The Contribution Of Homeostatically Expanded Donor Cd8 T Cells To Host Reconstitution Following Syngeneic Hematopoietic Cell Transplantation, Melinda Roskos Keith Jul 2008

The Contribution Of Homeostatically Expanded Donor Cd8 T Cells To Host Reconstitution Following Syngeneic Hematopoietic Cell Transplantation, Melinda Roskos Keith

Open Access Dissertations

During homeostatic expansion, peripheral T cells proliferate in response to lymphopenia, in the absence of cognate antigen or costimulatory signals. Host CD8 reconstitution following hematopoietic cell transplantation (HCT) involves the de novo-generation of T cells in addition to the homeostatic expansion of mature donor T cells present in the graft and donor lymphocyte infusion as well as host T cells that survive conditioning. Although it is well appreciated that CD8 homeostatic expansion contributes to host CD8 reconstitution following HCT, the factors governing the extent of the contribution by donor CD8 homeostatic expansion to host reconstitution have not been precisely and …

“Surreptitious sampling” may be police officers’ trump card in cracking otherwise unsolvable crimes as serious as murder, arson and rape. Law enforcement officers engage in surreptitious sampling when they covertly collect DNAsamples from unsuspecting people, who inadvertently leave behind hair, skin cells, saliva or other biological materials.Surreptitious sampling is a terrific crime-resolution tool. It allows diligent law enforcement officers to collect proof-positive evidence of guilt or innocence without the hassle of obtaining a warrant and absent probable cause or reasonable suspicion to believe that the contributor of the biological evidence committed a crime. Provided an officer has the energy and …

An Examination Of The Motivations Of Sporting Event Volunteers At Clemson University, Blair Schuyler Jul 2008

An Examination Of The Motivations Of Sporting Event Volunteers At Clemson University, Blair Schuyler

All Theses

In 2006, The Corporation for National and Community Service partnered with the Bureau of Labor Statistics to add a volunteer section to the Current Population Survey (CPS) that is sent out to 60,000 households each year. That year it was reported that 61.2 million people volunteered their time, effort, and labor through or for an organization. An additional 5.3 million people did volunteer work informally through helping their communities. Organizations across the world, specifically sports organizations, rely heavily on volunteers for success. Volunteers at the 2004 Summer Olympics (45,000), based off the assumption of working a full 8 hour for …

Matoaka: Pocahontas In The Age Of Identity, Matthew Shifflett Jul 2008

Matoaka: Pocahontas In The Age Of Identity, Matthew Shifflett

Theses and Dissertations

This thesis details the labors of research and judgment that informed the writing of Matoaka, a Play in Three Acts. Specifically, the thesis explores the historical puzzles surrounding the life of Pocahontas and justifies the decisions made in dramatizing her life in the aforementioned play. Non-fictional works of the last four hundred years are considered, as well as popular dramatic performances of the nineteenth century. These works are examined closely in order to reveal the Pocahontas story as a point of contact between many concurrent social discourses. Reflections are also offered on the production of the aforementioned play and its …

A Novel And Simple Rule Of Thumb For Multiplicity Control In Equivalence Testing Using Two One-Sided Tests, Carolyn Lauzon, Brian S. Caffo Jul 2008

A Novel And Simple Rule Of Thumb For Multiplicity Control In Equivalence Testing Using Two One-Sided Tests, Carolyn Lauzon, Brian S. Caffo

Johns Hopkins University, Dept. of Biostatistics Working Papers

Equivalence testing is growing in use in scientific research outside of its traditional role in the drug approval process. Largely due to its ease of use and recommendation from the United States Food and Drug Administration guidance, the most common statistical method for testing (bio)equivalence is the two one-sided tests procedure (TOST). Like classical point-null hypothesis testing, TOST is subject to multiplicity concerns as more comparisons are made. In this manuscript, a condition that bounds the family-wise error rate (FWER) using TOST is given. This condition then leads to a simple solution for controlling the FWER. Specifically, we demonstrate that …

Breeding And Foraging Ecology Of American Oystercatchers In The Cape Romain Region, South Carolina, Janet Thibault Jul 2008

Breeding And Foraging Ecology Of American Oystercatchers In The Cape Romain Region, South Carolina, Janet Thibault

All Theses

The majority of American Oystercatchers (Haematopus palliatus) in South Carolina make their nests on narrow, elevated mounds of oyster shells deposited naturally along the edges of bays by storms or formed artificially by boat wakes along the Atlantic Intracoastal Waterway (AICW). I compared reproductive success of American Oystercatchers nesting along the AICW with those nesting on naturally formed shell mounds in Bulls Bay within the Cape Romain Region of South Carolina during the breeding seasons of 2006 and 2007. I identified timing and causes of nest failure and attributes of re-nesting. Hatching success (15%) and productivity estimates (0.25 chicks per …

Risedronate Prevents Early Bone Loss Resulting From Whole-Body Irradiation, Eric Livingston Jul 2008

Risedronate Prevents Early Bone Loss Resulting From Whole-Body Irradiation, Eric Livingston

All Theses

Osteoporosis is a condition characterized by a reduction in bone strength resulting in an overall increase in the risk of fracture. There are many factors that contribute to the development of this condition, including ionizing radiation exposure. Declines in bone volume and trabecular micro-architecture have been found following exposure to multiple types of radiation. Past research has implicated reduction of osteoblast function and changes to vasculature as the primary sources of bone deterioration. Recently, an early increase in osteoclast number was observed following exposure to low-energy X-rays, identifying an increase in resorption as a possible cause and potential target for …

Fecal Coliform Source Identification Using Chemical Tracers, Nicole Fahrenfeld Jul 2008

Fecal Coliform Source Identification Using Chemical Tracers, Nicole Fahrenfeld

All Theses

Fecal sterols and fluorinated whitening agents (FWA) were used as chemical tracers for fecal coliform source tracking in Cane and Little Cane creeks in Walhalla, SC. Fecal sterols were quantifiable in both stream sediments and suspended particulates using an extraction, clean-up, and analytical method modified from Isobe et al. (2002) with a N,O-Bis(trimethylsilyl)trifluoroacetamide (BSTFA) derivatization scheme. Only human sterol source identification ratios were able to be calculated using the chosen sterol suite. Human fecal signatures were seen in both the sediment and the water column at various points along both creeks, indicating human fecal pollution is contributing to the fecal …

Manipulations Of Macrophage Activation Through Targeted Delivery Of Interferon Regulatory Factor Genes Mediated By Recombinant Adenovirus-Conjugated Zymosan, Mengmeng Zhang Jul 2008

Manipulations Of Macrophage Activation Through Targeted Delivery Of Interferon Regulatory Factor Genes Mediated By Recombinant Adenovirus-Conjugated Zymosan, Mengmeng Zhang

All Theses

According to the different phenotypes and distinct patterns of gene expression, macrophages can be subdivided into two types: M1 and M2. The M1 macrophages, which are activated through the classical pathway, have a proinflammatory characteristic, providing an in-front defense line against different kinds of pathogens as well as malignant cells. On the other hand, the M2 macrophages are immunosuppressive and instead can promote tumor growth and dissemination. The solid tumor tissues are often infiltrated with a large number of macrophages, the so-called tumor-associated macrophages (TAMs). TAMs, which are polarized into M2 phenotype under the tumor microenvironment, promote tumor progression by …

Self-Organization": A New Metaphor To Understand The Interdependences Of The Organizations And Their Environments, Tarek Mohamed Ali Jul 2008

Self-Organization": A New Metaphor To Understand The Interdependences Of The Organizations And Their Environments, Tarek Mohamed Ali

Business Administration

Self-organization metaphors have recently emerged to enrich research on organizational environment in particular, with respect to the networking view that presents organizational environment as a complex web of interactions among groups of organizations. The network analysis promotes the group members’ sensitivity to the variety in environmental demands. In an attempt to clarify how self-organization enhances the group’s sensitivity, we propose the interdisciplinary autopoietic and habitus-field models. The proposed self-organization models allow portraying business organizations as networks of decision communications and integrated partners. Self-referential and self-reflexivity processes have been utilized by the models to guide a group of decision makers and …

Spatial And Temporal Dynamics Of Lekking Behavior And Female Mate Choice In The Blue-Crowned Manakin (Lepidothrix Coronata, Aves: Pipridae), Renata Duraes Ribeiro Jul 2008

Spatial And Temporal Dynamics Of Lekking Behavior And Female Mate Choice In The Blue-Crowned Manakin (Lepidothrix Coronata, Aves: Pipridae), Renata Duraes Ribeiro

Dissertations

In lekking species, males aggregate at display grounds (?leks?) that females visit to copulate. Lekking systems are non-resource-based and males vary widely in reproductive output. This dissertation investigates how spatial and temporal factors influence lekking dynamics and female mate choice in the blue-crowned manakin Lepidothrix coronata. The first chapter deals with spatial dynamics of lekking behavior, and investigates whether leks are formed by the establishment of males on sites of high female density (?hotspot hypothesis?). Predictive maps of female spatial distribution were built based on field data and contrasted to the lek distribution. Leks were located in sites where males …

Physiological And Molecular Characterization Of Genetic Competence In Streptococcus Sanguinis, Alejandro Rodriguez Jul 2008

Physiological And Molecular Characterization Of Genetic Competence In Streptococcus Sanguinis, Alejandro Rodriguez

Theses and Dissertations

The ability of bacteria to assimilate free DNA from the environment is known as competence. Though many studies have focused on competence regulation in Streptococcus pneumoniae and Streptococcus gordonii, Streptococcus sanguinis has yet to be examined. Physiological characterization of competence in S. sanguinis strain SK36 and its comC mutant, JFP41, led to the genome-wide transcriptional analysis of cells induced to competence via addition of competence-stimulating peptide (CSP). A total of 128 genes were induced at least 2-fold, 74 of which were classified as either “early” or “late” based on their induction patterns. Expression patterns were verified using qRT-PCR. This study …

Faq #6: Are The Beijing Olympic Games Being Used As A Propaganda Campaign To Prop Up The Communist Party?

China Beat Blog: Archive 2008-2012

“Olympic education” is the IOC’s label for the educational efforts that are supposed to be an integral part of the Olympic Movement as required by Fundamental Principle #1 of the Olympic Charter, which states that Olympism is a philosophy of life that blends sport with culture and education. Since 2005 Chinahas been carrying out the largest-scale “Olympic education” campaign in history. There have been academic and professional conferences, textbooks and courses for public schools and universities, educational television and radio shows, magazine and newspaper essays, websites, and more.

In fall of 2007 I was added to the “experts team” of Beijing …

The Progymnasmata: New/Old Ways To Teach Reading, Writing, And Thinking In Secondary Schools, Natalie Sue Baxter Jul 2008

The Progymnasmata: New/Old Ways To Teach Reading, Writing, And Thinking In Secondary Schools, Natalie Sue Baxter

Theses and Dissertations

Within the past two decades, theorists have begun to look back to the classical rhetorical past for answers to modern dilemmas in composition teaching. Several textbooks have been published that focus their teaching of college composition on the classical tradition. While the movement to revitalize classical rhetoric is gaining strength in universities, however, the benefits of this movement have not yet reached, in any real way, the levels of elementary, middle school, or high school education. This thesis shows how the classical rhetorical curriculum generally, and a specific part of that curriculum, the progymnasmata, accomplish important aims of modern composition …

Selenoproteins Mediate T Cell Immunity Through An Antioxidant Mechanism, Rajeev K. Shrimali, Robert D. Irons, Bradley A. Carlson, Yasuyo Sano, Vadim N. Gladyshev, Jin Mo Park, Dolph L. Hatfield

Vadim Gladyshev Publications

Selenium is an essential dietary element with antioxidant roles in immune regulation, but there is little understanding of how this element acts at the molecular level in host defense and inflammatory disease. Selenium is incorporated into the amino acid selenocysteine (Sec), which in turn is inserted into selenoproteins in a manner dependent on Sec tRNA[Ser]Sec. To investigate the molecular mechanism that links selenium to T cell immunity, we generated mice with selenoprotein-less T cells by cell type-specific ablation of the Sec tRNA[Ser]Sec gene (trsp). Herein, we show that these mutant mice exhibit decreased pools of …

Duelling Dreams At The 2008 Beijing Olympics, James Leibold Jul 2008

Duelling Dreams At The 2008 Beijing Olympics, James Leibold

China Beat Blog: Archive 2008-2012

This year is China’s year. On the 8th day of the 8th month at 8pm, it will light the flame of the 29th Olympics Games—bringing not only the world’s athletes to Beijing but also thousands of foreign visitors and millions more through the massive global media contingency that will descend on the capital.

What will the media spotlight capture? The luster of an ancient civilization and emerging superpower as many in China hope; or the darker, seedier corners of an authoritarian yet fragile party-state as many in the West suspect.

According to the Chinese government, the Olympics games have never …

Factors Affecting Message Intelligibility Of Cued Speech Transliterators, Katherine Pelley Jul 2008

Factors Affecting Message Intelligibility Of Cued Speech Transliterators, Katherine Pelley

USF Tampa Graduate Theses and Dissertations

While a majority of deaf students mainstreamed in public schools rely on interpreters, little research has investigated interpreter skills and no research to date has focused on interpreter intelligibility (Kluwin & Stewart, 2001). This thesis is the second in a series of experiments designed to quantify the contribution of various factors affecting the intelligibility of interpreters (transliterators) who use English-based communication modes. In the first experiment, 12 Cued Speech transliterators were asked to transliterate an audio lecture. Two aspects of these transliterated performances were then analyzed: 1) accuracy, as measured as the percent-correct cues produced, and 2) lag time, the …

Luminex Microsphere Immunoassay Offers An Improved Method In Testing For Antibodies To Eastern Equine Encephalitis Virus In Sentinel Chicken Sera, Kelly Ann Fitzpatrick

USF Tampa Graduate Theses and Dissertations

Eastern Equine Encephalitis virus has a human mortality rate of 30% of those cases diagnosed, while 30% of those surviving infection remain with neurological sequelae for life (CDC.gov, 2007).

The use of sentinel chickens for surveillance of arboviruses that are known to use birds as a reservoir host, such as St. Louis Encephalitis (SLE), West Nile (WN) virus, Eastern Equine Encephalitis (EEE) and Highlands J (HJ) virus, in Florida began with the Sentinel Chicken Arboviral Surveillance Network in 1978 (Day and Stark, 1996). This network enables the activation of an early warning system for citizens, as well as, county epidemiologists …

Multilocus Virulence Typing Of Clinical And Environmental Vibrio Vulnificus Isolates, Katrina V. Gordon Jul 2008

Multilocus Virulence Typing Of Clinical And Environmental Vibrio Vulnificus Isolates, Katrina V. Gordon

USF Tampa Graduate Theses and Dissertations

The bacterium Vibrio vulnificus is an autochthonous inhabitant of estuarine waters and also found in shellfish such as oysters. It is a human pathogen of importance in the seafood industry, and can also infect recreational water users. Currently, recognized methods of detection rely upon isolation of pure cultures which requires at least 24 hours. To reduce the time needed for identification of the pathogen and simultaneously ascertain the virulence potential of the strains present, real-time PCR assays and sample processing procedures were developed (Chapter 1). These assays discriminate between type A (environmental, generally lower virulence) and type B (clinical, higher …
