We have previously reported that Potato virus X-expressed coat protein of Cucumber mosaic virus (CMV) formed virus-like particles (VLPs), which served as carriers for display of different neutralizing epitopes of Newcastle disease virus (NDV). In this work, we further modified the purification protocol of recombinant VLPs carrying short neutralizing epitopes of the NDV proteins and demonstrated that self-contained capsid protein subunits of CMV transiently expressed from heterologous virus packaged into individual virions morphologically resembling and/or indistinguishable from wild type CMV particles. Homogeneity of the final preparation represents an advance over our previous study, where VLPs were found to be of …

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

The lobate lac scale Paratachardina pseudolobata Kondo & Gullan (Kerriidae) is a polyphagous pest of woody plants in Florida (U.S.A.) the Bahamas, Christmas Island (Australia) and it has been reported from Cuba. Its recent appearance as a pest in these places indicates that this scale is introduced; however, its native range is unknown. Until 2006, this pest species was identified mistakenly as Paratachardina lobata (Chamberlin) [now P. silvestri (Mahdihassan)], which is native to India and Sri Lanka. Quarantine laboratory acceptance trials with Indian P. silvestri parasitoids indicated a strong immune response from P. pseudolobata. Gregarious development of encyrtid wasps …

A Core Set Of Microsatellite Markers For Western Corn Rootworm (Coleoptera: Chrysomelidae) Population Genetics Studies, Kyung Seok Kim, Uwe Stolz, Nicholas J. Miller, Miller R. Waits, Thomas Guillermaud, Douglas V. Sumerford, Thomas W. Sappington

United States Department of Agriculture-Agricultural Research Service / University of Nebraska-Lincoln: Faculty Publications

Interest in the ecological and population genetics of the western corn rootworm, Diabrotica virgifera virgifera LeConte, has grown rapidly in the last few years in North America and Europe. This interest is a result of a number of converging issues related to the increasing difficulty in managing this pest and the need to characterize and understand gene ßow in the context of insect resistance management. One of the key components needed for successful population genetics studies is the availability of suitable molecular markers. Using a standard group of microsatellite markers enables researchers from different laboratories to directly compare and share …

Background: In 2006, an atypical U.S. case of bovine spongiform encephalopathy (BSE) was discovered in Alabama and later reported to be polymorphic for glutamate (E) and lysine (K) codons at position 211 in the bovine prion protein gene (Prnp) coding sequence. A bovine E211K mutation is important because it is analogous to the most common pathogenic mutation in humans (E200K) which causes hereditary Creutzfeldt – Jakob disease, an autosomal dominant form of prion disease. The present report describes a high-throughput matrix-associated laser desorption/ionization-time-of-flight mass spectrometry assay for scoring the Prnp E211K variant and its use to determine an upper limit …

The Effects Of An Offender's Ethnicity As Well As The Type Of Crime Committed, On Public Perceptions Regarding Seriousness, And The Sentencing Principles Seen As Most Appropriate, Andrea Kysely Jan 2008

The Effects Of An Offender's Ethnicity As Well As The Type Of Crime Committed, On Public Perceptions Regarding Seriousness, And The Sentencing Principles Seen As Most Appropriate, Andrea Kysely

Theses : Honours

Public opinion has been evidenced as exerting significant influence over the development of, and alteration to, policies dealing with offenders (Roberts, 1992). Research suggests that an offender's ethnicity, as well as the seriousness of a crime, have a significant effect on public opinion regarding the appropriateness of an imposed sentence, and the goals of sentencing seen as most important (Herzog & Rattner, 2003). However whilst research in the United States and Europe has continued to expand, there is a significant lack of research directly related to the Australian context. The significant over-representation of Indigenous Australians in Australian prisons, potentially reflects …

The Role Of Sex, Gender Role, And Extraversion-Introversion In Explaining The Experience, Expression And Control Of Anger, James Oliver

Theses : Honours

Anger is a frequently experienced emotion that has been shown to influence perceptions, beliefs, ideas, reasoning, and ultimately choices and actions. It has the potential to become a serious problem if it reaches dysfunctional levels. This study examines the role of biological sex, gender role, and extraversion-introversion in the expression, experience and control of anger. A sample of 110 persons drawn from the Australian community were administered the Staxi-2, EPQ-R and the BSRI. Results showed that extraversion-introversion accounted for most variance associated with anger expression, while gender role accounted for most variance associated with anger control. Biological sex was not …

Special Education and Rehabilitation Counseling Faculty Publications

The effects of choosing between two academic assignments on task completion and percent correct by a ninth grade student with a learning disability was investigated. This case study extended the efficacy of antecedent based intervention as an instructional modification. Further, the study extends previous research by investigating the effect of choice on academic achievement. The withdrawal design showed that percent completed and correct were highest when the participant was given a choice between two assignments when compared to baseline conditions. Results are discussed in term of efficacy of choice as an antecedent intervention, choice selection, and future research directions.

Motivational Factors Of Seasonal Volunteers: A Study Of The Student Conservation Association, Cory J. Roegner Jan 2008

Motivational Factors Of Seasonal Volunteers: A Study Of The Student Conservation Association, Cory J. Roegner

Graduate Research Papers

The use and efficacy of motivational factors in attracting and retaining employees has become an increasingly important concern within the realm of leisure-service management. Authors of leading text on managing leisure-service organizations have affirmed this dilemma, stating that more research is needed on how to attract, retain, supervise, and motivate employees (Henderson & Bialeschki, 1993; Hoff, Ellis & Crossley, 1988). In recent decades, this need has become particularly true with respect to the motives of seasonal and part-time volunteers (DeGraaf & Edginton, 1992). The field of outdoor recreation/resource management is of no exception. Thus, the purpose of this study was …

Social Politics And Freedom: Incorporating Civil Society Into Arendt's Political Thought, Grant J. Rozeboom Jan 2008

Social Politics And Freedom: Incorporating Civil Society Into Arendt's Political Thought, Grant J. Rozeboom

Honors Program Theses

The possibility of human freedom has captivated philosophers throughout the ages, often leading them to conclude that freedom is a unique capacity of humanity, exemplifying our potential for politics, contemplation, and/or religious salvation. In the modern age, beginning with the political writings of Thomas Hobbes and John Locke, freedom has been understood (at least within the early liberal tradition and common discourse) as unhindered and individuated economic movement, motivated by self-interest and actualized most fully in institutions such as the consumerist free market. Yet, we find that the more we devote our actions to fulfilling our individual interests and needs, …

Carpe Noctum, Mark Andrew Turnage Jan 2008

Carpe Noctum, Mark Andrew Turnage

Honors Program Theses

In movies, there are certain conventions that identify a film with a particular genre: a typical western has cowboys; a superhero movie may star a gifted protagonist; or an adventure film could feature exotic locations. Such is also the case with film noir, a movement characterized by storylines featuring criminal elements and their tensions with the police, stylized black-and-white cinematography, and a seedy, nightmarish cityscape. But just as an audience can easily identify genres like these, an author can also demythologize a genre by providing original themes within a familiar setting or inverting those themes that currently exist within the …

Female Hysteria Across Cultures And Periods In American Literature, Jessica Daine Droogsma Jan 2008

Female Hysteria Across Cultures And Periods In American Literature, Jessica Daine Droogsma

Honors Program Theses

Modernity would like us to believe we are in control: you can be whatever you want if you work hard enough; you are in charge of your own destiny; practice makes perfect; if you don’t like something, change it; you are what you eat. These popular aphorisms reflect our society’s addiction to self-determinism. We are completely set on the idea that we create and direct our own lives. However, there are larger influences in the world which are sometimes out of our control. The government, the media, society as a collective, and other such establishments have power over the individual. …

Reduced Dntp Binding Affinity Of 3tc-Resistant M184i Hiv-1 Reverse Transcriptase Variants Responsible For Viral Infection Failure In Macrophage, Varuni K. Jamburuthugoda, Jose M. Santos-Velazquez, Mark Skasko, Darwin J. Operario, Vandana Purohit, Pauline Chugh, Erika A. Szymanski, Joseph E. Wedekind, Robert A. Bambara, Baek Kim

Biology

We characterized HIV-1 reverse transcriptase (RT) variants either with or without the (-)-2′,3′-deoxy-3′-thiacytidine-resistant M184I mutation isolated from a single HIV-1 infected patient. First, unlike variants with wild-type M184, M184I RT variants displayed significantly reduced DNA polymerase activity at low dNTP concentrations, which is indicative of reduced dNTP binding affinity. Second, the M184I variant displayed a ∼10- to 13-fold reduction in dNTP binding affinity, compared with the Met-184 variant. However, the kpol values of these two RTs were similar. Third, unlike HIV-1 vectors with wild-type RT, the HIV-1 vector harboring M184I RT failed to transduce cell types containing low dNTP concentrations, …

Krepa6 Is An Rna-Binding Protein Essential For Editosome Integrity And Survival Of Trypanosoma Brucei, Salvador Zipagun Tarun Jr., Achim Schnauffer, Nancy Lewis Ernst, Rosemary Proff, Junpeng Deng, William Hol, Kenneth Stuart, K. Jan 2008

Krepa6 Is An Rna-Binding Protein Essential For Editosome Integrity And Survival Of Trypanosoma Brucei, Salvador Zipagun Tarun Jr., Achim Schnauffer, Nancy Lewis Ernst, Rosemary Proff, Junpeng Deng, William Hol, Kenneth Stuart, K.

Biology

Most mitochondrial mRNAs in kinetoplastid protozoa require post-transcriptional RNA editing that inserts and deletes uridylates, a process that is catalyzed by multiprotein editosomes. KREPA6 is the smallest of six editosome proteins that have predicted oligonucleotide-binding (OB) folds. Inactivation of KREPA6 expression results in disruption and ultimate loss of ∼20S editosomes and inhibition of procyclic form cell growth. Gel shift studies show that recombinant KREPA6 binds RNA, but not DNA, with a preference for oligo-(U) whether on the 3′ end of gRNA or as a (UU)12 homopolymer. Thus, KREPA6 is essential for the structural integrity and presence of ∼20S editosomes and …

Subordination And The Fortuity Of Our Circumstances, Sergio J. Campos Jan 2008

Subordination And The Fortuity Of Our Circumstances, Sergio J. Campos

Articles

The antisubordination principle exists at the margins of equality law. This Article seeks to revive the antisubordination principle by taking a fresh look at its structure and underlying justification. First, the Article provides an account of the harm of subordination that focuses on one's position in society, rejecting the focus on groups popular in the existing antisubordination literature. Second, it argues for a theory of state obligation that goes beyond both the existing state action doctrine of the Equal Protection Clause and the failure to protect doctrine associated with Charles Black. The Article argues instead that the antisubordination principle mandates …

Transcriptional Regulatory Network Analysis During Epithelial-Mesenchymal Transformation Of Retinal Pigment Epithelium., Craig H Pratt, Rajanikanth Vadigepalli, Praveen Chakravarthula, Gregory E Gonye, Nancy J Philp, Gerald B Grunwald

Department of Pathology, Anatomy, and Cell Biology Faculty Papers

PURPOSE: Phenotypic transformation of retinal pigment epithelial (RPE) cells contributes to the onset and progression of ocular proliferative disorders such as proliferative vitreoretinopathy (PVR). The formation of epiretinal membranes in PVR may involve an epithelial-mesenchymal transformation (EMT) of RPE cells as part of an aberrant wound healing response. While the underlying mechanism remains unclear, this likely involves changes in RPE cell gene expression under the control of specific transcription factors (TFs). Thus, the purpose of the present study was to identify TFs that may play a role in this process.

METHODS: Regulatory regions of genes that are differentially regulated during …

Inhibition Of Cellular Thioredoxin Reductase By The Anticancer Prodrug Cloretazine, Tyler R. Schleicher Jan 2008

Inhibition Of Cellular Thioredoxin Reductase By The Anticancer Prodrug Cloretazine, Tyler R. Schleicher

Honors Theses

Sulfonylhydrazines are a class of DNA alkylating drugs which also produce carbamoylating activity in situ. The carbamoylating species is of significance because it has shown to inhibit important thiol-containing enzymes such as glutathione reductase (GR) and thioredoxin reductase (TrxR). TrxR catalyzes dithiol-disulfide exchange reactions on thioredoxin (Trx), which in turn catalyzes other reductive processes such as deoxyribonucleotide biosynthesis. In this study, we demonstrate that TrxR activity is strongly inhibited by the anticancer prodrug Cloretazine both in purified form and in leukemia cell lysates. This inhibition is specific to the carbamoylating activity of methylisocyanate (MiC). In contrast, another important oxidoreductase, glutathione …

Man has an enduring subconscious connection with anthropological imagery and narrative. This association extends to the methods and practices that place importance on certain objects over others. In every culture throughout history the shared understanding of existence has been determined by images, symbols and archetypes. Intuitive logic and shared unconscious knowledge pervaded the daily lives of pre-modern man. Early people maintained a stronger connection with nature, spirituality and the subconscious. Archaic man acknowledged one stone among many as possessing spiritual properties. That stone could become a sacred receptacle that differentiated it from all others and saturated it with meaning and …

PREP Reports & Publications

Each year Granite State shellfishers search shallow briny waters in search of delicious mussels, clams, or oysters for the dinner table. Those who are skilled often are rewarded with full buckets, but few shellfishers realize that good harvests in New Hampshire’s Seacoast owe much to activities occurring far upstream.

The quality of the water and amount of available nutrients that sustain a clam or

oyster is directly related to the condition of the rivers and streams that drain the land. The Hampton-Seabrook Estuary is fed by approximately 46 square miles of surrounding land. An even larger system, the Piscataqua River …

Changing The Rules Of Establishment Clause Litigation: An Alternative To The Public Expression Of Religion Act, Christopher D. Tomlinson

Vanderbilt Law Review

In 2004, the American Civil Liberties Union ("ACLU") threatened to sue the city of Redlands, California, if it did not remove a small cross from its city seal.' The cross represented the city's religious heritage and its history as a city of churches. Instead of facing the possibility of litigation and the more daunting risk of losing in court and being forced to pay the ACLU's attorneys' fees in addition to its own, the Redlands City Council agreed to change the seal. The City of Redlands not only could ill afford the risk of paying the ACLU's attorneys' fees; it …
