Sphingosine-1-Phosphate Elicits Receptor-Dependent Calcium Signaling In Retinal Amacrine Cells, Scott Crousillac, Jeremy Colonna, Emily Mcmains, Jill Sayes Dewey, Evanna Gleason

Evidence is emerging indicating that sphingosine-1-phosphate (S1P) participates in signaling in the retina. To determine whether S1P might be involved in signaling in the inner retina specifically, we examine the effects of this sphingolipid on cultured retinal amacrine cells. Whole cell voltage-clamp recordings reveal that S1P activates a cation current that is dependent on signaling through Gi and phospholipase C. These observations are consistent with the involvement of members of the S1P receptor family of G-protein-coupled receptors in the production of the current. Immunocytochemistry and PCR amplification provide evidence for the expression of S1P1R and S1P3R in amacrine cells. The …

Herpes simplex virus type 1 (HSV-1)-induced cell fusion is mediated by viral glycoproteins and other membrane proteins expressed on infected cell surfaces. Certain mutations in the carboxyl terminus of HSV-1 glycoprotein B (gB) and in the amino terminus of gK cause extensive virus-induced cell fusion. Although gB is known to be a fusogenic glycoprotein, the mechanism by which gK is involved in virus-induced cell fusion remains elusive. An asymmetric distribution of the sexes within the left and right uterine horns has been described in multiple species. A series of experiments were conducted to evaluate the sex ratio (% male) of calves gestated in the left and right uterine horns, as well as the sex ratio of embryos originating from the left and right ovaries of cattle. The sex ratio of calves gestated in the right uterine horn of naturally mated cows was significantly higher compared with the sex ratio of calves gestated in the left uterine horn. One of the most persistent challenges in biology is explaining the distribution of animal taxa. Quantitative explanations for the distribution of organisms are challenged by the complexity of factors that potentially limit a species range, including topography, ecology, climate and biology. Because of limits on data and approaches, early biogeographers were hampered in their ability to explain patterns. Here, I benefit from two primary developments in biogeography. My research draws from the accumulation of data across several scientific disciplines and advances in spatial and statistical approaches to examine the distribution of animal taxa within an integrative biogeography framework. Background: Heterogeneous cell populations have previously been described as noisy. However, recent studies have demonstrated that heterogeneity can be biologically significant. We present here an approach for rapid and complete identification of heterogeneous cell populations from high-throughput flow cytometry data. We have developed a novel measure Slope Differentiation Identification (SDI) using flow cytometry-based protein expression, quantifying the rate of change in protein expression between two conditions (exponential and stationary phase) of yeast cells, as a function of cell size or cell granularity.

ABSTRACT
Nine halonitromethanes (HNMs) were among the more than fifty unregulated high-priority disinfection by-products (DBPs) that were monitored for and detected in drinking waters in a 2000-2002 nationwide occurrence study funded by the US Environmental Protection Agency (USEPA). The toxicology studies conducted in recent years have shown that HNMs are one of the most cytotoxic and genotoxic classes among the emerging DBPs, with orders of magnitude higher cytotoxicity and genotoxicity than any of the regulated organic DBPs. Furthermore, brominated HNMs were found to be more toxic than their chlorinated analogs, with dibromonitromethane being the most cyto- and genotoxic. Today, our …

This study investigates learning outcomes of four-year-old children at a preschool in P. R. China. The children are educated in a school ecology designed to address cognitive, social, linguistic and psychological development, where an instructional technique, “Integrating Thinking, Art and Language” (ITAL), is applied to support them in developing multiple intelligences. A total of 67 beginning or pre-level I Chinese preschoolers participated in the study, whose learning in the first eight months under ITAL instruction is examined. Scenarios of ITAL instruction, children's samples and assessment of learning outcomes are reported and analyzed. Results indicate that the preschoolers demonstrated significant growth …

The intention of this study was to estimate population density, time of spawning, and fecundity of the turquoise darter (Etheostoma inscriptum) in Six Mile creek of the Clemson SC experimental forest. Population density estimates were conducted on 18 riffles within the lower 1,524 m of the creek starting 304.8 m above its confluence with Lake Issaqueena from January to April 2008. Density was determined using the Leslie Depletion method, and darters/m2 was calculated. Random point collection was performed From October 14 to November 13, 2008, and 301 specimens were measured and released to determine length frequency of the population. …

Proteases, proteins which cleave peptide bonds in other proteins, are a large and varied group of proteins which regulate a variety of physiological processes. Methodologies to study proteases are often protease specific and often differ greatly from the roles proteases play in vivo. In vitro protease assays often use peptide based substrates, which do not take into account highly specific interactions distal from the proteolytic site of peptide cleavage on protease substrates. The present study documents the indigenous knowledge of medicinal plants used in the Alaknanda catchment of Uttarakhand state in India. Ethnomedicinal uses of 100 medicinal plant species along with botanical name, vernacular name, family, habit, part used and folk medicinal uses are presented. They belong to 91 genera and 51 families. These plants have been used to cure 60 types of different ailments out of the 58 plant species used to cure more than one disease. Telomeric DNA terminates with a single-stranded 3′ G-overhang that in vertebrates and fission yeast is bound by POT1 (Protection Of Telomeres). However, no in vitro telomeric DNA binding is associated with Arabidopsis POT1 paralogs. To further investigate POT1–DNA interaction in plants, we cloned POT1 genes from 11 plant species representing major branches of plant kingdom. Telomeric DNA binding was associated with POT1 proteins from the green alga Ostreococcus lucimarinus and two flowering plants, maize and Asparagus. Site-directed mutagenesis revealed that several residues critical for telomeric DNA recognition in vertebrates are functionally conserved in plant POT1 proteins. However, the plant proteins …

The vast majority of animal species do not tolerate severe water stress, but the encysted embryo of the brine shrimp Artemia franciscana is an exceptionally useful organism to investigate physiological mechanisms for enduring extreme environmental insults. Any substantial reduction in cellular water poses a threat to survival. Nevertheless anhydrobiotic animals survive virtually complete loss of cellular water. The mechanisms that govern " life without water" (anhydrobiosis) are still not well understood. With certain exceptions, it seems that a recurring strategy for tolerating severe water loss involves the accumulation of both low molecular weight solutes (e.g., trehalose or other polyol) and …

The southeastern portion of the Edwards Plateau of Texas, historically a stronghold of Rio Grande wild turkeys Meleagris gallopavo intermedia, has seen the numbers of turkeys declining since the 1970s. Because hen survival is a key parameter affecting turkey population dynamics, we monitored radio-tagged Rio Grande wild turkey hens on the Edwards Plateau during 2001-2007 to compare survival during the breeding season in areas where turkey abundance has declined vs areas with stable populations. This thesis explores the social identities and the work and business strategies of five farming couples who had long farming careers in the Midwest. Much of the literature presented within this thesis discusses the hardships that family farmers face in today’s economy. However, the research also points to a strong attachment to farming and the farming lifestyle despite its many challenges. Using a qualitative interviewing method, I was interested in studying what meaning these farming couples attached to their work, including any gendered division of labor, and what survival techniques they had employed. The hypothalamic suprachiasmatic nucleus (SCN), which in mammals serves as the master circadian pacemaker by synchronizing autonomous clocks in peripheral tissues, is composed of coupled single-cell oscillators that are driven by interlocking positive/negative transcriptional/translational feedback loops. Several studies have suggested that heme, a common prosthetic group that is synthesized and degraded in a circadian manner in the SCN, may modulate the function of several feedback loop components, including the REV-ERB nuclear receptors and PERIOD2 (PER2). We found that ferric heme (hemin, 3-100 μM) dose-dependently and reversibly damped luminescence rhythms in SCN explants from mice expressing a PER2::LUCIFERASE (PER2::LUC) fusion protein. …

Manganese (Mn) is essential for brain growth and metabolism, but in excess can be a neurotoxicant. The chemical form (species) of Mn influences its kinetics and toxicity. Significant Mn species entering the brain are the Mn2+ ion and Mn citrate which, along with Mn transferrin, enter the brain by carrier-mediated processes. Although the divalent metal transporter (DMT-1) was suggested to be a candidate for brain Mn uptake, brain Mn influx was not different in Belgrade rats, which do not express functional DMT-1, compared to controls. The Peaks of Otter Salamander, Plethodon hubrichti, is found along a 19 km length of the Blue Ridge Mountains, Virginia, USA, often in sympatry with the Eastern Red-backed Salamander, P. cinereus. In a sympatric area of Bedford County, Virginia, we conducted a mark-recapture study on a 10 × 10 m site. Surface densities of salamanders increased as the number of days without precipitation prior to a collection event increased. This suggests vertical movements in response to surface moisture. When salamanders returned to the surface after rain, individuals appeared to “shuffle” between rocks and likely, leaf litter. That is, we were …

The art of magic reigns as the premiere form of entertainment made possible through the strategic means of deception. An untrained individual (civilian) who desires personal gain at the expense of others, however, may use the same strategic gimmicks and manipulations that magicians employ. This study considered the theatrical elements used by magicians during the performance of magic but in an interpersonal civilian context. In order to examine civilian use of the premeditated theatrical elements, and to determine their presence in deceptive interpersonal communication, this study examined low versus high self-monitors and male versus female genders.
