Purpose: Hepatocyte growth factor (HGF), acting through its receptor, c-met, is a cytokine whose role in promoting cell migration has been extensively documented. HGF and c-met have been implicated in the progression of angiogenesis in many cancer models and in the mouse model of retinal neovascularization. However, HGF mediation of protease expression and of an endothelial cell migratory phenotype in the retina has not been explored. We determined if HGF and/or hypoxia effects cell migration and protease levels in retinal microvascular endothelial cells and in a mouse model of retinal neovascularization. Methods: Bovine retinal microvascular endothelial cells (BRMVEC) were grown …
Neuronal And Molecular Adaptations Of Gaba Neurons In The Ventral Tegmental Area To Chronic Alcohol, Kimberly Hales
Neuronal And Molecular Adaptations Of Gaba Neurons In The Ventral Tegmental Area To Chronic Alcohol, Kimberly Hales
Theses and Dissertations
The purpose of this thesis project was to examine the effects of chronic alcohol on the excitability and molecular adaptation of GABA neurons of the ventral tegmental area (VTA). GABA neurons are of interest with regards to ethanol intoxication, reinforcement, and dependence due to their widespread distribution and connectivity to mesocorticolimbic dopamine (DA) neurons implicated in alcohol reward and addiction. Since we have previously shown adaptation of VTA GABA neuron firing rate to chronic ethanol (Gallegos, Criado et al. 1999) and suppression of gap-junction (GJ) mediated coupling between these neurons by acute ethanol (Stobbs, Ohran et al. 2004), we wanted …

Air power has seen constant development from the Wright Flyer’s first flight at Kitty Hawk on December 17, 1903 via the advent of the jet age with the service entry of the Messerschmitt Me 262 in 1942, to today’s multirole fighters (F-35 Joint Strike Fighter) and stealth aircraft (B-2 Spirit multi-role bomber). As a result of this evolution of one hundred years air power has emerged as a central component in power projection. As General William Mitchell said: ”Neither armies nor navies can exist unless the air is controlled over them.” (Mitchell 1925, xv)We have witnessed a corresponding development in …

Sol–gel coatings which elute bioactive silver ions are presented as a potential solution to the problem of biofilm formation on indwelling surfaces. There is evidence that high-temperature processing of such materials can lead to diffusion of silver away from the coating surface, reducing the amount of available silver. In this study, we report the biofilm inhibition of a Staphylococcus epidermidis biofilm using a low-temperature processed silver-doped phenyltriethoxysilane sol–gel coating. The incorporation of a silver salt into a sol–gel matrix resulted in an initial high release of silver in de-ionised water and physiological buffered saline (PBS), followed by a lower sustained …

Synthesis Of Novel Cysteine Peptide Nucleic Acid (Cpna), Mehul J. Ajmera
USF Tampa Graduate Theses and Dissertations
Many diseases are caused due to abnormalities in production of specific protein. Across this protein the conventional lock and key mechanism shows binding at the specific cites of protein. However use of antisense technology can prevent formation of protein. It does so by binding to mRNA and prevents transcription. The structural modifications lead to synthetic molecules with 18-mer units which show significant improvement in binding properties, this gives birth to a new class of oligomers called Peptide Nucleic Acid (PNA). We herein report cysteine based PNA called CPNA.

Biological Sciences Faculty Research & Creative Works
Low Temperature and Desiccation Stress Are Thought to Be Mechanistically Similar in Insects, and Several Studies Indicate that There is a Degree of Cross-Tolerance between Them, such that Increased Cold Tolerance Results in Greater Desiccation Tolerance and Vice Versa. This Assertion is Tested at an Evolutionary Scale by Examining Basal Cold Tolerance, Rapid Cold-Hardening (RCH) and Chill Coma Recovery in Replicate Populations of Drosophila Melanogaster Selected for Desiccation Resistance (With Controls for Both Selection and Concomitant Starvation) for over 50 Generations. All of the Populations Display a RCH Response, and There is No Effect of Selection Regime on RCH or …

Tetraspanin Kai1/Cd82 Inhibits Cell Migration-Related Cellular Events Via Reorganizing Actin Network, Wei Liu
Tetraspanin Kai1/Cd82 Inhibits Cell Migration-Related Cellular Events Via Reorganizing Actin Network, Wei Liu
Theses and Dissertations (ETD)
To determine how tetraspanin KAI1/CD82, a tumor metastasis suppressor, inhibits cell migration, we assessed which motility-related cellular events are altered by KAI1/CD82 and how KAI1/CD82 regulates these events. We found that KAI1/CD82-overexpressing cancer cells exhibit various morphologies but typically display elongated cellular extensions and a lack of lamellipodia. Live imaging demonstrated that the formation of lamellipodia and retraction of extensions were deficient upon KAI1/CD82 overexpression.
The deficiency in developing motility-related cellular events was accompanied by defects in actin cortical network and stress fiber formations. Notably, actin polymerization was attenuated by KAI1/CD82. Although Rac1 activity was diminished upon KAI1/CD82 expression, Rac1 …
Enzyme Architecture And Flexibility Affect Dna Topoisomerase I Function, Mariè Van Der Merwe
Enzyme Architecture And Flexibility Affect Dna Topoisomerase I Function, Mariè Van Der Merwe
Theses and Dissertations (ETD)
DNA topoisomerase I (Top1) is a highly conserved enzyme composed of four domains: a positively charged N-terminus; a DNA binding/core domain that circumscribes duplex DNA; a non-conserved linker domain; and a C-terminal/catalytic domain. Top1 catalyzes changes in DNA topology by transient cleavage of a single DNA strand and the concomitant formation of a phosphotyrosyl linkage between the enzyme and the 3’ DNA end. This covalent Top1-DNA complex is the binding site for camptothecin (CPT), which selectively inhibits religation of the cleaved DNA strand. CPT binding stabilizes the covalent complex, while the collision of replication forks with CPT-Top1-DNA adducts produces DNA …

An Integral Philosophy And Definition Of Nursing, Olga F. Jarrin
An Integral Philosophy And Definition Of Nursing, Olga F. Jarrin
School of Nursing Scholarly Works
A unifying meta-theory of nursing is suggested, building from the foundation of the AQAL framework. A definition of nursing as situated caring is presented, along with a historical discussion of nursing epistemology and theory for context. Implications for practice, education, and research are also discussed. A unifying meta-theory will enable nurses at both the practical and academic levels to appreciate the complexity and simplicity of nursing, allowing them to articulate confidently what we do and why we do it.

A Regulator Of Dscam Mutually Exclusive Splicing Fidelity, Sara Olson, Yiannis Savva, Jung Park, Brenton R. Graveley
UCHC Articles - Research
The Down syndrome cell adhesion molecule (Dscam) gene has essential roles in neural wiring and pathogen recognition in Drosophila melanogaster. Dscam encodes 38,016 distinct isoforms via extensive alternative splicing. The 95 alternative exons in Dscam are organized into clusters that are spliced in a mutually exclusive manner. The exon 6 cluster contains 48 variable exons and uses a complex system of competing RNA structures to ensure that only one variable exon is included. Here we show that the heterogeneous nuclear ribonucleoprotein hrp36 acts specifically within, and throughout, the exon 6 cluster to prevent the inclusion of multiple exons. Moreover, hrp36 …

Tigecycline In The Treatment Of Complicated Intra-Abdominal And Complicated Skin And Skin Structure Infections, M. L. Townsend, M. W. Pound, R. H. Drew
Pharmacy Practice
Tigecycline, a glycylcycline related to the tetracycline class of antibiotics, represents a new option for the treatment of complicated intra-abdominal and complicated skin and skin structure infections. It displays favorable activity in vitro against the most common causative Gram-positive, Gram-negative and anaerobic pathogens. In addition, tigecycline demonstrates activity against drug-resistant pathogens such as methicillin-resistant Staphylococcus aureus, vancomycin-resistant enterococci, and organisms (such as Escherichia coli and Klebsiella pneumoniae) producing extended-spectrum beta-lactamases. Tigecycline lacks activity in vitro against Pseudomonas and Proteus spp. In randomized clinical trials, tigecycline administered intravenously twice daily has demonstrated efficacy similar to comparators for a variety …
Effects Of An Inserted Endoscope On Chyme Movement In Small Intestine – A Theoretical Model, V. P. Srivastava
Effects Of An Inserted Endoscope On Chyme Movement In Small Intestine – A Theoretical Model, V. P. Srivastava
Applications and Applied Mathematics: An International Journal (AAM)
The effects of an inserted endoscope on chyme movement in small intestine (gastrointestinal tract) have been investigated. The flow of chyme is induced by a progressive wave of area contraction along the length of intestinal wall under long wavelength approximation. It is found that the chyme movement is significantly influenced due to the presence of the endoscope. The pressure drop assumes lower values for higher values of the endoscope radius for small flow rates but the property reverses with increasing flow rates. The friction forces at intestinal wall and endoscope possess character similar to the pressure drop for any given …
Heterologous Hybridization Of Cotton Microarrays With Velvetleaf (Abutilon Theophrasti) Reveals Physiological Responses Due To Corn Competition, David P. Horvath, Danny Llewellyn, Sharon A. Clay
Heterologous Hybridization Of Cotton Microarrays With Velvetleaf (Abutilon Theophrasti) Reveals Physiological Responses Due To Corn Competition, David P. Horvath, Danny Llewellyn, Sharon A. Clay
Agronomy, Horticulture and Plant Science Faculty Publications
Microarray analysis was used to identify changes in gene expression in velvetleaf that result from competition with corn. The plants were grown in field plots under adequate N (addition of 220 kg N ha−1) to minimize stress and sampled at the V6 growth stage of corn (late June). Leaf area, dry weight, and N and P concentration were similar in velvetleaf plants grown alone or with corn. Competition, however, did influence velvetleaf gene expression. Genes involved in carbon utilization, photosynthesis, red light signaling, and cell division were preferentially expressed when velvetleaf was grown in competition with corn. A less clear …

Leadership For Today Effective Group Communication, Karla Trautman
SDSU Extension Extra Archives
Within a given day, human beings rely on the ability to communicate as a means of transferring information. Whether we speak, listen, write notes or letters, email, or text message, the process of communicating thoughts, ideas, and feelings is something we find to be important but typically give little thought to. In organized group situations, the art of good communication helps build trust and respect. It fosters a positive learning environment and can set the stage for the group to achieve its identified mission, vision, and goals. In any group setting, members may share a common interest and commitment, yet …

Leadership For Today Managing Conflict Within Groups, Karla Trautman
SDSU Extension Extra Archives
The human relationship is a complex and dynamic interaction. As living creatures, we need and crave the opportunity to interact with other humans by speaking, listening, and spending time with them. Most of this interaction tends to be mutual and cordial. Yet, at times, the interaction can be laced with tension and discord. If left alone, the tension can lead to conflict that may damage the relationship or even become volatile.
The Politics Of Vodou: Aids, Access To Health Care And The Use Of Culture In Haiti, Catherine Benoît
The Politics Of Vodou: Aids, Access To Health Care And The Use Of Culture In Haiti, Catherine Benoît
Anthropology Faculty Publications
During the past few years, the AIDS campaign in Haiti has been targeting Vodou officiants and organizations. These awareness and training programmes in- form officiants about the transmission and prevention of AIDS, tests for HIV and anti- retroviral drugs, or even try to encourage them to become involved in a medical referral system. These culturalist interventions are grounded in an essentialist concept of culture that can have harmful effects on the targeted groups. The concept of culture underlying such interventions is deconstructed along with the categories of tradi- tional medicine and the ‘tradipractitioner’. An approach to public health is advocated …
