When Invasion Increases Population Genetic Structure: A Study With Centaurea Diffusa, Robin A. Marrs, René Sforza, Ruth A. Hufbauer

Biological invasions offer excellent systems to study the evolutionary processes involved in introductions of species to new ranges. Molecular markers can reveal invasion histories and the effects of introductions on amounts and structuring of genetic variation. We used five polymorphic microsatellite loci to elucidate genetic diversity and population structure between native range and introduced range populations of a prominent North American rangeland weed, Centaurea diffusa (Asteraceae). We found that the total number of alleles and the number of private alleles was slightly higher in the native Eurasian range, and that allelic richness did not differ between the ranges, indicating overall …

Sixty-eight families of USDA303 channel catfish Ictalurus punctatus were evaluated for growth performance for 30 d. The fastest- and slowest-growing catfish families were further evaluated to examine the hypothesis that genes or gene products associated with the growth regulatory and stress axes can be used to describe differences in growth performance. Research examined mRNA levels of genes involved in the growth hormone–insulin-like growth factor (GH–IGF) network in fast- (family A) and slow-growing (family H) USDA303 catfish. The traditional approach to pest management is to treat the crop or commodity in a particular management unit before an economically significant infestation of the pest has developed. Determining the need to take corrective action is based on the economic threshold concept, which forms the basis of most integrated pest management programmes (Metcalf and Luckman, 1975). Areawide pest management (AWPM) can be contrasted with traditional pest management in that pest management tactics are used over a broad spatial area, often treating the whole area simultaneously to maintain the pest below economic levels or, in some cases, completely eradicate it. Salt cedar (Tamarix spp. (Tamaricaceae: Tamaricales)) is a group of exotic shrubs to small trees that have invaded many riparian areas and lake shores across western North America. Of the 54 species known worldwide (Baum, 1967, 1968), ten species of salt cedars have been introduced into the USA (Crins, 1989), primarily from their countries of origin across Europe and Central Asia. They are also native in Africa and the Indian subcontinent of Asia. The overall goal of our research is to characterize and identify gene expression profiles of porcine hepatic cells. In this study, we have prepared two-dimensional electrophoresis maps of cytosol and membrane fractions from freshly prepared hepatocytes which were pooled from three crossbred pigs (35–69 kg). Following isoelectric focusing with three pH range immobilized pH gradient strips (pH 3–6, 5–8 and 7–10) and staining the second dimension gels with colloidal Coomassie blue, 728 protein spots were picked and digested with trypsin. Transient expression of engineered reporter RNAs encoding an intron-containing green fluorescent protein (GFP) from a Potato virus X-based expression vector previously demonstrated the nuclear targeting capability of the 359 nucleotide Potato spindle tuber viroid (PSTVd) RNA genome. To further delimit the putative nuclear-targeting signal, PSTVd subgenomic fragments were embedded within the intron, and recombinant reporter RNAs were inoculated onto Nicotiana benthamiana plants. Appearance of green fluorescence in leaf tissue inoculated with PSTVd-fragment-containing constructs indicated shuttling of the RNA into the nucleus by fragments as short as 80 nucleotides in length. The Hessian fly (Mayetiola destructor) is a gall-forming insect (Harris et al. 2006) with several attributes that make it suitable for genetic analysis: a short life cycle (30 days), a small genome (158 Mb) (Johnston et al. 2004), and polytene chromosomes. The reproductive biology and behavior of the insect also make it an attractive model (Harris and Rose 1989,1990; Bergh et al. 1990; Kanno and Harris 2000; Morris et al. 2000; Harris et al. 2001). There has been an increase in the use of formal approaches to environmental management. Jakeman and Letcher (2003) and Jakeman et al. (2006) have demonstrated the importance of integrated models as a means of assessing the response of environmental systems to proposed enlargement options. Gunderson and Holling (2000), Cowie and Borrett (2005), Curtis et al. (2005) and Pahl-Wostl (2005) have highlighted the need for the incorporation of social and institutional aspects into decision-making processes.

We investigated the effects of temperature and relative humidity on population growth and development of the psocid Lepinotus reticulatus Enderlein. Part of this study assessed the effects of marking psocids by using methylene blue, chalk powder, and fluorescent powder to differentiate nymphal stages during development. We found that marking psocids by using methylene blue increased mortality and took twice as long to accomplish compared with marking by using fluorescent powder. Using chalk powder shortened the duration of third and fourth nymphal instars. Marking psocids by using fluorescent powder had no effect on mortality or duration of nymphal instars. Therefore, we …

Background: Mycobacterium avium (M. avium) subspecies vary widely in both pathogenicity and host specificity, but the genetic features contributing to this diversity remain unclear.

Results: A comparative genomic approach was used to identify large sequence polymorphisms among M. avium subspecies obtained from a variety of host animals. DNA microarrays were used as a platform for comparing mycobacterial isolates with the sequenced bovine isolate M. avium subsp. paratuberculosis (MAP) K-10. Open reading frames (ORFs) were classified as present or divergent based on the relative fluorescent intensities of the experimental samples compared to MAP K-10 DNA. Florida enjoys 825 miles of sandy beaches. These beaches serve as nesting habitat for five species of threatened or endangered sea turtles. Florida’s beaches host the densest sea turtle nesting in the United States, the largest aggregation of loggerhead nesting in the world, and the second highest density of green sea turtle nesting in the hemisphere. Florida’s beaches also provide habitat for hundreds of other species as well. In addition to providing recreational and esthetic values to residents, Florida’s beaches attract millions of tourists – and billions of dollars – each year.
