This thesis project explores repurposing a hydroelectric plant along Richmond Virginia's Canal Walk. The building has been redesigned to create a community-oriented space programmed as an indoor park, event venue space, and cafe. Throughout this thesis, it became important to create private niches within a public space while creating a flexible public venue that accommodates a variety of activities. Through a variety of spaces that offer users options, a flexible public venue is creating, yielding a community-oriented environment that reconnects local with the site.

My current work revels in a state of flux. I strive for the work to be electrically charged, conveying a feverish sense of immediacy and vitality that implies motion and frenetic energy. The work is an accretion of brightly colored biomorphic forms that extend out from the wall and onto the floor. Viscous parts ooze and drip while others are globular and bulbous. The hyper-organic forms suggest a paradoxical state of both ripening and rotting, becoming and unbecoming. ABSTRACT 1: Abrasive treatment and subsequent electroporation of the basidiomycete Pleurotus ostreatus in a hypertonic buffer was investigated as a potential method of transforming of intact hyphæ. Mycelia, which are not capable of being transformed via electroporation due to interference by the cell wall, were abraded in an attempt to mechanically degrade the cell wall prior to electroporation. An electroporation field strength of 12,500 V/cm for 500 μs to 1.25 ms was found to be optimal based upon mortality effects. A 32 μm carborundum abrasive was initially evaluated but was quickly found to be inappropriately large so a corundum abrasive …

Eight small businesses in the electronics industry from Rochester, NY were studied in order to determine whether the European Union’s directives, Waste Electrical and Electronic Equipment (WEEE) and Restriction of Hazardous Substances Directives (RoHS) are spurring innovation in the US. Innovation was defined as any change in the design and manufacturing of the products, in the internal organizational structure and management of the business, or in the market strategies pursued by the small businesses that created a benefit beyond RoHS and WEEE compliance. With high definition, high resolution, technology becoming ever more popular, the vast amount of input available to modern object recognition systems can become overwhelming. Given an image taken from a high resolution digital camera, a target object may be very small in comparison to the entire image. Additionally, any non-target objects in the input are considered unnecessary data, or clutter. While many modern object recognition systems have been created to be over 90% accurate in the recognition task, adding large amounts of clutter to an input quickly degrades both the speed and accuracy of many models. Elliptic curve cryptography (ECC) is an increasingly popular method for securing many forms of data and communication via public key encryption. The algorithm utilizes key parameters, referred to as the domain parameters. These parameters must adhere to specific characteristics in order to be valid for use in the algorithm. The American National Standards Institute (ANSI), in ANSI X9.62, provides the process for generating and validating these parameters. The National Institute of Standards and Technology (NIST) has identified fifteen sets of parameters; five for prime fields, five for binary fields, and five for Koblitz curves. The XML Expert System Shell (XESS) was designed to alleviate some of the difficulties associated with translating a knowledge base from one expert system to another. In the beginning, we had filing cabinets managed by individuals and departments who had policies for archiving and purging documents over time due to space limitations or organization. This paper explores possible solutions for archiving a network file system. When we switched from filing cabinets to digital data storage, two things were not added to file systems. Head and neck cancer ranks high among the most common cancers world wide. In addition, there is a high recurrence rate, as well as a high prevalence of loco-regional tumor spread. Among many factors contributing to metastasis is vascular endothelial cell growth factor C. VEGF-C is primarily an inducer of new lymph vessel formation, typically during embryogenesis; however, some advanced cancers show a significant increase in VEGF-C expression, suggesting a role in metastasis. Krop Senior High School in Miami, Florida. The following text explains elements of pre-production, presents relevant research associated with the play, describes the production process in detail, and states conclusions drawn by the author about the experience. The work challenged both the ensemble and the director to seek new avenues of expression and theatricality different from those traditionally explored in their respective educational settings.

The following thesis is a qualitative comparative case study which investigated the values, practices, perspectives, and strategies of Indian and American community organizers (practitioners and volunteers of non-profits and non-governmental organizations) who use microfinance, including savings schemes, as an instrument within the social intermediation process of developing disadvantaged women's capacities for self-sufficiency/empowerment. The focused inquiry was conducted through similarly structured in-depth interviews of directors, staff members and volunteers/community-based organizers of a women's business center in Northern Virginia, a large U.S. metropolitan area, and a women's federation in the rural Himalayas of Uttarakhand, India. Successful treatment of cancer with adoptive immunotherapy (AIT) is dependent on the ability to produce large numbers of tumor-specific, functional T cells. The purpose of this thesis is to explore ways in which T cell expansion could be augmented. We have focused on exploring alternate gamma chain cytokines as stimulators of T cell proliferation and differentiation in addition to investigating the potential usefulness of gemcitabine (GEM) in abrogating the immunosuppressive effects of myeloid derived suppressor cells (MDSCs). Catecholaminergic neurotransmission is regionally altered following injury, and drugs aimed at these systems offer promising avenues for post-TBI pharmacotherapies. Atomoxetine is a selective norepinephrine transporter (NET) inhibitor currently indicated for treatment of attention-deficit hyperactivity disorder (ADHD). The studies in this dissertation were designed to test the efficacy of atomoxetine for treating cognitive deficits following experimental TBI and the potential mechanism for any beneficial effect. The first part of the study focused on behavioral recovery following atomoxetine treatment. Transient intracellular elevations of Ca2+ are common signaling mechanisms used to allosterically regulate proteins. One potential target of Ca2+ is Ca2+/calmodulin dependent protein kinase type II (CaMK-II). CaMK-II is a multi-functional protein kinase known to influence cellular pathways such as cell motility and cell cycle progression. Within the cell cycle, CaMK-II promotes the expression of the regulator protein cyclin D1, which is necessary for cell cycle progression. To further understand CaMK-II’s role in cyclin D1 expression, the binding partners of cytosolic CaMK-II were studied using mass spectrometry. Transient elevations in Ca²⁺ have previously been shown to promote focal adhesion disassembly and cell motility. Yet the targets of these Ca²⁺ transients have not been fully examined. In this study, we demonstrate that CaMK-II, a Ca²⁺/calmodulin dependent protein kinase, is activated in response to β1 integrin engagement with fibronectin to influence fibroblast adhesion and motility. We also show that CaMK-II is dynamically localized to the cell surface using Total Internal Reflection Fluorescence microscopy (TIRFm) and that inhibition of CaMK-II with two mechanistically distinct, membrane permeant inhibitors accelerates spreading on fibronectin, enlarges paxillin-containing focal adhesions and …

Medicaid orthodontic patients have been shown to miss more appointments and break more appliances than self-pay patients, indicating a greater tendency toward non-compliance. Lack of compliance during the post-treatment phase can be detrimental to retention. The purpose of this study was to determine whether there is a difference in PAR score changes between Medicaid and non-Medicaid patients during and after orthodontic treatment. Over the past few decades, there has been growing interest and increased research on bio-implantable devices using RF telemetry links, enabling the continuous monitoring and recording of physiological data. However the dispersive properties of tissues make this a formidable task. In the present work, a novel technique for tissue characterization using an LC sensor is developed which allows for the extraction of the relative permittivity, and the conductivity of dispersive tissues. In 1952, during the Korean War, President Truman seized control of American steel mills to prevent a strike that threatened the war effort. This action, prompted by a labor dispute between steelworkers and operators, led to a landmark Supreme Court case, Youngstown Sheet & Tube Co. v. Sawyer. The Court ultimately ruled that the President lacked the authority to seize the steel mills without explicit congressional authorization, establishing limits on presidential power.

Understanding how environmental fluctuations affect population persistence is essential for predicting the ecological impacts of expected future increases in climate variability. However, two bodies of theory make opposite predictions about the effect of environmental variation on persistence. Single‐species theory, common in conservation biology and population viability analyses, suggests that environmental variation increases the risk of stochastic extinction. By contrast, coexistence theory has shown that environmental variation can buffer inferior competitors against competitive exclusion through a storage effect. We reconcile these two perspectives by showing that in the presence of demographic stochasticity, environmental variation can increase the chance of extinction while …

Empirical and theoretical evidence suggests that facilitation between plants, when it occurs, is more likely during periods of abiotic stress, while competition predominates under more moderate conditions. Therefore, one might expect the relative importance of competition vs. facilitation to vary seasonally in ecosystems characterized by pronounced dry (abiotically stressful) and wet (benign) seasons. Herbivory also varies seasonally and can affect the net outcome of plant-plant interactions, but the interactive effects of seasonality and herbivory on the competition-facilitation balance are not known. The Puerto Rican terrestrial frog (Eleutherodactylus coqui) has received considerable attention in Hawaii because of its rapid spread, loud mating calls, and its potential threat to native species. Thus far, its invasion potential on the Island of Hawaii remains poorly understood. Critical components for determining this potential are robust estimates of abundance and vital rates across habitat types. To address this lack of information, we used mark-recapture methods to estimate E. coqui survival and abundance, determine growth rates of adult male and female frogs, and relate densities to elevation, snout–vent length (SVL), habitat structure, and invertebrate abundance.
