Mechanistic Studies On The Electrochemistry Of Glutathione And Homocysteine, Olufemi Oyesanya Apr 2008

Mechanistic Studies On The Electrochemistry Of Glutathione And Homocysteine, Olufemi Oyesanya

Theses and Dissertations

This research work has investigated the electrochemistry of glutathione (GSH)and homocysteine (HCSH) in order to develop sensors for these biological thiols.Ru(bpy)33+ and IrCl62− have been used as mediators for the electrooxidation of GSH andHCSH because direct oxidation of these thiols is slow at most conventional electrodes. The electrochemical detection of GSH and HCSH has been pursued because of their biological roles.

Concerted proton electron transfer (CPET) and stepwise proton electron transfer(PT/ET) pathways have been observed in the electrooxidation of GSH and HCSH.Oxidation of GSH by Ru(bpy)33+ carried out in deuterated and undeuterated buffered (pH= pD …

A Central Function For Perlecan In Skeletal Muscle And Cardiovascular Development, Jason J. Zoeller, Angela Mcquillan, John Whitelock, Shiu-Ying Ho, Renato V. Iozzo Apr 2008

A Central Function For Perlecan In Skeletal Muscle And Cardiovascular Development, Jason J. Zoeller, Angela Mcquillan, John Whitelock, Shiu-Ying Ho, Renato V. Iozzo

Department of Pathology, Anatomy, and Cell Biology Faculty Papers

Perlecan's developmental functions are difficult to dissect in placental animals because perlecan disruption is embryonic lethal. In contrast to mammals, cardiovascular function is not essential for early zebrafish development because the embryos obtain adequate oxygen by diffusion. In this study, we use targeted protein depletion coupled with protein-based rescue experiments to investigate the involvement of perlecan and its C-terminal domain V/endorepellin in zebrafish development. The perlecan morphants show a severe myopathy characterized by abnormal actin filament orientation and disorganized sarcomeres, suggesting an involvement of perlecan in myopathies. In the perlecan morphants, primary intersegmental vessel sprouts, which develop through angiogenesis, fail …

Expression Of Nicotinic Acetylcholine Receptor Mrna As A Function Of Age In Whole Hippocampus Preparations From Wistar Rats, Kasey C. Welch Apr 2008

Expression Of Nicotinic Acetylcholine Receptor Mrna As A Function Of Age In Whole Hippocampus Preparations From Wistar Rats, Kasey C. Welch

Theses and Dissertations

Whole hippocampus preparations, isolated bilaterally, from untreated Wistar rats at various ages (10-90 days old) were analyzed for the mRNA expression of the alpha 2, alpha 3, alpha 4, alpha 5, alpha 7, beta 2, beta 3, and beta 4 neuronal nicotinic acetylcholine receptor subunits. To do so, RNA was isolated from acutely isolated hippocampal samples, converted to cDNA by means of a reverse transcription reaction, then analyzed with quantitative real-time PCR to determine the relative levels of the mRNAs the cells were expressing at the age when the samples were obtained. The relative expression of the levels of RNA …

On The Role Of Mitochondria In The Regulation Of Calcium In Motor Nerve Terminals During Repetitive Stimulation, Luis E. Garcia-Chacon Apr 2008

On The Role Of Mitochondria In The Regulation Of Calcium In Motor Nerve Terminals During Repetitive Stimulation, Luis E. Garcia-Chacon

Open Access Dissertations

During repetitive stimulation of motor nerve terminals, mitochondrial Ca2+ uptake limits increases in free cytosolic [Ca2+] and helps ensure faithful neuromuscular transmission. Changes in cytosolic [Ca2+] and in mitochondrial [Ca2+] as well as changes in mitochondrial membrane potential (Psi m) were studied in mouse motor nerve terminals using Ca2+ sensitive indicator and potentiometric dyes, respectively. Trains of action potentials (APs) at 50 to 100 Hz produced a rapid increase in mitochondrial [Ca2+] followed by a plateau which usually continued beyond the end of stimulation. After stimulation, mitochondrial [Ca2+] decayed back to baseline over the course of tens of seconds to …

Open Access Dissertations

The purpose of this project is to develop a retinal imaging system suitable for routine examination or screening of mouse models that acquires fundus and Optical Coherence Tomography (OCT) images. The imaging system is composed of a digital camera with an objective for biomicroscopic examination of the fundus, an OCT interferometer, an OCT beam delivery system designed for the mouse eye, and a mouse positioning stage. The image acquisition is controlled with software that displays the fundus and OCT images in real-time, and allows the user to control the position of the OCT beam spot on the fundus image display. …

Angiographic Assessment Of Flow Divertors As Treatment For Cerebral Aneurysms: Results In The Rabbit Elastase-Induced Aneurysm Model, Chandramouli Sadasivan Apr 2008

Angiographic Assessment Of Flow Divertors As Treatment For Cerebral Aneurysms: Results In The Rabbit Elastase-Induced Aneurysm Model, Chandramouli Sadasivan

Open Access Dissertations

Cerebral aneurysms are abnormal focal dilations of the cerebral vasculature that may rupture and cause a hemorrhagic stroke. More than a decade after seminal experimental studies showed the feasibility of successfully treating cerebral aneurysms by flow diversion, the potential of this modality is gradually being recognized in the clinical arena. A flow divertor is a metallic meshed tube that has its porosity (ratio of metal-free surface area to total surface area) and pore density (number of pores per unit surface area) optimized to facilitate occlusion of aneurysms. An appropriately designed flow divertor severely attenuates the flow exchange between the parent …

Occurrence And Fate Of Pesticides In Four Contrasting Agricultural Settings In The United States, Gregory V. Steele, Henry M. Johnson, Mark W. Sandstrom, Paul D. Capel, Jack E. Barbash Apr 2008

Occurrence And Fate Of Pesticides In Four Contrasting Agricultural Settings In The United States, Gregory V. Steele, Henry M. Johnson, Mark W. Sandstrom, Paul D. Capel, Jack E. Barbash

United States Geological Survey: Staff Publications

Occurrence and fate of 45 pesticides and 40 pesticide degradates were investigated in four contrasting agricultural settings—in Maryland, Nebraska, California, and Washington. Primary crops included corn at all sites, soybeans in Maryland, orchards in California and Washington, and vineyards in Washington. Pesticides and pesticide degradates detected in water samples from all four areas were predominantly from two classes of herbicides—triazines and chloroacetanilides; insecticides and fungicides were not present in the shallow ground water. In most samples, pesticide degradates greatly exceeded the concentrations of parent pesticide. In samples from Nebraska, the parent pesticide atrazine [6-chloro-N-ethyl-N′-(1-methylethyl)-1,3,5-triazine-2,4- diamine] was about the same concentration …

Altered Signaling By Fibronectin Fragments Creates A Potent Chondrolytic State, Lei Ding Apr 2008

Altered Signaling By Fibronectin Fragments Creates A Potent Chondrolytic State, Lei Ding

Theses and Dissertations

It has been shown that fibronectin fragments (Fn-fs) greatly augment cartilage proteoglycan (PG) depletion by up-regulating matrix metalloproteinases (MMPs) and catabolic cytokines. The activities of three different Fn-fs have been studied: an amino terminal 29-kDa which is most potent with a weak affinity to the α5β1, integrin, the receptor for native fibronetin (Fn); a 50-kDa gelatin-binding Fn-f with no known interaction with Fn receptor; and a 140-kDa Fn-f which is the least potent and has a RGDS sequence which can bind to the α5β1 integrin. Confocal studies showed that Fn-fs caused diffusion of α5β 1 integrin clusters which were seen …

Light Induced Changes In The Photoreceptor Outer Segments, Vikram Palamalai Apr 2008

Light Induced Changes In The Photoreceptor Outer Segments, Vikram Palamalai

Theses and Dissertations

Light induced retinal degeneration in animals has been used to study human retinal degenerative disorders. Apoptosis is the final common pathway for photoreceptor degeneration in both the experimental model and human disease but the mechanism is unknown. Oxidative stress plays a role because antioxidants have been shown to be protective against experimental degeneration. Inhibitors of nitric oxide synthases (NOS) are also protective suggesting a role for nitric oxide. Glyceraldehyde-3-phosphate dehydrogenase (GAPDH) a key enzyme in glycolysis was identified as one of the proteins undergoing significant nitration in our studies on light induced retinal degeneration. This study focuses on the changes …

Regulation Of Pro-Inflammatory Responses By Ligands Of The Nuclear Receptor Pparγ, Andrew P. Woster Apr 2008

Regulation Of Pro-Inflammatory Responses By Ligands Of The Nuclear Receptor Pparγ, Andrew P. Woster

Theses and Dissertations

Inflammation-related neurotoxicity is evident in a number of different models neurodegenerative conditions, including Alzheimer's and Parkinson's diseases, Multiple Sclerosis, and HIV-associated dementia. Peroxisome proliferator-activated receptor gamma (PPARγ) agonists inhibit a number of select pro-inflammatory changes in models of CNS and systemic inflammation. Recent reports suggest that these anti-inflammatory effects are due to mechanisms other than canonical nuclear receptor-mediated transcriptional alteration. Results reported here demonstrate that rosiglitazone, a PPARγ-activating thiazolidinedione, decreases tumor necrosis factor α (TNFα) secretion in both primary microglia and the monocytic cell line, THP-1. Cells were pretreated with various PPARγ ligands, including rosiglitazone, prior to stimulation with lipopolysaccharide …

Lonely Sounds: Recorded Popular Music And American Society, 1949–1979, Chris R. Rasmussen Apr 2008

Lonely Sounds: Recorded Popular Music And American Society, 1949–1979, Chris R. Rasmussen

Department of History: Dissertations, Theses, and Student Research

Abstract: Lonely Sounds: Popular Recorded Music and American Society, 1949-1979 Lonely Sounds: Popular Recorded Music and American Society, 1949-1979 examines the relationship between the experience of listening to popular music and social disengagement. It finds that technological innovations, the growth of a youth culture, and market forces in the post-World War II era came together to transform the normal musical experience from a social event grounded in live performance into a consumable recorded commodity that satisfied individual desires. The musical turn inward began in the late 1940s. Prior to the postwar era, the popular music experience was communal, rooted in …

Neural Characteristics Of Affectionate Communicators: Trait Affection And Asymmetry In The Prefrontal Cortex, Robert Joel Lewis Apr 2008

Neural Characteristics Of Affectionate Communicators: Trait Affection And Asymmetry In The Prefrontal Cortex, Robert Joel Lewis

Theses

Individual differences in thresholds for affectionate communication should be reflected by differences in neurological structure and function. A theoretical schema from several overlapping literatures including evolutionary psychology, social neuroscience, fundamental personality, and communication are examined to make the case that high-affection communicators have greater relative electrical activity in the left prefrontal cortex (PFC) versus the right PFC reflected in asymmetrical baseline EEG recordings. Participants (N=16) reported trait-affection levels using Floyd?s (2002) TAS-G, which measures an individual?s threshold for expressing affection. Participants? baseline electrical activity was then recorded. Asymmetry was operationalized as the difference between microvolt (μV) values of laterally opposed …

Toward A New Perspective In Literary Criticism: Transgenerational Psychoanalysis And Anasemic Procedures, Gustav Arnold Apr 2008

Toward A New Perspective In Literary Criticism: Transgenerational Psychoanalysis And Anasemic Procedures, Gustav Arnold

Theses and Dissertations

This dissertation—Toward a New Perspective in Literary Criticism: Transgenerational Psychoanalysis and Anasemic Procedures"—is a contribution to literary criticism. It offers a new perspective in psychoanalytical criticism on how characters in literary narratives can be interpreted. By examining key concepts in psychoanalysis, which have been used in the humanities for decades, the present inquiry draws on transgenerational psychology, as elaborated by German and French theorists, and anasemic interpretations generated by M. Torok and N. Abraham's psychoanalytical approach. Based on the assumption that a character's psychic life is determined by factors that cannot be confined to intrapsychic processes alone, the transgenerational perspective …
