This thesis details -in personal narrative form- the process by which the author directed a production of Elizabeth Wong's Amazing Adventures of the Marvelous Monkey King at Dr. Michael M. Krop Senior High School in Miami, Florida. The following text explains elements of pre-production, presents relevant research associated with the play, describes the production process in detail, and states conclusions drawn by the author about the experience. The work challenged both the ensemble and the director to seek new avenues of expression and theatricality different from those traditionally explored in their respective educational settings.

Successful treatment of cancer with adoptive immunotherapy (AIT) is dependent on the ability to produce large numbers of tumor-specific, functional T cells. The purpose of this thesis is to explore ways in which T cell expansion could be augmented. We have focused on exploring alternate gamma chain cytokines as stimulators of T cell proliferation and differentiation in addition to investigating the potential usefulness of gemcitabine (GEM) in abrogating the immunosuppressive effects of myeloid derived suppressor cells (MDSCs). Catecholaminergic neurotransmission is regionally altered following injury, and drugs aimed at these systems offer promising avenues for post-TBI pharmacotherapies. Atomoxetine is a selective norepinephrine transporter (NET) inhibitor currently indicated for treatment of attention-deficit hyperactivity disorder (ADHD). The studies in this dissertation were designed to test the efficacy of atomoxetine for treating cognitive deficits following experimental TBI and the potential mechanism for any beneficial effect. The first part of the study focused on behavioral recovery following atomoxetine treatment. Increased road construction associated with urbanization may result in fragmentation and loss offish populations in streams. In this study, we documented frequency of movement of fishes through three separate types of road-crossings (clear-span bridges, box culverts, and tube culverts) in six small streams using mark-recapture sampling. Upstream movement between areas separated by either box or tube culverts was lower than upstream movement between similar areas not separated by a road crossing. Downstream movement between areas separated by box culverts was also lower than downstream movement between areas without obstructions. In a series of articles and now in their new book, Religious Freedom and the Constitution, Lawrence Sager and Christopher Eisgruber (E&S) defend an interpretation of the religion clauses of the First Amendment which, they write, "denies that religion is a constitutional anomaly, a category of human experience that demands special benefits and/or necessitates special restrictions." While not a book review in the traditional sense, my essay takes E&S's defense of a secular First Amendment as a starting point and asks, how did we get to the point where an interpretation of the First Amendment which denies that religion is …

Docosahexaenoic acid (DHA), an omega-3 fatty acid found in cold-water fish, has positive health benefits. While the suggested amount of 220 mg/day for adults may be attained by consuming fish, this may increase exposure to environmental pollutants. The heterotrophic marine alga Crypthecodinium cohnii is an important source of DHA because C. cohnii can accumulate lipid greater than 20% of their biomass with a large fraction of DHA (30-50%). Commercially, DHA production by C. cohnii is conducted in large bioreactors (∼100 m3 capacity) using a batch-mode process. The purpose of the first study was to determine how age and ovarian hormones affect flow-induced vasodilation in the coronary microcirculation. Coronary arterioles were isolated from young, middle-aged, and old control, ovariectomized (OVX) and ovariectomized + estrogen replaced (OVE) female Fischer-344 rats to assess vasodilation to increases in flow. L-NAME, a nitric oxide synthase (NOS) inhibitor, was used to assess NO contribution to flow-induced dilation (FID). FID of coronary arterioles was impaired with advancing age. Ovariectomy reduced FID in young females only; however, estrogen-replacement restored or improved FID in all age groups. FID was reduced after pretreatment with L-NAME in …

gamma-Secretase is an enzymatic complex critical to amyloid formation and other important pathophysiological processes. Non-specific gamma-secretase inhibitors are not ideal for intervention of familial Alzheimer's disease. The four essential include Presenilin (PS1 or PS-2), APH-1, Pen-2, nicastrin (Nct). Also gamma-secretase cleaves a variety of substrates (over 30). But different sites are cleaved even in a single substrate and no consensus sequence for cleavage by gamma-secretase is identified. We hypothesize that consensus compositional sequence (or functional domain) in components of gamma-secretase and/or its substrates should exist during gamma-secretase regulation and specific gamma-secretase inhibitor based on such consensus sequence (or functional domain) …

According to the United States Environmental Protections Agency (USEPA), nearly one in four households in the United States depends on an individual septic system (commonly referred as an onsite system or a decentralized wastewater system) to treat and disperse wastewater. More than half of these systems are over 30 years old, and surveys indicate at least 10 to 20% might not be functioning properly. The USEPA concluded in its 1997 report to Congress that adequately managed decentralized wastewater systems (DWS) are a cost-effective and long-term option for meeting public health and water quality goals, particularly in less densely populated areas.;The …

This thesis involved two components: identification of the parasite Anguillicola crassus in American eels, Anguilla rostrata and PIT tag retention in eels in a laboratory setting. The recent spread of the swim bladder nematode parasite, Anguillicola crassus, in American eels has caused concern among biologists and fishery managers. A total of 244 yellow-phase American eels were collected at the Millville Dam eel ladder on the Shenandoah River, WV. Swim bladders were removed and examined for the presence of the nematode parasite. The number of parasites in each eel was recorded, and prevalence, intensity, and mean intensity were calculated. This paper assesses the private remedies available under Rule 10b-5 to retail investors who have been defrauded by false corporate disclosures. After comparing the treatment received by retail investors to the treatment received by institutional investors, I identify several areas in which the federal securities laws disfavor retail investors who have been defrauded by false corporate disclosures, including the creation of a two-tiered system of investor remedies for securities fraud. Institutional investors are permitted to pick and choose which law and forum offers them the most attractive chance for recovery, but retail investors typically do not have this opportunity. They …

Motor proteins and filaments are essential elements in living cells. They are employed in skeletal muscles to generate forces, they transport cargos such as organelles to specific locations in the cells or they reorganize themselves to change a cell's structure. Moreover, motor proteins and filaments use hydrolysis of adenosine triphosphate (ATP) as chemical fuel to generate mechanical movement in their interaction. This article was authored by a university educator and a twelve year-old child. The first author was a visiting teacher to a small, rural, state primary school in Queensland Australia. She spent one day per week for nineteen weeks fostering an inquiry-based stance to teaching and learning. The second author is a student within the researched school. Because research generated and co-authored by a child is a largely unprecedented approach to articles published in scholarly journals, the topics of this inquiry are both the within-school research and the complexity of researching and writing about it. Many sections of the northern WA coastline have become heavily industrialised in the last three decades particularly throughout the Pilbara region where offshore and onshore oil and gas exploration and processing facilities are abundant. There have been recent moves to develop similar industry along the Kimberley coast to access the Browse Basin, a 140,000 Km2 offshore gas field approximately 300km off the Kimberley Coast. Throughout history, those arrested for vagrancy have generally been poor men and women, often young, able-bodied, unemployed, and homeless. Most histories of vagrancy have focused on the European and American experiences. Cast Out: Vagrancy and Homelessness in Global and Historical Perspective is the first book to consider the shared global heritage of vagrancy laws, homelessness, and the historical processes they accompanied.

Groundwater quality was monitored in four Spanish caves using concentrations of nitrate, potassium, phosphorus and in some cases total organic carbon. Three of the caves are located in NW Spain and contain prehistoric cave paintings and hence have special conservation interest. Of these, two are open show caves (Tito Bustillo and Pindal Caves), while the other one (Herrerías Cave) is not managed for tours and is partially closed off to public access. Villars Cave is a typical shallow cave from South-West France (45.44°N; 0.78°E; 175 m asl) that has provided several speleothem palaeoclimatic records such as the millennial scale variability of the Last Glacial period and the Last Deglaciation. Monitoring the Villars cave environment over a 13-year period has helped in the understanding of the stable isotopic speleothem content and in the hydrology. For example, it was demonstrated that most of the calcite CaCO3 carbon comes from the soil CO2, which explains the sensitivity of the δ13C to any vegetation and climatic changes. Molecular Characterization Of Dopamine Transporter Phosphorylation And Regulation, Balachandra Kumar Gorentla

The dopamine transporter (DAT) is a presynaptic membrane phosphoprotein that terminates dopaminergic synaptic transmission by clearing dopamine (DA) back into presynaptic neuron by a reuptake process. DAT is also a molecular target for psychostimulants such as cocaine, amphetamine (AMPH) and methamphetamine (METH) that increase DA synaptic levels either by blocking reuptake or by inducing DA efflux. DAT phosphorylation and transport down regulation are best demonstrated with the treatment of Protein kinase C (PKC) activators such as (phorbol 12-myristate 13-acetate) PMA. Serine(s) and threonine(s) are known to be involved in DAT phosphorylation.

Deer–Vehicle Collision Trends At A Suburban Immunocontraception Site, Allen T. Rutberg, Ricky E. Naugle
Human–Wildlife Interactions
Observations of extended estrus seasons in female white-tailed deer (Odocoileus virginianus) vaccinated with porcine zona pellucida (PZP) immunocontraceptives have led to speculation that management use of PZP would, perversely, increase the rate of deer–vehicle collisions. To test this hypothesis, we studied PZP-treated female deer at the National Institute of Standards and Technology, Gaithersburg, Maryland. PZP-treated female deer did not differ from untreated female deer in risk of death by vehicle collision, and the number of deer killed in vehicle collisions was uncorrelated with the number of deer treated with PZP, whether or not population size was controlled for. …
