The dimeric Mycobacterium tuberculosis FabH (mtFabH) catalyses a Claisen-type condensation between an acyl-CoA and malonyl-acyl carrier protein (ACP) to initiate the Type II fatty acid synthase cycle. To analyze the initial covalent acylation of mtFabH with acyl-CoA, we challenged it with mixture of C6-C20 acyl-CoAs and the ESI-MS analysis showed reaction at both subunits and a strict specificity for C12 acyl CoA. Crystallographic and ESI-MS studies of mtFabH with a decyl-CoA disulfide inhibitor revealed a decyl chain bound in acyl-binding channels of both subunits through disulfide linkage to the active site cysteine. Administrators, classroom educators, counselors, and other support staff attempt to attend to students through a cooperative effort of connecting with the community beyond the school building, as well as the families represented within it. As Christians, there is a higher calling to truly embrace those often underserved in our learning communities. Africa, a continent full of abundance and potential, is ironically hindered in desolation, deprivation, and chaos. The growth, future, and potential success of Africa is solely dependent on the education of its children. After identifying a number of educational barriers, defining the need for educational improvement, and documenting the efficiency of Western aid, this paper concludes that the West needs to recognize that it is the Africans who must ultimately save their own countries. The study uses two representative English newspapers in Asia – the China Daily (“CD”) and the South China Morning Post (“SCMP”). The West Indian manatee (Trichechus manatus) is an herbivorous marine mammal that occupies freshwater, estuarine, and marine habitats. Despite being considered endangered, relatively little is known about the feeding ecology of either of the two recognized subspecies, the Florida manatee (T.m. latirostris) and Caribbean or Antillean manatee (T.m. manatus). A better understanding of their respective feeding preferences and habitat use is essential to establish criteria on which conservation plans can be based. This excited state is often referred to as singlet oxygen and can be generated in a photosensitized process under one- or two-photon excitation of a photosensitizer. In recent years, intertidal reefs of Crassostrea virginica (eastern oyster) along central Florida's east coast have suffered extensive losses due to wakes from recreational boats. These wakes have caused extensive shell movement and sediment resuspension which results in large piles of disarticulated shells along the seaward edges of reefs. Dead margins extend up to one meter above mean high water. The creation and enforcement of "no wake" zones in the area are unlikely. Simulation models are valuable for making predictions that may be tested in natural systems and for understanding observed patterns. The simulation model developed for this thesis evaluates the effects of spatial network architecture, including organism dispersal patterns and isolation of habitats, on metapopulations. Two fields were merged throughout this project: metapopulation biology and small-world network theory. Small-world networks are characterized in their extremes as scale-free or single-scale. These models potentially simulate the networks of habitats and corridors in which metapopulations operate. Raman micro-spectroscopy provides a non-destructive probe with potential applications as a diagnostic tool for cellular disorders. This study presents micro-Raman spectra of live erythrocytes infected with a malaria parasite and investigates the potential of this probe to monitor molecular changes which occur during differentiation of the parasite inside the cell. At an excitation wavelength of 633 nm the spectral bands are dominated by hemoglobin vibrations yielding information the on structure and spin state of the heme moiety. Three-dimensional (3D) optical data storage based on two-photon processes provides highly confined excitation in a recording medium and a mechanism for writing and reading data with less cross talk between multiple memory layers, due to the quadratic dependence of two photon absorption (2PA) on the incident light intensity. The capacity for highly confined excitation and intrinsic 3D resolution affords immense information storage capacity (up to 1012 bits/cm3). Recent research in counselor training has focused on the difficulties and challenges facing counseling practitioners that result in high stress, burnout, compassion fatigue, vicarious trauma, and counselor impairment. The American Counseling Association's (ACA) Code of Ethics has addressed the ethical issues inherent in counselor impairment. Further, the Council for Accreditation of Counseling and Related Educational Programs (CACREP) has focused on the development of counselors that will assist them in being resilient to workplace stressors. Understanding how migration, genetic drift, and natural selection interact to maintain the genetic and phenotypic variation we observe in natural populations is a central goal of population genetics. Amphibians provide excellent model organisms for investigating the interplay between these evolutionary forces because amphibians are generally characterized by limited dispersal abilities, high philopatry, and are obligately associated with the areas around suitable habitats (e.g. breeding ponds). The southeastern coastal plain of the United States is a center of endemism for plants in temperate North America and second only to California among the states. In the southeast, Florida has the largest number of these endemic plants. The largest number of these Florida endemics can be found in the fire maintained scrub and sandhill communities located on sandy ridges in Central Florida. One such endemic is Clitoria fragrans, a rare perennial herb. C. fragrans reproduces via a mixed mating system. It produces both open, chasmogamous flowers and closed, selfed, cleistogamous flowers. First, the underline physics uses Quick Collision Response Calculation. Many species naturally occupy discrete habitat patches within a mosaic of habitats that vary in quality. The Florida scrub-jay (Aphelocoma coerulescens) is endemic to Florida scrub, a habitat that is naturally patchy and greatly reduced in area over recent decades owing to development and urbanization. Because of this habitat loss, future management of Florida scrub-jays will focus on smaller, fragmented tracts of land. My study examines such a tract, Lyonia Preserve, southwest Volusia County, FL. This preserve was unoccupied by scrub-jays prior to habitat restoration. The preserve is now frequently managed exclusively for scrub-jays as a habitat island surrounded by …

This study explores the ways cookbooks and their rhetorical dimensions have been re-imagined using hypertext and Web technology. Using the tenets of postmodern feminist rhetoric and Web design theory, the study considers how commercial cooking hypertexts construct users' identities. Although hypertext is a potentially empowering technology, democratizing rhetoric and knowledge making practices, commercial hypertext often circumscribes agency formation and prohibits participation. Participatory, constructive hypertexts are difficult to design and costly to maintain. Of the three sites studied, Epicurious.com, BettyCrocker.com, and FoodNetwork.com, only Epicurious.com encourages meaningful communication between users and between users and designers. High birefringence (Δn > 0.4) nematic liquid crystals are particularly attractive for infrared applications because they enable a thinner cell gap to be used for achieving fast response time and improved diffraction efficiency. In this thesis, the mesomorphic and electro‐optic properties of several new fluorinated isothiocyanate (NCS) terphenyl and phenyl tolane single compounds and mixtures are reported. The single compounds demonstrated Δn~0.35‐0.52 in the visible spectral region at room temperature and exhibit relatively low viscosity. It was found that lateral fluorine substitutions and short alkyl chains eliminate smectic phase and lower the melting temperature of the single compounds. However, the consequence …

Stress has been linked to increased illness in several biologically based studies. In contrast, only a limited number of studies have assessed psychological variables related to stress, with self-efficacy and locus of control serving as potentially important variables. Thus, the current study investigated the mediating effects of self-efficacy and locus of control in the relationship between stress, psychological and physical symptoms, and the utilization of health services in college students. Results suggested that stress was correlated positively with symptoms. Pain assessment continues to be poorly managed in the clinical arena. A review of the communication process in pain assessment is carried out and the hierarchical approach often recommended in the literature -with self-report as its "gold-standard," is criticized as limited and simplistic. A comprehensive approach to pain assessment is recommended and a model that conceptualizes pain assessment as a complex transaction with various patient and clinician dependant factors is proposed. In sea urchin development, cell fate specification appears by the 60-cell stage embryo when several embryonic territories are recognized: the small micromeres, the large micromeres which will generate primary mesenchyme cells, the vegetal2 layer that will give rise to pigment cells, immunocytes, and muscle cells, the vegetal1 layer, as well as the oral and aboral ectoderm. A Delta-Notch signaling event is required for the differential specification of mesodermal cells that will give rise to secondary mesenchyme cells (SMCs). Proper habitat management is essential for the survival and reproduction of species, especially those listed under state or federal laws as endangered, threatened or of special concern, and those with small local populations. Land managers use a combination of mechanical cutting and prescribed burning to manage and restore degraded scrub habitat in east central Florida. This approach improves habitat for the endangered Florida scrub-jay (Aphelocoma coerulescens), but little is known about its effects on other taxa, especially the threatened southeastern beach mouse (Peromyscus polionotus niveiventris).
