ICER is a member of the CREM family of basic leucine zipper transcription factors that acts as a dominant negative regulator of gene transcription. Four different isoforms of ICER (I, Iγ, II and IIγ) are transcribed from the P2 promoter of the Crem gene. We previously found that each of the ICER isoforms is induced by parathyroid hormone in osteoblasts. The goal of the present study was to assess the function of ICER in bone by overexpressing ICER in osteoblasts of transgenic mice. ICER I and ICER II cDNAs, each containing an N-terminal FLAG epitope tag, were cloned downstream of …

Platforms based on thin enzyme/DNA films were used in two-tier screening of chemicals for reactive metabolites capable of producing toxicity. Microsomes were used for the first time as sources of cytochrome (cyt) P450 enzymes in these devices. Initial rapid screening involved electrochemiluminescent (ECL) arrays featuring spots containing ruthenium poly(vinylpyridine), DNA, and rat liver microsomes or bicistronically expressed human cyt P450 2E1 (h2E1). Cyt P450 enzymes were activated via the NADPH/reductase cycle. When bioactivation of substrates in the films gives reactive metabolites, they are trapped by covalent attachment to DNA bases. The rate of increase in ECL with enzyme reaction time …

Coral Ultrastructural Response To Elevated Pco2 And Nutrients During Tissue Repair And Regeneration, Dorothy-Ellen A. Renegar, Patricia Blackwelder, Alison L. Moulding

Marine & Environmental Sciences Faculty Proceedings, Presentations, Speeches, Lectures

Corals and coral reefs have recently experienced widespread decline attributed to anthropogenic pressure on reef systems. Studies have demonstrated that nutrient and pCO2 stress effect coral growth and calcification, but study of specific effects on coral tissue is lacking. The objective of this research was to examine wound healing in corals and how it is affected by exposure to elevated nutrients and pCO2. Coral tissue repair and regeneration during wound healing in Montastraea cavernosa and Porites astreoides were assessed histologically and ultrastructurally by examining colony fragments exposed to elevated nitrate, phosphate, and pCO2. In M. …

Bovine Viral Diarrhea Virus Multiorgan Infection In Two White-Tailed Deer In Southeastern South Dakota, Christopher C.L. Chase, Lyle J. Braun, Pamela Leslie-Steen, Tanya Graham, Dale Miskimins, Julia F. Ridpath

Veterinary and Biomedical Sciences Faculty Publications

The susceptibility of wild ruminants, especially cervids, to bovine viral diarrhea virus (BVDV) has remained an enigma. Two white-tailed deer (Odocoileus virginianus) were submitted to the Animal Disease Research and Diagnostic Laboratory (ADRDL) in the fall of 2003 by the South Dakota Game Fish and Parks for chronic wasting disease (CWD) testing. Both animals were CWD negative. The animals were necropsied and histopathology, viral antigen detection, and virus isolation were performed. A noncytopathic (NCP) BVDV was isolated from the lungs and several other tissues of both animals. Formalin-fixed ear notches from both animals were positive for BVDV antigen by immunohistochemistry. …

One method to teach a multicultural class: a teacher walks in to a room calls the class to order, steps to the lectern and begins to deliver knowledge. Socially acquired information is disseminated; that gained over years of study and experience. Another view: I walk into a class where my students are sitting quietly at their desks pen and paper at the ready and quietly ask: shall we form a circle? The students agree and after having done so I follow up with a generative (Freire, 1970): what are your current understandings of Diversity? In following the work of Paulo …

A Simplified Experimental Method To Evaluate Equivalent Roughness Of Vegetated River Beds, S. De Felice, P. Gualtieri, G. Pulci Doria Jul 2008

A Simplified Experimental Method To Evaluate Equivalent Roughness Of Vegetated River Beds, S. De Felice, P. Gualtieri, G. Pulci Doria

International Congress on Environmental Modelling and Software

In this paper a new method to experimentally evaluate roughness of vegetated river beds is proposed. Unlike the usual method of attaining uniform flow, the new method is based on measurements performed in boundary layer flows, conditions that in flow experiments with plants can be attained easier then in uniform flows. The described method can be applied either in case of smooth or vegetated walls. The friction factor f and Manning’s n values so obtained are in agreement with literature results.

Developments In Urban Environmental Information Perception And Communication, Kostas Karatzas, J. Lee

International Congress on Environmental Modelling and Software

Urban environmental information (EI) is related to knowledge concerning the status, the pressures and the consequences of environmental quality parameters to human life and the urban surroundings. One of the most important aspects in the life cycle of EI is knowledge generation and flow, data management and information dissemination between ICT systems and environmental actors, players, or interested parties. In the present paper, urban air quality serves as the application domain, for which the following issues are discussed, • Analysis of the way that environmental information is related to urban structures (roads, buildings, etc) • Investigation of the spatial and …

The Brachistochrone Problem: Mathematics For A Broad Audience Via A Large Context Problem, Jeff Babb, James Currie

The Mathematics Enthusiast

Large context problems (LCP) are useful in teaching the history of science. In this article we consider the brachistochrone problem in a context stretching from Euclid through the Bernoullis. We highlight a variety of results understandable by students without a background in analytic geometry. By a judicious choice of methods and themes, large parts of the history of calculus can be made accessible to students in Humanities or Education.

Graphical User Interface Design For Climate Impact Research Data Retrieval, Markus Wrobel, T. Nocke, Michael Flechsig, A. Glauer

International Congress on Environmental Modelling and Software

Complex, heterogeneous environmental data sets are produced and stored in many scientific disciplines. For managing complex structured data, database and data warehouse technologies have been established, providing access via standardized interfaces. Nevertheless, there are some obstacles for researchers in applying the full functionality of sophisticated data management systems. Here, graphical user interfaces (GUI) are essential for bridging the gap between such systems and scientific users. However, designing / developing user interfaces for complex, environmental data sets is not trivial, facing user diversity and a variety of tasks. Consequently, for the example of climate impact research, this paper introduces an approach …

Towards A Simulation-Based Study Of Grassland And Animal Diversity Management In Livestock Farming Systems, G. Martin, Michel Duru, Roger Martin-Clouaire, Jp Rellier, Jp Theau, O. Thérond, L. Hossard

International Congress on Environmental Modelling and Software

In less-favored areas, livestock production involves the management of native vegetation as a resource base. A challenging issue in such systems regards the design of efficient and sustainable management strategies that make it possible to exploit the diversity of grasslands and herd batches over different time scales. This paper describes the conceptual approach that supports the representation and simulation of farm-scale models of grassland-based livestock systems in the SEDIVER project. It enables the coupling of species-rich grasslands and livestock models with sophisticated decision-making models. The latter embed a representation of management strategies that specify in a flexible and adaptive manner …

Grassland Songbird Survival And Recruitment In Agricultural Landscapes: Implications For Source-Sink Demography, Noah G. Perlut, Allan M. Strong, Therese M. Donovan, Neil J. Buckley Jul 2008

Grassland Songbird Survival And Recruitment In Agricultural Landscapes: Implications For Source-Sink Demography, Noah G. Perlut, Allan M. Strong, Therese M. Donovan, Neil J. Buckley

Environmental Studies Faculty Publications

Population growth and decline are particularly sensitive to changes in three key life-history parameters: annual productivity, juvenile survival, and adult survival. However, for many species these parameters remain unknown. For example, although grassland songbirds are imperiled throughout North America, for this guild, only a small number of studies have assessed these parameters. From 2002 to 2006, in the agricultural landscape of the Champlain Valley of Vermont and New York, USA, we studied Savannah Sparrow (Passerculus sandwichensis) and Bobolink (Dolichonyx oryzivorus) demography on four grassland treatments: (1) early-hayed fields cut before 11 June and again in early- …

The Ethics Of Driving, Amy E. Marga Jul 2008

The Ethics Of Driving, Amy E. Marga

Faculty Publications

The concept of “neighbor” takes on entirely new meanings in our mobile society. Who becomes my neighbor when I am behind the wheel? What is the consequence of my driving to my neighbors around the globe? What about the impact on our nonhuman “neighbors”? The ethics of driving has many dimensions for the Christian person of goodwill.
