PURPOSE: Angiogenesis plays an important role in pancreas cancer pathobiology. Pancreatic tumor cells secrete vascular endothelial growth factor (VEGF), activating endothelial cell protein kinase C beta (PKC beta) that phosphorylates GSK3 beta to suppress apoptosis and promote endothelial cell proliferation and microvessel formation. We used Enzastaurin (Enz) to test the hypothesis that inhibition of PKC beta results in radiosensitization of endothelial cells in culture and in vivo. MATERIALS/METHODS: We measured PKC beta phosphorylation, VEGF pathway signaling, colony formation, and capillary sprout formation in primary human dermal microvessel endothelial cells (HDMECs) after Enz or radiation (RT) treatment. Background: The usefulness of available vaccine and serological tests for leptospirosis is limited by the low cross-reactivity of antigens from numerous serovars of pathogenic Leptospira spp. Identification of genus-specific protein antigens (GP-Ag) of Leptospira would be important for development of universal vaccines and serodiagnostic methods. OmpL1, a transmembrane porin of pathogenic leptospires, was identified as a possible GP-Ag, but its sequence diversity and immune cross-reactivity among different serovars of pathogenic leptospires remains largely unknown.

Ultra Dynamised And Undynamised Dilutions Of Alloxan At Micro-Doses Influence Selective Pancreatic Beta Cell And Hormonal Profile: An Experimental Approach, Sunil Kumar, C Nayak

Dynamised and Undynamised preparations of Alloxan 6x, 30x, 200x, and 1000x were examined for its anti-diabetic activities in Alloxan induced diabetes mellitus albino rats. Oral administration of dynamised potencies of Alloxan 6x, 30x, 200x, and 1000x at dose level of 50μ1/100 g.b.w. daily for 30 days regularly exhibited slow and steady fall in blood sugar level accompanied with perceptible increase in serum growth hormone (GH) i.e. p < 0.01 (less significant) and p < 0.001 (significant) respectively when compared to dynamised and Undynamised control as well as Undynamised Alloxan fed groups under identical conditions. Histological and histo-morphometric studies also revealed reactivation of Pancreatic β-cells. Dynamised dilutions of Alloxan acts steadily through hypothalamo-hypophysial- pancreatic β-cells axis producing selective reactivation of β-cells at micro-doses, steadily viz 6x < 30x < 200x < 1000x. The drug may indirectly release Releasing factors (RF) from hypothalamic neurons, stimulating the secretion of growth hormone which in turn triggers optimum insulin secretion from β-cells. The therapeutic action of the test drug in dynamised dilutions at micro dose and relatively high dilutions on pancreatic β-cells confirms the phenomenon of "Potentization" & "Similia Similibus Curentur" and lack of acute and sub-acute toxicity at fairly large dosage may open up new prospects in the treatment of diabetes mellitus and throw light in elucidating the mechanism of action at higher dilutions. It was noticed that the dynamised dilutions of alcohol fed control group is more toxic and lethal to animals than the dynamised and Undynamised dilutions of Alloxan and Undynamised alcohol fed control groups. Furthermore, it was also discernible that blood sugar and growth hormone levels were stabilized even after withdrawal of test drug in its 30x, 200x and 1000x potencies.

Sexual violence against women during war and genocide is a pressing problem. Rape is used as a tactic of war and genocide because of its physical and psychosocial consequences for individuals, families, and communities. The physical and emotional sequelae of individual assaults are magnified when rape is committed on a mass scale, as in Bosnia-Herzegovina (1992–1996), Rwanda (1994), and Darfur, Sudan (2003–present). The victimization of raped women affects the community through the collective responses of survivors and their families, friends, and neighbors. Forced intercourse and impregnation represent a symbolic conquest of the woman by the rapist. This conquest becomes generalized …

Several Gram-negative pathogenic bacteria have evolved a complex protein secretion system termed the Type Three Secretion System (TTSS) to deliver bacterial effector proteins into host-cells that then modulate host-cellular functions. These bacterial devices are evolutionarily related to the flagellar apparatus. Although the TTSSs are substantially conserved among different species, the effector molecules they deliver are species-unique. There exist three human pathogenic Yersiniae. Yersinia enterocolitica and Yersinia pseudotuberculosis cause self-limiting gastro-enteric diseases and infect mesenteric lymph nodes, while Yersinia pestis is transmitted by fleas and can be aerosolized, causing the lethal disease known as plague (also known as Black Death). Serial passage of axenically cultured Leishmania chagasi promastigotes results in a progressive diminution in resistance to complement-mediated lysis (CML), whereas high CML resistance is seen in infectious metacyclic promastigotes from the sandfly vector as well as metacyclic-like promastigotes within low-passage cultures at stationary growth phase. Selection of a minor viral genotype during perinatal transmission of human Immunodeficiency virus type 1 (HIV-1) has been observed, but there is a lack of information on the correlation of the restrictive transmission with biological properties of the virus, such as replicative fitness. Assessment of tissue regeneration is essential to optimize the stages of tissue engineering (cell proliferation, tissue development and implantation). Optical and X-ray imaging have been used in tissue engineering to provide useful information, but each has limitations: for example, poor depth penetration and radiation damage. Magnetic resonance imaging (MRI) largely overcomes these restrictions, exhibits high resolution (approximately 100 μm) and can be applied both in vitro and in vivo. The potential of a solution-based hybridization assay using peptide nucleic acid (PNA) molecular beacon (MB) probes to quantify 16S rRNA of specific populations in RNA extracts of environmental samples was evaluated by designing PNA MB probes for the genera Dechloromonas and Dechlorosoma. In a kinetic study with 16S rRNA from pure cultures, the hybridization of PNA MB to target 16S rRNA exhibited a higher final hybridization signal and a lower apparent rate constant than the hybridizations to nontarget 16S rRNAs. A concentration of 10 mM NaCl in the hybridization buffer was found to be optimal for maximizing the difference between …

OBJECTIVE: To test for synergy between daptomycin (DAP) and rifampin (RIF) against RIF-resistant methicillin-resistant Staphylococcus aureus (MRSA) isolates. METHODS: Synergy testing using time-kill assay (TKA) was performed on 6 clinically, and genetically unique RIF-resistant MRSA isolates. The isolates were identified out of 489 (1.2%) samples collected during April 2003 to August 2006, from patients at the Ochsner Medical Center in New Orleans, Louisiana, United States of America. RESULTS: Synergy testing of DAP plus RIF by TKA showed that 5 isolates were indifferent, but one isolate was antagonistic. OBJECTIVE: To assess the outcome of patients with extra-abdominal desmoid tumor treated with low dose chemotherapy (methotrexate and vinblastine) both for tumor response and treatment related toxicity. METHODS: We retrospectively reviewed the outcome of 12 patients who underwent low dose chemotherapy for extra abdominal desmoid of different locations. The study took place in the McGill University Health Center, Montreal, Canada between 1996 and 2003. OBJECTIVE: To investigate the possible relationships between plasma bilirubin levels and concentrations of nitric oxide (NO), malondialdehyde (MDA), and erythrocyte antioxidant enzyme activities in newborn infants with hyperbilirubinemia. METHODS: Thirty term (gestational age >/= 37 weeks) newborn infants with indirect hyperbilirubinemia aged less than 10 days were prospectively recruited in the Kahramanmaras Sutcu Imam University Neonatal Unit, Kahramanmaras, Turkey, between January and July 2007. Thirty randomly selected healthy newborns who had similar age and without clinical jaundice comprised the control group. Background—A system was developed for the simultaneous measurement in urine of free catecholamines (i.e., dopamine, norepinephrine, epinephrine) and creatinine (i.e., an indicator of urine output and volume). This method was based on the use of tandem affinity/ion-pair HPLC and flow injection analysis.

Methods—The free catecholamines were extracted directly from urine by using an on-line phenylboronic acid affinity column. The extracted solutes were then separated and measured by ionpair chromatography followed by amperometric detection. The contact of inland and coastal prehistoric groups in Brazil is believed to have been restricted to regions with no geographical barrier, as is the case in the Ribeira de Iguape valley. The inland osteological collection from the riverine shellmound Moraes (5800–4500 BP) represents a unique opportunity to test this assumption for this region. Despite cultural similarities between riverine and coastal shellmounds, important ecological and site distribution differences are expected to impact on lifestyle. Objectives—Recent advances in the design of MRI contrast agents have rendered the lanthanide complexes of DOTA-tetraamide ligands of considerable interest, both as responsive MR agents and paramagnetic chemical exchange saturation transfer agents. The potential utility of these complexes for in vivo applications is contingent upon them being well tolerated by the body. The present study aimed to evaluate (a) the extent to which different impulsivity measures would be related to each other and to a risk taking measure, (b) the extent to which impulsivity, risk taking, time perception and time perspective are related to each other, and (c) the extent to which these processes differ in Latino and White American students. Experiment I was conducted at Utah State University. One hundred and forty-three participants were exposed to the delay discounting, probability discounting and temporal bisection procedures, and answered the Barratt Impulsiveness Scale (BIS-11) and the Zimbardo Time Perspective Inventory (ZTPI). Determining the root uptake of xenobiotic organic chemicals into plants is critical for assessing the human and ecological health risks associated with the consumption of plants growing in contaminated environments. Root uptake of xenobiotic organics occurs passively in conjunction with transpiration and the transport from root to shoot is ultimately controlled by passage through one or more lipid root membranes.
