Fabrication And Characterization Of Molecular Spintronics Devices, Pawan Tyagi Jan 2008

Fabrication And Characterization Of Molecular Spintronics Devices, Pawan Tyagi

University of Kentucky Doctoral Dissertations

Fabrication of molecular spin devices with ferromagnetic electrodes coupled with a high spin molecule is an important challenge. This doctoral study concentrated on realizing a novel molecular spin device by the bridging of magnetic molecules between two ferromagnetic metal layers of a ferromagnetic-insulator-ferromagnetic tunnel junction on its exposed pattern edges. At the exposed sides, distance between the two metal electrodes is equal to the insulator film thickness; insulator film thickness can be precisely controlled to match the length of a target molecule. Photolithography and thin-film deposition were utilized to produce a series of tunnel junctions based on molecular electrodes of …

Colloid Mediated Transport Of Heavy Metals In Soils Following Reclamation With And Without Biosolid Application, Jarrod O. Miller Jan 2008

Colloid Mediated Transport Of Heavy Metals In Soils Following Reclamation With And Without Biosolid Application, Jarrod O. Miller

University of Kentucky Doctoral Dissertations

Soils disturbed by strip mining practices may have increased colloid loads moving to groundwater resources, also enhancing the transport of contaminants into our water resources. We hypothesize that contaminant transport within soils following mining is enhanced by colloid mobility. Two sites were chosen for this study, a 30-year old reclaimed strip mine in southwest Virginia and a recently mined area from eastern Kentucky. Intact reclaimed soil monoliths were retrieved from sandstone derived soils in southwestern Virginia. Reclaimed monoliths from eastern Kentucky were recreated in the lab. Intact undisturbed (native) soil monoliths representing the soils before mining were also sampled for …

Algebraic Properties Of Edge Ideals, Rachelle R. Bouchat Jan 2008

Algebraic Properties Of Edge Ideals, Rachelle R. Bouchat

University of Kentucky Doctoral Dissertations

Given a simple graph G, the corresponding edge ideal IG is the ideal generated by the edges of G. In 2007, Ha and Van Tuyl demonstrated an inductive procedure to construct the minimal free resolution of certain classes of edge ideals. We will provide a simplified proof of this inductive method for the class of trees. Furthermore, we will provide a comprehensive description of the finely graded Betti numbers occurring in the minimal free resolution of the edge ideal of a tree. For specific subclasses of trees, we will generate more precise information including explicit formulas for …

Loyalists In War, Americans In Peace: The Reintegration Of The Loyalists, 1775-1800, Aaron N. Coleman Jan 2008

Loyalists In War, Americans In Peace: The Reintegration Of The Loyalists, 1775-1800, Aaron N. Coleman

University of Kentucky Doctoral Dissertations

After the American Revolution a number of Loyalists, those colonial Americans who remained loyal to England during the War for Independence, did not relocate to the other dominions of the British Empire. Instead, they sought to return to their homes and restart their lives. Despite fierce opposition to their return from all across the Confederation, their attempts to become part of a newly independent America were generally successful. Thus, after several years of struggle most former Loyalists who wanted to return were able to do so.

Various studies have concentrated on the wartime activities of Loyalists, but few have examined …

Design, Synthesis, And Pharmacological Evaluation Of A Series Of Novel, Guanidine And Amidine-Containing Neonicotinoid-Like Analogs Of Nicotine: Subtype-Selective Interactions At Neuronal Nicotinic-Acetylcholine Receptor., Aaron Joseph Haubner Jan 2008

Design, Synthesis, And Pharmacological Evaluation Of A Series Of Novel, Guanidine And Amidine-Containing Neonicotinoid-Like Analogs Of Nicotine: Subtype-Selective Interactions At Neuronal Nicotinic-Acetylcholine Receptor., Aaron Joseph Haubner

University of Kentucky Doctoral Dissertations

The current project examined the ability of a novel series of guandine and amidine-containing nicotine analogs to interact with several native and recombinantlyexpressed mammalian neuronal nicotinic-acetylcholine receptor (nAChR) subtypes. Rational drug design methods and parallel organic synthesis was used to generate a library of guanidine-containing nicotine (NIC) analogs (AH compounds). A smaller series of amidine-containing nicotine analogs (JC compounds) were also synthesized. In total, >150 compounds were examined. Compounds were first assayed for affinity in a high-throughput [3H]epibatidine radioligand-binding screen. Lead compounds were evaluated in subtype-selective binding experiments to probe for affinity at the α4β2 …

The H-Vectors Of Matroids And The Arithmetic Degree Of Squarefree Strongly Stable Ideals, Erik Stokes Jan 2008

The H-Vectors Of Matroids And The Arithmetic Degree Of Squarefree Strongly Stable Ideals, Erik Stokes

University of Kentucky Doctoral Dissertations

Making use of algebraic and combinatorial techniques, we study two topics: the arithmetic degree of squarefree strongly stable ideals and the h-vectors of matroid complexes.

For a squarefree monomial ideal, I, the arithmetic degree of I is the number of facets of the simplicial complex which has I as its Stanley-Reisner ideal. We consider the case when I is squarefree strongly stable, in which case we give an exact formula for the arithmetic degree in terms of the minimal generators of I as well as a lower bound resembling that from the Multiplicity Conjecture. Using this, we can …

Agglomeration Economies And Geographic Concentration Of Manufacturing In Ukraine, Volodymyr Vakhitov Jan 2008

Agglomeration Economies And Geographic Concentration Of Manufacturing In Ukraine, Volodymyr Vakhitov

University of Kentucky Doctoral Dissertations

As a post-Soviet economy, Ukraine has inherited substantial production assets and qualified personnel. However, the economy was dominated by large-scale enterprises designed for much bigger markets. After the collapse of the Soviet Union Ukrainian firms faced lack of planning, breaks in contacts with their former suppliers and customers, and distortion of prices. There was a clear need in restructuring of the entire economy. Restructuring included splitting firms into smaller parts and privatization. The first phase of transition was completed by 2000 when the output grew for the first time after a long recession in nineties, and most firms became private …

Efficient And Scalable Network Security Protocols Based On Lfsr Sequences, Saikat Chakrabarti Jan 2008

Efficient And Scalable Network Security Protocols Based On Lfsr Sequences, Saikat Chakrabarti

University of Kentucky Doctoral Dissertations

The gap between abstract, mathematics-oriented research in cryptography and the engineering approach of designing practical, network security protocols is widening. Network researchers experiment with well-known cryptographic protocols suitable for different network models. On the other hand, researchers inclined toward theory often design cryptographic schemes without considering the practical network constraints. The goal of this dissertation is to address problems in these two challenging areas: building bridges between practical network security protocols and theoretical cryptography. This dissertation presents techniques for building performance sensitive security protocols, using primitives from linear feedback register sequences (LFSR) sequences, for a variety of challenging networking applications. …

Endothelial Cell Dysfunction By Environmental Contaminants, Elizabeth Grace Oesterling Jan 2008

Endothelial Cell Dysfunction By Environmental Contaminants, Elizabeth Grace Oesterling

University of Kentucky Doctoral Dissertations

Within the last few decades, epidemiological evidence has linked exposure to air pollution, both its particles and its organic components, with cardiovascular disease (CVD) progression. CVD is a life long disease with the disruption of the endothelium being the inaugural event in this inflammatory process. The vascular endothelium is extremely susceptible to environmental insults given its tremendous surface area and that it is in constant contact with blood and components circulating within the blood, including xenobiotics. The endothelium is important as a barrier from blood constituents however, dysfunction of this barrier leads to the influx of lymphocytes and granulocytes that …

Charge Density Wave Polarization Dynamics, Luis Alejandro Ladino Gaspar Jan 2008

Charge Density Wave Polarization Dynamics, Luis Alejandro Ladino Gaspar

University of Kentucky Doctoral Dissertations

We have studied the charge density wave (CDW) repolarization dynamics in blue bronze (K0.3MoO3) by applying symmetric bipolar square-wave voltages of different frequencies to the sample and measuring the changes in infrared transmittance, proportional to CDW strain. The frequency dependence of the electro-transmittance was fit to a modified harmonic oscillator response and the evolution of the parameters as functions of voltage, position, and temperature are discussed. We found that resonance frequencies decrease with distance from the current contacts, indicating that the resulting delays are intrinsic to the CDW with the strain effectively flowing from the contact. …

Metabolite Analysis Of Clostridium Thermocellum Using Capillary Electrophoresis Based Techniques, Anup P. Thakur Jan 2008

Metabolite Analysis Of Clostridium Thermocellum Using Capillary Electrophoresis Based Techniques, Anup P. Thakur

University of Kentucky Doctoral Dissertations

Clostridium thermocellum is a thermophilic bacterium that converts biomass to ethanol directly; however, high sensitivity of this bacterium toward ethanol limits its commercial utility. To elucidate the effect of ethanol on the growth of this bacterium a metabolite analysis of C. thermocellum was performed. The hypothesis of the project was that exogenous ethanol alters the metabolite profile of C. thermocellum. For metabolite analysis, capillary electrophoresis-electrospray ionization-mass spectrometry method (CE-ESI-MS) was developed due to highly polar and charged nature of metabolites. To increase the sensitivity of CE-ESI-MS, several parameters at the ESI interface were optimized. The application of 50% isopropanol …

University of Kentucky Doctoral Dissertations

The biology of seed dormancy and germination of 46 species representing 11 of the 12 tribes in Convolvulaceae were compared in laboratory (mostly), field and greenhouse experiments. Seeds were tested for kind of dormancy and storage behavior; artificial or simulated natural treatments were applied to break physical dormancy (PY); the initial route of water entry (“water gap”) into seeds was identified; the morphoanatomy of the water gap was compared in seeds of 17 species; ontogenetical differences between water gap and seed coat away from the hilum were described in Ipomoea lacunosa seeds; cycling of sensitivity to dormancy break was elucidated …

Gentrification Moves To The Global South: An Analysis Of The Programa De Rescate, A Neoliberal Urban Policy In México City's Centro Histórico, David M. Walker Jan 2008

Gentrification Moves To The Global South: An Analysis Of The Programa De Rescate, A Neoliberal Urban Policy In México City's Centro Histórico, David M. Walker

University of Kentucky Doctoral Dissertations

This dissertation argues that urban neoliberal programs currently formulating in the Global South are unprecedented in historical México as well as in examined practices of gentrification and globalization. In this dissertation I specifically focus on the Programa de Rescate – an urban policy being amassed in México City’s Centro Histórico as a nexus of processes of gentrification, neoliberalization, and globalization. This work re-theorizes how gentrification functions when it is implemented in the Global South – as the neoliberalization of space.

Regulation Of L-Type Voltage-Dependnet Calcium Channels By The Rem Gtpase, Chunyan Pang Jan 2008

Regulation Of L-Type Voltage-Dependnet Calcium Channels By The Rem Gtpase, Chunyan Pang

University of Kentucky Doctoral Dissertations

The Rem, Rem2, Rad, and Gem/Kir GTPases, comprise a novel subfamily of the small Ras-related GTP-binding proteins known as the RGK GTPases, and have been shown to function as potent negative regulators of high voltage-activated (HVA) Ca2+ channels upon overexpression. HVA Ca2+ channels modulate Ca2+ influx in response to membrane depolarization to regulate a wide variety of cellular functions and they minimally consist of a pore-forming α1 subunit, an intracellular β subunit, and a transmembrane complex α2/δ subunit. While the mechanisms underlying RGK-mediated Ca2+ channel regulation remain poorly defined, it appears that both membrane …

Role Of Reactive Oxygen Species Peroxynitrite In Traumatic Spinal Cord Injury, Yiqin Xiong Jan 2008

Role Of Reactive Oxygen Species Peroxynitrite In Traumatic Spinal Cord Injury, Yiqin Xiong

University of Kentucky Doctoral Dissertations

Peroxynitrite (PN, ONOO-), formed by nitric oxide radical (•NO) and superoxide radical (O2•-), plays an important role in post-traumatic oxidative damage. In the early work, we determined the temporal characteristics of PN-derived oxidative damage in a rat spinal cord injury (SCI) model. Our results showed 3-nitrotyrosine (3-NT), a specific marker for PN, rapidly accumulated at early time points (1 hr, 3 hrs), after when it plateaued and the high level was sustained to 1 week post injury. The co-localization of 3-NT and lipid peroxidation derived-4-HNE observed in immunohistochemistry indicates PN is involved in lipid peroxidative as …

Thermal Sensitivity Of Vagal Pulmonary Sensory Neurons: Role Of Transient Receptor Potential Vanilloid Channels, Dan Ni Jan 2008

Thermal Sensitivity Of Vagal Pulmonary Sensory Neurons: Role Of Transient Receptor Potential Vanilloid Channels, Dan Ni

University of Kentucky Doctoral Dissertations

Hyperthermia can occur in lungs and airways during both physiological and pathophysiological conditions. A previous study carried out in our laboratory showed that hyperthermia activates and sensitizes vagal bronchopulmonary Cfiber afferents, whether this effect is through a direct action of hyperthermia on sensory nerves is not known. This dissertation study was aimed to investigate the thermal-sensitivity of pulmonary sensory neurons, and the roles of thermalsensitive transient receptor potential vanilloid (TRPV) channels. Whole-cell patch-clamp recordings of neurons isolated from nodose/jugular ganglia were applied in the study. Results of this study showed that hyperthermia directly activates pulmonary sensory neurons, and this effect …

Genetic Regulation Of Hematopoietic Stem Cell Aging, Erin J. Oakley Jan 2008

Genetic Regulation Of Hematopoietic Stem Cell Aging, Erin J. Oakley

University of Kentucky Doctoral Dissertations

It is well documented that both quantitative and qualitative changes in the murine hematopoietic stem cell (HSC) population occur with age. In mice, the effect of aging on stem cells is highly strain-specific, thus suggesting genetic regulation plays a role in HSC aging. In C57BL/6 (B6) mice, the HSC population steadily increases with age, whereas in DBA/2 (D2) mice, this population declines. Our lab has previously mapped a quantitative trait locus (QTL) to murine chromosome 2 that is associated with the variation in frequency of HSCs between aged B6 and D2 mice. In these dissertation studies, I first aim to …

A Systematic Study Of Thermodynamic And Transport Properties Of Layered CaN+1(Ru1-XCrX)NO3n+1, Vinobalan Durairaj Jan 2008

A Systematic Study Of Thermodynamic And Transport Properties Of Layered CaN+1(Ru1-XCrX)NO3n+1, Vinobalan Durairaj

University of Kentucky Doctoral Dissertations

Orbital degrees of freedom play vital role in prompting novel phenomena in ruthenium based Ruddlesden-Popper compounds through coupling of orbits to spin and lattice. Physical properties are then particularly susceptible to small perturbations by external magnetic fields and/or slight structural changes. Current study pertains to the impact when a more-extended 4d Ruthenium ion is replaced by a less-extended 3d Chromium ion.

Perovskite CaRuO3 (n=∞) is characterized by borderline magnetism and non- Fermi liquid behavior – common occurrences in quantum critical compounds. Remarkably, Cr substitution as low as x=0.05 abruptly drives CaRu1−xCrxO3 from a paramagnetic …

Nitration And Inactivation Of Manganese Superoxide Dismutase Plays A Critical Role In Metabolic Switch, Muthuswamy Anantharaman Jan 2008

Nitration And Inactivation Of Manganese Superoxide Dismutase Plays A Critical Role In Metabolic Switch, Muthuswamy Anantharaman

University of Kentucky Doctoral Dissertations

Alzheimer’s disease (AD) is a multifactorial, progressive, age-related neurodegenerative disease. Oxidative stress hypothesis is most prevalent and is gaining significant support. Inspite of the progress achieved on oxidative stress related damages in AD brain; the modification occurring on the various cellular antioxidant enzymes antioxidant has not been identified. Tyrosine nitration, a marker for peroxynitrite induced oxidative damage to protein is widespread in AD brain and Manganese superoxide dismutase (MnSOD), primary mitochondrial antioxidant enzyme is prone to peroxynitrite induced nitration and inactivation. Nitration of proteins involved in energy metabolism has been demonstrated in AD brain, which may explain the altered glucose …

Multiscale Dynamic Monte Carlo / Continuum Modeling Of Drying And Curing In Sol-Gel Silica Films, Xin Li Jan 2008

Multiscale Dynamic Monte Carlo / Continuum Modeling Of Drying And Curing In Sol-Gel Silica Films, Xin Li

University of Kentucky Doctoral Dissertations

Modeling the competition between drying and curing processes in polymerizing films is of great importance to many existing and developing materials synthesis processes. These processes involve multiple length and time scales ranging from molecular to macroscopic, and are challenging to fully model in situations where the polymerization is non-ideal, such as sol-gel silica thin film formation. A comprehensive model of sol-gel silica film formation should link macroscopic flow and drying (controlled by process parameters) to film microstructure (which dictates the properties of the films).

This dissertation describes a multiscale model in which dynamic Monte Carlo (DMC) polymerization simulations are coupled …

Role Of The Reactive Oxygen Species Peroxynitrite In Traumatic Brain Injury, Ying Deng Jan 2008

Role Of The Reactive Oxygen Species Peroxynitrite In Traumatic Brain Injury, Ying Deng

University of Kentucky Doctoral Dissertations

Reactive oxygen species (ROS) is cytotoxic to the cell and is known to contribute to secondary cell death following primary traumatic brain injury (TBI). We described in our study that PN is the main mediator for both lipid peroxidation and protein nitration, and occurred almost immediately after injury. As a downstream factor to oxidative damage, the peak of Ca2+-dependent, calpainmediated cytoskeletal proteolysis preceded that of neurodegeneration, suggesting that calpain-mediated proteolysis is the common pathway leading to neuronal cell death. The time course study clearly elucidated the interrelationship of these cellular changes following TBI, provided window of opportunity for pharmacological intervention. …

Surfactant And Metal Sorption Studies By Functionalized Membranes And Quartz Crystal Microbalance, Abhay R. Ladhe

University of Kentucky Doctoral Dissertations

Functionalized membranes provide an elegant platform for selective separations and sorptions. In this dissertation, application of functionalized membranes for surfactant and metal sorption studies are discussed. Sorption behavior of surfactants is also studied using quartz crystal microbalance (QCM) and other techniques.

Adsorption of the ethoxylated surfactants on polymeric materials (cotton and polyester) and model gold surface was quantified from a non-aqueous siloxane based solvent (D5) and water. The role of ethylene oxide group and the effect of nature of polymeric materials on adsorption behavior was quantified and established. In the case of gold-water interface, the adsorption data was fitted to …

The Underlying Mechanism(S) Of Fasting Induced Neuroprotection After Moderate Traumatic Brain Injury, Laurie Michelle Helene Davis Jan 2008

The Underlying Mechanism(S) Of Fasting Induced Neuroprotection After Moderate Traumatic Brain Injury, Laurie Michelle Helene Davis

University of Kentucky Doctoral Dissertations

Traumatic brain injury (TBI) is becoming a national epidemic, as it accounts for 1.5 million cases each year. This disorder affects primarily the young population and elderly. Currently, there is no treatment for TBI, which means that ~2% of the U.S. population is currently living with prolonged neurological damage and dysfunction. Recently, there have been many studies showing that TBI negatively impacts mitochondrial function. It has been proposed that in order to save the cell from destruction mitochondrial function must be preserved. The ketogenic diet, originally designed to mimic fasting physiology, is effective in treating epilepsy. Therefore, we have used …

Empirical Processes And Roc Curves With An Application To Linear Combinations Of Diagnostic Tests, Costel Chirila Jan 2008

Empirical Processes And Roc Curves With An Application To Linear Combinations Of Diagnostic Tests, Costel Chirila

University of Kentucky Doctoral Dissertations

The Receiver Operating Characteristic (ROC) curve is the plot of Sensitivity vs. 1- Specificity of a quantitative diagnostic test, for a wide range of cut-off points c. The empirical ROC curve is probably the most used nonparametric estimator of the ROC curve. The asymptotic properties of this estimator were first developed by Hsieh and Turnbull (1996) based on strong approximations for quantile processes. Jensen et al. (2000) provided a general method to obtain regional confidence bands for the empirical ROC curve, based on its asymptotic distribution.

Since most biomarkers do not have high enough sensitivity and specificity to …

Effect Of The SmartstepTm Stabilization System On Balance In Older Adults In An Independent Living Residence, Ann L. Livengood Jan 2008

Effect Of The SmartstepTm Stabilization System On Balance In Older Adults In An Independent Living Residence, Ann L. Livengood

University of Kentucky Doctoral Dissertations

An increase in postural sway is one of the risk factors that have been linked to an increased incidence of falls in the older adult population. Researchers have shown that peripheral sensation is crucial in maintaining a static posture for adults of all ages. It has been reported that older adults have decreased tactile sensation of the plantar surface of their feet. and when the sensory feedback was increased older adults had improved postural control. It was hypothesized that facilitation of the sole of the foot with the use of a semirigid foot orthotic would result in improved postural stability …

Matrix Decomposition For Data Disclosure Control And Data Mining Applications, Jie Wang Jan 2008

Matrix Decomposition For Data Disclosure Control And Data Mining Applications, Jie Wang

University of Kentucky Doctoral Dissertations

Access to huge amounts of various data with private information brings out a dual demand for preservation of data privacy and correctness of knowledge discovery, which are two apparently contradictory tasks. Low-rank approximations generated by matrix decompositions are a fundamental element in this dissertation for the privacy preserving data mining (PPDM) applications. Two categories of PPDM are studied: data value hiding (DVH) and data pattern hiding (DPH). A matrix-decomposition-based framework is designed to incorporate matrix decomposition techniques into data preprocessing to distort original data sets. With respect to the challenge in the DVH, how to protect sensitive/confidential attribute values without …

A Precision Instrument For Research Into Nanolithographic Techniques Using Field-Emitted Electron Beams, King-Fu Hii

University of Kentucky Doctoral Dissertations

Nanomanufacturing is an active research area in academia and industry due to the ever-growing demands for precision surface modifications of thin films or substrates with nanoscale features. Conventional lithographic techniques face many challenges as they approach their fundamental limits. Consequently, new nanomanufacturing tools, fabrication techniques, and precision instruments are being explored and developed to meet these challenges. It has been hypothesized that direct-write nanolithography might be achieved by using a field-emitted electron beam for nanomachining. This dissertation moves this research one step closer by developing a precision instrument that can enable the integration of direct-write nanolithography by a field-emitted electron …

The Role Of Spiders In The Detrital Food Web Of An Eastern Deciduous Forest, Erin Elizabeth Hladilek Jan 2008

The Role Of Spiders In The Detrital Food Web Of An Eastern Deciduous Forest, Erin Elizabeth Hladilek

University of Kentucky Doctoral Dissertations

Historically, terrestrial food web research has focused on describing the structure of aboveground grazing webs, and determining how interactions among plants, herbivores and higher trophic levels influence primary productivity. Detrital food webs however, play a significant role in regulation of ecosystem dynamics through direct impacts on decomposition. Unraveling the complex nature of detrital food web structure is critical to developing a better understanding of ecosystem function. Therefore the primary objective of this research was to describe the structure of the leaf-litter food web in a temperate deciduous forest, with emphasis on interactions between a community of generalist predators, the forest-floor …

A Novel Class Of Immunoproteasome Catalytic Subunit Lmp2 Inhibitor And Its Therapeutic Potentials In Cancer, Yik Khuan (Abby) Ho Jan 2008

A Novel Class Of Immunoproteasome Catalytic Subunit Lmp2 Inhibitor And Its Therapeutic Potentials In Cancer, Yik Khuan (Abby) Ho

University of Kentucky Doctoral Dissertations

The immunoproteasome, known to play an important role in MHC class I antigen processing and presentation, have been linked to neurodegenerative diseases and hematological cancers. However, the pathophysiological functions of the immunoproteasome in these diseases are still not very well established. This can be attributed mainly to the lack of appropriate molecular probes that selectively target the immunoproteasome catalytic subunits. Herein, we report the development of a small molecular inhibitor (AM) that selectively targets the major catalytic subunit, LMP2, of the immunoproteasome. We show that the compound covalently modifies the LMP2 subunit with high specificity in human prostate …

Linking The Structure And Mechanical Behavior Of Nanoporous Gold, Ye Sun

University of Kentucky Doctoral Dissertations

The structure of nanoporous gold (np-Au) provides a very limited volume for deformation to occur, and thus offers an opportunity to study the role of defects such as dislocations in nanoscale metal volumes. A practical goal is to understand mechanical properties of np-Au so that it can be can produced in stable form, for use in applications that require some mechanical integrity. Bulk and thin film np-Au have been fabricated and studied here.

Bulk np-Au was prepared by electrochemically dealloying Au-Ag alloys with 25 and 30 at.% Au. In the lower Au content material, cracks always formed during dealloying. When …
