We report the complete reconstruction of the firefly luciferase gene, fully codon optimized for expression in Neurospora crassa. This reporter enhances light output by approximately 4 log orders over that with previously available versions, now producing light that is visible to the naked eye and sufficient for monitoring the activities of many poorly expressed genes. Time lapse photography of strains growing in race tubes, in which the frq or eas/ccg-2 promoter is used to drive luciferase, shows the highest levels of luciferase activity near the growth front and newly formed conidial bands. Recent national efforts to stem the alarming growth of childhood overweight and obesity in Australia have highlighted the importance of preventative strategies that focus beyond the child-on parents, families, primary health care providers (PHCPs) and child care services. But, while such efforts have to date focused on school-based interventions, once poor eating habits and sedentary lifestyles have set in, a growing body of research is indicating that PHCPs can play a more influential role in monitoring and modifying factors affecting prevention and early intervention in preschool children. Since the discovery in the early eighties of the Human Immunodeficiency Virus (HIV) that causes acquired immunodeficiency syndrome (AIDS), there have been reports of people who were completely resistant to infection with HIV and others who progressed at slower rates to AIDS. The present article summarises the mechanisms involved in resistance against HIV infection and progression to AIDS. The paper will specifically focus on the role of immunological mechanisms, genetics, ethnicity and cultural practices such as male circumcision in mitigating infection. The current understanding on host natural resistance against HIV infection and progression to AIDS would potentially contribute to better …

The Link Between Emotional Recognition And Executive Functioning Ability, Alexandra Popa

This study investigated the link between executive functioning (e.g., planning, shifting, inhibiting) and emotional recognition ability based on facial expressions. In addition, differences in emotional recognition and executive functioning among college majors and year in school was assessed. Gender differences were studied as well. A sample of 53 volunteer undergraduate students were administered an abbreviated version of Ekman's Pictures of Facial Affect (AEFER) and the Wisconsin Card Sorting Test: Computer Version 4 (WCST:CV$). Findings indicate a moderate association between emotional recognition and executive functioning. Participants' performance varied on emotional recognition tasks based on year and major. 14-3-3 Proteins Interact With The Beta-Thymosin Repeat Protein Csp24, Terry Crow, Juan-Juan Xue-Bian, Joseph T Neary

Conditioned stimulus pathway protein 24 (Csp24) is a beta-thymosin-like protein that is homologous to other members of the family of beta-thymosin repeat proteins that contain multiple actin binding domains. Actin co-precipitates with Csp24 and co-localizes with it in the cytosol of type-B photoreceptor cell bodies. Several signal transduction pathways have been shown to regulate the phosphorylation of Csp24 and contribute to cellular plasticity. Here, we report the identification of the adapter protein 14-3-3 in lysates of the Hermissenda circumesophageal nervous system and its interaction with Csp24. All living cells contain the electrogenic enzyme Na/K ATPase, whose function is to pump ions against the electrochemical gradient, in order to provide potential energy which is later used for cellular processes such as action potentials, muscle contraction and facilitated transport. Using a technique developed in our lab, exploiting the molecule's voltage dependence, it is possible to increase this pump function by many folds. Optical measurement of the membrane potential of living cells was made using a potentiommetric dye, with successful manipulation of the ionic concentration and membrane potential reported. Natural killer (NK) cells are our initial immune defense against viral infections and cancer development. Thus, agents that are able to interfere with their function increase the risk of cancer and/or infection. A series of triorganotins (trimethyltin (TMT), dimethylphenyltin (DMPT), methyldiphenyltin (MDPT), and triphenyltin (TPT)) have been shown to decrease the lytic function of human NK cells. TPT and MDPT were much more effective than DMPT or TMT at reducing lytic function. This study investigates the role that decreased ATP levels may play in decreases in the lytic function of NK cells induced by these organotins (OTs). A 24h exposure …

This work examined cell composition gradients in maturing tillers of switchgrass (Panicum virgatuln L. ) with the aim of developing baseline information on this important forage and biomass crop. Flowering tillers were collected from plants raised from seeds in a greenhouse and field, harvested at soil level and separated into internodes beginning with the node subtending the peduncle. Internodes were analyzed using microscopy, by fiber digestion, high-performance liquid chromatography and by gas chromatography-mass spectrometry to obtain anatomical and compositional data.
