Identification of a reliable marker of skeletal precursor cells within calcified and soft tissues remains a major challenge for the field. To address this, we used a transgenic model in which osteoblasts can be eliminated by pharmacological treatment. Following osteoblast ablation a dramatic increase in a population of α-smooth muscle actin (α-SMA) positive cells was observed. During early recovery phase from ablation we have detected cells with the simultaneous expression of SMAA and a preosteoblastic 3.6GFP marker, indicating the potential for transition of α-SMA+ cells towards osteoprogenitor lineage. Based on remarkable success of PTH as an anabolic drug for osteoporosis, case reports of off-label use of teriparatide (1-34 PTH) in patients with complicated fractures and non-unions are emerging. We investigated the mechanisms underlying PTH accelerated fracture repair. Bone marrow cells from 7 days 40cg/kg of teriparatide treated or saline control mice were cultured and Osx and osteoblast phenotypic gene expression assessed by real time RT-PCR in these cells. Fractured animals injected daily with either saline or 40cg/kg of teriparatide for up to 21 days were X-rayed and histological assessment performed, as well as immunohistochemical analyses of the Osx …

There is a growing need for cellular imaging with fluorescent probes that emit at longer wavelengths to minimize the effects of absorption, autofluorescence and scattering from biological tissue. In this paper a series of new environmentally-sensitive hemicyanine dyes featuring amino(oligo)thiophene donors have been synthesized via aldol condensation between a 4-methylpyridinium salt and various amino(oligo)thiophene carboxaldehydes, which were, in turn, obtained from amination of bromo (oligo)thiophene carboxaldehyde. Side chains on these fluorophores impart a strong affinity for biological membranes. Compared with benzene analogues, these thiophene fluorophores show significant red shift in the absorption and emission spectra, offering compact red and near-infrared …

Bundling of rapidly polymerizing actin filaments underlies dynamics of filopodial protrusions that play an important role in cell migration and cell-cell interaction. Recently, formation of actin bundles has been reconstituted in vitro and two scenarios of bundle initiation, involving binding of two filament tips and, alternatively, linking of the tip of one filament to the side of the other, -have been discussed. A first theoretical analysis is presented indicating that the two mechanisms can be distinguished experimentally. While both of them result counter-intuitively in comparable numbers of bundles, these numbers scale differently with the average bundle length. We propose an …

A Crucial Role In Cell Spreading For The Interaction Of Abl Pxxp Motifs With Crk And Nck Adaptors, Susumu Antoku, Gonzalo M. Rivera, Bruce J. Mayer

The dynamic reorganization of actin structures helps to mediate the interaction of cells with their environment. The Abl non-receptor tyrosine kinase can modulate actin rearrangement during cell attachment. Here we report that the Abl PxxP motifs, which bind Src homology 3 (SH3) domains, are indispensable for the coordinated regulation of filopodium and focal adhesion formation and cell-spreading dynamics during attachment. Candidate Abl PxxP-motif-binding partners were identified by screening a comprehensive SH3-domain phage-display library. A combination of protein overexpression, silencing, pharmacological manipulation and mutational analysis demonstrated that the PxxP motifs of Abl exert their effects on actin organization by two distinct …

This study had three aims: to determine the prevalence of the risk-factors for contracting HPV (the Human Papillomavirus) and developing cervical cancer among young women; to establish if there are any links between the presence of these risk-factors, attendance for cervical screening and abnormal cervical screening results; and to ascertain the key barriers to the prevention of cervical cancer. The risk-factors were identified from literature as being sexually active at a young age, having increasing numbers of sexual partners for females and their partners, having had a sexually transmitted infection/disease (STI), smoking and long-term use of the oral contraceptive pill. …

A science education center at a university medical school had grant funding to develop a genetics curriculum unit, but needed a dissemination plan. A statewide science teacher organization that provided professional development training was facing decreased funding. These two groups combined their efforts, and created a unique partnership, called BEGIN (Biotechnology, Ethics and Genetics Instructional Network) that has brought together university medical and science faculty and high school biology teachers. A comprehensive literature review and modeling effort have been conducted in order to determine which vital rates are most important to determining the growth and sustainability of marine mammal populations. Also addressed are the impacts of life-history, ecological, and genetic variation on vital rates and population sustainability and how much each vital parameter can change before a change in population trend would be expected. Additionally, the influence of ecological energetics and foraging strategies on vital rates and their limits of sustainable change are examined, and the nature of how an increase in sound in the marine environment might influence marine …

Population Structure Of Coyote (Canis Latrans) In The Urban Landscape Of The Cleveland, Ohio Area, Rebecca M. Rashleigh, Robert A. Krebs, Harry Van Keulen

To obtain information on the population structure of coyote (Canis latrans) in an urban setting, a non-invasive genetic sampling technique was applied that consisted of DNA isolation from scat collected around the Cleveland metropolitan area. Muscle tissue was provided from two other coyotes. Amplifying the mitochondrial D-loop or control region produced 33 haplotypes from just 57 coyote sequences and two additional dog sequences from putative coyote-dog hybrids. The mitochondrial DNA genetic diversity in the Cleveland area was high, π 0.02, and composed of six distinct haplotype lineages. A New Species Of Cecidomyia (Diptera: Cecidomyiidae) Feeding On Resin Of Baldcypress, Raymond Gagne, Kenneth Hibbard

Cecidomyia lamellate Gagné, new species, is described from adults of both sexes, pupae, and larvae taken from resin in branchlet swellings of baldcypress, Taxodium distichum (L.) Rich. (Taxodiaceae), in Fort Pierce, Florida. Some anatomical characters of the new species depart from the previously known limits of the genus. Baldcypress is the first known non- Pinaceae host of Cecidomyia.

As an innovative community-based framework for science learning, the Big Sky Model is guiding high school and tribal college students from rural areas of Montana and Idaho in their understanding of chemical, physical, and environmental health concepts in the context of their own homes, schools, and communities. Students participate in classroom lessons and continue with systematic inquiry through actual field research to investigate a pressing, real-world issue: understanding the complex links between poor air quality and respiratory health outcomes. Changes in the envelope proteins of retroviruses can alter the ability of these viruses to infect the central nervous system (CNS) and induce neurological disease. In the present study, nine envelope residues were found to influence neurovirulence of the Friend murine polytropic retrovirus Fr98. When projected on a three-dimensional model, these residues were clustered in two spatially separated groups, one in variable region B of the receptor binding site and the other on the opposite side of the envelope. Further studies indicated a role for these residues in virus replication in the CNS, although the residues did not affect viral …

The study evaluates the role of the Senior Traveller Training Centres regarding further education. The centres provide basic compensatory education for Travellers aged 18 upwards. The aim of the centres is to provide Travellers with the knowledge, skills and attitudes required to successfully make the transition to work and adult life. It examines the rationale for the centres, the management structure, why people decide to go to the centres, and the curriculum. The study also highlights the difficulties the Traveller community have faced in the past and are still facing today in terms of equality of outcomes in education. Travellers …

The substantive aim of this study was to examine the adaptation of young-adult‟s to college life in Ireland today. The study also assessed the participant‟s levels of individuation-attachment and its consequences for college adjustment. A number of personal variables were examined in relation to both the participants‟ college adjustment and individuation-attachment, with gender being of significant interest. Data were collected from a sample of 125 (78 females and 47 males) first-year college students attending a number of Dublin based institutions. Participants‟ completed quantitative measures of college adjustment and individuation-attachment. Findings indicated that overall student adjustment was positive, with levels of …

“The Mother Church” analyzes the influence of literary sentimentalism on the writings and doctrine of Mary Baker Eddy, the founder of Christian Science. Having attempted a career as a sentimental poet in her early life, Eddy imported sentimental notions of motherhood and parent-child separation into Christian Science belief and iconography.
