Induced Hyper And Hypo Aminoacidemia In The Near Term Ovine Fetus: Effects On Electrocortical Activity And Cerebral Protein Synthesis, James Nakahara Maclachlan Jan 2009

Induced Hyper And Hypo Aminoacidemia In The Near Term Ovine Fetus: Effects On Electrocortical Activity And Cerebral Protein Synthesis, James Nakahara Maclachlan

Digitized Theses

We have utilized an amino acid infusate and an insulin euglycemic clamp

technique in the ovine fetus near term with increases and decreases in circulating amino acids, respectively, to determine the impact on electrocortical activity and cerebral protein synthesis. Fetal sheep were studied over a 2 hour control and subsequent 6 hour experimental period, during which animals received an infusion of Primene® 10%, or a co-infusion of insulin and 10% dextrose, as well as C13 leucine for determining cerebral protein synthesis. With Primene infusion, basic and neutral amino acids were increased, while acidic amino acids showed little change. With insulin/dextrose …

M Vivo Tracking Of Iron-Labeled Dendritic Cells In Mice Using Magnetic Resonance Imaging, Roja Rohani Jan 2009

M Vivo Tracking Of Iron-Labeled Dendritic Cells In Mice Using Magnetic Resonance Imaging, Roja Rohani

Digitized Theses

Successful migration of adequate numbers of dendritic cells (DC) to draining lymph nodes is crucial for effective DC immunotherapy. Since currently, less than 3% of injected DC migrate to lymph nodes, improving DC migration is necessary. A non- invasive method is required to monitor DC migration in animal models. This work presents the first report of using Micron-sized Superparamagnetic Iron-Oxide Particles (MPIO) to label DC for detection by MRI and of tracking their migration in a mouse model in vivo.

A quantitative approach was used to measure the signal loss in MR images and the fluorescence detected by microscopy. Moreover, …

Evaluation Of Pilo Substrate Specificity Using Normally Non-Glycosylated Proteins In Pseudomonas Aeruginosa, Matthew A. Henkel Jan 2009

Evaluation Of Pilo Substrate Specificity Using Normally Non-Glycosylated Proteins In Pseudomonas Aeruginosa, Matthew A. Henkel

Electronic Theses and Dissertations

P. aeruginosa 1244 (PA1244) possesses an O-linked glycosylation system by which the glycosyltransferase, PilO, transfers a single preassembled O-antigen repeating unit to the C-terminal serine residue of pilin as a posttranslational modification. Previous work has shown that the group II pilin of PA103 can be engineered for glycosylation by extending a C-terminal serine away from the surface with a short stretch of alanine residues (Horzempa et al., 2006a). As an extension of that study, several normally non-glycosylated proteins [PA683 pilin, PA1244 PilA-E. coli (EC) alkaline phosphatase (PhoA) fusion, and EC PhoA] were engineered with C-terminal amino acid extensions and assessed …

Demographics, Activity, And Habitat Selection Of The Eastern Box Turtle (Terrapene C. Carolina) In West Virginia, Justin Adam Weiss

Theses, Dissertations and Capstones

Little is known about the eastern box turtle, Terrapene c. carolina, in West Virginia. The purpose of this study was to compare demographic data between two populations of box turtles, to determine which environmental parameter(s) influence activity patterns, and to describe the optimal habitat of the box turtles at two study sites (Lake and State Park) at Beech Fork Wildlife Management Area. I observed male-skewed sex ratios and low numbers of juveniles in both populations. Male turtles were larger than females in carapace length, but females were larger in carapace height. Turtles at the Lake site exhibited greater body mass …

I'M A Highway Star: An Outline For A Federal Right Of Publicity, Sean D. Whaley Jan 2009

I'M A Highway Star: An Outline For A Federal Right Of Publicity, Sean D. Whaley

UC Law SF Communications and Entertainment Journal

The federal government has failed to codify a federal publicity right. This has caused numerous problems recently, as varying state laws unevenly restrain the commercial speech of many actors in interstate commerce. However, the nature of the publicity right as a property right and its essential function in interstate commerce calls for Congress to adopt a federal publicity right. Similar to the dual system of trademark protection, a federal publicity right will not preempt such rights at the state level. Rather, establishing such a right will detail proper justification and contours of the right. By establishing a federal right, the …

Nos-Based Biopolymers: Towards Novel Thromboresistant No-Release Materials, Charbel Abou Diwan Jan 2009

Nos-Based Biopolymers: Towards Novel Thromboresistant No-Release Materials, Charbel Abou Diwan

ETD Archive

Nitric Oxide releasing biopolymers have the potential to prolong vascular graft and stent potency without adverse systemic vasodilation. It was reported in literature that eNOS-overexpressing endothelial cell seeding of synthetic small diameter vascular grafts decreased human platelet aggregation by 46 and bovine aortic smooth muscle cell proliferation by 67.2 in vitro. We hypothesized that incorporating the enzyme nitric oxide synthase (NOS) in biocompatible polymeric matrix will provide a source of NO that utilizes endogenous compounds to maintain an unlimited supply of NO. To test this hypothesis, we have incorporated the enzyme nitric oxide synthase into a polyethyleneimine film using a …

Structural Studies Of Talin-Mediated Integrin Activation, Esen Bakhautdin Jan 2009

Structural Studies Of Talin-Mediated Integrin Activation, Esen Bakhautdin

ETD Archive

Activation of heterodimeric (alpha/beta) integrin transmembrane receptors by the cytoskeletal protein talin is essential for many important cell adhesive responses including cell-extracellular matrix contact, cell motility and survival. A key step in this process involves interaction of phosphotyrosine-binding (PTB) domain in the N-terminal head of talin (talin-H) with integrin beta3membrane-proximal cytoplasmic tails (beta-MP-CTs). Compared to talin-H, intact talin exhibits low potency in inducing integrin activation. Using TROSY NMR spectroscopy, we found that the large C-terminal rod domain of talin (talin-R) interacts with talin-H and allosterically restrains talin in a closed conformation. Structural, mutational, and biochemical analyses demonstrate that talin-R specifically …

Homeostatic Regulation Of Interleukin-4-Mediated Cell Signaling, Rikhia Chakraborty Jan 2009

Homeostatic Regulation Of Interleukin-4-Mediated Cell Signaling, Rikhia Chakraborty

ETD Archive

Cytokines are intracellular messengers that activate multiple signaling pathways and regulate cell survival, differentiation, migration, apoptosis and immune responses. We are investigating the homeostatic control of cytokine-mediated cell signaling using interleukin-4 (IL-4) as a model cytokine. Protein tyrosine phosphorylation is essential for cytokine-dependent signal transduction through the Janus Kinase-Signal Transducer and Activator of Transcription (Jak-STAT) pathway. Therefore, protein tyrosine dephosphorylation is a key mechanism providing the homeostatic control of the pathway. Here, we have shown for the first time that immediately following ligand-dependent activation, IL-4 receptor induces an intracellular calcium flux via insulin receptor substrate (IRS)-phosphoinositide 3-kinase (PI3K)-phospholipase C (PLC)-gamma …
