Cis-Regulatory Sequence And Co-Regulatory Transcription Factor Functions In Erα-Mediated Transcriptional Repression, Richard Leroy Smith
Cis-Regulatory Sequence And Co-Regulatory Transcription Factor Functions In Erα-Mediated Transcriptional Repression, Richard Leroy Smith
Theses and Dissertations
Estrogens exert numerous actions throughout the human body, targeting healthy tissue while also enhancing the proliferative capacity of breast cancers. Estrogen signaling is mediated by the estrogen receptor (ER), which binds DNA and ultimately affects the expression of adjacent genes. Current understanding of ER-mediated transcriptional regulation is mostly limited to genes whose transcript levels increase following estrogen exposure, though recent studies demonstrate that direct down-regulation of estrogen-responsive genes is also a significant feature of ER action. We hypothesized that differences in cis-regulatory DNA was a factor in determining target gene expression and performed computational and experimental studies to test this …

Role Of Endothelin-1 In The Regulation Of The Swelling-Activated Cl- Current In Atrial Myocytes, Wu Deng
Role Of Endothelin-1 In The Regulation Of The Swelling-Activated Cl- Current In Atrial Myocytes, Wu Deng
Theses and Dissertations
Swelling-activated Cl- current (ICl,swell) is an outwardly rectifying Cl- current that influences cardiac electric activities and acts as a potential effector of mechanoelectrical feedback that antagonizes the effects of stretch-activated cation channels. Persistent activation of ICl,swell has been observed in multiple models of cardiovascular diseases. Previously we showed that angiotensin II (AngII) signaling and reactive oxygen species (ROS) produced by NADPH oxidase (NOX) are involved in the activation of ICl, swell by both beta1-integrin stretch and osmotic swelling. Because endothelin-1 (ET-1) is a potential downstream mediator of AngII and ETA receptor blockade abrogates AngII-induced ROS generation, we studied how ET-1 …
A Cellular Mechanism For Dendritic Spine Loss Following Traumautic Brain Injury In Rat, Brian Low
A Cellular Mechanism For Dendritic Spine Loss Following Traumautic Brain Injury In Rat, Brian Low
Theses and Dissertations
Traumatic brain injury is a leading cause of death and disability in the United States. The injury is often composed of two processes: the primary injury, which can involve irreversible loss of tissue, and the secondary injury, which involves a cascade of reactive processes such as excitotoxicity that occur in the hours and days after the initial insult. Excitotoxic stimulation of neuronal circuits can lead to cellular dysfunction and modulation of neuronal sensitivity. One mechanism of dysfunction involves the calcium-regulated phosphatase, calcineurin. Calcineurin has been shown to be involved in the modulation of the neuronal post-synaptic structures known as dendritic …
Genetic Differentiation Of The Parasitoid, Cotesia Congregata (Say), Based On Host-Plant Complex, Georgia Karns
Genetic Differentiation Of The Parasitoid, Cotesia Congregata (Say), Based On Host-Plant Complex, Georgia Karns
Theses and Dissertations
Endoparasitoids of herbivorous lepidopterans have intimate relationships with their host species as well as the plant on which their host develops. Characteristics of both hosts and plants can affect parasitoid success in tri-trophic systems and thus, drive diversification. Genetic differentiation was estimated for Cotesia congregata (Say) collected from two distinct host-plant complexes, Manduca sexta L. on tobacco (Nicotiana tabacum L.) and Ceratomia catalpae, (Haworth) on catalpa (Catalpa bignonioides Walker), in the mid-Atlantic region of North America using seven microsatellite loci and the mitochondrial COI locus. Microsatellite allele frequencies were differentiated based on host-plant complex, and COI haplotypes from individuals on …

Innexons, Membrane Channels For Atp, Control Microglia Migration To Nerve Lesions, Stuart E. Samuels
Open Access Dissertations
ATP released upon nerve injury is an important chemotactic activator of microglia; however, the source of the extracellular ATP is uncertain. In large glial cells such as astrocytes, chemical or mechanical stimulation or injury causes an intracellular calcium wave that moves from cell to cell propagated by extracellular ATP through a regenerative ATP-induced ATP-release mechanism. In mammals a prime candidate for the ATP release channel is the pannexon, a hexamer of the invertebrate gap junction homologue pannexin1, although the pharmacologically similar protein connexin is also a candidate. I hypothesized that upon nerve injury, ATP released through pannexon channels activates microglia …

Crafting Authenticity, Allison N. Schumacher
Crafting Authenticity, Allison N. Schumacher
Theses and Dissertations
Authenticity is what we want from the world around us, from others, and crucially from ourselves and what we make. As it relates to graphic design, I define authenticity as a perceived match between form and purpose. For the designer, its quality is found in the process of simultaneously developing a concept and crafting the design/object.

Salmonella In Broiler Litter And Properties Of Soil At Farm Location., Victoriya V. Volkova, R. Hartford Bailey, Robert W. Wills
CVM Publications
Contamination of litter in a broiler grow-out house with Salmonella prior to placement of a new flock has been shown to be a precursor of the flock's Salmonella contamination further down the production continuum. In the southern USA, broiler grow-out houses are primarily built on dirt pad foundations that are placed directly on top of the native soil surface. Broiler litter is placed directly on the dirt pad. Multiple grow-out flocks are reared on a single litter batch, and the litter is kept in the houses during downtime between flocks. The effects of environmental determinants on conditions in broiler litter, …
Active Student Engagement By Using Cooperative Learning Strategies, Austin Martinez
Active Student Engagement By Using Cooperative Learning Strategies, Austin Martinez
Theses and Dissertations
The purpose of this study was to explore possible solutions to student interaction and product quality in a ninth grade English class by using strategies in cooperative learning. Reciprocal learning, jigsaw, and skills practice were used to promote student interaction and cooperation during a lesson on William Shakespeare's "Romeo and Juliet." The data reveals that the three student participants have the skills necessary to succeed and excel in cooperative learning strategies, and previous student achievements may not necessarily indicate an ability or inability to master a cooperative learning activity. Implications for using cooperative learning strategies in the classroom are discussed.

Species Composition, Seasonal Activity, And Aggregations Of Dung Beetles On Cattle Pastures In Arkansas, Justin George Fiene
Student Theses and Dissertations
The species composition, seasonal activities, and aggregations of dung beetles (Scarabaeidae and Geotrupidae) were monitored from April 2007 to September 2008 using ten dung-baited pitfall traps operated weekly at a site in northeastern Arkansas. Collections comprised 236,880 beetles, representing 22 species including Colobopterus erraticus L. and Onthophagus taurus Schreber, which are new state records. Labarrus pseudolividus Balthasar comprised 97.7 % of the total trap catch, with the majority being collected in June, July and August 2007 which involved two separate mass occurrences during that time. Colonizing dung beetles expressed strong intraspecific aggregation, while interspecific aggregation, though present, were typically of …

Theses and Dissertations
The objective of this thesis is to develop a sequential algorithm to determine accurately and quickly, at which point in time a product is well mixed or reaches a steady state plateau, in terms of the Refractive Index (RI). An algorithm using sequential non-linear model fitting and prediction is proposed. A simulation study representing typical scenarios in a liquid manufacturing process in pharmaceutical industries was performed to evaluate the proposed algorithm. The data simulated included autocorrelated normal errors and used the Gompertz model. A set of 27 different combinations of the parameters of the Gompertz function were considered. The results …
The Potential Role For Capb In Pathogenesis Of Francisella Tularensis, Eric Fleming
The Potential Role For Capb In Pathogenesis Of Francisella Tularensis, Eric Fleming
Theses and Dissertations
Francisella tularensis was a facultative intracellular pathogen and a gram-negative coccobacillus which has been categorized by the CDC as a potential class A select agent due to its highly infectious properties and high mortality rates. Francisella tularensis was also responsible for the zoonotic disease tularemia, which was usually transmitted by arthropod vectors or via contact with infected animals. Francisella tularensis subspecies novicida has been used by many researchers in genetic pathogenesis experiments to try to elucidate genes responsible for virulence factors. One of these virulence factors was a capsular material which has been thought to be involved in either increasing …

Consequences Of Weight Cycling: An Increase In Disease Risk?, Kelley Strohacker†, Katie C. Carpenter†, Brian K. Mcfarlin‡
International Journal of Exercise Science
Research indicates that weight cycling, or “yo-yo dieting” is a common occurrence in obese populations. The long term negative health consequences of weight cycling are debated and it is unclear whether or not this weight change pattern poses a greater disease risk compared to obesity maintenance. This review discusses the prevalence of weight cycling and physiological alterations occurring during weight loss that promotes weight regain. We also discuss the effect weight regain has upon adipose tissue in terms of rate and type of accumulation. Also within this review are discussions surrounding the previously published literature based upon human and rodent …

Physical Activity Is Associated With Percent Body Fat And Body Composition But Not Bmi In White And Black College Students, Michael Zanovec†, Anantha P. Lakkakula†, Lisa G. Johnson‡, Georgianna Tuuri‡
International Journal of Exercise Science
The objective of this study was to examine the association of self-reported physical activity (PA) with body composition in 290 college students (49% male, 60% White) 18-25 years of age. Outcome measures included: self-reported PA levels calculated in MET-hrs·wk-1 from the International Physical Activity Questionnaire (IPAQ); body mass index (BMI; in kg·m-2); and body composition variables estimated by dual-energy X-ray absorptiometry (DXA). Mean activity levels of the sample were 39.8 ± 23.8 MET-hrs·wk-1. Participants were divided into quartiles of PA levels: ≥0 to <24.0, ≥24.0 to <34.0, ≥34.0 to <51.25, and ≥51.25 MET-hrs·wk-1 and body composition variables were compared by group. Chi-square analyses revealed a significant difference for gender by PA quartile [χ2 (3, N=290) = 32.42, p < 0.0001], and for gender by race by PA quartile [χ2 (9, N=290) = 37.82, p < 0.0001]. MET-hrs·wk-1 was inversely correlated with %BF (r = -0.40, p < 0.0001) but not BMI (r = 0.05, p = 0.43). When comparing body composition variables across PA quartiles, no significant differences were observed for BMI; however, subjects in the highest quartile of PA had a lower percent body fat (%BF) and fat mass (FM), and a higher lean-tissue mass (LTM) compared to subjects in the other three groups. In this cohort of young adults, participants in the highest activity group had a more fit body composition profile (e.g., lower %BF, lower FM, and higher LTM) which was not reflected in BMI and was independent of gender and race.

Specific Levels Of Therapeutic Ultrasound Stimulate The Release Of Inflammatory And Angiogenic Mediators From Macrophages In Culture, Thomas Turner
Specific Levels Of Therapeutic Ultrasound Stimulate The Release Of Inflammatory And Angiogenic Mediators From Macrophages In Culture, Thomas Turner
Theses and Dissertations
Therapeutic ultrasound (TUS) is a treatment modality that is used to accelerate tissue healing. TUS is thought to affect cellular processes of tissue healing, especially those that occur in the inflammatory and early proliferative phases. TUS can be applied using various parameter selections including intensity, wavelength, duty cycle and treatment duration and no clear consensus exists on optimal parameters for healing enhancement. Macrophages are important mediators of inflammation and their actions are critical to normal progression into the proliferative phase of healing. They complete many functions during these periods of tissue healing, among those being release of cytokines and growth …
The Regulation Of Platelet Activating Factor Acetylhydrolase By Oxidized Phospholipids, Rachael Griffiths
The Regulation Of Platelet Activating Factor Acetylhydrolase By Oxidized Phospholipids, Rachael Griffiths
Theses and Dissertations
Platelet-activating factor acetylhydrolase (PAFAH) is elevated in atherosclerosis and may play a role in pathogenesis of this disease. Molecular mechanisms regulating the expression of this lipoprotein-associated PLA2 are indistinct. Mildy oxidized low density lipoprotein (oxLDL) and monocytes (the primary source of PAFAH) are co-localized in early atheromas. Monocytes are activated by oxidized phospholipids (oxPL) in the oxLDL particle. We hypothesized that oxPL-activated monocytes are the source of increased levels of PAFAH in atherosclerosis. We found that PAFAH expression is significantly induced by OxPAPC and in particular long-chain fractions of oxPAPC in monocytes and cytokine-differentiated DC, but not cytokine-differentiated MO. Furthermore, …
