Adaptive Management Report For The Clark County, Nevada Multiple Species Habitat Conservation Plan, Desert Conservation Program

Publications (C)

The Desert Conservation Program administers an incidental take permit for desert tortoise and 77 other species. The permit was issued in 2001 by the United States Fish and Wildlife Service in accordance with the Endangered Species Act. Compliance with the permit requires implementation of the Clark County Multiple Species Habitat Conservation Plan. This plan requires an adaptive management program and a biennial report by an independent science advisor to assess the status of the plan and make recommendations for future funding. The 2008 Adaptive Management Report reviews current data on land use trends, habitat loss, species status, plan implementation, programmatic …

Determining The Usefulness Of Aerobic And Anaerobic Enzyme Assays As Proxies For Rockfish Ecological Data., Erica M. Hudson Oct 2008

Determining The Usefulness Of Aerobic And Anaerobic Enzyme Assays As Proxies For Rockfish Ecological Data., Erica M. Hudson

USF Tampa Graduate Theses and Dissertations

Rockfish are commercially and recreationally important, yet due to the in habitat depths at which rockfish inhabit, little is known about their ecology. As a consequence, management of rockfish population as a fishery resource is a work in progress. In particular, changes in physiological condition aver the course of the year is poorly described. This study examined 19 different species of Sebastes from the Southern California Bight over four seasons (late summer, fall, winter, and spring) using metabolic enzyme assays. Enzymes used were lactate dehydrogenase (LDH), malate dehydrogenase (MDH), pyruvate kinase (PK), and citrate synthase (CS). Some muscle composition data …

Multilevel Latent Class Models With Dirichlet Mixing Distribution, Chongzhi Di, Karen Bandeen-Roche Oct 2008

Multilevel Latent Class Models With Dirichlet Mixing Distribution, Chongzhi Di, Karen Bandeen-Roche

Johns Hopkins University, Dept. of Biostatistics Working Papers

Latent class analysis (LCA) and latent class regression (LCR) are widely used for modeling multivariate categorical outcomes in social sciences and biomedical studies. Standard analyses assume data of different respondents to be mutually independent, excluding application of the methods to familial and other designs in which participants are clustered. In this paper, we develop multilevel latent class model, in which subpopulation mixing probabilities are treated as random effects that vary among clusters according to a common Dirichlet distribution. We apply the Expectation-Maximization (EM) algorithm for model fitting by maximum likelihood (ML). This approach works well, but is computationally intensive when …

High-Activity Mutants Of Butyrylcholinesterase For Cocaine Hydrolysis And Method Of Generating The Same, Chang-Guo Zhan, Hoon Cho, Hsin-Hsiung Tai

Pharmaceutical Sciences Faculty Patents

A novel computational method and generation of mutant butyrylcholinesterase for cocaine hydrolysis is provided. The method includes molecular modeling a possible BChE mutant and conducting molecular dynamics simulations and hybrid quantum mechanical/molecular mechanical calculations thereby providing a screening method of possible BChE mutants by predicting which mutant will lead to a more stable transition state for a rate determining step. Site-directed mutagenesis, protein expression, and protein activity is conducted for mutants determined computationally as being good candidates for possible BChE mutants, i.e., ones predicted to have higher catalytic efficiency as compared with wild-type BChE. In addition, mutants A199S/A328W/Y332G, A199S/F227A/A328W/Y332G, A199S/S287G/A328W/Y332G, …

A Schiff Base Connectivity Switch In Sensory Rhodopsin Signaling, Oleg A Sineshchekov, Jun Sasaki, Brian J Phillips, John L Spudich Oct 2008

A Schiff Base Connectivity Switch In Sensory Rhodopsin Signaling, Oleg A Sineshchekov, Jun Sasaki, Brian J Phillips, John L Spudich

Faculty, Staff and Student Publications

Sensory rhodopsin I (SRI) in Halobacterium salinarum acts as a receptor for single-quantum attractant and two-quantum repellent phototaxis, transmitting light stimuli via its bound transducer HtrI. Signal-inverting mutations in the SRI-HtrI complex reverse the single-quantum response from attractant to repellent. Fast intramolecular charge movements reported here reveal that the unphotolyzed SRI-HtrI complex exists in two conformational states, which differ by their connection of the retinylidene Schiff base in the SRI photoactive site to inner or outer half-channels. In single-quantum photochemical reactions, the conformer with the Schiff base connected to the cytoplasmic (CP) half-channel generates an attractant signal, whereas the conformer …

Estimating Kinetic And Thermodynamic Parameters From Single Molecule Enzyme-Inhibitor Interactions, Laura Porter-Peden, Sarah G. Kamper, Mark Vander Wal, Ronald Blankespoor

University Faculty Publications and Creative Works

We report the application of recently developed microscopic models to estimate the apparent kinetic and thermodynamic parameters in a single molecule force spectroscopy study of the carbonic anhydrase enzyme and a complementary sulfonamide inhibitor. The most probable rupture force for the enzyme-inhibitor interaction shows a nonlinear dependency on the log-loading rate. Estimates for the kinetic and thermodynamic parameters were obtained by fitting the nonlinear dependency to linear cubic potential and cusp potential models and compared to the standard Bell-Evans model. The reliability of the estimated parameters was verified by modeling the experimental rupture force distributions by the theoretically predicted distributions …

Formal Reasoning And Spatial Ability: A Step Towards "Science For All", Bo Jiang

USF Tampa Graduate Theses and Dissertations

This work conducts an evaluation of a non-majors science curriculum named Molecules of Life (MOL) that aims to provide effective science education to undergraduate students who are not majoring in scientific disciplines.

As part of the process of developing an assessment plan for MOL, three related studies were undertaken in order to help us choose assessment instruments for MOL. The first study examined the validity of student evaluations of teaching. The second study investigated the Test of Logical Thinking (TOLT) and Group Assessment of Logical Thinking (GALT), two widely-used instruments for measuring formal reasoning ability. GALT is very similar to …

Transcriptional Response Of Mexican Axolotls To Ambystoma Tigrinum Virus (Atv) Infection, Jennifer D. Cotter, Andrew Storfer, Robert B. Page, Christopher K. Beachy, S. Randal Voss

Biology Faculty Publications

Background

Very little is known about the immunological responses of amphibians to pathogens that are causing global population declines. We used a custom microarray gene chip to characterize gene expression responses of axolotls (Ambystoma mexicanum) to an emerging viral pathogen, Ambystoma tigrinum virus (ATV).

Result

At 0, 24, 72, and 144 hours post-infection, spleen and lung samples were removed for estimation of host mRNA abundance and viral load. A total of 158 up-regulated and 105 down-regulated genes were identified across all time points using statistical and fold level criteria. The presumptive functions of these genes suggest a robust …

On Measuring Progress In Animal Welfare, James A. Serpell Oct 2008

On Measuring Progress In Animal Welfare, James A. Serpell

Assessment of Animal Welfare Collection

Improvements in the welfare of animals arise from two primary sources: Changes in public attitudes and beliefs regarding what constitutes acceptable treatment of animals, and mandatory changes in behavior toward animals imposed by governments and regulatory authorities. During the last 30-40 years, public and political support for improvements in animal welfare has increased dramatically in many countries. Funding from both governmental and non-governmental sources continues to grow, and the relatively new field of animal welfare science has emerged and expanded to support and inform these efforts. Many experts in the field recognize, however, that there is still a distinct gap …
