Glutamate is the major excitatory neurotransmitter in the brain. Among the cognate ionotropic glutamate receptors, the subfamily selective for AMPA (alpha-amino-3-hydroxy-5-methyl-4-isoxazole propionic acid) is responsible for most fast excitatory synaptic signaling and plays key roles in synaptic plasticity. AMPA receptors (AMPA-Rs) have also been implicated in a number of neurological disorders. To investigate subunit-specific differences in the ligand binding and activation of AMPA-Rs, the GluR4 AMPA-R ligand-binding domain (LBD) was crystallized in complex with full and partial agonists. Group I intron-derived ribozymes are catalytic RNAs that have been engineered to catalyze a variety of different reactions, in addition to the native self-splicing reaction. One such ribozyme, derived from a group I intron of Pneumocystis carinii, can modify RNA transcripts through either the excision or insertion of RNA sequences. These reactions are mediated through the trans excision-splicing (TES) or trans insertionsplicing (TIS) reaction pathways. Atherosclerosis is a chronic inflammatory disease thought to be initiated by endothelial cell dysfunction. Research described in this dissertation is focused on the role of zinc deficiency in endothelial cell activation with an emphasis on the function of the transcription factors nuclear factor-κB (NF-κB), peroxisome proliferator activated receptor (PPAR), and the aryl hydrocarbon receptor (AhR), which all play critical roles in the early pathology of atherosclerosis. The temperature induced melting transition of a self-complementary DNA strand covalently attached at the 5' end to the surface of a gold interdigitated microelectrode (GIME) was monitored in a novel, label-free, manner. The structural state of the hairpin was assessed by measuring four different electronic properties of the GIME (capacitance, impedance, dissipation factor and phase angle) as a function of temperature from 25 degrees C to 80 degrees C. The antifungal phoslactomycins (PLM A-F), produced by Streptomyes sp HK803, are structurally unusual in that three of their four double bonds are in the cis form (Δ12,13, Δ14,15, Δ2,3). The PLM polyketide synthase (PKS) has the predicted dehydratase catalytic domain in modules 1,2 and 5 required for establishing two of these cis double bonds (Δ12,13Δ14,15), as well as the only trans Δ6,7double bond. Educational resilience is an important term for teacher candidates in Christian institutions to understand. Resilience can be nurtured and instilled in the lives of individuals with learning disabilities, and teacher candidates can play a vital role in that process. To assist in this course of action, a look at the definition and common characteristics of resilience are discussed. In addition, a closer look at four resilient individuals with learning disabilities is provided. Concrete suggestions on ways to incorporate resilient characteristics into students with learning disabilities are shared.

Researchers studying the affects of resource loss following a technological disaster have exclusively investigated the acute period directly after the event occurred. This study applied Hobfoll's (1988, 1989) Conservation of Resources model in order to examine the long term effects of resource loss on depression in Cordova, Alaska a decade after the Exxon Valdez Oil spill. Results suggest that resource loss was a more prominent predictor for depression than demographics, involvement in the on-going litigation, or commercial fishing jobs. This study combined documentation of four boat propeller scar types in Halodule wrightii seagrass beds in Mosquito Lagoon, Florida with manipulative field experiments to document scar recovery times with and without restoration. Scar types ranged from the most severe scar type (Type 1) with trench formation which had no roots or shoots in the trench, to the least severe (Type 4) scars that had no depth, intact roots and shoots shorter than the surrounding canopy. Prevalence And Severity Of Fibropapillomatosis In Juvenile Green Turtles (Chelonia Mydas) In Three Habitats On Florida's Eas, Kelly Borrowman

Fibropapillomatosis (FP) is a tumor-forming disease mainly found in juvenile green turtles (Chelonia mydas) inhabiting Florida's east coast. Despite increased research on the herpes virus that putatively causes it, long-term assessment is still needed of the distribution and severity of FP. Using the decades-long database compiled by the University of Central Florida Marine Turtle Research Group, I determined FP severity and distribution at three different sites: Indian River Lagoon (IRL), Sabellariid Worm Reef (SWR) and Trident Submarine Basin (TSB). Fibropapillomatosis occurred in > 50% of IRL turtles, 18% of SWR turtles and <1% of TSB turtles. Regression of FP tumors was correlated with its prevalence, i.e. when and where FP was common, tumor regression was common. The probability of a turtle being non-afflicted or mildly afflicted increased with both increasing straight-line carapace length (SCL) and increasing relative body condition (residuals of log body mass versus log SCL). Mean annual growth rates of IRL and SWR turtles did not vary with FP severity; however, mean annual growth rates in IRL (1.10 cm/y) were significantly higher than in SWR (1.05 cm/y) and TSB (1.04 cm/y). Annual apparent survival estimates for IRL and TSB turtles were 0.72 and 0.73, respectively, and were constant over time. Even with increased prevalence of FP, annual apparent survival estimates were constant and similar to survival estimates of juvenile green turtles in areas without FP. In IRL, survival rates among FP Categories 0, 1 and 2 were similar (0.74, 0.74 and 0.81 respectively) and lower only for FP Category 3 (0.63). Stomata are cellular structures that control water loss and gas exchange through the plant epidermis. Stomata arise from special stem cells called meristemoids through a series of programmed asymmetric divisions that are controlled by cell signaling, or via multitude of regulatory pathways and intercellular communication between epidermal cells. In Arabidopsis thaliana, stomata are spaced non-randomly in the epidermis by cell-cell signaling of the receptor-like protein TOO MANY MOUTHS (TMM) as well as other proteins. Increased production of amyloid beta (A-beta) peptide, via altered proteolytic cleavage of amyloid protein precursor (APP), and abnormalities in neuronal calcium homeostasis play central roles in the pathogenesis of Alzheimer's disease (AD). Notch1, a membrane receptor that controls cell fate decisions during development of the nervous system, has been linked to AD because it is a substrate for the gamma-secretase protein complex in which mutations cause early-onset inherited AD. This study evaluated the effectiveness of a school-based training that attempted to improve the reliability of holistic scores teachers assigned to the writings of elementary school students. Seventeen teachers at one suburban elementary school located in the Southeastern United States participated in three training sessions that allowed for scoring practice and group discussions. The trainers, or presenters, were "faculty-experts." A comparison of scores the participants assigned to students' writings before and after the training was conducted. Using a single subject Multiple Baseline design, this study examined the efficacy of the Text Talk vocabulary acquisition strategy with regard to the use of new vocabulary by pre-kindergarten students in an inclusive classroom within a public charter school. The typical types of changes include sequence variations [single nucleotide polymorphisms (SNP)] and length variations [short tandem repeats (STR)]. The steps for forensic DNA sample processing are similar for both types of changes but diverge at the point of detection. This dissertation examined various configurations of display systems on several measures of operator performance, physiological states, and perceptions of the task. Today's best intelligent, adaptive, multimedia trainers have shown excellent performance; however, their results still fall far-short of what good human tutors can achieve. The overarching thesis of this paper is that future intelligent, adaptive systems will be improved by taking into account relevant, consistent, and meaningful individual differences. In combat, soldiers encounter stress from multiple sources including loss of sleep, extremely high levels of physical and psychological discomfort, extended periods of increased vigilance, and intense danger. Therefore, it is imperative to train such personnel on how to cope with these stressors.
