1 in processing DNA end modifications, we generated a Tdp1 knockout mouse and characterized their behavior and specific repair deficiencies in extracts of embryonic fibroblasts …

A Privacy Framework For Mobile Health And Home-Care Systems, David Kotz, Sasikanth Avancha, Amit Baxi Nov 2009

A Privacy Framework For Mobile Health And Home-Care Systems, David Kotz, Sasikanth Avancha, Amit Baxi

Dartmouth Scholarship

In this paper, we consider the challenge of preserving patient privacy in the context of mobile healthcare and home-care systems, that is, the use of mobile computing and communications technologies in the delivery of healthcare or the provision of at-home medical care and assisted living. This paper makes three primary contributions. First, we compare existing privacy frameworks, identifying key differences and shortcomings. Second, we identify a privacy framework for mobile healthcare and home-care systems. Third, we extract a set of privacy properties intended for use by those who design systems and applications for mobile healthcare and home-care systems, linking them …

Part 2: Professional Development Curriculum: Integrating Molecular Biology And Microbiology Into The Existing Secondary Biology Curricula, Mary Ruth Greer

Theses and Dissertations

PART 1: Thirty-one medicinal plant species from Hawaii, Morocco, and the Sonoran Desert, USA have been shown in past studies to be highly inhibitory to pathogenic bacteria, fungi, and certain cancer cell lines. However, none were tested for antiviral activity. Acetone and methanol extracts from these species were bio-assayed for antiviral activity against herpes simplex virus types 1 and 2 (HSV 1 and HSV 2) and for cytotoxicity to the Vero C1008 cell line. Extracts from these species were tested in vitro for antiviral activity using an immunoperoxidase mini-plaque reduction assay to detect viral structural protein synthesis. Sulforhodamine B and …

A Study Of The Impact Of The Alternative Intervention Program On Improving Student Achievement, Attendance, And Discipline, Beverly Allen-Hardy Nov 2009

A Study Of The Impact Of The Alternative Intervention Program On Improving Student Achievement, Attendance, And Discipline, Beverly Allen-Hardy

Theses and Dissertations

This mixed methods study sought to examine student perceptions of the impact of an alternative intervention program (AIP), and provide a framework meant to inspire programs in other locations. Focusing on attendance, discipline, and academic achievement data, this study examined what factors motivate successful alternative education students to succeed upon their return to the comprehensive setting, and identify support factors that foster resiliency. Qualitative data were collected through three focus groups of students who were either new to the program, enrolled in the program for at least a nine-week period, and students who successfully completed a nine-week placement. Six adult …

The Effectiveness Of Religiously Tailored Couple Counseling, Joshua Hook Nov 2009

The Effectiveness Of Religiously Tailored Couple Counseling, Joshua Hook

Theses and Dissertations

Large numbers of couples seek treatment from religious counselors who integrate religion and spirituality (R/S) into counseling. The present dissertation reviewed the literature examining the effectiveness of R/S counseling. Several R/S treatments were helpful in treating psychological problems. There was little evidence that R/S treatments outperformed secular treatments. In Study 1, a nationwide survey was conducted that examined the beliefs of Christian counselors about integrating R/S into couple counseling. Christian counselors (N = 630) completed measures of religious commitment, experience in couple counseling, attitudes toward using religious techniques in couple counseling, and the use of theory in couple counseling. Counselors …

Designing Community: The Application Of New Urban Principles To Create Authentic Communities, Margaret Embry Nov 2009

Designing Community: The Application Of New Urban Principles To Create Authentic Communities, Margaret Embry

USF Tampa Graduate Theses and Dissertations

This research is an exploratory investigation of the potential of New Urbanist planning and design principles to create thriving and successful neighborhoods. New Urbanism is an urban design movement started in the early 1980s that promotes the development of walkable, compact, and diverse neighborhoods. It is the objective of this research to shed light on the value of creating a higher quality of life and more time for ourselves and our families. I hypothesize that New Urbanism may pave the way for recapturing commute time for time with family, creating authentic and successful communities, and engaging with neighbors. Furthermore, by …
